    Induced encystment improves resistance to preservation and storage of Acanthamoeba castellanii

    Several conditions that allow the preservation, storage and rapid, efficient recovery of viable Acanthamoeba castellanii organisms were investigated. The viability of trophozoites (as determined by time to confluence) significantly declined over a period of 12 months when stored at −70°C using dimethyl sulfoxide (DMSO; 5 or 10%) as cryopreservant. As A. castellanii are naturally capable of encystment, studies were undertaken to determine whether induced encystment might improve the viability of organisms under a number of storage conditions. A. castellanii cysts stored in the presence of Mg2+ at 4°C remained viable over the study period, although time to confluence was increased from approximately 8 days to approximately 24 days over the 12-month period. Storage of cysts at −70°C with DMSO (5 or 10%) or 40% glycerol, but not 80% glycerol as cryopreservants increased their viability over the 12-month study period compared with those stored at room temperature. Continued presence of Mg2+ in medium during storage had no adverse effects and generally improved recovery of viable organisms. The present study demonstrates that A. castellanii can be stored as a non-multiplicative form inexpensively, without a need for cryopreservation, for at least 12 months, but viability is increased by storage at −70°C

    The Toxoplasma gondii plastid replication and repair enzyme complex, PREX

    A plastid-like organelle, the apicoplast, is essential to the majority of medically and veterinary important apicomplexan protozoa including Toxoplasma gondii and Plasmodium. The apicoplast contains multiple copies of a 35 kb genome, the replication of which is dependent upon nuclear-encoded proteins that are imported into the organelle. In P. falciparum an unusual multi-functional gene, pfprex, was previously identified and inferred to encode a protein with DNA primase, DNA helicase and DNA polymerase activities. Herein, we report the presence of a prex orthologue in T. gondii. The protein is predicted to have a bi-partite apicoplast targeting sequence similar to that demonstrated on the PfPREX polypeptide, capable of delivering marker proteins to the apicoplast. Unlike the P. falciparum gene that is devoid of introns, the T. gondii prex gene carries 19 introns, which are spliced to produce a contiguous mRNA. Bacterial expression of the polymerase domain reveals the protein to be active. Consistent with the reported absence of a plastid in Cryptosporidium species, in silico analysis of their genomes failed to demonstrate an orthologue of prex. These studies indicate that prex is conserved across the plastid-bearing apicomplexans and may play an important role in the replication of the plastid genome

    Visible light reduces C. elegans longevity.

    The transparent nematode Caenorhabditis elegans can sense UV and blue-violet light to alter behavior. Because high-dose UV and blue-violet light are not a common feature outside of the laboratory setting, we asked what role, if any, could low-intensity visible light play in C. elegans physiology and longevity. Here, we show that C. elegans lifespan is inversely correlated to the time worms were exposed to visible light. While circadian control, lite-1 and tax-2 do not contribute to the lifespan reduction, we demonstrate that visible light creates photooxidative stress along with a general unfolded-protein response that decreases the lifespan. Finally, we find that long-lived mutants are more resistant to light stress, as well as wild-type worms supplemented pharmacologically with antioxidants. This study reveals that transparent nematodes are sensitive to visible light radiation and highlights the need to standardize methods for controlling the unrecognized biased effect of light during lifespan studies in laboratory conditions

    Recurrent glioma clinical trial, CheckMate-143: the game is not over yet

    Glioblastoma (GBM) is the most common, and aggressive, primary brain tumor in adults. With a median patient survival of less than two years, GBM represents one of the biggest therapeutic challenges of the modern era. Even with the best available treatment, recurrence rates are nearly 100% and therapeutic options at the time of relapse are extremely limited. Nivolumab, an anti-programmed cell death-1 (PD-1) monoclonal antibody, has provided significant clinical benefits in the treatment of various advanced cancers and represented a promising therapy for primary and recurrent GBM. CheckMate 143 (NCT 02017717) was the first large randomized clinical trial of PD pathway inhibition in the setting of GBM, including a comparison of nivolumab and the anti-VEGF antibody, bevacizumab, in the treatment of recurrent disease. However, preliminary results, recently announced in a WFNOS 2017 abstract, demonstrated a failure of nivolumab to prolong overall survival of patients with recurrent GBM, and this arm of the trial was prematurely closed. In this review, we discuss the basic concepts underlying the rational to target PD pathway in GBM, address implications of using immune checkpoint inhibitors in central nervous system malignancies, provide a rationale for possible reasons contributing to the failure of nivolumab to prolong survival in patients with recurrent disease, and analyze the future role of immune checkpoint inhibitors in the treatment of GBM

    YarpTp Notebooks a Tool for Teaching Programming

    This article shows the design, structure, and part of the functioning of YarpTP Notebooks, as a pedagogical proposal that has been evolving from the experience in the teaching practice of the course Fundamentals of Programming. It seeks to encourage students in the teaching of programming through interactive booklets designed with Jupyter Notebooks which, when executed, can control physical components (in this case, vehicles on a reduced scale). The student assumes a participatory role through these interactive booklets that leads them to develop a process of reflection on what, how, when it does it and what results it achieves with these. Analyzing the information received allowing you to identify and apply variables, expressions and general syntax of the Python language that leads them to understand a program, test the program, evaluate the data and the associated behavior to continue improving their development until formulating coherent and/or associated conclusions with the desired competencies in the course of Fundamentals of Programmin

    Ocorrência de incidentes de Segurança do Paciente e Carga de Trabalho de Enfermagem

    Indexación: Web of Science; Scopus; Scielo.Objective: to identify the relationship between the workload of the nursing team and the occurrence of patient safety incidents linked to nursing care in a public hospital in Chile. Method: quantitative, analytical, cross-sectional research through review of medical records. The estimation of workload in Intensive Care Units (ICUs) was performed using the Therapeutic Interventions Scoring System (TISS-28) and for the other services, we used the nurse/patient and nursing assistant/patient ratios. Descriptive univariate and multivariate analysis were performed. For the multivariate analysis we used principal component analysis and Pearson correlation. Results: 879 post-discharge clinical records and the workload of 85 nurses and 157 nursing assistants were analyzed. The overall incident rate was 71.1%. It was found a high positive correlation between variables workload (r = 0.9611 to r = 0.9919) and rate of falls (r = 0.8770). The medication error rates, mechanical containment incidents and self-removal of invasive devices were not correlated with the workload. Conclusions: the workload was high in all units except the intermediate care unit. Only the rate of falls was associated with the workload.Objetivo: identificar la relación entre la carga laboral del equipo de enfermería y la ocurrencia de incidentes de seguridad del paciente relacionados con los cuidados de enfermería en un hospital público de Chile. Método: investigación cuantitativa, analítica, transversal, realizada por revisión de fichas clínicas. La estimación de carga laboral en las Unidades de Cuidados Intensivos (UCI) se realizó utilizando el Índice de Intervenciones Terapéuticas (TISS-28) y, para el resto de los servicios, por la relación enfermera/paciente y auxiliar/paciente. Se realizó análisis descriptivo univariado y multivariado. Para el análisis multivariado se utilizó análisis de componentes principales y el test de correlación de Pearson. Resultados: se revisaron 879 fichas clínicas posteriores al alta y la carga laboral de 85 enfermeras y 157 auxiliares de enfermería. La tasa global de incidentes fue 71,1%. Se encontró correlación positiva alta entre las variables de carga laboral (r = 0,9611 a r = 0,9919) y tasa de caídas (r = 0,8770). Las tasas de error de medicación, incidente por contención mecánica y auto retiro de invasivos no se correlacionaron con la carga laboral. Conclusiones: la carga laboral fue elevada en todas las unidades a excepción de la unidad de cuidados intermedios. Sólo la tasa de caídas se asoció a la carga laboral.Objetivo: identificar a relação entre a carga de trabalho da equipe de enfermagem e a ocorrência de incidentes de segurança dos pacientes ligados aos cuidados de enfermagem de um hospital público no Chile. Método: pesquisa transversal analítica quantitativa através de revisão de prontuários médicos. A estimativa da carga de trabalho em Unidade de Terapia Intensiva (UTI) foi realizada utilizando o Índice de Intervenções Terapêuticas-TISS-28 e para os outros serviços, foram utilizados os cocientes enfermeira/paciente e auxiliar de enfermagem/ paciente. Foram feitas análises univariada descritiva e multivariada. Para a análise multivariada utilizou-se análise de componentes principais e correlação de Pearson. Resultados: foram analisados 879 prontuáriosclínicos de pós-alta e a carga de trabalho de 85 enfermeiros e 157 auxiliares de enfermagem. A taxa global de incidentes foi de 71,1%. Foi encontrada uma alta correlação positiva entre as variáveis de carga de trabalho (r = 0,9611 para r = 0,9919) e a taxa de quedas (r = 0,8770). As taxas de erro de medicação, incidentes de contenção mecânica e auto remoção de dispositivos invasivos não foram correlacionadas com a carga de trabalho. Conclusões: a carga de trabalho foi elevada em todas as unidades, exceto na unidade de cuidados intermédios. Apenas a taxa de quedas foi associada com a carga de trabalho.http://ref.scielo.org/tv7s8
