    The impact of the reform of the milk quota regime on the Italian dairy sector

    This paper analyses the impact of the milk quota regime reform, actually under discussion, on the European countries with a detailed focus on the Italian milk and dairy sector. The dismantling of the milk quota regime is already on the EU agenda, but how and when to do it is still matter of debate. A possibility is to enlarge gradually the size of the national quotas, up to the full dismantling in 2015 (“soft landing”). Meanwhile, the discussion on Health Check of the CAP is under way. In this work we analyse the possible impacts of the reform of the milk quota regime on the basis of a Computable General Equilibrium (CGE) approach, using two models in sequence: the Global Trade Analysis Project (GTAP) model is used to evaluate the impacts of different scenarios of milk quota reform on the EU market and to compute the price changes outside Italy; these, in turn, are used as inputs for the MEG-D model, that focuses on the Italian milk and dairy market. The two models were run together with two specific objectives: the first was to avoid, in evaluating the impacts of reform of the milk sector regime in Italy, running the model with rough price estimates taken for secondary sources; the second, to have more specific results on the outcome at the national level. Particularly, the model takes in account the particular relevance of quality products (GDO) in Italian diary sector.     In the last few years the Pichia pastoris expression system has been gaining more and more interest for the expression of recombinant proteins. Many groups have employed fermentation technology in their investigations because the system is fairly easy to scale up and suitable for the production in the milligram to gram range. A large number of heterologous proteins from different sources has been expressed, but the fermentation process technology has been investigated to a lesser extent. A large number of fermentations are carried out in standard bioreactors that may be insufficiently equipped to meet the demands of high-cell-density fermentations of methylotrophic yeasts. In particular, the lack of on-line methanol analysis leads to fermentation protocols that may impair the optimal expression of the desired products. We have used a commercially available methanol sensor to investigate in detail the effects of supplementary glycerol feeding while maintaining a constant methanol concentration during the induction of a Mut+ strain of Pichia pastoris. Specific glycerol feed rates in the range of 38-4.2 mg × g(exp -1) × h(exp -1) (mg glycerol per gram fresh weight per hour) were investigated. Expression of the recombinant scFv antibody fragment was only observed at specific feed rates below 6 mg × g(exp -1) × h(exp -1). At low specific feed rates, growth was even lower than with methanol as the sole carbon source and the harvest expression level of the scFv was only half of that found in the control fermentation. These results show that glycerol inhibits expression driven by the AOX1 promoter even at extremely limited availability and demonstrate the benefits of on-line methanol control in Pichia fermentation research

    Electrode thickness measurement of a Si(Li) detector for the SIXA array

    Cathode electrodes of the Si(Li) detector elements of the SIXA X-ray spectrometer array are formed by gold-palladium alloy contact layers. The equivalent thickness of gold in one element was measured by observing the characteristic L-shell X-rays of gold excited by monochromatised synchrotron radiation with photon energies above the L3 absorption edge of gold. The results obtained at 4 different photon energies below the L2 edge yield an average value of 22.4(35) nm which is consistent with the earlier result extracted from detection efficiency measurements.     A symmetric method for weakly imposing Dirichlet boundary conditions in embedded finite element meshes

    In this paper, we propose a way to weakly prescribe Dirichlet boundary conditions in embedded finite element meshes. The key feature of the method is that the algorithmic parameter of the formulation which allows to ensure stability is independent of the numerical approximation, relatively small, and can be fixed a priori. Moreover, the formulation is symmetric for symmetric problems. An additional element‐discontinuous stress field is used to enforce the boundary conditions in the Poisson problem. Additional terms are required in order to guarantee stability in the convection–diffusion equation and the Stokes problem. The proposed method is then easily extended to the transient Navier–Stokes equations
