    Intergenerational change and familial aggregation of body mass index

    The relationship between parental BMI and that of their adult offspring, when increased adiposity can become a clinical issue, is unknown. We investigated the intergenerational change in body mass index (BMI) distribution, and examined the sex-specific relationship between parental and adult offspring BMI. Intergenerational change in the distribution of adjusted BMI in 1,443 complete families (both parents and at least one offspring) with 2,286 offspring (1,263 daughters and 1,023 sons) from the west of Scotland, UK, was investigated using quantile regression. Familial correlations were estimated from linear mixed effects regression models. The distribution of BMI showed little intergenerational change in the normal range (\25 kg/m2), decreasing overweightness (25– \30 kg/m2) and increasing obesity (C30 kg/m2). Median BMI was static across generations in males and decreased in females by 0.4 (95% CI: 0.0, 0.7) kg/m2; the 95th percentileincreased by 2.2 (1.1, 3.2) kg/m2 in males and 2.7 (1.4, 3.9) kg/m2 in females. Mothers’ BMI was more strongly associated with daughters’ BMI than was fathers’ (correlation coefficient (95% CI): mothers 0.31 (0.27, 0.36), fathers 0.19 (0.14, 0.25); P = 0.001). Mothers’ and fathers’ BMI were equally correlated with sons’ BMI (correlation coefficient: mothers 0.28 (0.22, 0.33), fathers 0.27 (0.22, 0.33). The increase in BMI between generations was concentrated at the upper end of the distribution. This, alongside the strong parent-offspring correlation, suggests that the increase in BMI is disproportionally greater among offspring of heavier parents.     Refactoring the Whitby Intelligent Tutoring System for Clean Architecture

    Whitby is the server-side of an Intelligent Tutoring System application for learning System-Theoretic Process Analysis (STPA), a methodology used to ensure the safety of anything that can be represented with a systems model. The underlying logic driving the reasoning behind Whitby is Situation Calculus, which is a many-sorted logic with situation, action, and object sorts. The Situation Calculus is applied to Ontology Authoring and Contingent Scaffolding: the primary activities within Whitby. Thus many fluents and actions are aggregated in Whitby from these two sub-applications and from Whitby itself, but all are available through a common situation query interface that does not depend upon any of the fluents or actions. Each STPA project in Whitby is a single situation term, which is queried for fluents that include the ontology, and to determine what pedagogical interventions to offer. Initially Whitby was written in Prolog using a module system. In the interest of a cleaner architecture and implementation with improved code reuse and extensibility, the initial application was refactored into Logtalk. This refactoring includes decoupling the Situation Calculus reasoner, Ontology Authoring framework, and Contingent Scaffolding framework into third-party libraries that can be reused in other applications. This extraction was achieved by inverting dependencies via Logtalk protocols and categories, which are reusable interfaces and components that provide functionally cohesive sets of predicate declarations and predicate definitions.     Cholesterol and the risk of grade-specific prostate cancer incidence: evidence from two large prospective cohort studies with up to 37 years' follow up

    <b>Background</b> High cholesterol may be a modifiable risk factor for prostate cancer but results have been inconsistent and subject to potential "reverse causality" where undetected disease modifies cholesterol prior to diagnosis.<p></p> <b>Methods</b> We conducted a prospective cohort study of 12,926 men who were enrolled in the Midspan studies between 1970 and 1976 and followed up to 31st December 2007. We used Cox-Proportional Hazards Models to evaluate the association between baseline plasma cholesterol and Gleason grade-specific prostate cancer incidence. We excluded cancers detected within at least 5 years of cholesterol assay.<p></p> <b>Results</b> 650 men developed prostate cancer in up to 37 years' follow-up. Baseline plasma cholesterol was positively associated with hazard of high grade (Gleason score[greater than or equal to]8) prostate cancer incidence (n=119). The association was greatest among men in the 4th highest quintile for cholesterol, 6.1 to <6.69 mmol/l, Hazard Ratio 2.28, 95% CI 1.27 to 4.10, compared with the baseline of <5.05 mmol/l. This association remained significant after adjustment for body mass index, smoking and socioeconomic status.<p></p> <b>Conclusions</b> Men with higher cholesterol are at greater risk of developing high-grade prostate cancer but not overall risk of prostate cancer. Interventions to minimise metabolic risk factors may have a role in reducing incidence of aggressive prostate cancer

    Exclusion of known gene for enamel development in two Brazilian families with amelogenesis imperfecta

    Amelogenesis imperfecta (AI) is a genetically heterogeneous group of diseases that result in defective development of tooth enamel. Mutations in several enamel proteins and proteinases have been associated with AI. The object of this study was to evaluate evidence of etiology for the six major candidate gene loci in two Brazilian families with AI. Genomic DNA was obtained from family members and all exons and exon-intron boundaries of the ENAM, AMBN, AMELX, MMP20, KLK4 and Amelotin gene were amplified and sequenced. Each family was also evaluated for linkage to chromosome regions known to contain genes important in enamel development. The present study indicates that the AI in these two families is not caused by any of the known loci for AI or any of the major candidate genes proposed in the literature. These findings indicate extensive genetic heterogeneity for non-syndromic AI

    Thermal effects on electron-phonon interaction in silicon nanostructures

    Raman spectra from silicon nanostructures, recorded using excitation laser power density of 1.0 kW/cm^2, is employed here to reveal the dominance of thermal effects at temperatures higher than the room temperature. Room temperature Raman spectrum shows only phonon confinement and Fano effects. Raman spectra recorded at higher temperatures show increase in FWHM and decrease in asymmetry ratio with respect to its room temperature counterpart. Experimental Raman scattering data are analyzed successfully using theoretical Raman line-shape generated by incorporating the temperature dependence of phonon dispersion relation. Experimental and theoretical temperature dependent Raman spectra are in good agreement.     Observation of the Peach Fruit Moth, Carposina sasakii, Larvae in Young Apple Fruit by Dedicated Micro-Magnetic Resonance Imaging

    Infestation of young apple fruits by the larvae of the peach fruit moth, Carposina sasakii Matsumura (Lepidoptera: Carposinidae), was studied by a small dedicated micro-magnetic resonance imaging (MRI) apparatus using the three-dimensional (3D) gradient-echo method and the two-dimensional (2D) and 3D spin-echo methods. Changes from a young larva at 1.8 mm in length to a mature one ready to leave the fruit were observed in relation to the progression of infestation of the fruit tissues. The trace of larva intrusion was demonstrated by a series of sliced images in the 3D image data of an infested fruit, where it entered from outside the calyx, and migrated to near the vasculature around the carpel through the core. The small, dedicated MRI device was proven useful for ecological studies of the growth and movement of insect larvae in their food fruits. It can also be applied to detect the infestation of small fruits by insect larvae
