    The potential for constructed wetlands to treat alkaline bauxite-residue leachate: phragmites australis growth

    High alkalinity (pH > 12) of bauxite-residue leachates presents challenges for the long-term storage and managements of the residue. Recent evidence has highlighted the potential for constructed wetlands to effectively buffer the alkalinity, but there is limited evidence on the potential for wetland plants to establish and grow in soils inundated with residue leachate. A pot-based trial was conducted to investigate the potential for Phragmites australis to establish and grow in substrate treated with residue leachate over a pH range of 8.6–11.1. The trial ran for 3 months, after which plant growth and biomass were determined. Concentrations of soluble and exchangeable trace elements in the soil substrate and also in the aboveground and belowground biomass were determined. Residue leachate pH did not affect plant biomass or microbial biomass. With the exception of Na, there was no effect on exchangeable trace elements in the substrate; however, increases in soluble metals (As, Cd and Na) were observed with increasing leachate concentration. Furthermore, increases in Al, As and V were observed in belowground biomass and for Cd and Cr in aboveground biomass. Concentrations within the vegetation biomass were less than critical phytotoxic levels. Results demonstrate the ability for P. australis to grow in bauxite-residue leachate-inundated growth media without adverse effects

    Hydrologic Properties of Pervious Concrete

    Pervious concrete is concrete made by eliminating most or all of the fine aggregate (sand) in the concrete mix, which allows interconnected void spaces to be formed in the hardened product. These interconnected void spaces allow the concrete to transmit water at relatively high rates. The main objective of this project was to conduct research on the potential application of pervious concrete in agricultural settings, specifically for use in animal feed lots, manure storage pads, animal manure and bedding compost facilities, or floor systems in animal buildings. Laboratory tests were conducted on replicated samples of pervious concrete formed from two rock sources (river gravel and limestone) for coarse aggregates and different size fractions to determine hydrologic relationships. Linear relationships were found between density and porosity, density and permeability, porosity and permeability, and porosity and specific yield. The results suggest that properties such as permeability, porosity, and specific yield are not significantly affected by different aggregate types. However, density and porosity can be effective methods for predicting porosity, specific yield, and permeability. In addition, t-tests were conducted to determine the effect of aggregate types on the solid/liquid separation properties of the pervious concrete after adding composted beef cattle manure and bedding to the surface of the specimens. The amount of composted beef cattle manure and bedding retained within the specimens was significantly less (p = 0.012) when samples constructed of #8 river gravel were used rather than the other aggregates. The #8 river gravel also had significantly less reduction in permeability compared to other aggregates. Although the #8 river gravel had a different effect on the compost retained and the reduction in permeability for the specimens, all four aggregates exhibited a significant reduction in the permeability after the compost was applied

    Aneuploidy in oocytes is prevented by sustained CDK1 activity through degron masking in cyclin B1

    Successful mitosis requires that cyclin B1:CDK1 kinase activity remains high until chromosomes are correctly aligned on the mitotic spindle. It has therefore been unclear why, in mammalian oocyte meiosis, cyclin B1 destruction begins before chromosome alignment is complete. Here, we resolve this paradox and show that mouse oocytes exploit an imbalance in the ratio of cyclin B1 to CDK1 to control CDK1 activity; early cyclin B1 destruction reflects the loss of an excess of non-CDK1-bound cyclin B1 in late prometaphase, while CDK1-bound cyclin B1 is destroyed only during metaphase. The ordered destruction of the two forms of cyclin B1 is brought about by a previously unidentified motif that is accessible in free cyclin B1 but masked when cyclin B1 is in complex with CDK1. This protects the CDK1-bound fraction from destruction in prometaphase, ensuring a period of prolonged CDK1 activity sufficient to achieve optimal chromosome alignment and prevent aneuploidy

    Deliberate Practice in Simulation-Based Surgical Skills Training: A Scoping Review

    BackgroundIn recent years there has been a shift from traditional Halstedian methods towards more simulation-based medical education (SBME) for developing surgical skills. Questions remain about the role and value of SBME, although feedback and engagement in repetitive practice have been associated with positive learning outcomes. Regardless of approach, the principles of deliberate practice align with both the Halstedian traditions and ways of implementing SBME. Whilst deliberate practice is well described in the wider literature, the extent to which it is an effective instructional approach in surgical training remains unknown.ObjectiveTo explore the effectiveness of deliberate practice as an instructional design for developing surgical skills through SBME interventions, as assessed by improvements in trainee performance and/or patient outcomes.MethodsA combined search was conducted in PUBMED, CINAHL, EMBASE, MEDLINE, PSYCHINFO and Google Scholar. 301 articles were screened and 17 met the inclusion criteria for analysis.ResultsThere was heterogeneity of study methods with six randomised control trials, seven pre-test/post-test design, two non-randomised comparisons and two observational studies. All articles demonstrated positive learner outcomes following SBME with deliberate practice, although there was no direct comparison to another instructional method. Two studies demonstrated skill transfer to the clinical environment and one demonstrated improved patient outcomes. ConclusionDeliberate practice informed SBME interventions appeared effective for developing surgical skills among trainee surgeons, however the reliability of these conclusions was limited by the modest quality of the research studies and the design elements of deliberate practice were inconsistently applied. There was little evidence that deliberate practice leads to skills retention beyond 30 days, although participant numbers were low and the quality of studies was modest

    Trypanosoma brucei gambiense group 1 is distinguished by a unique amino acid substitution in the HpHb receptor implicated in human serum resistance

    Trypanosoma brucei rhodesiense (Tbr) and T. b. gambiense (Tbg), causative agents of Human African Trypanosomiasis (sleeping sickness) in Africa, have evolved alternative mechanisms of resisting the activity of trypanosome lytic factors (TLFs), components of innate immunity in human serum that protect against infection by other African trypanosomes. In Tbr, lytic activity is suppressed by the Tbr-specific serum-resistance associated (SRA) protein. The mechanism in Tbg is less well understood but has been hypothesized to involve altered activity and expression of haptoglobin haemoglobin receptor (HpHbR). HpHbR has been shown to facilitate internalization of TLF-1 in T.b. brucei (Tbb), a member of the T. brucei species complex that is susceptible to human serum. By evaluating the genetic variability of HpHbR in a comprehensive geographical and taxonomic context, we show that a single substitution that replaces leucine with serine at position 210 is conserved in the most widespread form of Tbg (Tbg group 1) and not found in related taxa, which are either human serum susceptible (Tbb) or known to resist lysis via an alternative mechanism (Tbr and Tbg group 2). We hypothesize that this single substitution contributes to reduced uptake of TLF and thus may play a key role in conferring serum resistance to Tbg group 1. In contrast, similarity in HpHbR sequence among isolates of Tbg group 2 and Tbb/Tbr provides further evidence that human serum resistance in Tbg group 2 is likely independent of HpHbR functio

    Towards Structure-Property-Function Relationships for Eumelanin

    We discuss recent progress towards the establishment of important structure-property-function relationships in eumelanins - key functional bio-macromolecular systems responsible for photo-protection and immune response in humans, and implicated in the development of melanoma skin cancer. We focus on the link between eumelanin's secondary structure and optical properties such as broad band UV-visible absorption and strong non-radiative relaxation; both key features of the photo-protective function. We emphasise the insights gained through a holistic approach combining optical spectroscopy with first principles quantum chemical calculations, and advance the hypothesis that the robust functionality characteristic of eumelanin is related to extreme chemical and structural disorder at the secondary level. This inherent disorder is a low cost natural resource, and it is interesting to speculate as to whether it may play a role in other functional bio-macromolecular systems.Comment: 19 pages, 8 figures, Invited highlight article for Soft Matte

    Hepatocyte membrane potential regulates serum insulin and insulin sensitivity by altering hepatic GABA release

    Hepatic lipid accumulation in obesity correlates with the severity of hyperinsulinemia and systemic insulin resistance. Obesity-induced hepatocellular lipid accumulation results in hepatocyte depolarization. We have established that hepatocyte depolarization depresses hepatic afferent vagal nerve firing, increases GABA release from liver slices, and causes hyperinsulinemia. Preventing hepatic GABA release or eliminating the ability of the liver to communicate to the hepatic vagal nerve ameliorates the hyperinsulinemia and insulin resistance associated with diet-induced obesity. In people with obesity, hepatic expression of GABA transporters is associated with glucose infusion and disposal rates during a hyperinsulinemic euglycemic clamp. Single-nucleotide polymorphisms in hepatic GABA re-uptake transporters are associated with an increased incidence of type 2 diabetes mellitus. Herein, we identify GABA as a neuro-hepatokine that is dysregulated in obesity and whose release can be manipulated to mute or exacerbate the glucoregulatory dysfunction common to obesity

    Environmental risk factors of pregnancy outcomes: A summary of recent meta-analyses of epidemiological studies.

    Background Various epidemiological studies have suggested associations between environmental exposures and pregnancy outcomes. Some studies have tempted to combine information from various epidemiological studies using meta-analysis. We aimed to describe the methodologies used in these recent meta-analyses of environmental exposures and pregnancy outcomes. Furthermore, we aimed to report their main findings. Methods We conducted a bibliographic search with relevant search terms. We obtained and evaluated 16 recent meta-analyses. Results The number of studies included in each reported meta-analysis varied greatly, with the largest number of studies available for environmental tobacco smoke. Only a small number of the studies reported having followed meta-analysis guidelines or having used a quality rating system. Generally they tested for heterogeneity and publication bias. Publication bias did not occur frequently. The meta-analyses found statistically significant negative associations between environmental tobacco smoke and stillbirth, birth weight and any congenital anomalies; PM2.5 and preterm birth; outdoor air pollution and some congenital anomalies; indoor air pollution from solid fuel use and stillbirth and birth weight; polychlorinated biphenyls (PCB) exposure and birth weight; disinfection by-products in water and stillbirth, small for gestational age and some congenital anomalies; occupational exposure to pesticides and solvents and some congenital anomalies; and agent orange and some congenital anomalies. Conclusions The number of meta-analyses of environmental exposures and pregnancy outcomes is small and they vary in methodology. They reported statistically significant associations between environmental exposures such as environmental tobacco smoke, air pollution and chemicals and pregnancy outcomes
