Sleep and circadian rhythms are considered to be important determinants of mental and physical health. Epidemiological studies have established the contribution of self‐reported sleep duration, sleep quality and chronotype to health outcomes. Mental health and sleep problems are more common in women and men are more likely to be evening types. Few studies have compared the relative strength of these contributions and few studies have assessed these contributions separately in men and women. Furthermore, sleep and circadian characteristics are typically assessed with a limited number of instruments and a narrow range of variables is considered, leaving the understanding of the relative contribution of different predictors somewhat fractionary. We compared sleep quality, sleep duration and chronotype as predictors for self‐reported mental and physical health and psychological characteristics in 410 men and 261 women aged 18 to 30. To ascertain that results were not dependent on the use of specific instruments we used a multitude of validated instruments including the Morningness‐Eveningness‐Questionnaire, Munich‐ChronoType‐Questionnaire, Pittsburgh‐Sleep‐Quality‐Index, British‐Sleep‐Survey, Karolinska‐Sleep‐Diary, Insomnia‐Severity‐Index, SF‐36‐Health Survey, General‐Health‐Questionnaire, Dutch‐Eating‐Behaviour‐Questionnaire, Big‐Five‐Inventory, Behaviour‐Inhibition‐System‐Behaviour‐Activation‐System, and the Positive‐Affect‐Negative‐Affect‐Schedule. Relative contributions of predictors were quantified as local effect sizes derived from multiple regression models. Across all questionnaires, sleep quality was the strongest independent predictor of health and in particular mental health and more so in women than in men. The effect of sleep duration and social jetlag was inconspicuous. A greater insight into the independent contributions of sleep quality and chronotype may aid the understanding of sleep‐health interactions in women and men

Three-dimensional Character of the Magnetization Dynamics in Magnetic Vortex Structures - Hybridization of Flexure Gyromodes with Spin Waves

Three-dimensional linear spin-wave eigenmodes of a Permalloy disk having finite thickness are studied by micromagnetic simulations based on the Landau-Lifshitz-Gilbert equation. The eigenmodes found in the simulations are interpreted as linear superpositions (hybridizations) of 'approximate' three-dimensional eigenmodes, which are the fundamental gyromode G0G_0, the spin-wave modes and the higher-order gyromodes GNG_N (flexure modes), the thickness dependence of which is represented by perpendicular standing spin waves. This hybridization leads to new and surprising dependencies of the mode frequencies on the disk thickness. The three-dimensional character of the eigenmodes is essential to explain the recent experimental results on vortex-core reversal observed in relatively thick Permalloy disks

A set of forty wheat cultivars comprising bread wheat, durum and triticale identified during 2010–2014 were tested for resistance to Indian pathotypes of leaf, stem and yellow rusts at seedling stage under controlled conditions. Eight Lr genes (Lr1, Lr3, Lr10, Lr13, Lr14a, Lr23, Lr24 and Lr26) were characterized based on differential interactions with specific rust races. Genes Lr23, Lr26 and Lr13 conferred leaf rust resistance in most of the accessions. Three Yr genes (YrA, Yr2 and Yr9) were inferred in 40 genotypes, where Yr2 followed by Yr9 were most frequent in conferring stripe rust resistance. Ten Sr genes, namely, Sr2, Sr5, Sr8a, Sr7b, Sr9b, Sr9e, Sr11, Sr13, Sr24 and Sr31, were postulated in these lines with predominance of Sr11, Sr31 and Sr2. These Lr, Sr and Yr genes were observed singly or in combination. Robust DNA markers were used to identify adult plant resistance genes Yr18/Lr34/Sr57, Lr68 and Sr2 and all stage resistance genes Lr24/Sr24, Sr28 and Yr9/Lr26/Sr31. STS marker iag95 showed presence of Yr9 in four additional cultivars which were resistant to one or more rusts. Gene Sr28 was identified in seven durum cultivars with the wPt7004 marker. This is first report of Sr28 being present in many Indian wheat cultivars. CsGs-STS marker identified Lr68 in nine cultivars

Phase resolved observation of spin wave modes in antidot lattices

Antidot lattices have proven to be a powerful tool for spin wave band structure manipulation. Utilizing time-resolved scanning transmission x-ray microscopy, we are able to experimentally image edge-localized spin wave modes in an antidot lattice with a lateral confinement down to < 80 nm x 130 nm. At higher frequencies, spin wave dragonfly patterns formed by the demagnetizing structures of the antidot lattice are excited. Evaluating their relative phase with respect to the propagating mode within the antidot channel reveals that the dragonfly modes are not directly excited by the antenna but need the propagating mode as an energy mediator. Furthermore, micromagnetic simulations reveal that additional dispersion branches exist for a tilted external field geometry. These branches correspond to asymmetric spin wave modes that cannot be excited in a non-tilted field geometry due to the symmetry restriction. In addition to the band having a negative slope, these asymmetric modes also cause an unexpected transformation of the band structure, slightly reaching into the otherwise empty bandgap between the low frequency edge modes and the fundamental mode. The presented phase resolved investigation of spin waves is a crucial step for spin wave manipulation in magnonic crystals

Suspension High Velocity Oxy-Fuel (SHVOF)-sprayed alumina coatings: microstructure, nanoindentation and wear

Suspension High Velocity Oxy Fuel Spraying (SHVOF) can be used to produce thermally sprayed coatings from powdered feedstocks too small to be processed by mechanical feeders, allowing formation of nanostructured coatings with improved density and mechanical properties. Here, alumina coatings were produced from sub-micron sized feedstock in aqueous suspension, using two flame combustion parameters yielding contrasting microstructures. Both coatings were tested in dry sliding wear conditions with an alumina counterbody. The coating processed with high combustion power of 101 kW contained 74 wt% amorphous phase and 26 wt% crystalline phase (95 wt% gamma and 3 wt% alpha alumina) while the 72 kW coating contained lower 58 wt% amorphous phase and 42 wt% crystalline phases (73 wt% was alpha and 26 wt % gamma). The 101 kW coating had a dry sliding specific wear rate between 4-4.5 x 10-5 mm3/Nm, 2 orders of magnitude higher than the 72 kW coating wear rate of 2-4.2 x 10-7 mm3/Nm. A severe wear regime dominated by brittle fracture and grain pull out of the coating was responsible for the wear of the 101 kW coating, explained by mean fracture toughness three times lower than the 72 kW coating, owing to the almost complete absence of alpha alumina

BACKGROUND: Caspases are important regulators and executioners in apoptosis pathway and have been defined as either tumor suppressors or oncogenes. Polymorphisms in promoter and exon of caspase 9 were shown to confer genetic susceptibility to multiple cancers, but the results were inconsistent. To accomplish a more precise estimation of the relationship, a meta-analysis was performed. METHODOLOGY/PRINCIPAL FINDINGS: We assessed published studies of the association between caspase 9 polymorphisms and cancer risk from nine studies with 5,528 subjects for rs4645978, six studies with 2,403 subjects for rs105276 and two studies for rs4645981. Overall meta-analysis indicated that no evidence of an association between rs4645978 and cancers was found. Through the stratified analysis, statistically significant reduced cancer risks were observed among Caucasians (AG vs AA: OR = 0.81, 95% CI = 0.66-0.99, P(heterogeneity) = 0.150 and the dominant model: OR = 0.86, 95% CI = 0.75-0.99, P(heterogeneity) = 0.290) and prostate cancer. As for rs105276, Ex5+32G>A polymorphism was found with protective effect in overall meta-analysis (AA vs GG: OR = 0.75, 95% CI = 0.60-0.92, P(heterogeneity) = 0.887; A vs G: OR = 0.85, 95% CI = 0.77-0.95, P(heterogeneity) = 0.739 and the recessive model: OR = 0.68, 95% CI = 0.56-0.82, P(heterogeneity) = 0.309) and Asians group. While for rs4645981, a statistically significant increase in risk of lung cancer was shown in Asians (T vs C: OR = 1.23, 95% CI = 1.07-1.42, P(heterogeneity) = 0.399 and the dominant model: OR = 1.22, 95% CI = 1.04-1.43, P(heterogeneity) = 0.660). CONCLUSIONS/SIGNIFICANCE: Our meta-analysis suggests that the caspase 9 rs4645978 most likely contributes to decreased susceptibility to cancer in Caucasians and prostate cancer. The A allele of rs105276 might be a protective factor for cancer, especially for Asians. However, it seems that rs4645981 confers increased susceptibility to lung cancer in Asians

Structural Modeling and DNA Binding Autoinhibition Analysis of Ergp55, a Critical Transcription Factor in Prostate Cancer

    Get PDF
BACKGROUND: The Ergp55 protein belongs to Ets family of transcription factor. The Ets proteins are highly conserved in their DNA binding domain and involved in various development processes and regulation of cancer metabolism. To study the structure and DNA binding autoinhibition mechanism of Ergp55 protein, we have produced full length and smaller polypeptides of Ergp55 protein in E. coli and characterized using various biophysical techniques. RESULTS: The Ergp55 polypeptides contain large amount of α-helix and random coil structures as measured by circular dichorism spectroscopy. The full length Ergp55 forms a flexible and elongated molecule as revealed by molecular modeling, dynamics simulation and structural prediction algorithms. The binding analyses of Ergp55 polypeptides with target DNA sequences of E74 and cfos promoters indicate that longer fragments of Ergp55 (beyond the Ets domain) showed the evidence of auto-inhibition. This study also revealed the parts of Ergp55 protein that mediate auto-inhibition. SIGNIFICANCE: The current study will aid in designing the compounds that stabilize the inhibited form of Ergp55 and inhibit its binding to promoter DNA. It will contribute in the development of drugs targeting Ergp55 for the prostate cancer treatment
