    The Effects of Grace on Self-Forgiveness with a Religious Community

    Genuine self-forgiveness entails accepting responsibility for wrongdoing while experiencing a continued sense of self-worth (Enright & Human Development Study Group, 1996; Fisher & Exline, 2006; Hall & Fincham, 2005; Martin, 2008; Szablowinski, 2012; Vitz & Meade, 2011; Wenzel, Woodyatt, & Hedrick, 2012; Woodyatt & Wenzel, 2013a; Woodyatt & Wenzel, 2013b). Previous research has demonstrated that a benevolent concept of God and a personal sense of God’s forgiveness facilitate self-forgiveness (Exline, Yali, & Lobel,1999; Hall & Fincham, 2008; Martin, 2008; McConnell & Dixon, 2012), suggesting that those who accept responsibility for the offense and believe God can forgive that offense will not become stuck in self-condemnation. The theological concept of grace is closely related to self-forgiveness; people must acknowledge that they have sinned while accepting God’s unmerited favor (McMinn, Ruiz, Marx, Wright, & Gilbert, 2006; Sells, Bechenbach, & Patrick, 2009). This study examined the effects of a grace intervention on self-forgiveness within two Friends (Quaker) churches. The grace intervention was developed in collaboration with church leaders and psychological researchers and included a 9-week sermon series, group Bible studies, and weekly grace practices. All church attendees were asked to complete a trait self-forgiveness scale, while a smaller portion of each church completed a more extensive battery of questionnaires, which were completed before and after each church experienced the grace intervention they developed. The study utilized a quasi-experimental crossover design for statistical analyses. Both congregations were assessed again at the conclusion of the second congregation’s grace intervention. Significant changes over time and an interaction effect were found in trait self-forgiveness, intrinsic religiosity, and daily spiritual experiences. Changes over time without interaction effects were found with spiritual wellbeing, grace to self, selfforgiveness feelings and actions, and self-forgiveness beliefs. Group differences were found with daily spiritual experiences, authoritarian God concept, grace to others, and genuine selfforgiveness. This study suggests that an intervention focused on the theological concept of grace may increase people’s ability to forgive themselves for offenses they have committed against other people. Future research should look at the implications this could have for those experiencing psychological distress

    Phenomenological Experiences of Masters Students with University-Based Supervisors using a Cognitive Development Process

    The researchers qualitatively investigated the individual supervision experiences of nine masters’ students who were supervised by university-based supervisors, who used a cognitive developmental supervision process. The participants indicated that supervisors using Deliberate Psychological Education (a) provided a supportive environment, (b) cultivated counselor identity development, (c) encouraged personal growth, and (d) offered a space for concurrent development.     The Effects of Grace Interventions in Church Communities

    Grace remains little studied though several recent studies have developed promising ways to measure grace. Here we report two studies involving grace interventions as part of an ongoing investigation of positive psychology in the context of Christian church communities, Study One used a crossover design in which two congregations were measured on grace and other variables at the outset, and again after phases one and two. One congregation received a grace intervention during phase one and the second during phase two. Results showed increased scores on grace but not an expected increase in marital satisfaction among married participants. Study Two used a similar design with two additional congregations to assess effects of a grace intervention on selfforgiveness. As expected, congregants receiving the grace intervention showed increases in trait self-forgiveness when compared to those in the wait-list

    Explaining asymmetry between weakening and recovery of the AMOC in a coupled climate model

    The Atlantic meridional overturning circulation (AMOC) is projected to weaken in the coming century due to anthropogenic climate change. Various studies have considered AMOC weakening and collapse, with less research focusing on the processes and timescales of the recovery phase. This study uses a coupled climate model to explore the roles of salinity and temperature in AMOC recovery after a weakening. The North Atlantic and Arctic region was hosed with freshwater for 200 years. The mean Atlantic salinity increased strongly during recovery, and remained elevated for ~ 600 years post hosing. The behaviour of the AMOC was well reconstructed by applying “rotated geostrophy” to meridional density gradient profiles between 50°N and 30°S. This makes it possible to determine the role of overturning, gyre, and surface fluxes in the North and South Atlantic. Changes at 50°N dominate the weakening and early recovery. The magnitude of the overshoot to high AMOC transports in the recovery phase was related to density changes in the South Atlantic

    N-Acetylglutamate Synthase Deficiency Due to a Recurrent Sequence Variant in the N-acetylglutamate Synthase Enhancer Region

    N-acetylglutamate synthase deficiency (NAGSD, MIM #237310) is an autosomal recessive disorder of the urea cycle that results from absent or decreased production of N-acetylglutamate (NAG) due to either decreased NAGS gene expression or defective NAGS enzyme. NAG is essential for the activity of carbamylphosphate synthetase 1 (CPS1), the first and rate-limiting enzyme of the urea cycle. NAGSD is the only urea cycle disorder that can be treated with a single drug, N-carbamylglutamate (NCG), which can activate CPS1 and completely restore ureagenesis in patients with NAGSD. We describe a novel sequence variant NM_153006.2:c.-3026C > T in the NAGS enhancer that was found in three patients from two families with NAGSD; two patients had hyperammonemia that resolved upon treatment with NCG, while the third patient increased dietary protein intake after initiation of NCG therapy. Two patients were homozygous for the variant while the third patient had the c.-3026C > T variant and a partial uniparental disomy that encompassed the NAGS gene on chromosome 17. The c.-3026C > T sequence variant affects a base pair that is highly conserved in vertebrates; the variant is predicted to be deleterious by several bioinformatics tools. Functional assays in cultured HepG2 cells demonstrated that the c.-3026C > T substitution could result in reduced expression of the NAGS gene. These findings underscore the importance of analyzing NAGS gene regulatory regions when looking for molecular causes of NAGSD

    Prenatal diagnosis of congenital adrenal hyperplasia caused by P450 oxidoreductase deficiency

    CONTEXT: Mutations in the electron donor enzyme P450 oxidoreductase (POR) result in congenital adrenal hyperplasia with apparent combined 17α-hydroxylase/17,20 lyase and 21-hydroxylase deficiencies, also termed P450 oxidoreductase deficiency (PORD). Major clinical features present in PORD are disordered sex development in affected individuals of both sexes, glucocorticoid deficiency, and multiple skeletal malformations. OBJECTIVE: The objective of the study was to establish a noninvasive approach to prenatal diagnosis of PORD including assessment of malformation severity to facilitate optimized prenatal diagnosis and timely treatment. DESIGN: We analyzed 20 pregnancies with children homozygous or compound heterozygous for disease-causing POR mutations and 1 pregnancy with a child carrying a heterozygous POR mutation by recording clinical and biochemical presentations and fetal ultrasound findings. In 4 of the pregnancies (3 homozygous and 1 heterozygous for disease-causing POR mutations), prenatal analysis of steroid metabolite excretion in maternal urine was carried out by gas chromatography/mass spectrometry during gestational weeks 11–23. RESULTS: Pregnancy complications in our cohort included maternal virilization (6 of 20) with onset in the second trimester. Seven pregnant women presented with low unconjugated estriol at prenatal screening (triple or quadruple antenatal screening test). Overt dysmorphic features were noted in 19 of the 20 babies at birth but observed in only 5 by prenatal ultrasound. These 5 had the most severe malformation phenotypes and poor outcome, whereas the other babies showed normal development. Steroid profiling of maternal urine revealed significantly increased steroids of fetal origin, namely the pregnenolone metabolite epiallopregnanediol and the androgen metabolite androsterone, with concomitant low values for estriol. Diagnostic steroid ratios conclusively indicated PORD as early as gestational week 12. In the heterozygous pregnancy, steroid ratios were only slightly elevated and estriol excretion was normal. CONCLUSION: Prenatal diagnosis in PORD is readily established via urinary steroid metabolite analysis of maternal urine. Visible malformations at prenatal ultrasound predict a severe malformation phenotype

    Abstinence-only-until-marriage : An Updated review of U.S. policies and programs and their impact

    Adolescence is marked by the emergence of human sexuality, sexual identity and the initiation of intimate relations; within this context, abstinence from sexual intercourse can be a healthy choice. However, programs that promote abstinence-only-until-marriage (AOUM) or sexual risk avoidance (SRA), are scientifically and ethically problematic and—as such—have been widely rejected by medical and public health professionals. Although abstinence is theoretically effective, in actual practice, intentions to abstain from sexual activity often fail. Given a rising age at first marriage around the world, a rapidly declining percentage of young people remain abstinent until marriage. Promotion of AOUM policies by the United States (U.S.) government has undermined sexuality education in the U.S. and in U.S. foreign aid programs; funding for AOUM continues in the U.S. The weight of scientific evidence finds that AOUM programs are not effective in delaying initiation of sexual intercourse or changing other sexual risk behaviors. AOUM programs, as defined by U.S. federal funding requirements, inherently withhold information about human sexuality and may provide medically inaccurate and stigmatizing information. Thus, AOUM programs threaten fundamental human rights to health, information, and life. Young people need access to accurate and comprehensive sexual health information to protect their health and lives

    A cross-sectional national investigation of COVID-19 outbreaks in nurseries during rapid spread of the Alpha (B.1.1.7) variant of SARS-CoV-2 in England

    Background: In England, the emergence the more transmissible SARS-CoV-2 variant Alpha (B.1.1.7) led to a third national lockdown from December 2020, including restricted attendance at schools. Nurseries, however, remained fully open. COVID-19 outbreaks (≥ 2 laboratory-confirmed cases within 14 days) in nurseries were investigated to assess the risk of SARS-CoV-2 infection and cumulative incidence in staff and children over a three-month period when community SARS-CoV-2 infections rates were high and the Alpha variant was spreading rapidly across England. Methods: This was a cross-sectional national investigation of COVID-19 outbreaks in nurseries across England. Nurseries reporting a COVID-19 outbreak to PHE between November 2020 and January 2021 were requested to complete a questionnaire about their outbreak. Results: Three hundred and twenty-four nurseries, comprising 1% (324/32,852) of nurseries in England, reported a COVID-19 outbreak. Of the 315 (97%) nurseries contacted, 173 (55%) reported 1,657 SARS-CoV-2 cases, including 510 (31%) children and 1,147 (69%) staff. A child was the index case in 45 outbreaks (26%) and staff in 125 (72%) outbreaks. Overall, children had an incidence rate of 3.50% (95%CI, 3.21–3.81%) and was similar irrespective of whether the index case was a child (3.55%; 95%CI, 3.01–4.19%) or staff (3.44%; 95%CI, 3.10–3.82%). Among staff, cumulative incidence was lower if the index case was a child (26.28%; 95%CI, 23.54–29.21%%) compared to a staff member (32.98%; 95%CI, 31.19–34.82%), with the highest cumulative incidence when the index case was also a staff member (37.52%; 95%CI, 35.39–39.70%). Compared to November 2020, outbreak sizes and cumulative incidence was higher in January 2021, when the Alpha variant predominated. Nationally, SARS-CoV-2 infection rates in < 5 year-olds remained low and followed trends in older age-groups, increasing during December 2020 and declining thereafter. Conclusions: In this cross-sectional study of COVID-19 outbreaks in nurseries, one in three staff were affected compared to one in thirty children. There was some evidence of increased transmissibility and higher cumulative incidence associated with the Alpha variant, highlighting the importance of maintaining a low level of community infections

    Nucleolar Proteins Suppress Caenorhabditis elegans Innate Immunity by Inhibiting p53/CEP-1

    The tumor suppressor p53 has been implicated in multiple functions that play key roles in health and disease, including ribosome biogenesis, control of aging, and cell cycle regulation. A genetic screen for negative regulators of innate immunity in Caenorhabditis elegans led to the identification of a mutation in NOL-6, a nucleolar RNA-associated protein (NRAP), which is involved in ribosome biogenesis and conserved across eukaryotic organisms. Mutation or silencing of NOL-6 and other nucleolar proteins results in an enhanced resistance to bacterial infections. A full-genome microarray analysis on animals with altered immune function due to mutation in nol-6 shows increased transcriptional levels of genes regulated by a p53 homologue, CEP-1. Further studies indicate that the activation of innate immunity by inhibition of nucleolar proteins requires p53/CEP-1 and its transcriptional target SYM-1. Since nucleoli and p53/CEP-1 are conserved, our results reveal an ancient immune mechanism by which the nucleolus may regulate immune responses against bacterial pathogens
