From patterned response dependency to structured covariate dependency: categorical-pattern-matching
Data generated from a system of interest typically consists of measurements
from an ensemble of subjects across multiple response and covariate features,
and is naturally represented by one response-matrix against one
covariate-matrix. Likely each of these two matrices simultaneously embraces
heterogeneous data types: continuous, discrete and categorical. Here a matrix
is used as a practical platform to ideally keep hidden dependency among/between
subjects and features intact on its lattice. Response and covariate dependency
is individually computed and expressed through mutliscale blocks via a newly
developed computing paradigm named Data Mechanics. We propose a categorical
pattern matching approach to establish causal linkages in a form of information
flows from patterned response dependency to structured covariate dependency.
The strength of an information flow is evaluated by applying the combinatorial
information theory. This unified platform for system knowledge discovery is
illustrated through five data sets. In each illustrative case, an information
flow is demonstrated as an organization of discovered knowledge loci via
emergent visible and readable heterogeneity. This unified approach
fundamentally resolves many long standing issues, including statistical
modeling, multiple response, renormalization and feature selections, in data
analysis, but without involving man-made structures and distribution
assumptions. The results reported here enhance the idea that linking patterns
of response dependency to structures of covariate dependency is the true
philosophical foundation underlying data-driven computing and learning in

How the Design Leadership and Strategic Design Drive New Value in Enterprises and Organizations
Today's design has shifted from the original primary pursuit of appearance and function to processes and systems, creating new meanings for corporate strategies, products and services. Nowadays, enterprises are increasingly paying attention to the power of design, and regard design as the main innovation method and incorporate it into the organizations. Design began to be considered a key role at the top of large organizations. Although people recognize that design can bring good effects to enterprises and organizations, and there is growing interest in cultivating design thinking, what are the leaders who can lead enterprises, organizations and have design thinking? Their characteristics and how to formulate and plan strategic design still not fully elucidated. Therefore, this research uses related theories to understand the way of thinking and characteristics of design leaders, only in this way we formulate good strategic design for enterprises and organizations, and become an indispensable and important help in today rapidly changing world

Crystallization of Adenylylsulfate Reductase from Desulfovibrio gigas: A Strategy Based on Controlled Protein Oligomerization
Adenylylsulfate reductase (adenosine 5′-phosphosulfate reductase, APS reductase or APSR, E.C.1.8.99.2) catalyzes the conversion of APS to sulfite in dissimilatory sulfate reduction. APSR was isolated and purified directly from massive anaerobically grown Desulfovibrio gigas, a strict anaerobe, for structure and function investigation. Oligomerization of APSR to form dimers–α_2β_2, tetramers–α_4β_4, hexamers–α_6β_6, and larger oligomers was observed during purification of the protein. Dynamic light scattering and ultracentrifugation revealed that the addition of adenosine monophosphate (AMP) or adenosine 5′-phosphosulfate (APS) disrupts the oligomerization, indicating that AMP or APS binding to the APSR dissociates the inactive hexamers into functional dimers. Treatment of APSR with β-mercaptoethanol decreased the enzyme size from a hexamer to a dimer, probably by disrupting the disulfide Cys156—Cys162 toward the C-terminus of the β-subunit. Alignment of the APSR sequences from D. gigas and A. fulgidus revealed the largest differences in this region of the β-subunit, with the D. gigas APSR containing 16 additional amino acids with the Cys156—Cys162 disulfide. Studies in a pH gradient showed that the diameter of the APSR decreased progressively with acidic pH. To crystallize the APSR for structure determination, we optimized conditions to generate a homogeneous and stable form of APSR by combining dynamic light scattering, ultracentrifugation, and electron paramagnetic resonance methods to analyze the various oligomeric states of the enzyme in varied environments
Red Teaming Language Model Detectors with Language Models
The prevalence and strong capability of large language models (LLMs) present
significant safety and ethical risks if exploited by malicious users. To
prevent the potentially deceptive usage of LLMs, recent works have proposed
algorithms to detect LLM-generated text and protect LLMs. In this paper, we
investigate the robustness and reliability of these LLM detectors under
adversarial attacks. We study two types of attack strategies: 1) replacing
certain words in an LLM's output with their synonyms given the context; 2)
automatically searching for an instructional prompt to alter the writing style
of the generation. In both strategies, we leverage an auxiliary LLM to generate
the word replacements or the instructional prompt. Different from previous
works, we consider a challenging setting where the auxiliary LLM can also be
protected by a detector. Experiments reveal that our attacks effectively
compromise the performance of all detectors in the study with plausible
generations, underscoring the urgent need to improve the robustness of

Early Evolutionary Selection of NAD Biosynthesis Pathway in Bacteria
Bacteria use two alternative pathways to synthesize nicotinamide adenine dinucleotide (NAD) from nicotinamide (Nam). A short, two-step route proceeds through nicotinamide mononucleotide (NMN) formation, whereas the other pathway, a four-step route, includes the deamidation of Nam and the reamidation of nicotinic acid adenine dinucleotide (NAAD) to NAD. In addition to having twice as many enzymatic steps, the four-step route appears energetically unfavourable, because the amidation of NAAD includes the cleavage of ATP to AMP. Therefore, it is surprising that this pathway is prevalent not only in bacteria but also in yeast and plants. Here, we demonstrate that the considerably higher chemical stability of the deamidated intermediates, compared with their amidated counterparts, might compensate for the additional energy expenditure, at least at elevated temperatures. Moreover, comprehensive bioinformatics analyses of the available >6000 bacterial genomes indicate that an early selection of one or the other pathway occurred. Correlation of the composite equilibrium score of computerized dynamic posturography and clinical balance tests
AbstractBackgroundThe computerized dynamic posturography has been widely used to access balance control in patients with balance dysfunction. A composite-equilibrium score (CS) can be calculated from the sensory organization test using the computerized dynamic posturography. However, the correlation between the composite equilibrium score and clinical tests and its ability to predict falls has rarely been explored in the past.MethodsA total of 60 patients with chief complaint of dizziness were enrolled in our study, and clinical assessments were done including the sensory organization test (SOT), Timed Up and Go test (TUG), Tinetti Performance-Oriented Mobility Assessment (POMA), and the dynamic gait index (DGI). The age and the subjective feeling of the severity of dizziness quantified by the visual analog scale (VAS) of each patient were also recorded.ResultsStatistical analysis revealed significant correlation between the composite equilibrium score and the TUG, POMA (gait, balance and total scores), and the DGI. However, there is statistically significant correlation between neither the CS and the age nor the VAS of dizziness. When grouping the DGI, POMA (total score), and the TUG cutoff to predict fall risks, the correlations to the CS can still be established except the TUG.ConclusionFrom the results of our study, the validity of the clinical tests was established in assessment of balance function, and clinicians can utilize these tools for preliminary evaluation of patient balance when computerized dynamic posturography is not available. Relationships between serum HER2 ECD, TIMP-1 and clinical outcomes in Taiwanese breast cancer
<p>Abstract</p> <p>Background</p> <p>Serum levels of the extracellular domain of HER2/neu (HER2 ECD) have been demonstrated to be associated with clinical outcomes. A disintegrin and metalloproteinase-10, a sheddase of HER2/neu, can drive cancer progression and its activity is inhibited by tissue inhibitor of metalloproteinase-1 (TIMP-1). However, elevated TIMP-1 expression has been associated with a poor prognosis of breast cancer. Therefore, this study was performed to explore the relationships between serum HER2 ECD, TIMP-1 and clinical outcomes.</p> <p>Methods</p> <p>One hundred and eighty-five female breast cancer patients, who received curative mastectomy without neo-adjuvant chemotherapy at Chang-Gung Memorial Hospital, were recruited with informed consent for this study. Pre-operative serum levels of HER2 ECD and TIMP-1 were measured using an enzyme-linked immunosorbent assay.</p> <p>Results</p> <p>Twenty-three cases (12.4%) were classified HER2 ECD positive. HER2 ECD positivity was significantly associated with age, lymph node involvement, histological grade, estrogen receptor status, progesterone receptor status, tissue HER2/neu overexpression, and disease-free survival (DFS). In an age, stage, ER and HER2/neu status matched subgroup (N = 41), the serum level of TIMP-1 was significantly associated with HER2 ECD positivity and DFS.</p> <p>Conclusions</p> <p>A high serum TIMP-1 was significantly associated with HER2 ECD positivity and a poorer DFS among Taiwanese primary breast cancer patients with HER2 overexpression.</p
