    Genetic transformation of Paulownia elongata S. Y. Hu., mediated by Agrobacterium tumefaciens and biolistic system

    Objective: The most appropriate conditions for genetic transformation through direct (bioballistic) and indirect (Agrobacterium tumefaciens) transformation systems in Paulownia elongata were established. Design/methodology/approach: Starting from in vitro propagation through both direct and indirect organogenesis, internodal stem segments with 0.5 to 1 cm length were determined as the best explant. The optimum dose for selection media was determined to be 15 mg L-1 of kanamycin. It was possible to obtain transgenic plants under both transformation systems. In the case of Agrobacterium tumefaciens, two hours of incubation, 48 h of co-cultivation, and optical density of 0.9 were used; while for bioballistics, the best conditions were 120 PSI of shot pressure, shot height at level 6 (16 cm), and vacuum pressure of 22 Hg mm, with particle inflow gun system (PIG). Results: Both systems produced complete transformants, chimeras, as well as those confirmed by histochemical X-GLUC and PCR analysis, producing a total of 14 positive plants by A. tumefaciens transformation from 26 trials and ten positive plants by the bioballistic system from 30 trials; a construction with chitinase and glucanase, NPT II selection gene and the GUS reporter gene were used. Findings/conclusions: So far, this has been the first report including integration of chitinase and glucanase genes

    Race diversity in dryland maize (Zea mays L.) landraces from southern Nuevo León, Mexico

    This study was done for reviewing the diversity and distribution of native maize grown at southern Nuevo León, Mexico, in 40 accessions collected in the area. Information obtained was on the production unit and on the ears traits, according to the CONABIO guidelines. In this area maize is usually grown during the Spring-Summer period, and using little technology, without fertilization or pest control. The 40 accessions correspond to seven races and seven interracial crosses. The most frequent maize races were Ratón and Cónico Norteño, especially in dry areas with little rain. Races Celaya, Tablilla de Ocho and Elotes Cónicos found in this study, had not been previously reported; but the Tabloncillo and Olotillo races that had previously been reported were no longer found. Three collections stood out for producing large ears with large kernels, thus having a high yield potential. Native maize harvested in dryland areas might offer advantages to be grown under harsh environments.Objective: To analyze the race diversity and geographic distribution of the native maize landraces currently cropped at southern Nuevo León, México. Study Limitations/Implications: Three races, Celaya, Tablilla de Ocho and Elotes Cónicos, had not been previously reported; while two formerly reported races Tabloncillo and Olotillo, were no longer found. This study did not include the grain-colored accessions. Findings/Conclusions: Three collections stood out for producing large ears with large kernels, thus showing a high yield potential. The maize landraces harvested in dryland areas might offer advantages to be grown under harsh environments or be used as gene donors for drought tolerance

    Morphological variability of native maize (Zea mays L.) of the west highland of Puebla and east highland of Tlaxcala, Mexico

    The objective of this research was to assess the morphological variability of maize landraces native of the west highland of Puebla and east highland of Tlaxcala, México, in order to, besides define it, related it to races, commercial varieties and with the altitude of the localities of seed collection. The genetic resources evaluated were 134 accessions collected in 34 localities, along with 10 controls. Experiments were established on three localities using a Lattice 12 x 12. Thirty-two morphological traits were analyzed, 27 of them resulting with highly significant differences, reflecting the high variability at the level of morphological characters, many of them of agronomic interest. From variance analysis 16 traits were selected for use in a cluster analysis through the Modified Localization Method, which joint the populations in six groups, most of them in the group 1, with morphological traits of long cycle varieties: high plants, with greater primary branches of the tassel, greater ear diameter and length, and greater grain length and width.     Actualidad y prospectiva de la investigación científica en el Centro Universitario Amecameca de la Universidad Autónoma del Estado de México

    Con responsabilidad, se organizó un programa cuya finalidad fuera publicitar con transparencia dichos avances, a través de un esfuerzo de rendición de cuentas a la comunidad inmediata, la universitaria, y a la comunidad abierta, la sociedad que la principal referencia para tal efecto. El programa se concretiza a través del presente libro, conformado con una inspiración de investigación multidisciplinaria; sin embargo, para llegar a tal fin, el reto es realizar el proceso de búsqueda y generación de conocimiento transitando hacia la colaboración de los cuerpos académicos, que puedan construir nuevos conocimientos fortalecidos por la convergencia de diferentes campos del saber. En consecuencia, la primera etapa de esta estrategia es la publicidad de los trabajos investigativos ejercidos, para hacer un balance al día, pero también proyectar el futuro de cada campo y área del conocimiento. La organización explicativa está organizada por tres bloques representativos del quehacer en la generación de conocimiento del Centro Universitario, un primer bloque centra el interés en las humanidades, educación y sustentabilidad; el segundo bloque lo integra la reflexión científica sobre la construcción democrática, derechos humanos y equidad de género; en el tercer segmento se destina a la seguridad alimentaria, salud pública y sistemas agropecuarios. La actualidad de la investigación eleva la producción lograda y lo que en el momento se encuentra en construcción y los alcances que produce para la docencia, la investigación misma, y para la sociedad en general. La prospectiva es un área que todos los capítulos desarrollan con el propósito de delinear los alcances innovadores por andar en teoría, metodología e incluso en los saberes mismo

    Reproducibility of fluorescent expression from engineered biological constructs in E. coli

    We present results of the first large-scale interlaboratory study carried out in synthetic biology, as part of the 2014 and 2015 International Genetically Engineered Machine (iGEM) competitions. Participants at 88 institutions around the world measured fluorescence from three engineered constitutive constructs in E. coli. Few participants were able to measure absolute fluorescence, so data was analyzed in terms of ratios. Precision was strongly related to fluorescent strength, ranging from 1.54-fold standard deviation for the ratio between strong promoters to 5.75-fold for the ratio between the strongest and weakest promoter, and while host strain did not affect expression ratios, choice of instrument did. This result shows that high quantitative precision and reproducibility of results is possible, while at the same time indicating areas needing improved laboratory practices.Peer reviewe
