    In the last few years the Pichia pastoris expression system has been gaining more and more interest for the expression of recombinant proteins. Many groups have employed fermentation technology in their investigations because the system is fairly easy to scale up and suitable for the production in the milligram to gram range. A large number of heterologous proteins from different sources has been expressed, but the fermentation process technology has been investigated to a lesser extent. A large number of fermentations are carried out in standard bioreactors that may be insufficiently equipped to meet the demands of high-cell-density fermentations of methylotrophic yeasts. In particular, the lack of on-line methanol analysis leads to fermentation protocols that may impair the optimal expression of the desired products. We have used a commercially available methanol sensor to investigate in detail the effects of supplementary glycerol feeding while maintaining a constant methanol concentration during the induction of a Mut+ strain of Pichia pastoris. Specific glycerol feed rates in the range of 38-4.2 mg × g(exp -1) × h(exp -1) (mg glycerol per gram fresh weight per hour) were investigated. Expression of the recombinant scFv antibody fragment was only observed at specific feed rates below 6 mg × g(exp -1) × h(exp -1). At low specific feed rates, growth was even lower than with methanol as the sole carbon source and the harvest expression level of the scFv was only half of that found in the control fermentation. These results show that glycerol inhibits expression driven by the AOX1 promoter even at extremely limited availability and demonstrate the benefits of on-line methanol control in Pichia fermentation research

    Rapid Formation and Flexible Expression of Memories of Subliminal Word Pairs

    Our daily experiences are incidentally and rapidly encoded as episodic memories. Episodic memories consist of numerous associations (e.g., who gave what to whom where and when) that can be expressed flexibly in new situations. Key features of episodic memory are speed of encoding, its associative nature, and its representational flexibility. Another defining feature of human episodic memory has been consciousness of encoding/retrieval. Here, we show that humans can rapidly form associations between subliminal words and minutes later retrieve these associations even if retrieval words were conceptually related to, but different from encoding words. Because encoding words were presented subliminally, associative encoding, and retrieval were unconscious. Unconscious association formation and retrieval were dependent on a preceding understanding of task principles. We conclude that key computations underlying episodic memory – rapid encoding and flexible expression of associations – can operate outside consciousness

    Abstract Hibernation, a hypometabolic state associated with low body temperature and reduced metabolic and activity rates, represents one adaptation to harsh seasonal environmental conditions. As a consequence of hypometabolism, energetically costly neuronal processes also ought to be reduced. Since active neuronal pathways are prerequisites for learning and memory, and because previous studies revealed variable patterns, it remains unclear whether and how hibernating animals retain memories, however. Here, we investigated the effect of seasonally reduced activity on memory retention in 36 wild grey mouse lemurs (Microcebus murinus). Data from activity loggers confirmed that female grey mouse lemurs entered hibernation during the cool dry season, whereas males exhibited episodic bursts of activity throughout the austral winter. Thus, compared to males, we predicted females to show lower memory retention of visual and spatial stimulus?reward associations learned before hibernation. In contrast to our prediction, all individuals performed worse in the post-hibernation testing session in both types of tests, compared to the pre-hibernation learning session, and males (N =?11) performed even worse than females (N =?14) in the post-hibernation testing session. Although females (N =?9) equipped with activity loggers tended to be less active than males (N =?4), sex-specific activity levels were unrelated to interindividual differences in memory retention. Hence, the post-hibernation decrease in performance of grey mouse lemurs may reflect a more general disability to retain stimulus?reward associations than a lack of memory retention due to seasonal hypometabolism, as suggested for some species of bats or squirrels

    The interstellar oxygen-K absorption edge as observed by XMM-Newton

    High resolution X-ray spectra of the Reflection Grating Spectrometer (RGS) on board the XMM satellite are used to resolve the oxygen K absorption edge. By combining spectra of low and high extinction sources, the observed absorption edge can be split in the true interstellar (ISM) extinction and the instrumental absorption. The detailed ISM edge structure closely follows the edge structure of neutral oxygen as derived by theoretical R-matrix calculations. However, the position of the theoretical edge requires a wavelength shift. In addition the detailed instrumental RGS absorption edge structure is presented.     Using a task-based approach in evaluating the usability of BoBIs in an e-book environment

    This paper reports on a usability evaluation of BoBIs (Back-of-the-book Indexes) as searching and browsing tools in an e-book environment. This study employed a task-based approach and within-subject design. The retrieval performance of a BoBI was compared with a ToC and Full-Text Search tool in terms of their respective effectiveness and efficiency for finding information in e-books. The results demonstrated that a BoBI was significantly more efficient (faster) and useful compared to a ToC or Full-Text Search tool for finding information in an e-book environment

    Vasopressinergic modulation of stress responses in the central amygdala of the Roman high-avoidance and low-avoidance rat

    The central nucleus of the amygdala (CEA) is selectively involved in the passive component of the behavioral (immobility) and the accompanying parasympathetic response during conditioned, stressful environmental challenges. Vasopressinergic mechanisms in the brain seem to play a role in these stress responses. The effects of the neuropeptides arginine-8-vasopressin (AVP) and oxytocin (OXT) on modulating CEA activity during conditioned stress of inescapable footshock were studied in male Roman high-avoidance (RHA/Verh) and low-avoidance (RLA/Verh) rats, psychogenetically selected on the basis of shuttle-box acquisition behavior. In RLA/Verh rats, the cardiac and behavioral responses to the conditioned emotional stressor were bradycardia and immobility, suggesting an important role for the CEA in these rats. The RHA/Verh rats, however, failed to show any change in heart rate or immobility in response to a conditioned stress situation. The low dose of AVP (20 pg) in the CEA of conscious RLA/Verh rats caused an enhancement of the stress-induced bradycardiac and immobility response. However, the high dose of AVP (2 ng) and OXT (200 pg) attenuated the bradycardiac and immobility responses in the RLA/Verh rats. Infusion of AVP and OXT in the RHA/Verh rats failed to induce any change in heart rate nr immobility. Binding studies revealed that the AVP receptor selectively binds AVP with high affinity. In contrast, the OXT receptor recognizes both AVP and OXT with a similar (but lower) affinity. This suggests that the behavioral and autonomic responses of the high dose of AVP may be caused by OXT receptor stimulation. In conclusion, on the basis of the present results one may hypothesize that CEA differences in AVP and OXT innervation and/or receptor densities may contribute to the differences in coping strategy found in these animals.

    LET spectra measurements of charged particles in the P0006 experiment on LDEF

    Measurements are under way of the charged particle radiation environment of the Long Duration Exposure Facility (LDEF) satellite using stacks of plastic nuclear track detectors (PNTD's) placed in different locations of the satellite. In the initial work the charge, energy, and linear energy transfer (LET) spectra of charged particles were measured with CR-39 double layer PNTD's located on the west side of the satellite (Experiment P0006). Primary and secondary stopping heavy ions were measured separately from the more energetic particles. Both trapped and galactic cosmic ray (GCR) particles are included, with the latter component being dominated by relativistic iron particles. The results from the P0006 experiment will be compared with similar measurements in other locations on LDEF with different orientation and shielding conditions. The remarkably detailed investigation of the charged particle radiation environment of the LDEF satellite will lead to a better understanding of the radiation environment of the Space Station Freedom. It will enable more accurate prediction of single event upsets (SEU's) in microelectronics and, especially, more accurate assessment of the risk - contributed by different components of the radiation field (GCR's, trapped protons, secondaries and heavy recoils, etc.) - to the health and safety of crew members

    Vanishing Fe 3d orbital moments in single-crystalline magnetite

    We show detailed magnetic absorption spectroscopy results of an in situ cleaved high quality single crystal of magnetite. In addition the experimental setup was carefully optimized to reduce drift, self absorption, and offset phenomena as far as possible. In strong contradiction to recently published data, our observed orbital moments are nearly vanishing and the spin moments are quite close to the integer values proposed by theory. This very important issue supports the half metallic full spin polarized picture of magnetite.Comment: 7 pages, 4 figure
