    A proposal to develop rural tourism in the municipalities of Zacatecas, Mexico: agro-cultural routes

    El trabajo evaluó el potencial de los municipios de Zacatecas para desarrollar turismo de Rutas Agro-Culturales (RAC) en base a una serie de recursos mediante el Índice del Evaluación de Rutas Agro-Culturales (IERAC); dando por resultado la Ruta del chile (R1); Ruta del maíz (R2); Ruta ganadera (R3); Ruta láctea (R4) y Ruta frutal (R5), donde los municipios con mayor potencial para éste tipo de turismo son: Zacatecas, Fresnillo, Jerez y Sombrerete mientras que en el caso contrario están de Pinos, Moyahua y Concepción del Oro. Así, ésta propuesta permite evaluar el potencial de un sitio para desarrollar turismo RAC en base a una serie de recursos y puede ser aplicado en diferentes regiones de México u otros países.The study evaluated the potential of the municipalities of Zacatecas to develop tourism Agro-Cultural Tour (RAC) based on a number of resources through the Evaluation Index Agro-Cultural Tour (IERAC) resulting in the he Pepper Route (R1); Corn Route (R2); Cattle Route (R3); Dairy Route (R4), and Fruit Route (R5);, where the municipalities with the greatest potential for this type of tourism are: Zacatecas, Fresnillo, Jerez and Sombrerete while are in the opposite case of Pinos, and Concepcion del Oro Moyahua Thus, this proposal to assess the potential of a site to develop tourism RAC based on a series of resources and can be applied in different regions of Mexico or other countries

    Influence of the academic background for the entry to a mexican public university: A case study

    Este estudio se orientó a analizar la influencia de los antecedentes académicos en los aspirantes para ingresar a la Facultad de Contaduría y Administración (FCA) de una Universidad Pública en México (UPM); tomando en cuenta las condiciones de aquellos quienes fueron aceptados y de los que no aprobaron los requisitos para ingresar a las licenciaturas que ofrece la dependencia, para determinar sí existe igualdad académica en las oportunidades de ingreso. La metodología aplicada fue un diseño mixto de triangulación, con las variantes de convergencia y transformación de datos. La población participante en esta investigación fueron 2173 aspirantes que realizaron sus trámites de ingreso a la FCA en el ciclo escolar 2011-2012, de los cuales se tomó una muestra aleatoria simple de 385 aspirantes. Los hallazgos mostraron que el tipo de bachillerato sí influyó en el promedio del examen de admisión por áreas de conocimiento de los aspirantes que ingresaron a la FCAThis study was oriented to evaluate the Academic background, influencing in the aspiring students to enter the School of Accounting and Administration (FCA) of a public university in Mexico (UPM); taking into account, the conditions of those who were accepted and those who did not pass the requirements to enter the degrees, offered by the dependency, to determine whether if there academic equality in the income oportunities. The methodology applied, was a mixed triangulation design with variants of convergence and data transformation. The participating population in this research were 2173 applicants, who made their admission process to the FCA in the 2011-2012 school year, of which a simple random sample was taken, out of 385 applicants.     Percepción de la población frente al cambio climático en áreas naturales protegidas de Baja California Sur, México

    Este trabajo muestra la percepción de la población humana asentada en las siete áreas naturales protegidas federales de Baja California Sur, desde el punto de vista del conocimiento empírico sobre los cambios en el medio ambiente y recursos naturales ante efectos del Cambio Climático (CC). Se recolectaron datos a partir de la aplicación de 250 encuestas cualitativas en 2011. Los resultados muestran que la mayor parte de la población conoce el significado de CC y que los efectos que perciben son reducción en la superficie forestal por cambio en uso de suelos, reducción de la actividad pesquera por cambios en las condiciones del mar, efectos negativos en el hato ganadero y frecuencia e intensidad de huracanes, aumento en sequías, falta de agua y reducción en la actividad ecoturística por cambios en las playas. Estudios han demostrado que la valoración subjetiva de la población humana contribuye a cambios en el comportamiento mediante una estrategia de adaptación y toma de conciencia del ciudadano.Este artigo mostra a percepção da população humana estabelecida em sete áreas protegidas federais de Baja California Sur, do ponto de vista do conhecimento empírico sobre as mudanças no meio ambiente e dos recursos naturais para efeitos das mudanças climáticas (MC) . Os dados foram coletados a partir de aplicação de 250 pesquisas qualitativas em 2011. Os resultados mostram que a maioria da população sabe o significado da MC e que os efeitos percebidos são a redução na área da floresta por mudança no uso da terra, a redução da actividade de pesca por mudanças nas condições do mar, os efeitos negativos além do rebanho e da freqüência e intensidade dos furacões, aumento das secas, falta de água e redução de ecoturismo por mudanças nas praias. Estudos têm demonstrado que a avaliação subjetiva da população humana contribui para a mudança de comportamento através de uma estratégia de adaptação e consciência cidadã.The work shows the perception of the human population settled in the seven federal natural protected areas of Baja California Sur, from the point of view of empirical knowledge about changes in the environment and natural resources to effects of climate change (CC). Data were collected from the application of 250 qualitative surveys in 2011. The results show that most of the people know the meaning of CC and that the effects are perceived reduction in forest area by land use change, reduction of fishing activity by changes in sea conditions, negative effects on the cattle herd and frequency and intensity of hurricanes, increased droughts, lack of water and reduction in ecotourism by changes in the beaches. Studies have shown that the subjective assessment of the human population contributes to behavior change through a strategy of adaptation and citizen awareness

    Guanosine inhibits hepatitis C virus replication and increases indel frequencies, associated with altered intracellular nucleotide pools

    In the course of experiments aimed at deciphering the inhibition mechanism of mycophenolic acid and ribavirin in hepatitis C virus (HCV) infection, we observed an inhibitory effect of the nucleoside guanosine (Gua). Here, we report that Gua, and not the other standard nucleosides, inhibits HCV replication in human hepatoma cells. Gua did not directly inhibit the in vitro polymerase activity of NS5B, but it modified the intracellular levels of nucleoside di- and tri-phosphates (NDPs and NTPs), leading to deficient HCV RNA replication and reduction of infectious progeny virus production. Changes in the concentrations of NTPs or NDPs modified NS5B RNA polymerase activity in vitro, in particular de novo RNA synthesis and template switching. Furthermore, the Gua-mediated changes were associated with a significant increase in the number of indels in viral RNA, which may account for the reduction of the specific infectivity of the viral progeny, suggesting the presence of defective genomes.     Amino acid substitutions associated with treatment failure of hepatitis C virus infection

    Trabajo presentado en el XVI Congreso Nacional de Virología, celebrado en Málaga (España) del 06 al 09 de septiembre de 2022.Despite the high sustained virological response rates achieved with current directly-acting antiviral agents (DAAs) against hepatitis C virus (HCV), around 2% to 5% of patients do not achieve such a response. Identification of amino acid substitutions associated with treatment failure requires analytical designs, such as subtype-specific ultra-deep sequencing (UDS) methods for HCV characterization and patient management. By deep sequencing analysis of 220 subtyped HCV samples from infected patients who failed therapy, collected from 39 Spanish hospitals, we determined amino acid sequences of the DAA-target proteins NS3, NS5A and NS5B, by UDS of HCV patient samples, in search of resistanceassociated substitutions (RAS). Using this procedure, we have identified six highly represented amino acid substitutions (HRSs) in NS5A and NS5B of HCV, which are not bona fide RAS. They were present frequently in basal and post-treatment virus of patients who failed therapy to different DAA-based therapies. Contrary to several RAS, HRSs belong to the acceptable subset of substitutions according to the PAM250 replacement matrix. Coherently, their mutant frequency, measured by the number of deep sequencing reads within the HCV quasispecies that encode the relevant substitutions, ranged between 90% and 100% in most cases. Also, they have limited predicted disruptive effects on the threedimensional structures of the proteins harboring them.     Vaccine breakthrough infections with SARS-CoV-2 Alpha mirror mutations in Delta Plus, Iota, and Omicron

    Replication of SARS-CoV-2 in the human population is defined by distributions of mutants that are present at different frequencies within the infected host and can be detected by ultra-deep sequencing techniques. In this study, we examined the SARS-CoV-2 mutant spectra of amplicons from the spike-coding (S-coding) region of 5 nasopharyngeal isolates derived from patients with vaccine breakthrough. Interestingly, all patients became infected with the Alpha variant, but amino acid substitutions that correspond to the Delta Plus, Iota, and Omicron variants were present in the mutant spectra of the resident virus.     Long runs of homozygosity are associated with Alzheimer's disease

The presence of recessive loci is suggested for Alzheimer's disease (AD); however, their search has been poorly assessed to date. To investigate homozygosity in AD, here we performed a fine-scale ROH analysis using 10 independent cohorts of European ancestry (11,919 AD cases and 9181 controls.) We detected an increase of homozygosity in AD cases compared to controls [ β (CI 95%) = 0.070 (0.037-0.104); P = 3.91 × 10 −5 ; β (CI95%) = 0.043 (0.009-0.076); P = 0.013]. ROHs increasing the risk of AD (OR > 1) were significantly overrepresented compared to ROHs increasing protection (p < 2.20 × 10 −16). A significant ROH association with AD risk was detected upstream the HS3ST1 locus (chr4:11,189,482‒11,305,456), (β (CI 95%) = 1.09 (0.48 ‒ 1.48), p value = 9.03 × 10 −4), previously related to AD. Next, to search for recessive candidate variants in ROHs, we constructed a homozygosity map of inbred AD cases extracted from an outbred population and explored ROH regions in whole-exome sequencing data (N = 1449). We detected a candidate marker, rs117458494, mapped in the SPON1 locus, which has been previously associated with amyloid metabolism. Here, we provide a research framework to look for recessive variants in AD using outbred populations. Our results showed that AD cases have enriched homozygosity, suggesting that recessive effects may explain a proportion of AD heritability

    SARS-CoV-2 mutant spectra reveal differences between COVID-19 severity categories

    Trabajo presentado en el XVI Congreso Nacional de Virología, celebrado en Málaga (España) del 06 al 09 de septiembre de 2022.RNA virus populations are composed of complex mixtures of genomes that are termed mutant spectra. SARS-CoV-2 replicates as a viral quasispecies, and mutations that are detected at low frequencies in a host can be dominant in subsequent variants. We have studied mutant spectrum complexities of SARS-CoV-2 populations derived from thirty nasopharyngeal swabs of patients infected during the first wave (April 2020) in the Hospital Universitario Fundación Jiménez Díaz. The patients were classified according to the COVID-19 severity in mild (non-hospitalized), moderate (hospitalized) and exitus (hospitalized with ICU admission and who passed away due to COVID-19). Using ultra-deep sequencing technologies (MiSeq, Illumina), we have examined four amplicons of the nsp12 (polymerase)-coding region and two amplicons of the spike-coding region. Ultra-deep sequencing data were analyzed with different cut-off frequency for mutation detection. Average number of different point mutations, mutations per haplotype and several diversity indices were significantly higher in SARS-CoV-2 isolated from patients who developed mild disease. A feature that we noted in the SARS-CoV-2 mutant spectra from diagnostic samples is the remarkable absence of mutations at intermediate frequencies, and an overwhelming abundance of mutations at frequencies lower than 10%. Thus, the decrease of the cut-off frequency for mutation detection from 0.5% to 0.1% revealed an increasement (50- to 100 fold) in the number of different mutations. The significantly higher frequency of mutations in virus from patients displaying mild than moderate or severe disease was maintained with the 0.1% cut- off frequency. To evaluate whether the frequency repertoire of amino acid substitutions differed between SARS-CoV-2 and the well characterized hepatitis C virus (HCV), we performed a comparative study of mutant spectra from infected patients using the same bioinformatics pipelines. HCV did not show the deficit of intermediate frequency substitutions that was observed with SARS-CoV-2. This difference was maintained when two functionally equivalent proteins, the corresponding viral polymerases, were compared. In conclusion, SARS-CoV-2 mutant spectra are rich reservoirs of mutants, whose complexity is not uniform among clinical isolates.     Effectiveness of a strategy that uses educational games to implement clinical practice guidelines among Spanish residents of family and community medicine (e-EDUCAGUIA project):A clinical trial by clusters

    This study was funded by the Fondo de Investigaciones Sanitarias FIS Grant Number PI11/0477 ISCIII.-REDISSEC Proyecto RD12/0001/0012 AND FEDER Funding.Background: Clinical practice guidelines (CPGs) have been developed with the aim of helping health professionals, patients, and caregivers make decisions about their health care, using the best available evidence. In many cases, incorporation of these recommendations into clinical practice also implies a need for changes in routine clinical practice. Using educational games as a strategy for implementing recommendations among health professionals has been demonstrated to be effective in some studies; however, evidence is still scarce. The primary objective of this study is to assess the effectiveness of a teaching strategy for the implementation of CPGs using educational games (e-learning EDUCAGUIA) to improve knowledge and skills related to clinical decision-making by residents in family medicine. The primary objective will be evaluated at 1 and 6months after the intervention. The secondary objectives are to identify barriers and facilitators for the use of guidelines by residents of family medicine and to describe the educational strategies used by Spanish teaching units of family and community medicine to encourage implementation of CPGs. Methods/design: We propose a multicenter clinical trial with randomized allocation by clusters of family and community medicine teaching units in Spain. The sample size will be 394 residents (197 in each group), with the teaching units as the randomization unit and the residents comprising the analysis unit. For the intervention, both groups will receive an initial 1-h session on clinical practice guideline use and the usual dissemination strategy by e-mail. The intervention group (e-learning EDUCAGUIA) strategy will consist of educational games with hypothetical clinical scenarios in a virtual environment. The primary outcome will be the score obtained by the residents on evaluation questionnaires for each clinical practice guideline. Other included variables will be the sociodemographic and training variables of the residents and the teaching unit characteristics. The statistical analysis will consist of a descriptive analysis of variables and a baseline comparison of both groups. For the primary outcome analysis, an average score comparison of hypothetical scenario questionnaires between the EDUCAGUIA intervention group and the control group will be performed at 1 and 6months post-intervention, using 95% confidence intervals. A linear multilevel regression will be used to adjust the model. Discussion: The identification of effective teaching strategies will facilitate the incorporation of available knowledge into clinical practice that could eventually improve patient outcomes. The inclusion of information technologies as teaching tools permits greater learning autonomy and allows deeper instructor participation in the monitoring and supervision of residents. The long-term impact of this strategy is unknown; however, because it is aimed at professionals undergoing training and it addresses prevalent health problems, a small effect can be of great relevance.
