    PRIME International School, PRIS in short is an international private school that will be located in the suggested area, Kepong as it is one of the high density areas with 360,000 population in that area. It has only one international school which is targeting to a higher income group segment compared with PRIS. There are other neighbouring area such as Gombak, Selayang, Sentul and Batu Caves can be the potential targeted area as well. PRIS will be running the British syllabus which is one of the world’s popular and widely acceptable syllabus. There will be Nursery (3 years), Primary (6 Years) and Secondary (3 years) up till form 3 offered by PRIS at the moment due to the high set up cost. PRIS aims to become a cost leader among the international school in Kuala Lumpur, by charging reasonable tuition fee, focus on IT enhanced learning and emphasise on leadership and character building. Leadership and character building are seriously lacking in most of the international school as students are seen as clients instead of future leaders and school disciplinary policy is over liberal. PRIS aims to create a difference especially in this area as one of its own core competencies. Being a cost leader will allow PRIS to offer opportunities to students to be able to experience and receive good and structural education system within an affordable fee range. The required investment is within RM160,000 - RM170,000 to complete the education from nursery to year 9, which is affordable to the major targeted middle class income family in the targeted areas. In addition, PRIS would embrace the concept as a ‘One-Stop’ Centre that allows the parents to choose the additional services such as music classes, public speaking classes, extra supervisory classes to complete homework etc to offer convenience to parents who wish to save time and willing to pay extra fees for the services provided. Though the fee charged is lower compared with most of the international schools in Kuala Lumpur, there will be profit in the second year onwards if there is a consistent number of 150 students enrol with PRIS in a span of three years. An approximate of 3.6 million is required as a set-up fee for an international school, however, it is recommended to get a bank loan with investors to become guarantors as it is tax deductible. There will be a total of RM960,901 profit in the second year though there will be a negative 1.8 million in the first year because of the high set up cost. The profit will continue if the set targeted number of students can be achieved for 3 years while PRIS gaining its credibility in the market. There will be a possible expansion to another state if not overseas within 10 years if the business model is successful. The company will be registered under Sdn Bhd because it does not need to disclose its financial position to the public and with the permission to raise fund by selling equity to investors. The principal owner will be me, the entrepreneur by holding the majority equity, however will consider to release the majority equity to investors who share the same vision of emphasising on leadership and character building; and if releasing the equity means benefiting the school’s overall development and expansion. The key personnel who will sit in the school educational quality panel will be educationist as the Head of School who is qualified, experienced and passionate, entrepreneur and some external education advisory to ensure making a difference in providing quality education

    Targeting a Designer TIMP-1 to the Cell Surface for Effective MT1-MMP Inhibition: A Potential Role for the Prion Protein in Renal Carcinoma Therapy

    Renal carcinoma cells express Membrane Type 1-Matrix Metalloproteinase (MT1-MMP, MMP-14) to degrade extracellular matrix components and a range of bioactive molecules to allow metastasis and cell proliferation. The activity of MT1-MMP is modulated by the endogenous inhibitors, Tissue Inhibitor of Metalloproteinases (TIMPs). In this study, we describe a novel strategy that would enable a “designer” TIMP-1 tailored specifically for MT1-MMP inhibition (V4A/P6V/T98L; Kiapp 1.66 nM) to be targeted to the plasma membrane for more effective MT1-MMP inhibition. To achieve this, we fuse the designer TIMP-1 to the glycosyl-phosphatidyl inositol (GPI) anchor of the prion protein to create a membrane-tethered, high-affinity TIMP variant named “T1Pr αMT1” that is predominantly located on the cell surface and co-localised with MT1-MMP. Confocal microscopy shows that T1Pr αMT1 is found throughout the cell surface in particular the membrane ruffles where MT1-MMP is most abundant. Expression of T1Pr αMT1 brings about a complete abrogation of the gelatinolytic activity of cellular MT1-MMP in HT1080 fibrosarcoma cells whilst in renal carcinoma cells CaKi-1, the GPI-TIMP causes a disruption in MMP-mediated proteolysis of ECM components such as fibronectin, collagen I and laminin that consequently triggers a downstream senescence response. Moreover, the transduced cells also suffer from an impairment in proliferation and survival in vitro as well as in NOD/SCID mouse xenograft. Taken together, our findings demonstrate that the GPI anchor of prion could be exploited as a targeting device in TIMP engineering for MT1-MMP inhibition with a potential in renal carcinoma therapy

    Compartment syndrome due to extravasation of peripheral parenteral nutrition: extravasation injury of parenteral nutrition

    Compartment syndrome is a rare but devastating condition that can result in permanent neuromuscular or soft tissue injuries. Extravasation injuries, among the iatrogenic causes of compartment syndrome, occur under a wide variety of circumstances in the inpatient setting. Total parenteral nutrition via a peripheral route is an effective alternative for the management of critically ill children who do not obtain adequate nutrition via the oral route. However, there is an inherent risk of extravasation, which can cause compartment syndrome, especially when detected at a later stage. Herein, we report a rare case of compartment syndrome and skin necrosis due to extravasation, requiring emergency fasciotomy and skin graft in a 7-month-old boy who was treated with peripheral parenteral nutrition via a pressurized infusion pump. Although we cannot estimate the exact time at which extravasation occurred, the extent and degree of the wound suggest that the ischemic insult was prolonged, lasting for several hours. Pediatric clinicians and medical teams should carefully examine the site of insertion of the intravenous catheter, especially in patients receiving parenteral nutrition via a peripheral intravenous catheter with a pressurized infusion pump

    Pentosan polysulfate increases affinity between ADAMTS-5 and TIMP-3 through formation of an electrostatically driven trimolecular complex

    The semi-synthetic sulfated polysaccharide PPS (pentosan polysulfate) increases affinity between the aggrecan-degrading ADAMTSs (adamalysins with thrombospondin motifs) and their endogenous inhibitor, TIMP (tissue inhibitor of metalloproteinases)-3. In the present study we demonstrate that PPS mediates the formation of a high-affinity trimolecular complex with ADAMTS-5 and TIMP-3. A TIMP-3 mutant that lacks extracellular-matrix-binding ability was insensitive to this affinity increase, and truncated forms of ADAMTS-5 that lack the Sp (spacer) domain had reduced PPS-binding ability and sensitivity to the affinity increase. PPS molecules composed of 11 or more saccharide units were 100-fold more effective than those of eight saccharide units, indicating the involvement of extended or multiple protein-interaction sites. The formation of a high-affinity trimolecular complex was completely abolished in the presence of 0.4 M NaCl. These results suggest that PPS enhances the affinity between ADAMTS-5 and TIMP-3 by forming electrostatically driven trimolecular complexes under physiological conditions

    Guru besar masih kurang jelas tentang kepimpinan distributif yang cenderung kepada pertanggungjawaban bersama lalu menjejaskan komitmen guru. Kajian ini bertujuan untuk mengenal pasti tahap amalan kepimpinan distributif guru besar dan hubungannya dengan tahap komitmen guru SJKC. Informasi kajian dapat dijadikan rujukan KPM dalam menilai keberkesanan amalan kepimpinan distributif mengikut kondisi Malaysia. Kajian ini berbentuk tinjauan dan mengaplikasikan pendekatan kuantitatif. Borang soal selidik dalam bentuk Google Form telah diedarkan kepada 291 orang guru daripada 1195 populasi secara rawak mudah. Data kuantitatif yang dikumpulkan telah ditafsirkan secara deskriptif dan inferensi dengan perisian SPSS versi 26. Kajian melaporkan tahap amalan kepimpinan distributif guru besar (min=4.17, sp =0.44) pada tahap tinggi. Manakala tahap komitmen guru (min=3.57, sp =0.71) pada tahap sederhana tinggi. Analisis Spearman Rho menunjukkan terdapat hubungan positif kuat yang signifikan antara amalan kepimpinan distributif guru besar dengan tahap komitmen guru (r = 0.617, p > 0.05). Kesimpulannya kajian ini membuktikan wujudnya hubungan positif kuat antara amalan kepimpinan distributif guru besar dengan komitmen guru SJKC. Amalan kepimpinan distributif terbukti sesuai dengan perubahan dan tuntutan semasa demi memupuk pemimpin berkualiti serta berupaya memupuk komitmen guru. Hasil kajian memberi mesej kepada KPM bahawa amalan kepimpinan distributif boleh meningkat kualiti pemimpin lalu memupuk komitmen guru. KPM perlu menyebarkan amalan kepimpinan distributif secara meluas kepada pemimpin sekolah mahupun pemimpin pertengahan dengan program yang sesuai. Kajian lanjutan boleh mengkaji ciri-ciri demografi yang mempengaruhi tahap amalan kepimpinan distributif dan komitmen guru
