Study of chiral symmetry restoration in linear and nonlinear O(N) models using the auxiliary field method

We consider the O(N) linear {\sigma} model and introduce an auxiliary field to eliminate the scalar self-interaction. Using a suitable limiting process this model can be continuously transformed into the nonlinear version of the O(N) model. We demonstrate that, up to two-loop order in the CJT formalism, the effective potential of the model with auxiliary field is identical to the one of the standard O(N) linear {\sigma} model, if the auxiliary field is eliminated using the stationary values for the corresponding one- and two-point functions. We numerically compute the chiral condensate and the {\sigma}- and {\pi}-meson masses at nonzero temperature in the one-loop approximation of the CJT formalism. The order of the chiral phase transition depends sensitively on the choice of the renormalization scheme. In the linear version of the model and for explicitly broken chiral symmetry, it turns from crossover to first order as the mass of the {\sigma} particle increases. In the nonlinear case, the order of the phase transition turns out to be of first order. Under the Law No. 23 of 2006 in Article 27 paragraph 1 explained that everybirth must be reported to the institution birth registration in birth place of occurrence ofthe event no later than 60 (sixty) days after birth. But in reality there are many people inthe district Kuantan Singingi not have a birth certificate. Therefore, the authors intend toconduct research in order to determine how the implementation of population policies inthe District Kuantan Singingi (Kuantan Singingi Regional Regulation No. 12 of 2011 onthe Implementation of Administration) and how public support for government policiesregarding the administration, especially processing of birth certificates in the DistrictKuantan Singingi in 2012 and the factors that influence it.This research sites in the district Kuantan Singingi. This study includesqualitative research, the technique of collecting data through interviews, observation,and research informants determined by purposive sampling technique. In this research,the theory is derived from the theory put forward by Grindle, the successfulimplementation of these policies can be seen from the benefits that would result,deployed resources, compliance and responsiveness to the program.The results of this research concluded that the implementation of governmentpolicy Kuantan Singingi about obtaining a birth certificate has not been run inaccordance with the expected goals and many people who do not support thegovernments policy in regard to obtaining a birth certificate. Nesting habitats and rates of cell parasitism in some bee species of the genera ancyloscelis, centris and euglossa (Hymenoptera: Apidae) from Colombia

We describe a dense nest aggregation of Ancyloscelis aff. apiformis, two nests of Centris flavifrons, and six nests of Euglossa. We found a nest of E. analis built inside a cavity of a living tree, an aerial nest of E. cybelia attached under a leaf of a palm tree and four nests of E. nigropilosa associated with timber buildings and abandoned nests of carpenter bees (Xylocopa sp.). We report for the first time wasps of the family Eurytomidae (Hymenoptera: Chalcidoidea) and a cuckoo bee, Coelioxys (Rhinocoelioxys) sp. (Hymenoptera: Megachilidae), attacking the brood cells of E. cybelia and E. nigropilosa. We also briefly discuss the variation found in the nest structure of C. flavifrons; unlike those nests studied in Costa Rica and northeastern Brazil, the nests in Colombia were shallow (~12 cm) and consisted of a main tunnel with three short branches, each one ending in a single cell. Further data from other localities are needed to detect other variations in the nesting biology and if they are correlated with the morphological variation exhibited by this species or with local differences in parasite pressure

The use of digital tools by general practitioners in Finnish public health centres

Electronic health services (eHealth) have increased rapidly in the last few years. These services hold the potential to facilitate today’s challenging primary care. The aim of the study was to examine how digitalisation manifests in Finnish health centres in 2021. We aimed to find out which digital tools are used in health centres and how they are exploited. In addition, we were interested in general practitioners’ attitudes towards digitalisation in their everyday work. An online survey was distributed to general practitioners (GPs) working in primary health care centres throughout Finland, and 265 GPs replied. A health portal and various digital calculators were used daily. In remote communication with their patients, general practitioners preferred telephone calls over new tools (chat/video). Attitudes towards eHealth were positive, but digital tools were not yet commonly used. The implementation of digital solutions still needs more effort

Depletion of Mediator Kinase Module Subunits Represses Superenhancer-Associated Genes in Colon Cancer Cells

In cancer, oncogene activation is partly mediated by acquired superenhancers, which therefore represent potential targets for inhibition. Superenhancers are enriched for BRD4 and Mediator, and both BRD4 and the Mediator MED12 subunit are disproportionally required for expression of superenhancer-associated genes in stem cells. Here we show that depletion of Mediator kinase module subunit MED12 or MED13 together with MED13L can be used to reduce expression of cancer-acquired superenhancer genes, such as the MYC gene, in colon cancer cells, with a concomitant decrease in proliferation. Whereas depletion of MED12 or MED13/MED13L caused a disproportional decrease of superenhancer gene expression, this was not seen with depletion of the kinases cyclin-dependent kinase 9 (CDK8) and CDK19. MED12-MED13/MED13L-dependent superenhancer genes were coregulated by beta-catenin, which has previously been shown to associate with MED12. Importantly, beta-catenin depletion caused reduced binding of MED12 at the MYC superenhancer. The effect of MED12 or MED13/MED13L depletion on cancer-acquired superenhancer gene expression was more specific than and partially distinct from that of BRD4 depletion, with the most efficient inhibition seen with combined targeting. These results identify a requirement of MED12 and MED13/MED13L for expression of acquired superenhancer genes in colon cancer, implicating these Mediator subunits as potential therapeutic targets for colon cancer, alone or together with BRD4.Peer reviewe

A novel partial de novo duplication of JARID2 gene causing a neurodevelopmental phenotype

Molecular Genetics & Genomic Medicine published by Wiley Periodicals LLC.Background: Deletions covering the entire or partial JARID2 gene as well as pathogenic single nucleotide variants leading to haploinsufficiency of JARID2 have recently been shown to cause a clinically distinct neurodevelopmental syndrome. Here, we present a previously undescribed partial de novo duplication of the JARID2 gene in a patient displaying features similar to those of patients with JARID2 loss-of-function variants. Case report: The index patient presents with abnormalities in gross motor skills and speech development as well as neuropsychiatric disorders. The patient has markedly dark infraorbital circles and slightly prominent supraorbital ridges.Whole-genome sequencing and array comparative genomic hybridization revealed a novel disease-causing variant type, a partial tandem duplication of JARID2, covering the exons 1–7. Furthermore, RNA sequencing validated the increased expression of these exons. Expression alterations were also detected in target genes of the PRC2 complex, in which JARID2 acts as an essential member. Conclusion: Our data add to the variety of different pathogenic variants associated with JARID2 neurodevelopmental syndrome.Peer reviewe

Group A Streptococcal S Protein Utilizes Red Blood Cells as Immune Camouflage and Is a Critical Determinant for Immune Evasion.

Group A Streptococcus (GAS) is a human-specific pathogen that evades the host immune response through the elaboration of multiple virulence factors. Although many of these factors have been studied, numerous proteins encoded by the GAS genome are of unknown function. Herein, we characterize a biomimetic red blood cell (RBC)-captured protein of unknown function-annotated subsequently as S protein-in GAS pathophysiology. S protein maintains the hydrophobic properties of GAS, and its absence reduces survival in human blood. S protein facilitates GAS coating with lysed RBCs to promote molecular mimicry, which increases virulence in vitro and in vivo. Proteomic profiling reveals that the removal of S protein from GAS alters cellular and extracellular protein landscapes and is accompanied by a decrease in the abundance of several key GAS virulence determinants. In vivo, the absence of S protein results in a striking attenuation of virulence and promotes a robust immune response and immunological memory

Symptom Burden Is Associated with Psychological Wellbeing and Mortality in Older Adults

Importance Over half of outpatient visits are due to physical symptoms; yet, the significance of symptoms in relation to older people's wellbeing and prognosis has gained very little research attention. Objectives This study aims to analyze the prognostic value of symptom burden, derived from symptom count and frequency, in an older cohort aged 75 to 95. We also explore the association between symptom burden and psychological wellbeing. Design Randomly assigned cohorts of community-dwelling people aged 75-95 filled in the postal questionnaire of the Helsinki Aging Study in 2009. Setting Community-based, postal questionnaires (survey response rate 74%). Participants 1583 community-dwelling people aged 75-95 in the urban Helsinki area. Main outcomes and measures The inquired symptoms were dizziness, back pain, joint pain, chest pain or discomfort, shortness of breath, leg pain when walking, loss of appetite, and urinary incontinence. Symptom burden was calculated according to the number of symptoms and their frequency (score range: 0-8). The participants were subdivided into four groups according to their symptom burden. Mortality data was extracted from the Finnish Population Register in 2014. Psychological wellbeing (PWB) was measured using the validated PWB score. Results Of 1583 participants, 18% reported no symptoms over the past 2 weeks (Group 0), 31% scored 0.5-1 in the symptom burden score (Group 1), 23% scored 1.5-2 (Group 2), and 28% scored 2.5-8 (Group 3). There was a linear relationship between symptom burden and comorbidities, functional status, falls, and PWB.
