The aim of this study was to verify if radiotherapy is able to modify the salivary PLUNC expression and if they are associated with radiotherapy side-effects. MATERIALS AND METHODS: Unstimulated whole-mouth saliva of 65 voluntaries (45 cancer patients and 20 controls) was collected. In the study group the collection was performed one week before the beginning of radiotherapy, in the middle of the treatment and one week after finishing. SPLUNC1 and SPLUNC2A expression were detected by western blotting and was analyzed with clinicopathological data and radiotherapy side-effects. RESULTS: Reduction of salivary flow rates was observed during and after conclusion of radiotherapy, being more accentuated in patients who underwent radiotherapy involving the facial region. Facial radiation field was correlated with collateral effects, mainly with the presence (p=0.0110) and severity (p=0.0143) of mucositis. SPLUNC1 and SPLUNC2A were detected in saliva of patients without treatment in variable concentrations. The study group showed levels of SPLUNC2A significantly higher than the control group, while SPLUNC1 did not show differences. Concentration of PLUNC was modified by radiotherapy, observing decreasing of glycosilated form of SPLUNC2A levels (p<.0001) and increasing of SPLUNC1 levels (p=0.0081) in the second and third collections. The only association between collateral effects of radiotherapy and PLUNC was the presence of mucositis (p=0.0363) and its severity (p=0.0500) with higher levels of SPLUNC1. CONCLUSIONS: The present study reported that levels of SPLUNC1 and glycosilated SPLUNC2A are affected by the radiotherapy, suggesting that these proteins may have importance in the oral microenvironment of irradiated head and neck cancer patientsDoutoradoEstomatologiaDoutor em Estomatopatologi

Mucoepidermoid carcinomas (MECs) are the most frequent malignant neoplasia of the salivary glands and are histologically classified as low, intermediate and high-grade. Histochemical stains such as PAS (Periodic Acid-Schiff) or mucicarmine are very useful to indentify mucine in tumors and to make the diagnosis of MEC. Recently, the expression of PLUNC family proteins has been described in salivary glands tumors, such as papillary cystoadenocarcinoma and MEC, suggesting that these proteins can be useful for the diagnosis in some difficult cases. The aim of this study was to evaluate the expression of PLUNC family proteins in 30 cases of MEC of the salivary glands and test the hypothesis that these proteins could be useful in the diagnosis of MEC. The tumors were reviewed and classified according to the grade of malignancy. Histochemical stains (PAS and mucicarmine) and immunohistochemical reactions for SPLUNC1, LPLUNC1, SPLUNC2A, SPLUNC2B and LPLUNC2 were performed. Patient's mean age was 44.07 years old. The majority of tumors (19) was histologically classified as low grade, 4 tumors were intermediate grade and 7 were high grade. All cases were positive for PAS and mucicarmine. Immunohistochemical stain was classified according to the quantity and intensity of stained cells in the tumor. Most of the MECs were positive for SPLUNC1 (90%) and LPLUNC1 (93.33%), particularly in mucous cells, mucin plugs and intermediary cells. SPLUNC2A, SPLUNC2B and LPLUNC2 did not present significant expression in the tumors. LPLUNC2 presented positivity for cells that remember mast cells in 83.33% of the tumors, expression that seems not to be previously reported in the literature. The expression pattern of SPLUNC1 and LPLUNC1 was similar to PAS and mucicarmine. The expression in intermediary cells for SPLUNC1 and LPLUNC1, suggest that these proteins could be useful in the diagnosis of high grade MECsMestradoEstomatologiaMestre em Estomatopatologi

The Giraffe Inner Bulge Survey (GIBS) II. Metallicity distributions and alpha element abundances at fixed Galactic latitude

High resolution (R\sim22,500) spectra for 400 red clump giants, in four fields within 4.8b3.4\rm -4.8^{\circ} \lesssim b \lesssim -3.4^{\circ} and 10l+10\rm -10^{\circ} \lesssim l \lesssim +10^{\circ}, were obtained within the GIRAFFE Inner Bulge Survey (GIBS) project. To this sample we added another \sim 400 stars in Baade's Window, observed with the identical instrumental configuration. We constructed the metallicity distributions for the entire sample, as well as for each field individually, in order to investigate the presence of gradients or field-to-field variations in the shape of the distributions. The metallicity distributions in the five fields are consistent with being drawn from a single parent population, indicating the absence of a gradient along the major axis of the Galactic bar. The global metallicity distribution is well fitted by two Gaussians. The metal poor component is rather broad, with a mean at =0.31\rm =-0.31 dex and σ=0.31\sigma=0.31 dex. The metal-rich one is narrower, with mean =+0.26\rm =+0.26 and σ=0.2\sigma=0.2 dex. The [Mg/Fe] ratio follows a tight trend with [Fe/H], with enhancement with respect to solar in the metal-poor regime, similar to the one observed for giant stars in the local thick disc. [Ca/Fe] abundances follow a similar trend, but with a considerably larger scatter than [Mg/Fe]. A decrease in [Mg/Fe] is observed at [Fe/H]=0.44\rm [Fe/H]=-0.44 dex. This \textit{knee} is in agreement with our previous bulge study of K-giants along the minor axis, but is 0.1 dex lower in metallicity than the one reported for the Bulge micro lensed dwarf and sub-giant stars. We found no variation in α\alpha-element abundance distributions between different fields.Comment: Accepted for publication in A&

In oral and oropharyngeal squamous cell carcinoma (OCSCC and OPSCC) exist an association between clinical and histopathological parameters with cell proliferation, basal lamina, connective tissue degradation and surrounding stroma markers. We evaluated these associations in Chilean patients. A convenience sample of 37 cases of OCSCC (n=16) and OPSCC (n=21) was analyzed clinically (TNM, clinical stage) and histologically (WHO grade of differentiation, pattern of tumor invasion). We assessed the expression of p53, Ki67, HOXA1, HOXB7, type IV collagen (ColIV) and carcinoma-associated fibroblast (a-SMA-positive cells). Additionally we conducted a univariate/bivariate analysis to assess the relationship of these variables with survival rates. Males were mostly affected (56.2% OCSCC, 76.2% OPSCC). Patients were mainly diagnosed at III/IV clinical stages (68.8% OCSCC, 90.5% OPSCC) with a predominantly infiltrative pattern invasion (62.9% OCSCC, 57.1% OPSCC). Significant association between regional lymph nodes (N) and clinical stage with OCSCC-HOXB7 expression (Chi-Square test P < 0.05) was observed. In OPSCC a statistically significant association exists between p53, Ki67 with gender (Chi-Square test P < 0.05). In OCSCC and OPSCC was statistically significant association between ki67 with HOXA1, HOXB7, and between these last two antigens (Pearson's Correlation test P < 0.05). Furthermore OPSCC-p53 showed significant correlation when it was compared with a-SMA (Kendall's Tau-c test P < 0.05). Only OCSCC-pattern invasion and OPSCC-primary tumor (T) pattern resulted associated with survival at the end of the follow up period (Chi-Square Likelihood Ratio, P < 0.05). Clinical, histological and immunohistochemical features are similar to seen in other countries. Cancer proliferation markers were associated strongly from each other. Our sample highlights prognostic value of T and pattern of invasion, but the conclusions may be limited and should be considered with caution (small sample). Many cases were diagnosed in the advanced stages of the disease, which suggests that the diagnosis of OCSCC and OPSCC is made late.7959685977Wang, Q., Gao, P., Wang, X., Duan, Y., Investigation and The GIRAFFE Inner Bulge Survey (GIBS). I. Survey Description and a kinematical map of the Milky Way bulge

The Galactic bulge is a massive, old component of the Milky Way. It is known to host a bar, and it has recently been demonstrated to have a pronounced boxy/peanut structure in its outer region. Several independent studies suggest the presence of more than one stellar populations in the bulge, with different origins and a relative fraction changing across the bulge area. This is the first of a series of papers presenting the results of the Giraffe Inner Bulge Survey, carried out at the ESO-VLT with the multifibre spectrograph FLAMES. Spectra of ~5000 red clump giants in 24 bulge fields have been obtained at resolution R=6500, in the infrared Calcium triplet wavelength region at 8500 {\AA}. They are used to derive radial velocities and metallicities, based on new calibration specifically devised for this project. Radial velocities for another ~1200 bulge red clump giants, obtained from similar archive data, have been added to the sample. Higher resolution spectra have been obtained for 450 additional stars at latitude b=-3.5, with the aim of investigating chemical abundance patterns variations with longitude, across the inner bulge. In total we present here radial velocities for 6392 RC stars. We derive a radial velocity, and velocity dispersion map of the Milky Way bulge, useful to be compared with similar maps of external bulges, and to infer the expected velocities and dispersion at any line of sight. The K-type giants kinematics is consistent with the cylindrical rotation pattern of M-giants from the BRAVA survey. Our sample enables to extend this result to latitude b=-2, closer to the Galactic plane than probed by previous surveys. Finally, we find strong evidence for a velocity dispersion peak at (0,-1) and (0,-2), possibly indicative of a high density peak in the central 250 pc of the bulgeComment: A&A in pres

Purpose: The main goal of this study was to evaluate the impact of different ionizing radiation doses on the mineral (carbonate/phosphate ratio, crystallinity index [CI]) and organic (amide III/phosphate, amide I sub-band ratios) structures, as well as the microhardness, of enamel and dentin, along with their influence on the bonding strength stability of the etch-and-rinse (ER) and self-etch (SE) dental adhesive strategies. Materials and methods: Enamel and dentin human tissue specimens were irradiated (with 0, 20, 40, and 70 Gy radiation doses, respectively) and sectioned to perform an attenuated total reflection-Fourier transform IR spectroscopy assay (ATR-FTIR) and the Vickers microhardness (VHN) test to conduct a biochemical and biomechanical evaluation of the tissues. Regarding the adhesive properties, restored enamel and dentin specimens exposed to the same radiation doses were submitted to microshear bond strength (μSBS) tests for enamel in immediate time (IM) and to microtensile bond strength (μTBS) tests after for IM and 12-month (12 M) period of time, Mann–Whitney U tests were implemented, using the ATR-FTIR data for significant differences (α \u3c 0.05), and three- and two-way analyses of variance, along with post-testing, were performed on the μTBS and μSBS data (MPa), respectively (Tukey post hoc test at α = 0.05). Results: The ATR-FTIR results showed a significant decrease (p Conclusions: It is possible to conclude that structural alterations of enamel and dentin are generated by all radiation doses, decreasing the microhardness of dental hard tissues and influencing bond strength over time, starting at 40 Gy radiation dose. The etch-and-rinse strategy demonstrates better adhesive performance but generates cohesive fractures in the enamel
