    Generalised framework of limit equilibrium methods for slope stability analysis

    A generalised framework is proposed in this paper incorporating almost all of the existing limit equilibrium methods of slices for slope stability analysis with general slip surfaces. The force and moment equilibrium equations are derived in terms of the factor of safety and the initially assumed normal stress distribution over the slip surface, multiplied by a modification function involving two auxiliary unknowns. These equations are then analytically solved to yield explicit expressions for the factor of safety. Various assumptions regarding the interslice forces can be transformed into a unified form of expression for the normal stress distribution along the slip surface. An iterative procedure is developed to expedite the convergence of the solution for the factor of safety. Experience to date indicates that the process generally converges within a few iterations. Computation schemes are suggested to avoid numerical difficulty, especially in computing the factor of safety associated with the rigorous Janbu method. The present framework can be readily implemented in a computer program, giving solutions of slope stability associated with a number of conventional methods of slices.published_or_final_versio

    Staged and non-staged anaerobic filters : microbial activity segregation, hydrodynamic behaviour and performance

    This work describes a comparative study of staged and non-staged anaerobic flters for treating a synthetic dairy waste under similar operating conditions. The effect of increasing the substrate concentration from 3 to 12 g COD dmĖ‰Ā³ at a constant hydraulic residence time (HRT) of 2 days was evaluated with respect to overall reactor performance, biogas production, volatile fatty acids profiles along the height, methanogenic and acidogenic activity distribution, and hydrodynamic behaviour. The potential maximum specific methanogenic activity against acetate, hydrogen, propionate and butyrate and the lactose specific activity were determined for sludge sampled from three different points in each reactor, under two operating conditions (influent COD of 3 and 9 g COD dmĖ‰Ā³). Although all trophic groups involved in the anaerobic process were found throughout the reactors, it was possible to identify different specific sludges at different heights in both reactors.     The interplay of DNA and lipid biomarkers in the detection of tuberculosis and leprosy in mummies and other skeletal remains

    The detection of ancient DNA and lipid biomarkers has become established for the detection of tuberculosis and leprosy in archaeological material, including mummies. Recording the characteristic profiles of long-chain mycolic acids by fluorescence high-performance liquid chromatography (HPLC) has been developed to provide a highly specific method for the detection of ancient tuberculosis and leprosy. Initially, mycolate methylanthryl esters were analysed but these derivatives have been superseded by pyrenebutyric acid derivatives of pentafluorobenzyl esters. Long-chain compounds are released by an efficient non-aqueous alkaline extraction and acidic components are converted to stable pentafluorobenzyl esters, which can be preserved for immediate or future analysis. These long-chain components are fractionated into non-hydroxylated esters, mycolates and characteristic phthiocerols. The mycolate pyrenebutyrates are analyzed by fluorescence HPLC to produce profiles characteristic of mycobacterial disease. It is shown in this study that residual material from DNA determinations, on mainly Turkish and Hungarian skeletons, can be used for the detection of mycolic acid biomarkers for tuberculosis and leprosy. The correlation between DNA and mycolic acids biomarker results was not precise, confirming the importance of using complementary methods. In one particular Turkish skeleton, with poor DNA preservation, mycolic acid analysis supported pathological changes indicative of leprosy

    Late widespread skeletal metastases from myxoid liposarcoma detected by MRI only

    Background Myxoid liposarcoma is the second most commonly occurring sub-type of liposarcomas. In contrast to other soft tissue sarcomas, it is known to have a tendency to spread toward extrapulmonary sites, such as soft tissues, retroperitoneum, and the peritoneal surface. Bony spread, however, is not as common. Case presentation We report an unusual case of diffuse skeletal metastases from myxoid liposarcoma occurring 13 years after treatment of the primary tumour in the left lower limb. The skeletal spread of the disease was demonstrated on MRI only after other imaging modalities (plain radiography, CT and TC99 bone scans) had failed to detect these metastases. Conclusion MRI is an extremely sensitive and specific screening tool in the detection of skeletal involvement in these types of sarcomas, and therefore, should be a part of the staging proces

    Selection of chromosomal DNA libraries using a multiplex CRISPR system.

    The directed evolution of biomolecules to improve or change their activity is central to many engineering and synthetic biology efforts. However, selecting improved variants from gene libraries in living cells requires plasmid expression systems that suffer from variable copy number effects, or the use of complex marker-dependent chromosomal integration strategies. We developed quantitative gene assembly and DNA library insertion into the Saccharomyces cerevisiae genome by optimizing an efficient single-step and marker-free genome editing system using CRISPR-Cas9. With this Multiplex CRISPR (CRISPRm) system, we selected an improved cellobiose utilization pathway in diploid yeast in a single round of mutagenesis and selection, which increased cellobiose fermentation rates by over 10-fold. Mutations recovered in the best cellodextrin transporters reveal synergy between substrate binding and transporter dynamics, and demonstrate the power of CRISPRm to accelerate selection experiments and discoveries of the molecular determinants that enhance biomolecule function

    On the force field optimisation of Ī² -lactam cores using the force field Toolkit

    When employing molecular dynamics (MD) simulations for computer-aided drug design, the quality of the used force fields is highly important. Here we present reparametrisations of the force fields for the core molecules from 9 different Ī²-lactam classes, for which we utilized the force field Toolkit and Gaussian calculations. We focus on the parametrisation of the dihedral angles, with the goal of reproducing the optimised quantum geometry in MD simulations. Parameters taken from CGenFF turn out to be a good initial guess for the multiplicity of each dihedral angle, but the key to a successful parametrisation is found to lie in the phase shifts. Based on the optimised quantum geometry, we come up with a strategy for predicting the phase shifts prior to the dihedral potential fitting. This allows us to successfully parameterise 8 out of the 11 molecules studied here, while the remaining 3 molecules can also be parameterised with small adjustments. Our work highlights the importance of predicting the dihedral phase shifts in the ligand parametrisation protocol, and provides a simple yet valuable strategy for improving the process of parameterising force fields of drug-like molecules

    Area-level deprivation and adiposity in children: is the relationship linear?

    OBJECTIVE: It has been suggested that childhood obesity is inversely associated with deprivation, such that the prevalence is higher in more deprived groups. However, comparatively few studies actually use an area-level measure of deprivation, limiting the scope to assess trends in the association with obesity for this indicator. Furthermore, most assume a linear relationship. Therefore, the aim of this study was to investigate associations between area-level deprivation and three measures of adiposity in children: body mass index (BMI), waist circumference (WC) and waist-to-height ratio (WHtR). DESIGN: This is a cross-sectional study in which data were collected on three occasions a year apart (2005-2007). SUBJECTS: Data were available for 13,333 children, typically aged 11-12 years, from 37 schools and 542 lower super-output areas (LSOAs). MEASURES: Stature, mass and WC. Obesity was defined as a BMI and WC exceeding the 95th centile according to British reference data. WHtR exceeding 0.5 defined obesity. The Index of Multiple Deprivation affecting children (IDACI) was used to determine area-level deprivation. RESULTS: Considerable differences in the prevalence of obesity exist between the three different measures. However, for all measures of adiposity the highest probability of being classified as obese is in the middle of the IDACI range. This relationship is more marked in girls, such that the probability of being obese for girls living in areas at the two extremes of deprivation is around half that at the peak, occurring in the middle. CONCLUSION: These data confirm the high prevalence of obesity in children and suggest that the relationship between obesity and residential area-level deprivation is not linear. This is contrary to the 'deprivation theory' and questions the current understanding and interpretation of the relationship between obesity and deprivation in children. These results could help make informed decisions at the local level

    A numerical study of the bearing capacity factor NĪ³

    Values of the bearing capacity factor NĪ³ are numerically computed using the method of triangular slices. Three assumptions of the value of Ļˆ, the base angle of the active wedge, are analyzed, corresponding to the following three cases: (1) Ļˆ = Ļ†, the internal friction angle; (2) Ļˆ = 45Ā° + Ļ†/2; and (3) Ļˆ has a value such that NĪ³ is a minimum. The location of the critical failure surface is presented and the numerical solutions to NĪ³ for the three cases are approximated by simple equations. The influence of the base angle on the value of NĪ³ is investigated. Comparisons of the present solutions are made with those commonly used in foundation engineering practice.published_or_final_versio

    Load-Balancing for Parallel Delaunay Triangulations

    Computing the Delaunay triangulation (DT) of a given point set in RD\mathbb{R}^D is one of the fundamental operations in computational geometry. Recently, Funke and Sanders (2017) presented a divide-and-conquer DT algorithm that merges two partial triangulations by re-triangulating a small subset of their vertices - the border vertices - and combining the three triangulations efficiently via parallel hash table lookups. The input point division should therefore yield roughly equal-sized partitions for good load-balancing and also result in a small number of border vertices for fast merging. In this paper, we present a novel divide-step based on partitioning the triangulation of a small sample of the input points. In experiments on synthetic and real-world data sets, we achieve nearly perfectly balanced partitions and small border triangulations. This almost cuts running time in half compared to non-data-sensitive division schemes on inputs exhibiting an exploitable underlying structure.Comment: Short version submitted to EuroPar 201
