    The Interactive Effects of Multiple Stressors on Lithobates catesbeianus and Anaxyrus americanus

    Amphibian populations worldwide have experienced dramatic declines, and many species have already become locally, regionally, or globally extirpated with thousands more being threatened with extinction. These declines have occurred more rapidly in amphibians than any other group of vertebrates, which is especially concerning to scientists because amphibians serve as indicator species of overall environmental health. Major causes for amphibian declines are discussed in Chapter 1 and include: habitat modification and destruction, commercial over-exploitation, introduced species, environmental contaminants, global climate change, and infectious diseases. Chapter 2 discusses the major research aspects of the thesis by examining the interactive effects of multiple stressors on two species of larval amphibians. The study investigated the individual and combined effects of a major environmental contaminant (Glyphosate, commercial Roundup ®), increased temperatures, and predatory cues on survival, growth, and development of tadpoles from two species (Lithobates catesbeianus and Anaxyrus americanus). Glyphosate reduced tadpole survival in both amphibian species and becomes more toxic to tadpoles as temperature increases. Increased temperature reduced survivorship over time in both species; however, survivorship decreased only when temperature interacted with glyphosate. Increased temperature also caused a decrease in growth in L. catesbeianus and an increase in growth and development in A. americanus. Accelerated growth and development caused by temperature may ameliorate the adverse effects of glyphosate by reducing larval period and increasing size at metamorphosis. Glyphosate caused significant anatomical shape variation in L. catebeianus, while increased temperature caused significant anatomical shape variation in A. americanus. The shape variations caused by the different stressors may lead to further developmental and behavioral abnormalities. Predatory cues had no effect on A. americanus survival, and only decreased growth and development at intermediate glyphosate concentrations and temperatures; therefore, the effects of temperature and glyphosate concentration may have been enhanced in the presence of predatory cues. The study highlighted the importance of examining the interactions between multiple stressors on amphibian declines. Chapter 3 focuses on potential solutions for global amphibian declines. Conservation efforts such as educational outreach, effective land management and water quality regulation guidelines, captive breeding programs, and several others are discussed

    In this paper, we propose a model where N strategic informed traders who are endowed with heterogeneous noisy signals with different precisions compete in a market with a single risky asset. We explicitly describe the unique linear equilibrium that exists in this setup and derive its properties. Moreover, we focus on the effects of noise on the competition between traders. We show that noise softens the competition between traders. In particular, for N exceeding three and for certain sets of noise in traders' signals, each trader's individual profit is greater than the one obtained in the case of perfect information

    The Application of Three-Dimensional Collagen-Scaffolds Seeded with Myoblasts to Repair Skeletal Muscle Defects

    Three-dimensional (3D) engineered tissue constructs are a novel and promising approach to tissue repair and regeneration. 3D tissue constructs have the ability to restore form and function to damaged soft tissue unlike previous methods, such as plastic surgery, which are able to restore only form, leaving the function of the soft tissue often compromised. In this study, we seeded murine myoblasts (C2C12) into a collagen composite scaffold and cultured the scaffold in a roller bottle cell culture system in order to create a 3D tissue graft in vitro. The 3D graft created in vitro was then utilized to investigate muscle tissue repair in vivo. The 3D muscle grafts were implanted into defect sites created in the skeletal muscles in mice. We detected that the scaffolds degraded slowly over time, and muscle healing was improved which was shown by an increased quantity of innervated and vascularized regenerated muscle fibers. Our results suggest that the collagen composite scaffold seeded with myoblasts can create a 3D muscle graft in vitro that can be employed for defect muscle tissue repair in vivo

    Correction of a Depth-Dependent Lateral Distortion in 3D Super-Resolution Imaging

    Three-dimensional (3D) localization-based super-resolution microscopy (SR) requires correction of aberrations to accurately represent 3D structure. Here we show how a depth-dependent lateral shift in the apparent position of a fluorescent point source, which we term 'wobble', results in warped 3D SR images and provide a software tool to correct this distortion. This system-specific, lateral shift is typically > 80 nm across an axial range of similar to 1 mu m. A theoretical analysis based on phase retrieval data from our microscope suggests that the wobble is caused by non-rotationally symmetric phase and amplitude aberrations in the microscope's pupil function. We then apply our correction to the bacterial cytoskeletal protein FtsZ in live bacteria and demonstrate that the corrected data more accurately represent the true shape of this vertically-oriented ring-like structure. We also include this correction method in a registration procedure for dual-color, 3D SR data and show that it improves target registration error (TRE) at the axial limits over an imaging depth of 1 mu m, yielding TRE values of < 20 nm.     Comprehensive Immune Monitoring of Clinical Trials to Advance Human Immunotherapy

    The success of immunotherapy has led to a myriad of clinical trials accompanied by efforts to gain mechanistic insight and identify predictive signatures for personalization. However, many immune monitoring technologies face investigator bias, missing unanticipated cellular responses in limited clinical material. We present here a mass cytometry (CyTOF) workflow for standardized, systems-level biomarker discovery in immunotherapy trials. To broadly enumerate immune cell identity and activity, we established and extensively assessed a reference panel of 33 antibodies to cover major cell subsets, simultaneously quantifying activation and immune checkpoint molecules in a single assay. This assay enumerates &gt;= 98% of peripheral immune cells with &gt;= 4 positively identifying antigens. Robustness and reproducibility are demonstrated on multiple samples types, across two research centers and by orthogonal measurements. Using automated analysis, we identify stratifying immune signatures in bone marrow transplantation-associated graft-versus-host disease. Together, this validated workflow ensures comprehensive immunophenotypic analysis and data comparability and will accelerate biomarker discovery

    Investor Competition Over Information and the Pricing of Information Asymmetry

    Whether the information environment affects the cost of capital is a fundamental question in accounting and finance research. Relying on theories about competition between informed investors as well as the pricing of information asymmetry, we hypothesize a cross-sectional variation in the pricing of information asymmetry that is conditional on competition. We develop and validate empirical proxies for competition using the number and concentration of institutional investor ownership. Using these proxies, we find a lower pricing of information asymmetry when there is more competition. Overall, our results suggest that competition between informed investors has an important effect on how the information environment affects the cost of capital.Deloitte Foundatio
