    The static magnetic field accelerates the osteogenic differentiation and mineralization of dental pulp cells

    Dental pulp cells (DPCs) can differentiate into osteoblasts and are deemed a promising cell source for bone regeneration. Static magnetic field (SMF) stimulates osteoblast differentiation but the effect in DPCs remains unknown. The aim of this study was to investigate the effect of SMF exposure on the osteogenic differentiation and mineralization of rat DPCs in vitro. Cells were continuously exposed to SMF at 290 mT in the presence/absence of osteogenic induction [dexamethasone (Dex)/beta-glycerophosphate (beta-GP)]. Results showed that SMF alone did not impair the cell cycle and proliferation. On the other hand, obvious condensation in the metachromatic staining of the extracellular matrix with toluidine blue was observed for SMF-exposed cells as well as the Dex/beta-GP treated cells. SMF in combination with Dex/beta-GP significantly increased the mRNA expression of osteogenic genes, as well as the ALP activity and extracellular calcium concentration at the early stage, followed by obvious calcium deposits later. Besides, SMF exposure increased the activity of extracellular signal-regulated kinase 1/2 (ERK1/2) at 3 h and accelerated the mRNA expression of osteogenic transcription factor, Cbfa1, advancing its activation time from 168 to 72 h under osteogenic induction. In summary, SMF exposure in combination of Dex/beta-GP induction could significantly accelerate the osteogenic differentiation and mineralization of DPCs

    Quantitative evaluation of motor function before and after engraftment of dopaminergic neurons in a rat model of Parkinson's disease

    Although gait change is considered a useful indicator of severity in animal models of Parkinson's disease, systematic and extensive gait analysis in animal models of neurological deficits is not well established. The CatWalk-assisted automated gait analysis system provides a comprehensive way to assess a number of dynamic and static gait parameters simultaneously. In this study, we used the Catwalk system to investigate changes in gait parameters in adult rats with unilateral 6-OHDA-induced lesions and the rescue effect of dopaminergic neuron transplantation on gait function. Four weeks after 6-OHDA injection, the intensity and maximal area of contact were significantly decreased in the affected paws and the swing speed significantly decreased in all four paws. The relative distance between the hind paws also increased, suggesting that animals with unilateral 6-OHDA-induced lesions required all four paws to compensate for loss of balance function. At 8 weeks post-transplantation, engrafted dopaminergic neurons expressed tyrosine hydroxylase. In addition, the intensity, contact area, and swing speed of the four limbs increased and the distance between the hind paws decreased.     Canopy CO2 concentrations and Crassulacean acid metabolism in Hoya carnosa in a subtropical rain forest in Taiwan: consideration of CO2 avallability and the evolution of CAM in epiphytes

    The potential importance of CO2 derived from host tree respiration at night as a substrate for night time CO2 uptake during CAM was investigated in the subtropical and tropical epiphytic vine Hoya carnosa in a subtropical rainforest in north-eastern Taiwan. Individuals were examined within the canopies of host trees in open, exposed situations, as well as in dense forests. Although night time CO2 concentrations were higher near the epiphytic vines at night, relative to those measured during the day, presumably the result Of CO2 added to the canopy air by the host tree, no evidence for substantial use of this CO2 was found. In particular, stable carbon isotope ratios of H. carnosa were not substantially lower than those of many other CAM plants, as would be expected if host-respired CO2 were an important source Of CO2 for these CAM epiphytes. Furthermore, laboratory measurements of diel CO2 exchange revealed a substantial contribution of daytime CO2 uptake in these vines, which should also result in lower carbon isotope values than those characteristic of a CAM plant lacking daytime CO2 uptake. Overall, we found that host-respired CO2 does not contribute substantially to the carbon budget of this epiphytic CAM plant. This finding does not support the hypothesis that CAM may have evolved in tropical epiphytes in response to diel changes in the CO2 concentrations within the host tree canopy

    An Inflationary Scenario in Intersecting Brane Models

    We propose a new scenario for D-term inflation which appears quite straightforwardly in the open string sector of intersecting brane models. We take the inflaton to be a chiral field in a bifundamental representation of the hidden sector and we argue that a sufficiently flat potential can be brane engineered. This type of model generically predicts a near gaussian red spectrum with negligible tensor modes. We note that this model can very naturally generate a baryon asymmetry at the end of inflation via the recently proposed hidden sector baryogenesis mechanism. We also discuss the possibility that Majorana masses for the neutrinos can be simultaneously generated by the tachyon condensation which ends inflation.     Non-Perturbative Effects in Matrix Models and D-branes

    The large order growth of string perturbation theory in c1c\le 1 conformal field theory coupled to world sheet gravity implies the presence of O(e1gs)O(e^{-{1\over g_s}}) non-perturbative effects, whose leading behavior can be calculated in the matrix model approach. Recently it was proposed that the same effects should be reproduced by studying certain localized D-branes in Liouville Field Theory, which were constructed by A. and Al. Zamolodchikov. We discuss this correspondence in a number of different cases: unitary minimal models coupled to Liouville, where we compare the continuum analysis to the matrix model results of Eynard and Zinn-Justin, and compact c=1 CFT coupled to Liouville in the presence of a condensate of winding modes, where we derive the matrix model prediction and compare it to Liouville theory. In both cases we find agreement between the two approaches. The c=1 analysis also leads to predictions about properties of D-branes localized in the vicinity of the tip of the cigar in SL(2)/U(1) CFT with c=26.Comment: 27 pages, lanlmac; minor change

    One new acetogenin, 6-hydroxy-8-pentadecyloxocane-2,7-dione (1), and four new prenylated flavonoids, 4 '' a,5 '',6 '',7 '',8 '',8 '' a-hexahydro-5,3',4'-trihydroxy-5 '',5 '',8 '' a-trimethyl-4H-chromeno[2 '',3 '': 7,6] flavone (2), 4 '' a,5 '',6 '',7 '',8 '',8 '' a-hexahydro5,3', 4',-trihydroxy-5 '',5 '',8 '' a-trimethyl-4Hchromeno[2 '', 3 '': 7,8] flavone (3), 2-(3,4-dihydroxyphenyl)6-((2,2-dimethyl-6-methylenecyclohexyl) methyl)-5,7-dihydroxy-chroman-4-one (4), and 2-(3,4-dihydroxy-2-[(2,6,6-trimethylcyclohex-2-enyl) methyl] phenyl)-3,5,7-trihydroxy-4H-chromen-4-one (5), together with six known compounds, were isolated and purified from the rhizomes of Helminthostachys zeylanica by column chromatography and high performance liquid chromatography (HPLC) via bioactivity-guided fractionation isolation. The structures of the new isolates were elucidated by spectroscopic methods. Compounds 1, 3, and 5 showed inhibitory activities on either superoxide anion generation or elastase release by human neutrophils in response to formyl-L-methionyl-L-leucyl-L-phenylalanine/cytochalasin B (FMLP/CB)

    Identification and phylogenetic analysis of orf virus from goats in Taiwan

    An outbreak of contagious ecthyma in goats in central Taiwan was investigated. The disease was diagnosed by physical and histopathologic examinations, and the etiology of the disease was identified as orf virus by electron microscopy and polymerase chain reaction (PCR) and sequence of major envelope protein (B2L) gene. The entire protein-coding region of B2L gene were cloned and sequenced. Phylogenetic analysis of B2L amino acid sequences showed that the orf virus identified in this outbreak was closer to the Indian ORFV-Mukteswar 59/05 isolate. This is the first report on the molecular characterization of orf virus in Taiwan

Guidance does not specify which typing technique is most practical to base decisions on. Aim: We explore the usefulness of whole genome sequencing (WGS) in the investigation of a Pseudomonas aeruginosa outbreak describing how it compares with pulsed-field gel electrophoresis (PFGE) and variable number tandem repeat (VNTR) analysis. Methods: Six patient isolates and six environmental samples from an Intensive Care Unit (ICU) positive for P. aeruginosa over two years underwent VNTR, PFGE and WGS. Findings:  VNTR and PFGE were required to fully determine the potential source of infection and rule out others. WGS results unambiguously distinguished linked isolates giving greater assurance of the transmission route between wash hand basin (WHB) water and two patients supporting control measures employed. Conclusion:  WGS provided detailed information without need for further typing. When allied to epidemiological information it can be used to understand outbreak situations rapidly and with certainty. Implementation of WGS in real-time would be a major advance in day-to-day practice. It could become a standard of care as it becomes more widespread due to its reproducibility and reduction in costs.Publisher PDFPeer reviewe
