Putative Equine Neorickettsiosis in a Mare from Southern Brazil
Background: This report describes the occurrence of equine neorickettsiosis (EN) in the northern region of Paraná, southern Brazil. EN is a non-contagious infectious disease caused by the Gram-negative bacterium, Neorickettsia risticii. Equine neorickettsiosis was previously known as Potomac horse fever and monocytic ehrlichiosis. The disease occurs predominantly in the USA and Canada; data relative to EN in Brazil is scarce. The aim of this study was to report the first case of putative EN in the state of Paraná due to a combination of IHC and molecular testing.Case: A 2-year-old Quarter Horse was referred to a Veterinary Hospital with episodes of abdominal discomfort, fever, anorexia, tachycardia, and tachypnea. The animal reportedly demonstrated episodes of blackened and fetid diarrhea after the ingestion of hay. A treatment was established upon arrival at the veterinary hospital, but the mare died after 12 hours of monitoring. An autopsy examination performed soon after death revealed severe hyperemia of the mucosa of the cecum and colon, with multifocal cecal erosions and ulcerations. The principal histological lesion observed was necrotizing enterocolitis. Additional significant histopathologic lesions included widespread lymphoid depletion affecting the spleen, tonsils, and lymph nodes. An IHC assay designed to identify the antigens of N. helminthoeca (NH) in formalin fixed paraffin embedded (FFPE) tissues, identified antigens of intralesional neorickettsial organisms within macrophages of the mucosa of the colon. Additionally, a PCR assay designed to amplify the 16S rRNA gene of Neorickettsia, amplified the desired amplicon, but sequencing was frustrating.Discussion: A putative diagnosis of equine neorickettsiosis was established due to the combination of epidemiological evidence, pathologic findings, immunohistochemical identification of intralesional antigens of neorickettsial agents, and amplification of the 16S gene of Neorickettsia spp. by PCR. The gross and microscopic lesions described must be differentiated from lesions observed in equine salmonellosis and Clostridium difficile associated disease of horses. IHC was performed on FFPE intestinal tissue using an anti-NH hyperimmune serum as primary antibody due to high antigenic cross-reactivity between Neorickettsia spp. There was positive intracytoplasmic immunolabeling of macrophages in the mucosa of the large colon indicating the presence of neorickettsial antigens. The PCR amplified the desired amplicon from the colon fragment, but sequencing was frustrating due to poor quality product that resulted in an organism consistent with Methylobacterium spp., probably, a contaminant. EN show a seasonal and endemic tendency in USA, Uruguay and in southern Brazil, occurring in warmer months, as in the case reported. Evidence suggests that an ancient disease named “churrido” known for at least 100 years in southern Brazil and Uruguay is, in fact, equine neorickettsiosis and might have been introduced to North America from South America. Considering the epidemiological evidence and the worldwide distribution of NR, we suggest that EN is probably underdiagnosed in Brazil and that the etiology of equine enterocolitis should be more thoroughly investigated. Therefore, this report broadens the knowledge of EN in our region. In conclusion, considering the epidemiologic, pathologic, immunohistochemical and molecular evidence, and the specie-specificity of Neorickettsia spp., we report a putative case of EN in southern Brazil and the first report in Paraná state. IHC is a useful technique, less technically demanding and time consuming than bacterial culture and isolation, considered the gold standard; and can be used to diagnose EN when compatible pathologic evidence is present concomitantly
Effect of treatment of clinical seizures vs electrographic seizures in full-term and near-term neonates : a randomized clinical trial
Importance: Seizures in the neonatal period are associated with increased mortality and morbidity. Bedside amplitude-integrated electroencephalography (aEEG) has facilitated the detection of electrographic seizures; however, whether these seizures should be treated remains uncertain.
Objective: To determine if the active management of electrographic and clinical seizures in encephalopathic term or near-term neonates improves survival free of severe disability at 2 years of age compared with only treating clinically detected seizures.
Design, Setting, and Participants: This randomized clinical trial was conducted in tertiary newborn intensive care units recruited from 2012 to 2016 and followed up until 2 years of age. Participants included neonates with encephalopathy at 35 weeks’ gestation or more and younger than 48 hours old. Data analysis was completed in April 2021.
Interventions: Randomization was to an electrographic seizure group (ESG) in which seizures detected on aEEG were treated in addition to clinical seizures or a clinical seizure group (CSG) in which only seizures detected clinically were treated.
Main Outcomes and Measures: Primary outcome was death or severe disability at 2 years, defined as scores in any developmental domain more than 2 SD below the Australian mean assessed with Bayley Scales of Neonate and Toddler Development, 3rd ed (BSID-III), or the presence of cerebral palsy, blindness, or deafness. Secondary outcomes included magnetic resonance imaging brain injury score at 5 to 14 days, time to full suck feeds, and individual domain scores on BSID-III at 2 years.
Results: Of 212 randomized neonates, the mean (SD) gestational age was 39.2 (1.7) weeks and 122 (58%) were male; 152 (72%) had moderate to severe hypoxic-ischemic encephalopathy (HIE) and 147 (84%) had electrographic seizures. A total of 86 neonates were included in the ESG group and 86 were included in the CSG group. Ten of 86 (9%) neonates in the ESG and 4 of 86 (4%) in the CSG died before the 2-year assessment. The odds of the primary outcome were not significantly different in the ESG group compared with the CSG group (ESG, 38 of 86 [44%] vs CSG, 27 of 86 [31%]; odds ratio [OR], 1.83; 95% CI, 0.96 to 3.49; P = .14). There was also no significant difference in those with HIE (OR, 1.77; 95% CI, 0.84 to 3.73; P = .26). There was evidence that cognitive outcomes were worse in the ESG (mean [SD] scores, ESG: 97.4 [17.7] vs CSG: 103.8 [17.3]; mean difference, −6.5 [95% CI, −1.2 to −11.8]; P = .01). There was little evidence of a difference in secondary outcomes, including time to suck feeds, seizure burden, or brain injury score.
Conclusions and Relevance: Treating electrographic and clinical seizures with currently used anticonvulsants did not significantly reduce the rate of death or disability at 2 years in a heterogeneous group of neonates with seizures
The Long-Baseline Neutrino Experiment: Exploring Fundamental Symmetries of the Universe
The preponderance of matter over antimatter in the early Universe, the
dynamics of the supernova bursts that produced the heavy elements necessary for
life and whether protons eventually decay --- these mysteries at the forefront
of particle physics and astrophysics are key to understanding the early
evolution of our Universe, its current state and its eventual fate. The
Long-Baseline Neutrino Experiment (LBNE) represents an extensively developed
plan for a world-class experiment dedicated to addressing these questions. LBNE
is conceived around three central components: (1) a new, high-intensity
neutrino source generated from a megawatt-class proton accelerator at Fermi
National Accelerator Laboratory, (2) a near neutrino detector just downstream
of the source, and (3) a massive liquid argon time-projection chamber deployed
as a far detector deep underground at the Sanford Underground Research
Facility. This facility, located at the site of the former Homestake Mine in
Lead, South Dakota, is approximately 1,300 km from the neutrino source at
Fermilab -- a distance (baseline) that delivers optimal sensitivity to neutrino
charge-parity symmetry violation and mass ordering effects. This ambitious yet
cost-effective design incorporates scalability and flexibility and can
accommodate a variety of upgrades and contributions. With its exceptional
combination of experimental configuration, technical capabilities, and
potential for transformative discoveries, LBNE promises to be a vital facility
for the field of particle physics worldwide, providing physicists from around
the globe with opportunities to collaborate in a twenty to thirty year program
of exciting science. In this document we provide a comprehensive overview of
LBNE's scientific objectives, its place in the landscape of neutrino physics
worldwide, the technologies it will incorporate and the capabilities it will

Eclectic approach to anxiety disorders among rural children Abordagem ecletica a transtornos de ansiedade em criancas de zona rural
Introduction: Anxiety disorders in primary school-aged children negatively affect their mental health and psychological development. Available non-medical treatments for these conditions are time-consuming and expensive. In this context, eclectic therapy is a therapeutic approach that incorporates some therapeutic techniques and philosophies to create the ideal treatment. In this study, eclectic therapy consisted of art therapy and cognitive-behavioral therapy designed for children suffering from high level of anxiety in their middle childhood years. The therapy also included group guidance sessions for their mothers. The effectiveness of this intervention was examined in the study. Methods: 61 students aged 9-12 years with high levels of anxiety participated in the study. Intervention A (n = 20) consisted of 9-hour eclectic therapy for children with 3-hour group guidance sessions for their mothers. Intervention B (n = 20) consisted of 9-hour eclectic therapy for children. There was also a control group (n = 21). Results: Teacher ratings of children’s mental health difficulties and self-report ratings of anxiety disorders indicated a significant difference from pretest to posttest, revealing a large effect size between the two interventions. Higher levels of pretest scores significantly predicted higher posttest scores for all domains of anxiety and mental health difficulties. Furthermore, age, gender, mothers working a 15-hour day, mother’s educational level, parental divorce rates, parental death, and family monthly income predicted therapy outcomes. Conclusion: Results provide support for the effectiveness of eclectic art and CBT to improve children’s mental health and reduce anxiety through changing thoughts, beliefs, emotions, and behaviors that may cause fear and anxiety

The article reveals that between the mid-17th century and the mid-18th century, in the ecclesiastical world of the religious orders (Jesuits, franciscans, Carmelites, Cistercians, Augustinian, etc.) and the episcopate, there was the progressive adoption of the imperial symbol, the double-headed eagle, attribute of the Christian Empire, the germanic Holy Roman Empire emblem. However, in the religious field, this imperial eagle of the baroque time appears without the political insignia (sword, scepter and the imperial orb), adorning altars, monstrances, trumphal archs, facades of temples, doors, walls, domes, pulpits, sacred washbasins, sculptures and paintings of the Virgin and Child, liturgical robes, etc.; therefore related to the cult and the dogmas of the Catholic faith - in artistic works, the association between the double eagle and the flesh-spirit or human-divine unity, axial principle of the Catholic faith, represented by Virgin-Mother and Christ, is often indicated directly. The research has located and identified numerous remnants of the double eagle emblem in religious field in Portugal and Spain and in their conquests and dominions in America, Asia and Africa, and also in Italy, and the historiography on painful birth of the modern era in the West has never realized this phenomenon. A constrained extended kalman filter for state-of-charge estimation of a vanadium redox flow battery with crossover effects
One of the main issues with vanadium redox flow batteries is that vanadium ions travel across the membrane during operation which leads to a concentration imbalance and capacity loss after long-term cycling. Precise state-of-charge (SOC) monitoring allows the operator to effectively schedule electrolyte rebalancing and devise a control strategy to keep the battery running under optimal conditions. However, current SOC monitoring methods are too expensive and impractical to implement on commercial VRFB systems. Furthermore, physical models alone are neither reliable nor accurate enough to predict long-term capacity loss due to crossover. In this paper, we present an application of using an extended Kalman filter (EKF) to estimate the total vanadium concentration in each half-cell by combining three voltage measurements and a state prediction model without crossover effects. Simulation results show that the EKF can accurately predict capacity loss for different crossover patterns over a few hundred cycles. © 2014 by ASME

Development and Experimental Validation of a Physics-Based PEM Fuel Cell Model for Cathode Humidity Control Design
In large polymer electrolyte membrane (PEM) fuel cell stacks, monitoring and control of the local changes in membrane humidity inside the cathode channel is critical. In this study, a control-oriented dynamic model capable of describing the spatial distribution of voltage and relative humidity (RH) in a large fuel cell stack is developed and experimentally validated. The model tracks energy and mass flow inside the cathode, anode, and coolant channels, as well as the fuel cell stack body. Validation tests show that the model agrees well with the experimental data. The new modeling framework developed in this study can be used to predict the localized effects of humidity on the performance of a fuel cell stack. Also, given its accurate prediction of RH in the stack, this model can be used as an observer to predict local humidity variations that are, otherwise, not available. This capability would allow PEM fuel cells to avoid membrane damage due to low operating humidities as well as efficiency losses due to catalyst layer flooding
