    First Calorimetric Measurement of OI-line in the Electron Capture Spectrum of 163^{163}Ho

    The isotope 163^{163}Ho undergoes an electron capture process with a recommended value for the energy available to the decay, QECQ_{\rm EC}, of about 2.5 keV. According to the present knowledge, this is the lowest QECQ_{\rm EC} value for electron capture processes. Because of that, 163^{163}Ho is the best candidate to perform experiments to investigate the value of the electron neutrino mass based on the analysis of the calorimetrically measured spectrum. We present for the first time the calorimetric measurement of the atomic de-excitation of the 163^{163}Dy daughter atom upon the capture of an electron from the 5s shell in 163^{163}Ho, OI-line. The measured peak energy is 48 eV. This measurement was performed using low temperature metallic magnetic calorimeters with the 163^{163}Ho ion implanted in the absorber. We demonstrate that the calorimetric spectrum of 163^{163}Ho can be measured with high precision and that the parameters describing the spectrum can be learned from the analysis of the data. Finally, we discuss the implications of this result for the Electron Capture 163^{163}Ho experiment, ECHo, aiming to reach sub-eV sensitivity on the electron neutrino mass by a high precision and high statistics calorimetric measurement of the 163^{163}Ho spectrum.Comment: 5 pages, 3 figure

    A statutory minimum wage has been introduced in Germany, in the face of business opposition but abetted by union support. The political coalition in favour of minimum wage regulation brought together the centre-left and the centre-right with the argument that regulation is needed to prevent disfunctional interaction between low wages and the social security system. Thus the dualization which characterises Germany’s inegalitarian form of coordinated capitalism has provoked a corrective political response. The paper traces the long path to government intervention and assesses why employers were unable, or unwilling, to pre-empt intervention by maintaining the coverage of collective bargaining. It is argued that market liberalization has had a paradoxical effect on employer power: intense domestic as well as international competition has reduced employers’ capacity to act strategically to fend off regulation by the government

    Arsenic trioxide down-regulates antiapoptotic genes and induces cell death in mycosis fungoides tumors in a mouse model

    Background: Mycosis fungoides (MF) is the most frequent cutaneous T-cell lymphoma (CTCL). Arsenic trioxide (As2O3) has recently been shown to be effective against leukemias, so we studied whether As2O3 induces apoptosis of CTCL cells in vitro. We further investigated if As2O3 is effective in a MF mouse model. Material and methods: Annexin V/7-amino-actinomycin-D stainings were carried out to investigate if As2O3 induced apoptosis of CTCL cell lines. To study the underlying mechanisms, the effects of As2O3 on various transcription factors and apoptosis regulating proteins were analyzed by western blots, electrophoretic mobility shift assays and transcription factor enzyme-linked immunosorbent assays. The ability of As2O3 to induce tumor regression was investigated in a MF mouse model. Results: As2O3-induced apoptosis was paralleled by a reduction of the DNA-binding activities of transcription factors of the NFkB and signal transducer and activator of transcription gene families and reduced expression of the antiapoptotic proteins bcl-1, bcl-xL and mcl-1. Local injections of 200 μM As2O3 into tumors caused complete remissions in five of six mice and one partial remission. Conclusions: As2O3 induced apoptosis of CTCL cells by the down-regulation of transcription factors that stimulate the expression of antiapoptotic genes.     Characterization of the 163^{163}Ho Electron Capture Spectrum: A Step Towards the Electron Neutrino Mass Determination

    The isotope 163^{163}Ho is in many ways the best candidate to perform experiments to investigate the value of the electron neutrino mass. It undergoes an electron capture process to 163^{163}Dy with an energy available to the decay, QEC_{EC}, of about 2.8 keV. According to the present knowledge, this is the lowest QEC_{EC} value for such transitions. Here we discuss a newly obtained spectrum of 163^{163}Ho, taken by cryogenic metallic magnetic calorimeters with 163^{163}Ho implanted in the absorbers and operated in anticoincident mode for background reduction. For the first time, the atomic deexcitation of the 163^{163}Dy daughter atom following the capture of electrons from the 5s shell in 163^{163}Ho, the OI line, was observed with a calorimetric measurement. The peak energy is determined to be 48 eV. In addition, a precise determination of the energy available for the decay QEC_{EC}=(2.858±0.010stat_{stat}±0.05syst_{syst})  keV was obtained by analyzing the intensities of the lines in the spectrum. This value is in good agreement with the measurement of the mass difference between 163^{163}Ho and 163^{163}Dy obtained by Penning-trap mass spectrometry, demonstrating the reliability of the calorimetric technique

    Effects of oxidized lipids (4,5 (E)-epoxy-2(E)-heptenal and 4,5 (E)-epoxy-2 (E) -decenal) and lysine reaction products on zinc and calcium utilization: assays in Caco-2 cells

    The influence of the presence of brown products from the reaction between two oxidized lipids (4,5 (E)-epoxy-2(E)-heptenal, EH, and 4,5 (E)-epoxy-2 (E)-decenal, ED) and lysine (EH-L and ED-L) on zinc and calcium utilization was studied, and compared with a fructosyl-lysine mixture (F-L). Assays were carried out in Caco-2 cells grown in bicameral chambers. The Zn transported across the cell monolayer was significantly lower in the presence of the EH-L, ED-L and F-L samples, specially with EH-L. Significant decreases in Zn uptake were also observed, with no differences between samples. However, calcium transport was not modified. Thus, the assayed lipid-aminoacid brown products seem to have negative effects on Zn availability, whereas Ca availability appears to be unaffected.Se estudió la influencia de la presencia de productos obtenidos en la reacción de dos lípidos oxidados (4,5(E)-epoxy-2(E)- heptenal, EH, y 4,5(E)-epoxy-2(E)-decenal, ED) con el aminoácido lisina (EH-L y ED-L), sobre la absorción de zinc y calcio, comparándolos frente a una mezcla de fructosil-lisina (F-L). Los ensayos se realizaron con células Caco-2 sembradas en placas bicamerales. La adición de las muestras EH-L, ED-L y F-L al medio de cultivo supuso una reducción significativa en el Zn transportado a través de la monocapa de células, mucho más marcada ante la presencia de EH-L. También se redujo significativamente la captación celular de Zn, sin diferencias entre las distintas muestras ensayadas. Sin embargo, el transporte de Ca no se vio modificado.     Primary melanoma of the prostate: case report and review of the literature

    Background: Primary melanoma of the prostate has an extremely rare incidence. Only five cases have been reported in the literature and prognosis is poor. The most likely origin of prostatic melanoma is the transitional epithelium of the prostatic urethra. Surgical care for primary melanoma of mucosal sites is less well established than for primary cutaneous melanoma, but excision of the primary is recommended if the patient has no systemic disease. Case presentation: Here, we describe a case of primary malignant melanoma of the prostate. A 37-year-old male patient with history of both chemo- and radiation therapy for Hodgkin’s disease was admitted to the University Hospital Heidelberg on suspicion of pleomorphic sarcoma of the bladder. In-house diagnostic work-up revealed a malignant melanoma of the prostate. We then performed radical prostatectomy with extended lymphadenectomy. Despite presumably curative surgery, the patient suffered from early relapse of disease with pulmonary metastasis. Systemic chemotherapy and subsequent immuno-oncologic treatment was thereafter initiated. Conclusion: Since prostatic melanoma is a rare disease and a melanoma metastasis of unknown primary is the differential diagnosis, a multidisciplinary approach including early imaging to rule out possible metastases and to search for another potentially existing primary is advisable. To prevent complications related to local tumor progression and to receive tissue for mutational analysis, we recommend complete surgical resection to reduce the tumor mass. Novel immune and targeted oncologic therapies can lead to an improved survival in some cases and support of clinical trials is needed

    Jet formation and jet velocity fluctuations in a flue organ pipe

    Flow visualization of the initial transient in a small recorderlike flue organ pipe is presented and the various stages of the jet formation are related to measurements of the acoustic response of the pipe. An initial acoustic signal, due to the unsteady volume flow of the jet, appears before the forming jet reaches the labium. This signal can easily be modeled using a low-frequency approximation. The initial trajectory of the jet makes a curve towards the exterior of the pipe. Under certain conditions, the jet may even, at first, miss the labium. This effect is related to the steepness of the pressure rise in the foot of the pipe. The initial impact of the jet with the labium appears to be a crucial factor in the triggering of the transient. Moving the labium towards the exterior of the pipe, using a steep pressure rise or putting ears around the mouth increase the chance that the jet will hit the labium. This initial impact is followed by an impulsive vortex shedding at the labium and subsequently a high-frequency varicoselike oscillation is observed on the jet. This oscillation is also observed without labium. After about three periods of the fundamental mode of the pipe, turbulence appears therefore destroying these coherent structures. Whereas the time dependency of the jet velocity dominates the first stage of the starting transient, the jet velocity fluctuations during steady-state result in a non-negligible damping. This loss mechanism is, for the fundamental mode of our experimental organ pipe, of the same order of magnitude as the radiation or visco-thermal damping.</p

    Connecting Berry's phase and the pumped charge in a Cooper pair pump

    The properties of the tunnelling-charging Hamiltonian of a Cooper pair pump are well understood in the regime of weak and intermediate Josephson coupling, i.e. when EJ≲ECE_{\mathrm{J}}\lesssim E_{\mathrm{C}}. It is also known that Berry's phase is related to the pumped charge induced by the adiabatical variation of the eigenstates. We show explicitly that pumped charge in Cooper pair pump can be understood as a partial derivative of Berry's phase with respect to the phase difference ϕ\phi across the array. The phase fluctuations always present in real experiments can also be taken into account, although only approximately. Thus the measurement of the pumped current gives reliable, yet indirect, information on Berry's phase. As closing remarks, we give the differential relation between Berry's phase and the pumped charge, and state that the mathematical results are valid for any observable expressible as a partial derivative of the Hamiltonian.Comment: 5 pages, 5 figures, RevTeX, Presentation has been clarifie

    Role of 2-5A-Dependent RNase-L in Senescence and Longevity

    Senescence is a permanent growth arrest that restricts the lifespan of primary cells in culture, and represents an in vitro model for aging. Senescence functions as a tumor suppressor mechanism that can be induced independent of replicative crisis by diverse stress stimuli. RNase-L mediates antiproliferative activities and functions as a tumor suppressor in prostate cancer, therefore, we examined a role for RNase-L in cellular senescence and aging. Ectopic expression of RNase-L induced a senescent morphology, a decrease in DNA synthesis, an increase in senescence-associated -galactosidase activity, and accelerated replicative senescence. In contrast, senescence was retarded in RNase-L-null fibroblasts compared with wild-type fibroblasts. Activation of endogenous RNase-L by 2-5A transfection induced distinct senescent and apoptotic responses in parental and Simian virus 40-transformed WI38 fibroblasts, respectively, demonstrating cell type specific differences in the antiproliferative response to RNase-L activation. Replicative senescence is a model for in vivo aging; therefore, genetic disruption of senescence effectors may impact lifespan. RNase-L-/- mice survived 31.7% (P\u3c0.0001) longer than strain-matched RNase-L+/+ mice providing evidence for a physiological role for RNase-L in aging. These findings identify a novel role for RNase-L in senescence that may contribute to its tumor suppressive function and to the enhanced longevity of RNase-L-/- mice
