    Capturing the realities of sports programmes: systematic ‘messiness’?

    Early workings based on traditional methods (Schuman, 1967, as cited in Clarke and Dawson, 1999) have given way to more pragmatic, social paradigms with scientific realism (Pawson and Tilley, 1997) and evaluation utility (Patton, 2002) establishing evaluation research as a specialist area of applied social research. There has been more pressure for those who work in community sport to deliver with evaluation in mind. This can be interpreted as the government demanding greater accountability for its investment, but it is more than that. Community sport needs to modernise. It needs to be able to fully explain not just what works but why it works. Given the current economic and political instabilities, sport needs to work harder than ever to establish itself as a mainstream function of our communities needs and development (Coalter, 2007). Evaluation may not be a panacea but it will provide support in terms of evidence based decisions and stronger rationales for community sport’s existence. Evaluation is not an exact science and draws on a number of disciplines, using an eclectic repertoire of concepts and methods (Rossi, et al. 2004). To this end, this positional paper will reflect on the processes of a six-year community sport and physical activity strategy evaluation. Recommendations based on the reflection will be acknowledged. The strategy and the evaluation (Daniels, 2016) were developed by the Community Sport Network for this region and involved expertise from the Manchester Metropolitan University, public sector sport development and third sector sports clubs whose projects where supported by Sport England’s Community Investment Fund. The paper will present outcome patterns for the strategy and explain the methods that helped build them

    Multi-dimensional super-resolution imaging enables surface hydrophobicity mapping

    We have developed a multi-dimensional super-resolution (md-SR) imaging technique to determine both the localization and the environmental properties of single-molecule fluorescent emitters. The method, termed sPAINT, exploits the solvatochromic and fluorogenic properties of nile red to extract both the emission spectrum and the position of each dye molecule to enable the mapping of hydrophobicity of biological structures. We first validated the sPAINT method by studying synthetic lipid vesicles of known composition, then applied it to measure the hydrophobicity of amyloid fibrils and oligomers implicated in neurodegenerative diseases, and of the plasma membrane of mammalian cells. sPAINT is easily implemented by inserting a transmission diffraction grating into the optical path of a localization-based super-resolution microscope, which enables all the necessary information to be extracted simultaneously from a single image plane. sPAINT enables the hydrophobicity of surfaces to be mapped at the nanoscale in a dynamic fashion.Medical Research Council (Grant ID: MR/K015850/1), Engineering and Physical Sciences Research Council, Royal Society (University Research Fellowship, Grant ID: UF120277), Augustus Newman Foundation, Cambridge Advanced Imaging Centre, Christ’s Colleg

    Lessons from the removal of lead from gasoline for controlling other environmental pollutants: A case study from New Zealand

    <p>Abstract</p> <p>Background</p> <p>It took over two decades to achieve the removal of leaded gasoline in this country. This was despite international evidence and original research conducted in New Zealand on the harm to child cognitive function and behaviour from lead exposure.</p> <p>Objective</p> <p>To identify lessons from the New Zealand experience of removing leaded gasoline that are potentially relevant to the control of other environmental pollutants.</p> <p>Discussion</p> <p>From the available documentation, we suggest a number of reasons for the slow policy response to the leaded gasoline hazard. These include: (1) industry power in the form of successful lobbying by the lead additive supplier, Associated Octel; (2) the absence of the precautionary principle as part of risk management policy; and (3) weak policymaking machinery that included: (a) the poor use of health research evidence (from both NZ and internationally), as well as limited use of expertise in academic and non-governmental organisations; (b) lack of personnel competent in addressing technically complex issues; and (c) diffusion of responsibility among government agencies.</p> <p>Conclusion</p> <p>There is a need for a stronger precautionary approach by policymakers when considering environmental pollutants. Politicians, officials and health workers need to strengthen policymaking processes and effectively counter the industry tactics used to delay regulatory responses.</p

    Using sequence data to identify alternative routes and risk of infection: a case-study of campylobacter in Scotland

    &lt;b&gt;Background:&lt;/b&gt; Genetic typing data are a potentially powerful resource for determining how infection is acquired. In this paper MLST typing was used to distinguish the routes and risks of infection of humans with Campylobacter jejuni from poultry and ruminant sources.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Methods:&lt;/b&gt; C. jejuni samples from animal and environmental sources and from reported human cases confirmed between June 2005 and September 2006 were typed using MLST. The STRUCTURE software was used to assign the specific sequence types of the sporadic human cases to a particular source. We then used mixed case-case logistic regression analysis to compare the risk factors for being infected with C. jejuni from different sources.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Results:&lt;/b&gt; A total of 1,599 (46.3%) cases were assigned to poultry, 1,070 (31.0%) to ruminant and 67 (1.9%) to wild bird sources; the remaining 715 (20.7%) did not have a source that could be assigned with a probability of greater than 0.95. Compared to ruminant sources, cases attributed to poultry sources were typically among adults (odds ratio (OR) = 1.497, 95% confidence intervals (CIs) = 1.211, 1.852), not among males (OR = 0.834, 95% CIs = 0.712, 0.977), in areas with population density of greater than 500 people/km(2) (OR = 1.213, 95% CIs = 1.030, 1.431), reported in the winter (OR = 1.272, 95% CIs = 1.067, 1.517) and had undertaken recent overseas travel (OR = 1.618, 95% CIs = 1.056, 2.481). The poultry assigned strains had a similar epidemiology to the unassigned strains, with the exception of a significantly higher likelihood of reporting overseas travel in unassigned strains.&lt;p&gt;&lt;/p&gt; &lt;b&gt;Conclusions:&lt;/b&gt; Rather than estimate relative risks for acquiring infection, our analyses show that individuals acquire C. jejuni infection from different sources have different associated risk factors. By enhancing our ability to identify at-risk groups and the times at which these groups are likely to be at risk, this work allows public health messages to be targeted more effectively. The rapidly increasing capacity to conduct genetic typing of pathogens makes such traced epidemiological analysis more accessible and has the potential to substantially enhance epidemiological risk factor studies

    Genetic nurse counsellors can be an acceptable and cost-effective alternative to clinical geneticists for breast cancer risk genetic counselling. Evidence from two parallel randomised controlled equivalence trials

    This study compared genetic nurse counsellors with standard services for breast cancer genetic risk counselling services in two regional genetics centres, in Grampian region, North East Scotland and in Cardiff, Wales. Women referred for genetic counselling were randomised to an initial genetic counselling appointment with either a genetic nurse counsellor (intervention) or a clinical geneticist (current service, control). Participants completed postal questionnaires before, immediately after the counselling episode and 6 months later to assess anxiety, general health status, perceived risk and satisfaction. A parallel economic evaluation explored factors influencing cost-effectiveness. The two concurrent randomised controlled equivalence trials were conducted and analysed separately. In the Grampian trial, 289 patients (193 intervention, 96 control) and in the Wales trial 297 patients (197 intervention and 100 control) returned a baseline questionnaire and attended their appointment. Analysis suggested at least likely equivalence in anxiety (the primary outcome) between the two arms of the trials. The cost per counselling episode was £11.54 less for nurse-based care in the Grampian trial and £12.50 more for nurse-based care in Cardiff. The costs were sensitive to the grade of doctor (notionally) replaced and the extent of consultant supervision required by the nurse. In conclusion, care based on genetic nurse counsellors was not significantly different from conventional cancer genetic services in both trial locations

    The Chemical Information Ontology: Provenance and Disambiguation for Chemical Data on the Biological Semantic Web

    Cheminformatics is the application of informatics techniques to solve chemical problems in silico. There are many areas in biology where cheminformatics plays an important role in computational research, including metabolism, proteomics, and systems biology. One critical aspect in the application of cheminformatics in these fields is the accurate exchange of data, which is increasingly accomplished through the use of ontologies. Ontologies are formal representations of objects and their properties using a logic-based ontology language. Many such ontologies are currently being developed to represent objects across all the domains of science. Ontologies enable the definition, classification, and support for querying objects in a particular domain, enabling intelligent computer applications to be built which support the work of scientists both within the domain of interest and across interrelated neighbouring domains. Modern chemical research relies on computational techniques to filter and organise data to maximise research productivity. The objects which are manipulated in these algorithms and procedures, as well as the algorithms and procedures themselves, enjoy a kind of virtual life within computers. We will call these information entities. Here, we describe our work in developing an ontology of chemical information entities, with a primary focus on data-driven research and the integration of calculated properties (descriptors) of chemical entities within a semantic web context. Our ontology distinguishes algorithmic, or procedural information from declarative, or factual information, and renders of particular importance the annotation of provenance to calculated data. The Chemical Information Ontology is being developed as an open collaborative project.     Comparative Transcriptional and Genomic Analysis of Plasmodium falciparum Field Isolates

    Mechanisms for differential regulation of gene expression may underlie much of the phenotypic variation and adaptability of malaria parasites. Here we describe transcriptional variation among culture-adapted field isolates of Plasmodium falciparum, the species responsible for most malarial disease. It was found that genes coding for parasite protein export into the red cell cytosol and onto its surface, and genes coding for sexual stage proteins involved in parasite transmission are up-regulated in field isolates compared with long-term laboratory isolates. Much of this variability was associated with the loss of small or large chromosomal segments, or other forms of gene copy number variation that are prevalent in the P. falciparum genome (copy number variants, CNVs). Expression levels of genes inside these segments were correlated to that of genes outside and adjacent to the segment boundaries, and this association declined with distance from the CNV boundary. This observation could not be explained by copy number variation in these adjacent genes. This suggests a local-acting regulatory role for CNVs in transcription of neighboring genes and helps explain the chromosomal clustering that we observed here. Transcriptional co-regulation of physical clusters of adaptive genes may provide a way for the parasite to readily adapt to its highly heterogeneous and strongly selective environment
