    MetaCook: FAIR Vocabularies Cookbook

    One of the prerequisites for FAIR data publication is the use of FAIR vocabularies. Currently, tools for the collaborative composition of such vocabularies are missing. For this reason, a universal manual and software for user-friendly vocabulary assembly is being composed in the HMC-funded MetaCook project. The project includes 4 separate test cases from 4 labs across KIT and Hereon, which will help strengthen the software\u27s universality and applicability to various domains. The components described in MetaCook will be implemented in the form of multiple software tools. The first one, a Python-based web application called VocPopuli, is the entry point for domain experts. The software, whose first version is being developed at the time of writing, enables the collaborative definition, and editing of metadata terms. Additionally, it annotates each term, as well as the entire vocabulary, with the help of the PROV Data Model (PROV-DM) - a schema used to describe the provenance of a given object. Finally, it assigns a unique ID to each term in the vocabulary, as well as a hash-based ID the vocabulary itself. The second software tool will facilitate the transformation of the vocabularies developed with the help of VocPopuli into ontologies. It will handle two distinct use cases – the from-scratch conversion of vocabularies into ontologies, and the augmentation of existing ontologies with the terms from a given thesaurus. Both software tools will be used by two semi-overlapping user groups: domain experts will input, edit, and discuss vocabulary terms in their area of interest, while vocabulary and ontology administrators will oversee the vocabulary creation, and ontology transformation. Both the controlled vocabularies and the corresponding ontologies offer the possibility to enrich data documented in Electronic Laboratory Notebooks (ELNs). As the simplest solution, terms used within the ELN are linked to the IDs of the related vocabulary and ontology for an unambiguous definition. Additionally, an export of the defined schemes can be used to automatically create a structured form in the ELNs for documenting the described processes.     TCF1(+) hepatitis C virus-specific CD8(+) T cells are maintained after cessation of chronic antigen stimulation.

    Differentiation and fate of virus-specific CD8(+) T cells after cessation of chronic antigen stimulation is unclear. Here we show that a TCF1(+)CD127(+)PD1(+) hepatitis C virus (HCV)-specific CD8(+) T-cell subset exists in chronically infected patients with phenotypic features of T-cell exhaustion and memory, both before and after treatment with direct acting antiviral (DAA) agents. This subset is maintained during, and for a long duration after, HCV elimination. After antigen re-challenge the less differentiated TCF1(+)CD127(+)PD1(+) population expands, which is accompanied by emergence of terminally exhausted TCF1-CD127-PD1(hi) HCV-specific CD8(+) T cells. These results suggest the TCF1(+)CD127(+)PD1(+) HCV-specific CD8(+) T-cell subset has memory-like characteristics, including antigen-independent survival and recall proliferation. We thus provide evidence for the establishment of memory-like virus-specific CD8(+) T cells in a clinically relevant setting of chronic viral infection and we uncover their fate after cessation of chronic antigen stimulation, implicating a potential strategy for antiviral immunotherapy

    Chronic infections promote the terminal differentiation (or "exhaustion") of T cells and are thought to preclude the formation of memory T cells. In contrast, we discovered a small subpopulation of virus-specific CD8(+) T cells that sustained the T cell response during chronic infections. These cells were defined by, and depended on, the expression of the transcription factor Tcf1. Transcriptome analysis revealed that this population shared key characteristics of central memory cells but lacked an effector signature. Unlike conventional memory cells, Tcf1-expressing T cells displayed hallmarks of an "exhausted" phenotype, including the expression of inhibitory receptors such as PD-1 and Lag-3. This population was crucial for the T cell expansion that occurred in response to inhibitory receptor blockade during chronic infection. These findings identify a memory-like T cell population that sustains T cell responses and is a prime target for therapeutic interventions to improve the immune response in chronic infections

Biochemical Systematics and Ecology. 2011;39(3):193-197.Besides the expected iridoid glucosides aucubin and catalpol as well as three known esters of the latter, Veronica beccabunga (brooklime) was shown to contain five carboxylated iridoid glucosides, namely gardoside, mussaenosidic acid, 8-epiloganic acid, arborescosidic acid and alpinoside. In addition to these compounds, the plant contained salidroside and a previously unknown caffeoyl phenylethanoid glycoside (CPG) which we have named chionoside J. The structure was elucidated mainly by 1D and 2D NMR spectroscopy to be 2 ''-(beta-glucopyranosyl)-plantamajoside. The distribution of plantamajoside and its derivatives as well as that of carbocyclic iridoids with an 8,9-double bond is briefly discussed, and it is noted that such compounds are mainly confined to the tribe Veroniceae of the Plantaginaceae.     Discutindo a educação ambiental no cotidiano escolar: desenvolvimento de projetos na escola formação inicial e continuada de professores

    A presente pesquisa buscou discutir como a Educação Ambiental (EA) vem sendo trabalhada, no Ensino Fundamental e como os docentes desta escola compreendem e vem inserindo a EA no cotidiano escolar., em uma escola estadual do município de Tangará da Serra/MT, Brasil. Para tanto, realizou-se entrevistas com os professores que fazem parte de um projeto interdisciplinar de EA na escola pesquisada. Verificou-se que o projeto da escola não vem conseguindo alcançar os objetivos propostos por: desconhecimento do mesmo, pelos professores; formação deficiente dos professores, não entendimento da EA como processo de ensino-aprendizagem, falta de recursos didáticos, planejamento inadequado das atividades. A partir dessa constatação, procurou-se debater a impossibilidade de tratar do tema fora do trabalho interdisciplinar, bem como, e principalmente, a importância de um estudo mais aprofundado de EA, vinculando teoria e prática, tanto na formação docente, como em projetos escolares, a fim de fugir do tradicional vínculo “EA e ecologia, lixo e horta”.Facultad de Humanidades y Ciencias de la Educació
