    Partial root zone drying irrigation in papaya (Carica papaya L.) for enhanced water use efficiency under limited water situations

    Field experiments were conducted during 2015-17 at ICAR-Indian Institute of Horticultural Research, Hessaraghatta, Bengaluru, to standardize the partial root zone drying irrigation in papaya with 12 treatments in RBD design. The results indicated that better soil moisture in the root zone could be maintained under drip irrigation by shifting laterals on either side at fortnightly intervals as compared to fixed laterals with thesame amount of water. Significantly more primary roots (16.5/plant) were observed when irrigation was scheduled on one side with single emitter meeting 60% of the evaporative demand. PRD irrigation through shifting of laterals recorded significantly higher transpiration rate especially at 50% of ER (8.05 m mol m-2 s-1) as compared to the control (3.95m mol m-2 s-1). Further, the same treatment recorded significantly lower fruit cavity index (0.26) with relatively higher fruit volume (1388 cm3). Irrigating papaya only on one side with single emitter resulted in significantly higher T.S.S (13.0%). Higher water productivity (23.7 kg/m3) could be obtained by scheduling the irrigation at 40% evaporation replenishment through shifting of laterals with saving of substantial water (1285m3/ha) resulting in higher water use efficiency (237.4 kg/ha.mm)

    Emerging wetlands from river diversions can sustain high denitrification rates in a Coastal Delta

However, this excess also represents a risk by increasing soil decomposition rates triggering peat collapse and wetland fragmentation. Here, we evaluated the role of %OM and temperature interactions controlling denitrification rates in eroding (Barataria Bay-BLC) and emerging (Wax Lake Delta-WLD) deltaic regions in coastal Louisiana using the isotope pairing (IPT) and N2:Ar techniques. We also assessed differences between total (direct denitrification + coupled nitrification-denitrification) and net (total denitrification minus nitrogen fixation) denitrification rates in benthic and wetland habitats with contrasting %OM and bulk density (BD). Sediment (benthic) and soil (wetland) cores were collected during summer, spring, and winter (2015–2016) and incubated at close to in-situ temperatures (30°C, 20°C, and 10°C, respectively). Denitrification rates were linearly correlated with temperature; maximum mean rates ranged from 40.1–124.1 μmol m−2 h−1 in the summer with lower rates (30 μM) and water temperature is >10°C. In coastal Louisiana, substrates under these regimes are represented by emergent supra-tidal flats or land created by sediment diversions under oligohaline conditions (<1 ppt).This study was supported by the NOAA-Sea Grant Program-Louisiana (Grant 2013R/E-24) to Victor H.     Writing a Research Paper for Journal of Nepal Agricultural Research Council

    Agricultural research findings are required to reach soon to the farmers, extensionists, media, researchers, policy makers, businessperson, students, teachers and so many other stakeholders. Among different types of publications, research paper is generally published in journal considered as standard type of publication in term of quality and recognition. Most of the journals follow similar pattern and framework; however, the style, format and process may be different with each other. A research (scientific) paper is a written describing original research result using standard methods and materials. The major sections in a journal paper are abstract, introduction, materials and methods, results, discussion, conclusion, and references. Accordingly Journal of Nepal Agricultural Research Council has its own style and format. Author needs to follow guidelines strictly on the use of punctuation marks such as comma (,), period (.), space, justification etc; otherwise submitted manuscripts could be immediately return to author without considering in review process. In general, we received manuscripts with many errors on citation and references, poor elaboration of results of experiments, weak discussion, missing to acknowledge funding agencies, submitting non-editable figures, very few numbers of citations of Nepalese researchers, statements not in logical order, etc. In general, the scientific papers should be written in simple way with new but sufficient justification backed up by data in the form of tables, graphs, flow diagrams etc so that readers can understand easily with high readability. The submitted manuscript in the journal office are sent to two to three reviewers for specific recommendation on the originality of the work, appropriateness of the approach and experimental design, adequacy of experimental techniques, soundness of conclusions and interpretations, relevance of discussion and importance of the research. The language clarity and organization of the article are also asked with the reviewers. In response to reviewer's comments all authors are expected to reply each and every comments and suggestions of reviewers, if such comments and suggestion are not acceptable, the author/s can argue for their points, if genuine.     Streamlined and Cost-Effective Genomic DNA Extraction Method for Lichens, Mushrooms, and Endolichenic Fungi: Enabling DNA Barcoding and Molecular Research

    Extraction of nucleic acids in pure form from organisms is of paramount importance for DNA based identification and other molecular studies. Over the past few decades, DNA-barcoding has emerged as a powerful technique, facilitating species identification across various ‘difficult to identify’ life-forms. Fungi, being an immensely diverse group of microorganisms, contribute significantly to global biodiversity, with estimates ranging from 2.2 to 3.8 million species. However, a vast majority of this diversity remains unidentified, and many fungal species are considered cryptic. Therefore, numerous large- and small-scale DNA-barcoding projects are being conducted worldwide to unravel this rich biodiversity. However, the rigidity and high complex polysaccharides content of fungal cell-wall presents a significant obstacle, making the extraction of high-quality genomic DNA a challenging task across varied fungal organisms. In this study, we employed a modified CTAB based method to isolate and purify high-quality PCR-amplifiable genomic DNA primarily from lichens and tested it on other fungal life forms as well, including, mushrooms, endolichenic fungi, and parasitic fungi. Remarkably, the isolated DNA proved successful as a template in PCR reactions, serving the purposes of DNA barcoding, RAPD as well as for metagenomic analysis effectively. This versatile protocol demonstrated its utility across all the fungal life forms investigated in this study, offering a universal, cost-effective, and efficient approach for fungal DNA isolation

    Coupling Between Magnetic and Transport Properties in Magnetic Layered Material Mn2-xZnxSb

    We synthesized single crystals for Mn2-xZnxSb and studied their magnetic and electronic transport properties. This material system displays rich magnetic phase tunable with temperature and Zn composition. In addition, two groups of distinct magnetic and electronic properties, separated by a critical Zn composition of x = 0.6, are discovered. The Zn-less samples are metallic and characterized by a resistivity jump at the magnetic ordering temperature, while the Zn-rich samples lose metallicity and show a metal-to-insulator transition-like feature tunable by magnetic field. Our findings establish Mn2-xZnxSb as a promising material platform that offers opportunities to study how the coupling of spin, charge, and lattice degrees of freedom governs interesting transport properties in 2D magnets, which is currently a topic of broad interest.Comment: 23 pages, 5 figures, Figures are at the end of the manuscrip

    Measurement of tamsulosin in human serum by liquid chromatography-tandem mass spectrometry

    AbstractA simple, sensitive and robust method to extract tamsulosin from human serum, and quantify by liquid chromatography–tandem mass spectrometry (LC–MS/MS) was developed and validated and is applicable as a measure of compliance in clinical research. Tamsulosin was extracted from human serum (100μL) via liquid–liquid extraction with methyl tert-butyl ether (2mL) following dilution with 0.1M ammonium hydroxide (100μL), achieving 99.9% analyte recovery. Internal standard, d9-finasteride, was synthesised in-house. Analyte and internal standard were separated on an Ascentis® Express C18 (100mm×3mm, 2.7μm) column using a gradient elution with mobile phases methanol and 2mM aqueous ammonium acetate (5:95, v/v). Total run-time was 6min. Tamsulosin was quantified using a triple quadrupole mass spectrometer operated in multi-reaction-monitoring (MRM) mode using positive electrospray ionisation. Mass transitions monitored for quantitation were: tamsulosin m/z 409→228 and d9-finasteride m/z 382→318, with the structural formulae of ions confirmed by Fourier transform ion cyclotron resonance mass spectrometry (within 10ppm). The limit of quantitation was 0.2ng/mL, and the method was validated in the linear range 0.2–50ng/mL with acceptable inter- and intra-assay precision and accuracy and stability suitable for routine laboratory practice. The method was successfully applied to samples taken from research volunteers in a clinical study of benign prostatic hyperplasia

    Physio-biochemical responses of polyembryonic mango (Mangifera indica L.) genotypes to varying levels of salinity stress

    Mango genotypes that are salinity tolerant can possibly be used as clonal rootstock for sustained production of salinity sensitive commercial mango cultivars in salt affected soils. Present study was carried out to elucidate the effect of salinity stress induced by salts of NaCl+CaCl2 (1:1) at concentrations of 0, 25, 50 and 100 mM on fifteen polyembryonic mango genotypes. The physio-biochemical parameters such as relative water content, chlorophyll content, epicuticular wax content, water potential (Ψ), carbohydrate content, lipid peroxidation, proline accumulation and antioxidant enzymes were determined at each level of salinity in all genotypes. On the basis of these physio-biochemical changes, the study illustrated that the polyembryonic genotypes, Turpentine, Deorakhio, Olour, Bappakkai, Vattam, Nekkare, Kurukkan, Kensington, Muvandan, EC-95862, Manipur, Sabre, Vellaikolamban, Kitchener and Mylepelian were in the decreasing order in response to salinity stress tolerance

    Integrated Landscape Approach: Closing the Gap between Theory and Application

    Recently, the integrated landscape approach has gained increasing interest of the scientific community, as well as of organizations active in the field of sustainable development. However, the enthusiastic welcome is challenged by little consensus on theory, terminology and definitions. Moreover, the operationalization of the approach into practice is a major challenge. In this paper, we present a framework to operationalize the integrated landscape approach in practice by putting a long-term collaboration between scientists and various stakeholder at center stage. Based on encompassing understanding of landscape-level processes and interactions, four pillars addressing different steps of a joint-learning circle are described and illustrated with examples. We consider the integrated landscape approach to be a prime way of targeting the Sustainable Development Goals (SDGs), but novel forms of collaboration between scientists and other stakeholders based on long-term commitments will be needed for operationalization in practice
