    How the brain represents the reward value of fat in the mouth.

    The palatability and pleasantness of the sensory properties of foods drive food selection and intake and may contribute to overeating and obesity. Oral fat texture can make food palatable and pleasant. To analyze its neural basis, we correlated humans’ subjective reports of the pleasantness of the texture and flavor of a high- and low-fat food with a vanilla or strawberry flavor, with neural activations measured with functional magnetic resonance imaging. Activity in the midorbitofrontal and anterior cingulate cortex was correlated with the pleasantness of oral fat texture and in nearby locations with the pleasantness of flavor. The pregenual cingulate cortex showed a supralinear response to the combination of high fat and pleasant, sweet flavor, implicating it in the convergence of fat texture and flavor to produce a representation of highly pleasant stimuli. The subjective reports of oral fattiness were correlated with activations in the midorbitofrontal cortex and ventral striatum. The lateral hypothalamus and amygdala were more strongly activated by high- versus low-fat stimuli. This discovery of which brain regions track the subjective hedonic experience of fat texture will help to unravel possible differences in the neural responses in obese versus lean people to oral fat, a driver of food intake

    The anticipation of events in time

    Humans anticipate events signaled by sensory cues. It is commonly assumed that two uncertainty parameters modulate the brain's capacity to predict: the hazard rate (HR) of event probability and the uncertainty in time estimation which increases with elapsed time. We investigate both assumptions by presenting event probability density functions (PDFs) in each of three sensory modalities. We show that perceptual systems use the reciprocal PDF and not the HR to model event probability density. We also demonstrate that temporal uncertainty does not necessarily grow with elapsed time but can also diminish, depending on the event PDF. Previous research identified neuronal activity related to event probability in multiple levels of the cortical hierarchy (sensory (V4), association (LIP), motor and other areas) proposing the HR as an elementary neuronal computation. Our results—consistent across vision, audition, and somatosensation—suggest that the neurobiological implementation of event anticipation is based on a different, simpler and more stable computation than HR: the reciprocal PDF of events in time

    Neural systems underlying decisions about affective odors.

    Decision making about affective value may occur after the reward value of a stimulus is represented and may involve different brain areas to those involved in decision-making about the physical properties of stimuli, such as intensity. In an fMRI study, we delivered two odors separated by a delay, with instructions on different trials to decide which odor was more pleasant or more intense or to rate the pleasantness and intensity of the second odor without making a decision. The fMRI signals in the medial pFC area 10 and in regions to which it projects, including the ACC and insula, were higher when decisions were being made compared with ratings, implicating these regions in decision-making. Decision-making about affective value was related to larger signals in the dorsal part of medial area 10 and the agranular insula, whereas decisions about intensity were related to larger activations in the dorsolateral pFC, ventral premotor cortex, and anterior insula. For comparison, the mid-OFC had activations related not to decision making but to subjective pleasantness ratings, providing a continuous representation of affective value. In contrast, areas such as medial area 10 and the ACC are implicated in reaching a decision in which a binary outcome is produced

    Attention-dependent modulation of cortical taste circuits revealed by granger causality with signal-dependent noise

    We show, for the first time, that in cortical areas, for example the insular, orbitofrontal, and lateral prefrontal cortex, there is signal-dependent noise in the fMRI blood-oxygen level dependent (BOLD) time series, with the variance of the noise increasing approximately linearly with the square of the signal. Classical Granger causal models are based on autoregressive models with time invariant covariance structure, and thus do not take this signal-dependent noise into account. To address this limitation, here we describe a Granger causal model with signal-dependent noise, and a novel, likelihood ratio test for causal inferences. We apply this approach to the data from an fMRI study to investigate the source of the top-down attentional control of taste intensity and taste pleasantness processing. The Granger causality with signal-dependent noise analysis reveals effects not identified by classical Granger causal analysis. In particular, there is a top-down effect from the posterior lateral prefrontal cortex to the insular taste cortex during attention to intensity but not to pleasantness, and there is a top-down effect from the anterior and posterior lateral prefrontal cortex to the orbitofrontal cortex during attention to pleasantness but not to intensity. In addition, there is stronger forward effective connectivity from the insular taste cortex to the orbitofrontal cortex during attention to pleasantness than during attention to intensity. These findings indicate the importance of explicitly modeling signal-dependent noise in functional neuroimaging, and reveal some of the processes involved in a biased activation theory of selective attention

    The cingulate cortex and limbic systems for emotion, action, and memory

    Evidence is provided for a new conceptualization of the connectivity and functions of the cingulate cortex in emotion, action, and memory. The anterior cingulate cortex receives information from the orbitofrontal cortex about reward and non-reward outcomes. The posterior cingulate cortex receives spatial and action-related information from parietal cortical areas. It is argued that these inputs allow the cingulate cortex to perform action–outcome learning, with outputs from the midcingulate motor area to premotor areas. In addition, because the anterior cingulate cortex connects rewards to actions, it is involved in emotion; and because the posterior cingulate cortex has outputs to the hippocampal system, it is involved in memory. These apparently multiple different functions of the cingulate cortex are related to the place of this proisocortical limbic region in brain connectivity

    Stuck in time – a new Chaenothecopsis species with proliferating ascomata from Cunninghamia resin and its fossil ancestors in European amber

    Resin protects wounded trees from microbial infection, but also provides a suitable substrate for the growth of highly specialized fungi. Chaenothecopsis proliferatus is described growing on resin of Cunninghamia lanceolata from Hunan Province, China. The new fungus is compared with extant species and two new fossil specimens from Eocene Baltic and Oligocene Bitterfeld ambers. The Oligocene fossil had produced proliferating ascomata identical to those of the newly described species and to other extant species of the same lineage. This morphology may represent an adaptation to growing near active resin flows: the proliferating ascomata can effectively rejuvenate if partially overrun by fresh, sticky exudate. Inward growth of fungal hyphae into resin has only been documented from Cenozoic amber fossils suggesting comparatively late occupation of resin as substrate by fungi. Still, resinicolous Chaenothecopsis species were already well adapted to their special ecological niche by the Eocene, and the morphology of these fungi has since remained remarkably constant

    Assimilation of healthy and indulgent impressions from labelling influences fullness but not intake or sensory experience

    Background: Recent evidence suggests that products believed to be healthy may be over-consumed relative to believed indulgent or highly caloric products. The extent to which these effects relate to expectations from labelling, oral experience or assimilation of expectations is unclear. Over two experiments, we tested the hypotheses that healthy and indulgent information could be assimilated by oral experience of beverages and influence sensory evaluation, expected satiety, satiation and subsequent appetite. Additionally, we explored how expectation-experience congruency influenced these factors. Results: Results supported some assimilation of healthiness and indulgent ratings—study 1 showed that indulgent ratings enhanced by the indulgent label persisted post-tasting, and this resulted in increased fullness ratings. In study 2, congruency of healthy labels and oral experience promoted enhanced healthiness ratings. These healthiness and indulgent beliefs did not influence sensory analysis or intake—these were dictated by the products themselves. Healthy labels, but not experience, were associated with decreased expected satiety. Conclusions: Overall labels generated expectations, and some assimilation where there were congruencies between expectation and experience, but oral experience tended to override initial expectations to determine ultimate sensory evaluations and intake. Familiarity with the sensory properties of the test beverages may have resulted in the use of prior knowledge, rather than the label information, to guide evaluations and behaviour

    Preferences for fat, sugar, and oral-sensory food qualities in monkeys and humans

    In humans and other primates, food intake depends on sophisticated, individualized preferences for nutrients and oral-sensory food qualities that guide decision-making and eating behavior. The neural and behavioral mechanisms for such primate-typical food preferences remain poorly understood, despite their importance for human health and their targeting by pharmacological obesity treatments. Here, we review a series of experiments that investigated how the biologically critical properties of foods—their nutrients (sugar, fat, protein) and oral-sensory qualities (viscosity, oral sliding friction)—influence food preferences in monkeys and humans. In an economic nutrient-choice paradigm, macaques flexibly trade nutrients and oral-sensory food qualities against varying food amounts, consistent with the assignment of subjective values. Nutrient-value functions that link objective nutrient content to subjective values accurately model these preferences, predict choices across contexts, and explain individual differences. The monkeys’ aggregated choice patterns resulting from their nutrient preferences lead to daily nutrient balances that deviate from dietary reference points, resembling suboptimal human eating patterns when exposed to high-calorie foods. To investigate the sensory basis underlying nutrient values, we developed novel engineering tools that quantify food textures on oral surfaces, using fresh pig tongues. Oral-texture (i.e., mouthfeel) parameters, including viscosity and sliding friction, were shown to mediate monkeys' preferences for high-fat foods. When translated to human subjects, this approach revealed a neural mechanism for preferring high-fat foods from oral texture in the orbitofrontal cortex (OFC)—a key reward system of the brain. Importantly, human OFC responses to oral sliding friction in individual subjects—measured in the MRI scanner—predicted subsequent fat intake in a naturalistic, life-like eating test. These findings suggest that a primate nutrient-reward paradigm offers a promising approach for investigating the behavioral and neural mechanisms for human-typical food reward and food choice, to advance understanding of human eating behavior, overeating, and obesity

    The amygdala and the pursuit of future rewards

    The successful pursuit of future rewards requires forming an internal goal, followed by planning, decision-making, and progress-tracking over multiple steps. The initial step—forming goals and the plans for obtaining them—involves the subjective valuation of an anticipated reward, considering both the reward’s properties and associated delay and physical-effort costs. Recent findings indicate individuals similarly evaluate cognitive effort over time (Johnson and Most, 2023). Success and failure in these processes have been linked to differential life outcomes and psychiatric conditions. Here we review evidence from single-neuron recordings and neuroimaging studies that implicate the amygdala—a brain structure long associated with cue-reactivity and emotion—in decision-making and the planned pursuit of future rewards (Grabenhorst et al., 2012, 2016, 2019, 2023; Hernadi et al., 2015; Zangemeister et al., 2016). The main findings are that, in behavioral tasks in which future rewards can be pursued through planning and stepwise decision-making, amygdala neurons prospectively encode the value of anticipated rewards and related behavioral plans. Moreover, amygdala neurons predict the stepwise choices to pursue these rewards, signal progress toward goals, and distinguish internally generated (i.e., self-determined) choices from externally imposed actions. Importantly, amygdala neurons integrate the subjective value of a future reward with delay and effort costs inherent in pursuing it. This neural evidence identifies three key computations of the primate amygdala that underlie the pursuit of future rewards: (1) forming a self-determined internal goal based on subjective reward-cost valuations, (2) defining a behavioral plan for obtaining the goal, (3) executing this plan through stepwise decision-making and progress-tracking. Based on this framework, we suggest that amygdala neurons constitute vulnerabilities for dysfunction that contribute to maladaptive reward pursuit in psychiatric and behavioral conditions. Consequently, amygdala neurons may also represent potential targets for behavioral-change interventions that aim to improve individual decision-making
