    Role of LRR-RLKS in stress response and developmental processes through systematic K.O. analysis in rice

    The function of a small number of Leucine-Rich Repeat Receptor-Like Kinase (LRR-RLK) genes has been studied, mostly in Arabidopsis thaliana. They are involved in several important biological processes, including development, resistance/defence and adaptation to biotic and abiotic stresses. We are interested in deciphering the function of these genes in rice. In this aim, we decided to take advantage of the availability of T-DNA and Tos17 insertion mutant libraries of rice cv. Nipponbare to start a systematic screen of knock-out mutants of these genes. The rice genome contains ~320 LRRRLK genes. Our search in international collections of insertion mutant lines which are catalogued by Flanking Sequence Tag (FST) information from most of the rice insertional mutagenesis projects around the world revealed that 238 insertional mutant lines are available for the 323 genes. Phenotypical evaluation of homozygous mutants is performed under various biotic and abiotic stresses and mutant plants are observed for phenotypes at several stages of growth, from seedlings in Petri dishes to flowering and grain development stages in the greenhouse. Thus, with this project, we aim first to decipher and explore new genes involved in defence and developmental responses to stresses. Second, our analysis will be of great benefit to provide potential targets for cereal improvement, notably in the breeding of drought-tolerant and pathogen-resistant crops.     Identification of precursor transcripts for 6 novel miRNAs expands the diversity on the genomic organisation and expression of miRNA genes in rice

    The plant miRNAs represent an important class of endogenous small RNAs that guide cleavage of an mRNA target or repress its translation to control development and adaptation to stresses. MiRNAs are nuclear-encoded genes transcribed by RNA polymerase II, producing a primary precursor that is subsequently processed by DCL1 an RNase III Dicer-like protein. In rice hundreds of miRNAs have been described or predicted, but little is known on their genes and precursors which are important criteria to distinguish them from siRNAs. Here we develop a combination of experimental approaches to detect novel miRNAs in rice, identify their precursor transcripts and genes and predict or validate their mRNA targets. We produced four cDNA libraries from small RNA fractions extracted from distinct rice tissues. By in silico analysis we selected 6 potential novel miRNAs, and confirmed that their expression requires OsDCL1. We predicted their targets and used 5'RACE to validate cleavage for three of them, targeting a PPR, an SPX domain protein and a GT-like transcription factor respectively. In addition, we identified precursor transcripts for the 6 miRNAs expressed in rice, showing that these precursors can be efficiently processed using a transient expression assay in transfected Nicotiana benthamiana leaves. Most interestingly, we describe two precursors producing tandem miRNAs, but in distinct arrays. We focus on one of them encoding osa-miR159a.2, a novel miRNA produced from the same stem-loop structure encoding the conserved osa-miR159a.1. We show that this dual osa-miR159a.2-osa-miR159a.1 structure is conserved in distant rice species and maize. Finally we show that the predicted mRNA target of osa-miR159a.2 encoding a GT-like transcription factor is cleaved in vivo at the expected site. The combination of approaches developed here identified six novel miRNAs expressed in rice which can be clearly distinguished from siRNAs. Importantly, we show that two miRNAs can be produced from a single precursor, either from tandem stem-loops or tandemly arrayed in a single stem-loop. This suggests that processing of these precursors could be an important regulatory step to produce one or more functional miRNAs in plants and perhaps coordinate cleavage of distinct targets in the same plant tissue.     Computation with perturbed dynamical systems

    This paper analyzes the computational power of dynamical systems robust to infinitesimal perturbations. Previous work on the subject has delved on very specific types of systems. Here we obtain results for broader classes of dynamical systems (including those systems defined by Lipschitz/analytic functions). In particular we show that systems robust to infinitesimal perturbations only recognize recursive languages. We also show the converse direction: every recursive language can be robustly recognized by a computable system. By other words we show that robustness is equivalent to decidability.     Transcultural engagement with Polish memory of the Holocaust while watching Leszek Wosiewicz's Kornblumenblau

    Kornblumenblau (Leszek Wosiewicz 1989) is a film that explores the experience of a Polish political prisoner interned at Auschwitz I. It particularly foregrounds issues related to Polish-Jewish relations during the Holocaust in its diegesis. Holocaust films are often discussed in relation to representation and the cultural specificity of their production context. However, this paper suggests thinking about film and topographies, the theme of this issue, not in relation to where a work is produced but in regards to the spectatorial space. It adopts a phenomenological approach to consider how, despite Kornblumenblau's particularly Polish themes, it might address the transcultural spectator and draw attention to the broader difficulties one faces when attempting to remember the Holocaust. Influenced particularly by the writing of Jennifer M. Barker and Laura U. Marks, this paper suggests that film possesses a body ¬¬- a display of intentionality, beyond those presented within the diegesis, which engages in dialogue with the spectator. During the experience of viewing Kornblumenblau, this filmic corporeality draws attention to the difficulties of confronting the Holocaust in particularly haptic ways, as the film points to the unreliability of visual historical sources, relates abject sensations to concentrationary spaces and breaks down as it confronts the scene of the gas chamber

    Strange Attractors in Dissipative Nambu Mechanics : Classical and Quantum Aspects

    We extend the framework of Nambu-Hamiltonian Mechanics to include dissipation in R3R^{3} phase space. We demonstrate that it accommodates the phase space dynamics of low dimensional dissipative systems such as the much studied Lorenz and R\"{o}ssler Strange attractors, as well as the more recent constructions of Chen and Leipnik-Newton. The rotational, volume preserving part of the flow preserves in time a family of two intersecting surfaces, the so called {\em Nambu Hamiltonians}. They foliate the entire phase space and are, in turn, deformed in time by Dissipation which represents their irrotational part of the flow. It is given by the gradient of a scalar function and is responsible for the emergence of the Strange Attractors. Based on our recent work on Quantum Nambu Mechanics, we provide an explicit quantization of the Lorenz attractor through the introduction of Non-commutative phase space coordinates as Hermitian N×N N \times N matrices in R3 R^{3}. They satisfy the commutation relations induced by one of the two Nambu Hamiltonians, the second one generating a unique time evolution. Dissipation is incorporated quantum mechanically in a self-consistent way having the correct classical limit without the introduction of external degrees of freedom. Due to its volume phase space contraction it violates the quantum commutation relations. We demonstrate that the Heisenberg-Nambu evolution equations for the Quantum Lorenz system give rise to an attracting ellipsoid in the 3N23 N^{2} dimensional phase space.Comment: 35 pages, 4 figures, LaTe

    The Epstein-Barr Virus G-Protein-Coupled Receptor Contributes to Immune Evasion by Targeting MHC Class I Molecules for Degradation

    Epstein-Barr virus (EBV) is a human herpesvirus that persists as a largely subclinical infection in the vast majority of adults worldwide. Recent evidence indicates that an important component of the persistence strategy involves active interference with the MHC class I antigen processing pathway during the lytic replication cycle. We have now identified a novel role for the lytic cycle gene, BILF1, which encodes a glycoprotein with the properties of a constitutive signaling G-protein-coupled receptor (GPCR). BILF1 reduced the levels of MHC class I at the cell surface and inhibited CD8+ T cell recognition of endogenous target antigens. The underlying mechanism involves physical association of BILF1 with MHC class I molecules, an increased turnover from the cell surface, and enhanced degradation via lysosomal proteases. The BILF1 protein of the closely related CeHV15 c1-herpesvirus of the Rhesus Old World primate (80% amino acid sequence identity) downregulated surface MHC class I similarly to EBV BILF1. Amongst the human herpesviruses, the GPCR encoded by the ORF74 of the KSHV c2-herpesvirus is most closely related to EBV BILF1 (15% amino acid sequence identity) but did not affect levels of surface MHC class I. An engineered mutant of BILF1 that was unable to activate G protein signaling pathways retained the ability to downregulate MHC class I, indicating that the immune-modulating and GPCR-signaling properties are two distinct functions of BILF1. These findings extend our understanding of the normal biology of an important human pathogen. The discovery of a third EBV lytic cycle gene that cooperates to interfere with MHC class I antigen processing underscores the importance of the need for EBV to be able to evade CD8+ T cell responses during the lytic replication cycle, at a time when such a large number of potential viral targets are expressed

    Nuevas perspectivas en los estudios de la motivación en las clases de Educación Física en contextos de vulnerabilidad

    En la presente ponencia relataremos algunos avances investigativos sobre el tema motivación en clases de Educación Física en contextos de vulnerabilidad. Nuestro punto de partida son observaciones realizadas en escuelas primarias en el marco de proyectos de investigación desarrollados en la UNLP (Argentina), en los cuales hemos intentado indagar en cuestiones tales como género, interacción grupal, significado de las prácticas motrices, y niveles de satisfacción y ansiedad ante la clase. En este trabajo que aquí presentamos, y que forma parte de otro mayor que está en curso de elaboración, hemos retomado como eje de análisis los núcleos de significación motriz (Gómez, 1997, 2002). Particularmente nuestro interés se ha centrado indagar en cómo, desde esta perspectiva, los profesores/as a cargo gestionan las clases y la resolución de las problemáticas que surgen, y preguntándonos asimismo si los/las docentes logran contemplar qué mueve a los niños/as a actuar, tanto a la hora del planeamiento como al momento de la prácticaMesa de Trabajo 6: Educación Física y DidácticaFacultad de Humanidades y Ciencias de la Educació
