    Evaluation of Inter-Mountain Labs infrasound sensors : July 2007.

    Sandia National Laboratories has tested and evaluated three Inter Mountain Labs infrasound sensors. The test results included in this report were in response to static and tonal-dynamic input signals. Most test methodologies used were based on IEEE Standards 1057 for Digitizing Waveform Recorders and 1241 for Analog to Digital Converters; others were designed by Sandia specifically for infrasound application evaluation and for supplementary criteria not addressed in the IEEE standards. The objective of this work was to evaluate the overall technical performance of the Inter Mountain Labs (IML) infrasound sensor model SS. The results of this evaluation were only compared to relevant noise models; due to a lack of manufactures documentation notes on the sensors under test prior to testing. The tests selected for this system were chosen to demonstrate different performance aspects of the components under test

    The Hartlepool Networked Learning Community and the Bransholme NLC are resourced by the National College for School Leadership. The paper describes how two of the leaders of these networks approached their tasks and how they came to terms with a challenging role that added to their own leadership learning. Emphasis is given to the search for locating network practices that provide high leverage for improvement, the design of organizational structures that support this learning, the alignment of the learning to the pressing challenges and compelling work that networked schools face and the problems of evaluating the successes of networked learning

    Seismic data acquisition at the FACT site for the CASPAR project.

    Since May 2010, we have been recording continuous seismic data at Sandia's FACT site. The collected signals provide us with a realistic archive for testing algorithms under development for local monitoring of explosive testing. Numerous small explosive tests are routinely conducted around Kirtland AFB by different organizations. Our goal is to identify effective methods for distinguishing these events from normal daily activity on and near the base, such as vehicles, aircraft, and storms. In this report, we describe the recording system, and present some observations of the varying ambient noise conditions at FACT. We present examples of various common, non-explosive, sources. Next we show signals from several small explosions, and discuss their characteristic features

    CoESPRIT: A Library-Based Construct Screening Method for Identification and Expression of Soluble Protein Complexes

    Structural and biophysical studies of protein complexes require multi-milligram quantities of soluble material. Subunits are often unstable when expressed separately so co-expression strategies are commonly employed since in vivo complex formation can provide stabilising effects. Defining constructs for subunit co-expression experiments is difficult if the proteins are poorly understood. Even more problematic is when subunit polypeptide chains co-fold since individually they do not have predictable domains. We have developed CoESPRIT, a modified version of the ESPRIT random library construct screen used previously on single proteins, to express soluble protein complexes. A random library of target constructs is screened against a fixed bait protein to identify stable complexes. In a proof-of-principle study, C-terminal fragments of the influenza polymerase PB2 subunit containing folded domains were isolated using importin alpha as bait. Separately, a C-terminal fragment of the PB1 subunit was used as bait to trap N-terminal fragments of PB2 resulting in co-folded complexes. Subsequent expression of the target protein without the bait indicates whether the target is independently stable, or co-folds with its partner. This highly automated method provides an efficient strategy for obtaining recombinant protein complexes at yields compatible with structural, biophysical and functional studies

    The Ground-Based Monitoring R&E Component Evaluation project performs testing on the hardware components that make up Seismic and Infrasound monitoring systems. The majority of the testing is focused on the Digital Waveform Recorder (DWR), Seismic Sensor, and Infrasound Sensor. In order to guarantee consistency, traceability, and visibility into the results of the testing process, it is necessary to document the test and analysis procedures that are in place. Other reports document the testing procedures that are in place (Kromer, 2007). This document serves to provide a comprehensive overview of the analysis and the algorithms that are applied to the Component Evaluation testing. A brief summary of each test is included to provide the context for the analysis that is to be performed

    Host Determinant Residue Lysine 627 Lies on the Surface of a Discrete, Folded Domain of Influenza Virus Polymerase PB2 Subunit

    Understanding how avian influenza viruses adapt to human hosts is critical for the monitoring and prevention of future pandemics. Host specificity is determined by multiple sites in different viral proteins, and mutation of only a limited number of these sites can lead to inter-species transmission. Several of these sites have been identified in the viral polymerase, the best characterised being position 627 in the PB2 subunit. Efficient viral replication at the relatively low temperature of the human respiratory tract requires lysine 627 rather than the glutamic acid variant found systematically in avian viruses. However, the molecular mechanism by which any of these host specific sites determine host range are unknown, although adaptation to host factors is frequently evoked. We used ESPRIT, a library screening method, to identify a new PB2 domain that contains a high density of putative host specific sites, including residue 627. The X-ray structure of this domain (denoted the 627-domain) exhibits a novel fold with the side-chain of Lys627 solvent exposed. The structure of the K627E mutated domain shows no structural differences but the charge reversal disrupts a striking basic patch on the domain surface. Five other recently proposed host determining sites of PB2 are also located on the 627-domain surface. The structure of the complete C-terminal region of PB2 comprising the 627-domain and the previously identified NLS-domain, which binds the host nuclear import factor importin alpha, was also determined. The two domains are found to pack together with a largely hydrophilic interface. These data enable a three-dimensional mapping of approximately half of PB2 sites implicated in cross-species transfer onto a single structural unit. Their surface location is consistent with roles in interactions with other viral proteins or host factors. The identification and structural characterization of these well-defined PB2 domains will help design experiments to elucidate the effects of mutations on polymerase–host factor interactions

    Strengthening the role of universities in addressing sustainability challenges: the Mitchell Center for Sustainability Solutions as an institutional experiment

    As the magnitude, complexity, and urgency of many sustainability problems increase, there is a growing need for universities to contribute more effectively to problem solving. Drawing upon prior research on social-ecological systems, knowledge-action connections, and organizational innovation, we developed an integrated conceptual framework for strengthening the capacity of universities to help society understand and respond to a wide range of sustainability challenges. Based on experiences gained in creating the Senator George J. Mitchell Center for Sustainability Solutions (Mitchell Center), we tested this framework by evaluating the experiences of interdisciplinary research teams involved in place-based, solutions-oriented research projects at the scale of a single region (i.e., the state of Maine, USA). We employed a multiple-case-study approach examining the experiences of three interdisciplinary research teams working on tidal energy development, adaptation to climate change, and forest vulnerability to an invasive insect. Drawing upon documents, observations, interviews, and other data sources, three common patterns emerged across these cases that were associated with more effective problem-solving strategies. First, an emphasis on local places and short-term dynamics in social-ecological systems research provides more frequent opportunities for learning while doing. Second, iterative stakeholder engagement and inclusive forms of knowledge co-production can generate substantial returns on investment, especially when researchers are dedicated to a shared process of problem identification and they avoid framing solutions too narrowly. Although these practices are time consuming, they can be accelerated by leveraging existing stakeholder relationships. Third, efforts to mobilize interdisciplinary expertise and link knowledge with action are facilitated by an organizational culture that emphasizes mutual respect, adaptability, and solutions. Participation of faculty associated with interdisciplinary academic programs, solutions-oriented fields, and units with partnership-oriented missions hastens collaboration within teams and between teams and stakeholders. The Mitchell Center also created a risk-tolerant culture that encouraged organizational learning. Solutions-focused programs at other universities can potentially benefit from the lessons we learned
