Fibrous Cap Smooth Muscle Cells in Atherosclerotic Coronary Arteries Do Not Express Pluripotent Stem Cell Markers
Rupture of the coronary artery fibrous cap is a common cause of myocardial infarction, and bone marrow derived cells could play a role in preventing plaque rupture. It is currently unknown whether smooth muscle cells within coronary artery fibrous cap formation are of mature phenotype. Objective. To characterize cells expressing bone marrow stem cells of embryonic type (ESC) markers in coronary artery fibrous cap formation. Design. New Zealand White rabbits were fed a diet supplemented with 0.5% cholesterol  +  1% methionine  +  5% peanut oil for 4 weeks and then a normal diet for 9 weeks. The left main coronary artery was excised from the heart, processed for paraffin and immunohistochemistry was performed by standard techniques. Results. Oct-4, SSEA 1, 3, and 4 were all present within in atherosclerotic plaque core, codistributed with RAM-11, and were sparingly found in the fibrous cap, but TRA-1-60 and TRA-1-81 positive cells were scarce. Core but not fibrous cap smooth muscle (SMC actin+) cells also showed codistribution with ESC markers. Conclusions. These results suggest that smooth muscle cells present in the fibrous cap do not express ESC markers, indicative of a mature cell
Eicosapentaenoic acid and oxypurinol in the treatment of muscle wasting in a mouse model of cancer cachexia
Cancer cachexia is a wasting condition, driven by systemic inflammation and oxidative stress. This study investigated eicosapentaenoic acid (EPA) in combination with oxypurinol as a treatment in a mouse model of cancer cachexia. Mice with cancer cachexia were randomized into 4 treatment groups (EPA (0.4 g/kg/day), oxypurinol (1 mmol/L ad-lib), combination, or control), and euthanized after 29 days. Analysis of oxidative damage to DNA, mRNA analysis of pro-oxidant, antioxidant and proteolytic pathway components, along with enzyme activity of pro- and antioxidants were completed on gastrocnemius muscle. The control group displayed earlier onset of tumor compared to EPA and oxypurinol groups (P<0.001). The EPA group maintained body weight for an extended duration (20 days) compared to the oxypurinol (5 days) and combination (8 days) groups (P<0.05). EPA (18.2±3.2 pg/ml) and combination (18.4±3.7 pg/ml) groups had significantly higher 8-OH-dG levels than the control group (12.9±1.4 pg/ml, P≤0.05) indicating increased oxidative damage to DNA. mRNA levels of GPx1, MURF1 and MAFbx were higher following EPA treatment compared to control (P≤0.05). Whereas oxypurinol was associated with higher GPx1, MnSOD, CAT, XDH, MURF1, MAFbx and UbB mRNA compared to control (P≤0.05). Activity of total SOD was higher in the oxypurinol group (32.2±1.5 U/ml) compared to control (27.0±1.3 U/ml, P<0.01), GPx activity was lower in the EPA group (8.76±2.0 U/ml) compared to control (14.0±1.9 U/ml, P<0.05), and catalase activity was lower in the combination group (14.4±2.8 U/ml) compared to control (20.9±2.0 U/ml, P<0.01). There was no change in XO activity. The increased rate of weight decline in mice treated with oxypurinol indicates that XO may play a protective role during the progression of cancer cachexia, and its inhibition is detrimental to outcomes. In combination with EPA, there was little significant improvement from control, indicating oxypurinol is unlikely to be a viable treatment compound in cancer cachexia.<br /
Educación para la animación en un contexto índígena
En este trabajo describimos los procesos y resultados de los talleres de animación y video que realizamos en el Centro Rural de Educación Superior, en Estipac, Jalisco, México. Los talleres se dirigieron a estudiantes de preparatoria y licenciatura, muchos de ellos huicholes, a quienes les dimos el equipo técnico, el entrenamiento y el apoyo necesario para permitir que contaran sus propias historias en su idioma a través de la animación cuadro por cuadro. Durante el proceso, no sólo aprendieron a valorar, preservar y transmitir sus tradiciones culturales en forma novedosa, sino que también se ejercitaron en ser consumidores más críticos de los medios de comunicación de la sociedad contemporánea. La idea que guió estos talleres es que la animación puede abrir las puertas al mundo de los medios visuales a partir del interés en las competencias sobre medios de comunicación y justicia social.

This article describes the processes and results of the animation and video workshops we held at Centro Rural de Educación Superior, en Estipac, Jalisco, Mexico. The workshops were directed to high school and college students, many of whom were Huicholes. We gave them technical equipment, training, and the necessary support to allow them to tell their own stories in their language, through frame–by–frame animation. During the process, they learned not only to value, preserve, and transmit their cultural traditions in a new way, but also to practice being more critical consumers of mass media in today's society. The idea that guided these workshops is that animation can open the door to the world of the visual media, based on interest in media skills and social justice
Environmental monitoring in the Mechara caves, Southeastern Ethiopia: implications for speleothem palaeoclimate studies
The interpretation of palaeoclimate records in speleothems depends on the understanding of the modern climate of the region, the geology, the hydrology above the caves, and the within-cave climate. Monitoring within-cave climate variability, geochemistry of speleothem-forming drip waters, and associated surface and groundwater, provides a modern baseline for interpretation of speleothem palaeohydrological and palaeoclimate records. Here, we present results of such monitoring of the Mechara caves in southeastern Ethiopia, conducted between 2004 and 2007. Results show nearly constant within-cave climate (temperature and humidity) in all caves, which generally reflects the surface climate. Groundwater and surface water geochemistry is similar across the region (except slight modification by local lithological variations) and modern drip water isotope data fall close to regional Meteoric Water Line, but speleothems further from equilibrium. Holocene and modern speleothems from these caves give high-resolution climate records, implying that the Mechara caves provide a suitable setting for the deposition of annually laminated speleothems that could record surface climate variability in a region where rainfall is sensitive to both the strength of the intertropical convergence zone as well as Indian Monsoon variability
Decreased NADPH oxidase expression and antioxidant activity in cachectic skeletal muscle
Background - Cancer cachexia is the progressive loss of skeletal muscle protein that contributes significantly to cancer morbidity and mortality. Evidence of antioxidant attenuation and the presence of oxidised proteins in patients with cancer cachexia indicate a role for oxidative stress. The level of oxidative stress in tissues is determined by an imbalance between reactive oxygen species production and antioxidant activity. This study aimed to investigate the superoxide generating NADPH oxidase (NOX) enzyme and antioxidant enzyme systems in murine adenocarcinoma tumour-bearing cachectic mice. Methods - Superoxide levels, mRNA levels of NOX enzyme subunits and the antioxidant enzymes superoxide dismutase (SOD), glutathione peroxidise (GPx) and catalase was measured in the skeletal muscle of mice with cancer and cancer cachexia. Protein expression levels of NOX enzyme subunits and antioxidant enzyme activity was also measured in the same muscle samples. Results - Superoxide levels increased 1.4-fold in the muscle of mice with cancer cachexia, and this was associated with a decrease in mRNA of NOX enzyme subunits, NOX2, p40phox and p67phox along with the antioxidant enzymes SOD1, SOD2 and GPx. Cancer cachexia was also associated with a 1.3-fold decrease in SOD1 and 2.0-fold decrease in GPx enzyme activity. Conclusion - Despite increased superoxide levels in cachectic skeletal muscle, NOX enzyme subunits, NOX2, p40phox and p67phox, were downregulated along with the expression and activity of the antioxidant enzymes. Therefore, the increased superoxide levels in cachectic skeletal muscle may be attributed to the reduction in the activity of endogenous antioxidant enzymes
The effect of cocoa supplementation on hepatic steatosis, reactive oxygen species and LFABP in a rat model of NASH
Background
Non alcoholic steatohepatitis is hypothesised to develop via a mechanism involving fat accumulation and oxidative stress. The current study aimed to investigate if an increase in oxidative stress was associated with changes in the expression of liver fatty acid binding protein in a rat model of non alcoholic steatohepatitis and whether cocoa supplementation attenuated those changes.
Methods
Female Sprague Dawley rats were fed a high fat control diet, a high fat methionine choline deficient diet, or one of four 12.5% cocoa supplementation regimes in combination with the high fat methionine choline deficient diet.
Results
Liver fatty acid binding protein mRNA and protein levels were reduced in the liver of animals with fatty liver disease when compared to controls. Increased hepatic fat content was accompanied by higher levels of oxidative stress in animals with fatty liver disease when compared to controls. An inverse association was found between the levels of hepatic liver fatty acid binding protein and the level of hepatic oxidative stress in fatty liver disease. Elevated NADPH oxidase protein levels were detected in the liver of animals with increased severity in inflammation and fibrosis. Cocoa supplementation was associated with partial attenuation of these pathological changes, although the severity of liver disease induced by the methionine choline deficient diet prevented complete reversal of any disease associated changes. Red blood cell glutathione was increased by cocoa supplementation, whereas liver glutathione was reduced by cocoa compared to methionine choline deficient diet fed animals.
Conclusion
These findings suggest a potential role for liver fatty acid binding protein and NADPH oxidase in the development of non alcoholic steatohepatitis. Furthermore, cocoa supplementation may have be of therapeutic benefit in less sever forms of NASH
Who consults chiropractors in Victoria, Australia?: Reasons for attending, general health and lifestyle habits of chiropractic patients
Background
COAST (Chiropractic Observational and Analysis STudy) reported the clinical practices of chiropractors. The aims of this study were to: 1) describe the chiropractic patient demographic and health characteristics; 2) describe patient-stated reasons for visiting a chiropractor; 3) describe chiropractic patient lifestyle characteristics; 4) compare, where possible, chiropractic patient characteristics to the general Australian population.
Methods
Fifty-two chiropractors in Victoria, Australia, provided information for up to 100 consecutive encounters. If patients attended more than once during the 100 encounters, only data from their first encounter were included in this study. Where possible patient characteristics were compared with the general Australian population.
Results
Data were collected from December 2010 to September 2012. Data were provided for 4464 encounters, representing 3287 unique individuals. The majority of chiropractic encounters were for musculoskeletal conditions or for wellness/maintenance. The majority of patient comorbidities were musculoskeletal, circulatory or endocrine/metabolic in nature. Eight hundred chiropractic patients (57 %, 95 % CI: 53–61) described their self-reported health as excellent or very good and 138 patients (10 %, 95 % CI: 8–12) as fair or poor. Seventy-one percent of adult male patients (18 years and older), and 53 % of adult female patients, were overweight or obese. Fourteen percent (n = 188, 95 % CI: 12–16) were current smokers and 27 % (n = 359, 95 % CI: 24–31) did not meet Australian alcohol consumption guidelines. Less than half of the chiropractic patients participated in vigorous exercise at least twice per week. Approximately 20 % ate one serving of vegetables or less each day, and approximately 50 % ate one serve of fruit or less each day. Compared to the general Australian population, chiropractic patients were less likely to smoke, less likely to be obese and more likely to describe their health in positive terms. However, many patients were less likely to meet alcohol consumption guidelines, drinking more than is recommended.
Conclusions
In general, chiropractic patients had more positive health and lifestyle characteristics than the Australian population. However, there were a significant proportion of chiropractic patients who did not meet guideline recommendations about lifestyle habits and there is an opportunity for chiropractors to reinforce public health messages with their patients
The SDSS Coadd: A Galaxy Photometric Redshift Catalog
We present and describe a catalog of galaxy photometric redshifts (photo-z's)
for the Sloan Digital Sky Survey (SDSS) Coadd Data. We use the Artificial
Neural Network (ANN) technique to calculate photo-z's and the Nearest Neighbor
Error (NNE) method to estimate photo-z errors for 13 million objects
classified as galaxies in the coadd with . The photo-z and photo-z
error estimators are trained and validated on a sample of
galaxies that have SDSS photometry and spectroscopic redshifts measured by the
SDSS Data Release 7 (DR7), the Canadian Network for Observational Cosmology
Field Galaxy Survey (CNOC2), the Deep Extragalactic Evolutionary Probe Data
Release 3(DEEP2 DR3), the VIsible imaging Multi-Object Spectrograph - Very
Large Telescope Deep Survey (VVDS) and the WiggleZ Dark Energy Survey. For the
best ANN methods we have tried, we find that 68% of the galaxies in the
validation set have a photo-z error smaller than . After
presenting our results and quality tests, we provide a short guide for users
