    In Vitro and In Vivo Effects of Tamoxifen against Larval Stage Echinococcus granulosus

    Cystic echinococcosis is a zoonotic infection caused by the larval stage of the cestode Echinococcus granulosus. Chemotherapy currently employs benzimidazoles; however, 40% of cases do not respond favorably. With regard to these difficulties, novel therapeutic tools are needed to optimize treatment in humans. The aim of this work was to explore the in vitro and in vivo effects of tamoxifen (TAM) against E. granulosus. In addition, possible mechanisms for the susceptibility of TAM are discussed in relation to calcium homeostasis, P-glycoprotein inhibition, and antagonist effects on a putative steroid receptor. After 24 h of treatment, TAM, at a low micromolar concentration range (10 to 50 μM), inhibited the survival of E. granulosus protoscoleces and metacestodes. Moreover, we demonstrated the chemotherapeutic and chemopreventive pharmacological effects of the drug. At a dose rate of 20 mg/kg of body weight, TAM induced protection against the infection in mice. In the clinical efficacy studies, a reduction in cyst weight was observed after the administration of 20 mg/kg in mice with cysts developed during 3 or 6 months, compared to that of those collected from control mice. Since the collateral effects of high TAM doses have been largely documented in clinical trials, the use of low doses of this drug as a short-term therapy may be a novel alternative approach for human cystic echinococcosis treatment.Fil: Nicolao, María Celeste.     Epicatechin induces NF-kappa B, activator rotein-1 (AP-1) and nuclear transcription factor erythroid 2p45-related factor-2 (Nrf2) via phosphatidylinositol-3-kinase/protein kinase B (PI3K/AKT) and extracellular regulated kinase (ERK) signalling in HepG2 cells

    The dietary flavonoid epicatechin has been reported to exhibit a wide range of biological activities. The objective of the present study was to investigate the time-dependent regulation by epicatechin on the activity of the main transcription factors (NF-kappa B, activator protein-1 (AP-1) and nuclear transcription factor erythroid 2p45-related factor (Nrf2)) related to antioxidant defence and survival and proliferation pathways in HepG2 cells. Treatment of cells with 10 mu M-epicatechin induced the NF-kappa B pathway in a time-dependent manner characterised by increased levels Of I kappa B kinase (IKK) and phosphorylated inhibitor Of kappa B subunit-a (p-I kappa B alpha) and proteolytic degradation Of I kappa B, which was consistent with an up-regulation of the NF-kappa B-binding activity. Time-dependent activation of the AP-1 pathway, in concert with enhanced c-Jun nuclear levels and induction of Nrf2 translocation and phosphorylation were also demonstrated. Additionally, epicatechin-induced NF-kappa B and Nrf2 were connected to reactive oxygen species intracellular levels and to the activation of cell survival and proliferation pathways, being phosphatidylinositol-3-kinase/protein kinase B (PI3K/AKT) and extracellular regulated kinase (ERK) associated to Nrf2 modulation and ERK to NF-kappa B induction. These data suggest that the epicatechin-induced survival effect occurs by the induction of redox-sensitive transcription factors through a tight regulation of survival and proliferation pathways

    Insulin-like growth factor-I gene delivery to astrocytes reduces their inflammatory response to lipopolysaccharide

    <p>Abstract</p> <p>Background</p> <p>Insulin-like growth factor-I (IGF-I) exerts neuroprotective actions in the central nervous system that are mediated at least in part by control of activation of astrocytes. In this study we have assessed the efficacy of exogenous IGF-I and IGF-I gene therapy in reducing the inflammatory response of astrocytes from cerebral cortex.</p> <p>Methods</p> <p>An adenoviral vector harboring the rat IGF-I gene and a control adenoviral vector harboring a hybrid gene encoding the herpes simplex virus type 1 thymidine kinase fused to <it>Aequorea victoria </it>enhanced green fluorescent protein were used in this study. Primary astrocytes from mice cerebral cortex were incubated for 24 h or 72 h with vehicle, IGF-I, the IGF-I adenoviral vector, or control vector; and exposed to bacterial lipopolysaccharide to induce an inflammatory response. IGF-I levels were measured by radioimmunoassay. Levels of interleukin 6, tumor necrosis factor-α, interleukin-1β and toll-like receptor 4 mRNA were assessed by quantitative real-time polymerase chain reaction. Levels of IGF-I receptor and IGF binding proteins 2 and 3 were assessed by western blotting. The subcellular distribution of nuclear factor κB (p65) was assessed by immunocytochemistry. Statistical significance was assessed by one way analysis of variance followed by the Bonferroni pot hoc test.</p> <p>Results</p> <p>IGF-I gene therapy increased IGF-I levels without affecting IGF-I receptors or IGF binding proteins. Exogenous IGF-I, and IGF-I gene therapy, decreased expression of toll-like receptor 4 and counteracted the lipopolysaccharide-induced inflammatory response of astrocytes. In addition, IGF-I gene therapy decreased lipopolysaccharide-induced translocation of nuclear factor κB (p65) to the cell nucleus.</p> <p>Conclusion</p> <p>These findings demonstrate efficacy of exogenous IGF-I and of IGF-I gene therapy in reducing the inflammatory response of astrocytes. IGF-I gene therapy may represent a new approach to reduce inflammatory reactions in glial cells.</p

    Efficacy of an oral health promotion program for infants in the public sector

    This study compared the caries index (dmft), presence of sucking habits and facial alterations in children attended by the preventive program for infants and children assisted by spontaneous demand, at the same age, at the public sector of Maringá, PR. A total of 100 children were evaluated (1-6 years), who were divided into 2 groups: GI - children attended by the infant program with educational and preventive care and bimonthly follow-up, since the 1st year of age; GII - children assisted for conventional treatment (preventive and restorative) according to the needs, and follow-up as requested by the parents. Data collection was performed by interview with the parents and clinical examination for evaluation of the dmft and facial alterations. Analysis of data (Mann-Whitney test for dmft comparison and chi-square test for all other comparisons) demonstrated a significant difference (pO objetivo do trabalho foi comparar o índice de cárie (ceo), presença de hábitos de sucção e alterações faciais em crianças atendidas no programa de atendimento preventivo a bebês, com crianças atendidas por demanda espontânea, na mesma idade, no serviço público de Maringá-PR. Foram avaliadas 100 crianças (1-5 anos), divididas em 2 grupos: GI-crianças atendidas no programa de bebês com atendimento educativo-preventivo e retornos bimestrais, desde o 1º ano de vida; GII-crianças atendidas para tratamento convencional (preventivo e curativo) de acordo com a necessidade, sendo o retorno responsabilidade dos familiares.     Goya Lopes – Trajetória de uma Criadora

    O artigo apresenta a criação de uma vertente da moda e decoração com a finalidade de atender à demanda de um mercado que sonhava com um produto com identidade cultural vinculado a matriz africana e/ou o afro-brasileiro. Trata-se de texto biográfico ilustrado com fotos e desenhos da produção artística da autora. Foram adotadas três etapas: motivação da autora desde a descoberta quando criança de que tinha uma maneira diferente de desenhar; formação como artista plástica e designer e a estratégia de ser empresária para estar no mercado e poder desenvolver um padrão de estamparia que tivesse personalidade própria. A contribuição desse trabalho pioneiro da moda afro-brasileira abriu caminhos para a discussão de um novo contexto ainda não muito discutido no campo da moda. Abordagem da experiência da artista/designer/empresária Goya Lopes, um retrato de sua trajetória, formação e de como a motivação ao longo de sua vida a fez construir esse trabalho autoral.Palavras Chaves: Motivação; Mercado Afro-brasileiro; Estamparia

    Circadian rhythms identified in Caenorhabditis elegans by in vivo long-term monitoring of a bioluminescent reporter

    Circadian rhythms are based on endogenous clocks that allow organisms to adjust their physiology and behavior by entrainment to the solar day and, in turn, to select the optimal times for most biological variables. Diverse model systems-including mice, flies, fungi, plants, and bacteria-have provided important insights into the mechanisms of circadian rhythmicity. However, the general principles that govern the circadian clock of Caenorhabditis elegans have remained largely elusive. Here we report robust molecular circadian rhythms in C elegans recorded with a bioluminescence assay in vivo and demonstrate the main features of the circadian system of the nematode. By constructing a luciferase-based reporter coupled to the promoter of the suppressor of activated let-60 Ras (sur-5) gene, we show in both population and single-nematode assays that C elegans expresses approximately 24-h rhythms that can be entrained by light/dark and temperature cycles. We provide evidence that these rhythms are temperature-compensated and can be re-entrained after phase changes of the synchronizing agents. In addition, we demonstrate that light and temperature sensing requires the photoreceptors LITE and GUR-3, and the cyclic nucleotide-gated channel subunit TAX-2. Our results shed light on C elegans circadian biology and demonstrate evolutionarily conserved features in the circadian system of the nematode

    Piconewton Mechanical Forces Promote Neurite Growth

    Investigations over half a century have indicated that mechanical forces induce neurite growth, with neurites elongating at a rate of 0.1–0.3 μm h−1 pN−1 when mechanical force exceeds a threshold, with this being identified as 400–1000 pN for neurites of PC12 cells. In this article, we demonstrate that neurite elongation of PC12 cells proceeds at the same previously identified rate on application of mechanical tension of ∼1 pN, which is significantly lower than the force generated in vivo by axons and growth cones. This observation raises the possibility that mechanical tension may act as an endogenous signal used by neurons for promoting neurite elongation

    Toward unified molecular surveillance of RSV: A proposal for genotype definition

    Background: Human respiratory syncytial virus (RSV) is classified into antigenic subgroups A and B. Thirteen genotypes have been defined for RSV-A and 20 for RSV-B, without any consensus on genotype definition. Methods: We evaluated clustering of RSV sequences published in GenBank until February 2018 to define genotypes by using maximum likelihood and Bayesian phylogenetic analyses and average p-distances. Results: We compared the patterns of sequence clustering of complete genomes; the three surface glycoproteins genes (SH, G, and F, single and concatenated); the ectodomain and the 2nd hypervariable region of G gene. Although complete genome analysis achieved the best resolution, the F, G, and G-ectodomain phylogenies showed similar topologies with statistical support comparable to complete genome. Based on the widespread geographic representation and large number of available G-ectodomain sequences, this region was chosen as the minimum region suitable for RSV genotyping. A genotype was defined as a monophyletic cluster of sequences with high statistical support (≥80% bootstrap and ≥0.8 posterior probability), with an intragenotype p-distance ≤0.03 for both subgroups and an intergenotype p-distance ≥0.09 for RSV-A and ≥0.05 for RSV-B. In this work, the number of genotypes was reduced from 13 to three for RSV-A (GA1-GA3) and from 20 to seven for RSV-B (GB1-GB7). Within these, two additional levels of classification were defined: subgenotypes and lineages. Signature amino acid substitutions to complement this classification were also identified. Conclusions: We propose an objective protocol for RSV genotyping suitable for adoption as an international standard to support the global expansion of RSV molecular surveillance.Fil: Goya, Stephanie.
