    Calcium signalling links MYC to NUAK1

    NUAK1 is a member of the AMPK-related family of kinases. Recent evidence suggests that NUAK1 is an important regulator of cell adhesion and migration, cellular and organismal metabolism, and regulation of TAU stability. As such, NUAK1 may play key roles in multiple diseases ranging from neurodegeneration to diabetes and metastatic cancer. Previous work revealed a crucial role for NUAK1 in supporting viability of tumour cells specifically when MYC is overexpressed. This role is surprising, given that NUAK1 is activated by the tumour suppressor LKB1. Here we show that, in tumour cells lacking LKB1, NUAK1 activity is maintained by an alternative pathway involving calcium-dependent activation of PKCĪ±. Calcium/PKCĪ±-dependent activation of NUAK1 supports engagement of the AMPK-TORC1 metabolic checkpoint, thereby protecting tumour cells from MYC-driven cell death, and indeed, MYC selects for this pathway in part via transcriptional regulation of PKCĪ± and ITPR. Our data point to a novel role for calcium in supporting tumour cell viability and clarify the synthetic lethal interaction between NUAK1 and MYC

    Prevalence of Presenting Conditions in Grey Seal Pups (Halichoerus grypus) Admitted for Rehabilitation

    A retrospective survey was performed on the presenting conditions of 205 live grey seal pups (Halichoerus grypus) admitted to the Cornish Seal Sanctuary in Gweek, United Kingdom between May 2005 and March 2011. The purpose of the survey was to examine the prevalence of various presenting signs at the sanctuary. The presenting signs were classified into nine non-mutually exclusive categories: ocular disorders, nasal disorders, oral disorders, respiratory disorders, orthopaedic disorders, puncture wounds, abrasions, netting injuries, and onychia. The sex ratio of seal pups in this study was 1.35 males per female. Of the 205 examined for rehabilitation, 22 (10.73%) did not survive to release. 68.78% of grey seal pups presented with puncture wounds, 47.80% with respiratory disorders, 46.34% with ocular disorders, 42.63% malnourished, 36.59% with abrasions, 25.37% with oral disorders, 23.90% with nasal disorders, 11.71% with orthopaedic disorders, 9.27% with onychia, and 3.41% presented with netting injuries. 52% were normothermic, 42% were hyperthermic, and 5% were hypothermic. Associations between gender, outcome of rehabilitation, hospitalisation time and presenting disorders were examined. In addition, admissions rates were found to display seasonality. The results of this study will aid in future preparation of grey seal rehabilitation facilities

    Adoption of Plantain within New Zealand Farming Systems

    In New Zealand, plantain has been incorporated into the pasture forage base in three ways: as a short-term pure crop; as a major component in ā€œherb mixesā€ that include clovers and chicory; and as a minor component in perennial grass-based swards. All types are typically grazed in-situ. Given the incentive to optimise the plantain proportion in livestock diet to mitigate nitrogen losses and the observed lack of persistence in grass-dominant pasture over time, there is a need to use various pasture types strategically across a farm system to ensure a long-term sustainable contribution of plantain to feed intake. The inclusion of plantain in the forage base should be relatively straightforward since farmers are already familiar with establishing and managing forage crops and mixed-species pastures. However, there are still technical questions to answer and incentives to implement before we can achieve the region-wide adoption necessary to result in a measurable impact on water quality. The Tararua District, on the east coast of the North Island of New Zealand, represents a useful case study of adoption in a strong dairy farming region. Some underpinning elements for successful adoption were in place: 1) national agronomic experience in using a plantain cultivar released in 1996; and 2) regulatory push in terms of reducing N leaching losses on farms. However, other initiatives were required to see meaningful progress: provision of data from local dairy farm systems, focused on the impact on forage supply/quality; a targeted extension program by a trusted sector leader agency (DairyNZ); individual farmer risk-taking and leadership in establishing and managing new crops and pastures. After three years, awareness is well embedded in the rural community and approximately 30% of dairy farms within the district have started to use plantain

    Periprocedural antithrombotic management for lumbar puncture: Association of British Neurologists clinical guideline

    Lumbar puncture (LP) is an important and frequently performed invasive procedure for the diagnosis and management of neurological conditions. There is little in the neurological literature on the topic of periprocedural management of antithrombotics in patients undergoing LP. Current practice is therefore largely extrapolated from guidelines produced by anaesthetic bodies on neuraxial anaesthesia, haematology groups advising on periprocedural management of antiplatelet agents and anticoagulants, and by neuroradiology on imaging-guided spinal procedures. This paper summarises the existing literature on the topic and offers recommendations to guide periprocedural antithrombotic management for LP, based on the consolidation of the best available evidence. ā€‹.     A paleomagnetic reconnaissance has been carried out on the Grupo DiabĆ”sico of the western Cordillera from the Colombian Andes, between El Tambo and El Penol, Departamento NariƱo. The Grupo DiabĆ”sico wich is of Cretaceous age is thought to be of submarine origin and to have undergone a low grade metamorphism.Ā  The geometric mean of the natural remanent magnetization JNRM = 8.8 x 10-5 G. The low field magnetic susceptibility geometric mean X= 8.87 x 10-4G/Oe gives a Koenigsberger ratio of 0.28 indicating that the dominant magnetization is a induced one.Some pilot samples have been progressively demagnetized by alternating fields up to 600 OĀ°. A viscous remanence test was applied to reject unstable sample.     Rheumatoid arthritis and incident fracture in women: a case-control study

    Background: To examine fracture incidence in women with rheumatoid arthritis (RA) for an entire geographical region of south-eastern Australia.Methods: Women aged 35 years and older, resident in the Barwon Statistical Division (BSD) and clinically diagnosed with RA 1994–2001 were eligible for inclusion as cases (n =1,008). The control population (n = 172,422) comprised the entire female BSD population aged 35 years and older, excluding those individuals identified as cases. Incident fractures were extracted from the prospective Geelong Osteoporosis Study Fracture Grid. We calculated rate ratios (RR) and 95% confidence intervals (CI) to compare the age- adjusted rate of fracture between the RA and non-RA populations, and used a chi-square test to compare proportions of fractures between women with and without RA, and a two-sided Mann–Whitney U-test to examine age-differences.Results: Among 1,008 women with RA, 19 (1.9%) sustained a fracture, compared to 1,981 fractures sustained by the 172,422 women without RA (1.2%). Fracture rates showed a trend for being greater among women diagnosed with RA (age-adjusted RR 1.43, 95%CI 0.98-2.09, p= 0.08). Women with RA sustained vertebral fractures at twice the expected frequency, whereas hip fractures were underrepresented in the RA population (p< 0.001). RA status was not associated with the likelihood of sustaining a fracture at sites adjacent to joints most commonly affected by RA (p= 0.22).Conclusion: Given that women with RA have a greater risk of fracture compared to women without RA, these patients may be a suitable target population for anti-resorptive agents; however, larger studies are warranted

    Nanoscale mobility mapping in semiconducting polymer films

    This work was supported by grant No 19-12-00066 of the Russian Science Foundation.Local electrical properties of thin films of the polymer PTB7 are studied by conductive atomic force microscopy (C-AFM). Non-uniform nanoscale current distribution in the neat PTB7 film is revealed and connected with the existence of ordered PTB7 crystallites. The shape of local I-V curves is explained by the presence of space charge limited current. We modify an existing semi-empirical model for estimation of the nanoscale hole mobility from our experimental C-AFM measurements. The procedure of nanoscale charge mobility estimation was described and applied to the PTB7 films. The calculated average C-AFM hole mobility is in good agreement with macroscopic values reported for this material. Mapping of nanoscale hole mobility was achieved using the described procedure. Local mobility values, influenced by nanoscale structure, vary more than two times in value and have a root-mean-square value 0.22 Ɨ 10āˆ’8 m2/(Vs), which is almost 20% from average hole mobility.PostprintPeer reviewe
