Methyl Jasmonate Affects Photosynthesis Efficiency, Expression of HvTIP Genes and Nitrogen Homeostasis in Barley
Jasmonates modulate many growth and developmental processes and act as stress hormones
that play an important role in plant tolerance to biotic and abiotic stresses. Therefore, there is a
need to identify the genes that are regulated through the jasmonate signalling pathway. Aquaporins,
and among them the Tonoplast Intrinsic Proteins (TIPs), form the channels in cell membranes that
are responsible for the precise regulation of the movement of water and other substrates between
cell compartments. We identified the cis-regulatory motifs for the methyl jasmonate (MeJA)-induced
genes in the promoter regions of all the HvTIP genes, which are active in barley seedlings, and thus we
hypothesised that the HvTIP expression could be a response to jasmonate signalling. In the presented
study, we determined the e ect of methyl jasmonate on the growth parameters and photosynthesis
e ciency of barley seedlings that had been exposed to di erent doses of MeJA (15–1000 M 120 h)
in a hydroponic solution. All of the applied MeJA concentrations caused a significant reduction of
barley seedling growth, which was most evident in the length of the first leaf sheath and dry leaf
weight. The observed decrease of the PSII parameters after the exposure to high doses of MeJA
(500 M or higher) was associated with the downregulation of HvPsbR gene encoding one of the
extrinsic proteins of the Oxygen Evolving Complex. The reduced expression of HvPsbR might lead
to the impairment of the OEC action, manifested by the occurrence of the K-band in an analysis of
fluorescence kinetics after MeJA treatment as well as reduced photosynthesis e ciency. Furthermore,
methyl jasmonate treatment caused a decrease in the nitrogen content in barley leaves, which was
associated with an increased expression the four tonoplast aquaporin genes (HvTIP1;2, HvTIP2;2,
HvTIP4;1 and HvTIP4;2) predicted to transport the nitrogen compounds from the vacuole to the
cytosol. The upregulation of the nitrogen-transporting HvTIPs might suggest their involvement in
the vacuolar unloading of ammonia and urea, which both could be remobilised when the nitrogen
content in the leaves decreases. Our research provides tips on physiological role of the individual TIP
subfamily members of aquaporins under methyl jasmonate action

Aberrant Orbitofrontal Cortex Reactivity to Erotic Cues in Compulsive Sexual Behavior Disorder
BACKGROUND AND AIMS: Compulsive Sexual Behavior Disorder (CSBD) is characterized by increased reactivity to erotic reward cues. Cue-encoded reward parameters, such as type (e.g. erotic or monetary) or probability of anticipated reward, shape reward-related motivational processes, increase the attractiveness of cues and therefore might enhance maladaptive behavioral patterns in CSBD. Studies on the neural patterns of cue processing in individuals with CSBD have been limited mainly to ventral striatal responses. Therefore, here we aimed to examine the cue reactivity of multiple key structures in the brains reward system, taking into account not only the type of predicted reward but also its probability. METHODS: Twenty Nine men seeking professional help due to CSBD and 24 healthy volunteers took part in an fMRI study with a modified Incentive Delay Task with erotic and monetary rewards preceded by cues indicating a 25%, 50%, or 75% chance of reward. Analyses of functional patterns of activity related to cue type and probability were conducted on the whole-brain and ROI levels. RESULTS: Increased anticipatory response to cues predictive of erotic rewards was observed among CSBD participants when compared to controls, in the ventral striatum and anterior orbitofrontal cortex (aOFC). The activity in aOFC was modulated by reward probability. DISCUSSION AND CONCLUSIONS: Type of anticipated reward (erotic vs monetary) affects reward-related behavioral motivation in CSBD more strongly than reward probability. We present evidence of abnormal aOFC function in CSBD by demonstrating the recruitment of additional subsections of this region by erotic reward cues
The assessment of postpartum mood disorders and its risk factors
W pracy wykorzystano skalę depresji Becka, Edynburską Skalę Depresji Poporodowej (EPDS) i kwestionariusz własny. Uzyskano 41 odpowiedzi zwrotnych. Badanie uzyskało zgodę Lokalnej Komisji Bioetycznej.
Wyniki: Wśród badanych pacjentek będących w 1.−7. dniu po porodzie: 11,96% uzyskało wynik świadczący o łagodnej depresji, a 0,54% o umiarkowanej według skali Becka.
W pracy nie wykazano istotnej statystycznie zależności pomiędzy uzyskanymi wynikami w skali Becka a badanymi czynnikami ocenianymi miesiąc od porodu, analizowana grupa była istotnie mniejsza (n = 41).Introduction: The aim of our study was to estimate the prevalence of mood disorders including postpartum depression and to analyze its potential risk factors.
Material and methods: The patients staying in maternity wards at the Gynecological Clinical Hospital of PUMS were examined during their postpartum period. Depressive symptoms were measured using the Edinburgh Postnatal Depression Scale (EPDS), Beck Depression Inventory (BDI) and our own questionnaire. The anonymous questionnaire and BDI were filled by the patients between 1st and 7th day after the delivery (n = 184), then they were asked to fill the BDI and EPDS after 30 days of the childbed (n = 41 patients refilled the survey). The research has obtained permission of the Local Bioethical Commission.
Results: During the first week of the childbed, 11.96% of the patients achieved score, which might imply they suffer from mild depression and 0.54% from major depression according to BDI. After the second examination the score increased to 19.51% and 2.44%, respectively.
Conclusions: The unplanned pregnancy, health issues during the gestation, hospitalization due to pregnancy complications, no stressful factors in the past like: history of miscarriage, stillbirth or death of the child, episodes of depression in the past, suicidal ideation in the history can be bracketing the risk factors of mood disorders after 1 to 7 days of the childbed. The significant correlation between score of the BDI and the answers in the questionnaire wasn’t demonstrated after 30 days of the postpartum period, however the analyzed group was significantly lower (n = 41)

An Empirical Study of Affective and Cognitive Functions in Compulsive Sexual Behavior Disorder
BACKGROUND AND AIMS: Despite the inclusion of the Compulsive Sexual Behavior Disorder (CSBD) in the International Classification of Diseases, very little is known about the underlying affective and cognitive processes. To fill this gap, we compared CSBD subjects and Healthy-Controls (HC) across negative/positive valence, cognitive and sensorimotor systems, as proposed by the Research Domain Criteria framework. METHODS: 74 heterosexual CSBD and 66 matched HC males were studied with 10 questionnaires and 8 behavioral tasks. Analyses were conducted with frequent and Bayesian statistics. RESULTS: CSBD individuals showed significantly higher (than HC) punishment sensitivity, anxiety, depression, compulsivity, and impulsivity symptoms. Frequentist statistical analysis revealed significant interaction between subject group and condition in Incentive Delay Task, concerning the strength of motivation and hedonic value of erotic rewards. Bayesian analysis produced evidence for the absence of group differences in Facial Discrimination Task, Risk-Ambiguity Task, and Learning Task. Also, Bayesian methods provided evidence for group differences in the Emotional Stroop Task and the Incentive Delay Task. Sexual Discounting Task, Attentional Network Task, and Stop Signal Task produced mixed results. CONCLUSIONS: Higher punishment sensitivity and impulsivity among CSBD subjects, along with significant interaction between these groups and erotic vs. non-erotic reward processing is in line with previous findings on negative/positive valence alterations in CSBD patients. This result shows that there are similarities to substance and behavioral addictions. The absence of group differences and mixed results related to cognitive and sensorimotor systems raise concerns to what extent CSBD resembles a wide spectrum of impairments observed in disorders, and demand further research
HorTILLUS - a rich and renewable source of induced mutations for forward/reverse genetics and pre-breeding programs in barley (Hordeum vulgare L.)
TILLING (Targeting Induced Local Lesions IN Genomes) is a strategy used for functional
analysis of genes that combines the classical mutagenesis and a rapid, high-throughput
identification of mutations within a gene of interest. TILLING has been initially developed
as a discovery platform for functional genomics, but soon it has become a valuable tool
in development of desired alleles for crop breeding, alternative to transgenic approach.
Here we present the HorTILLUS (Hordeum—TILLING—University of Silesia) population
created for spring barley cultivar “Sebastian” after double-treatment of seeds with
two chemical mutagens: sodium azide (NaN3) and N-methyl-N-nitrosourea (MNU). The
population comprises more than 9,600 M2 plants from which DNA was isolated, seeds
harvested, vacuum-packed, and deposited in seed bank. M3 progeny of 3,481 M2
individuals was grown in the field and phenotyped. The screening for mutations was
performed for 32 genes related to different aspects of plant growth and development.
For each gene fragment, 3,072–6,912 M2 plants were used for mutation identification
using LI-COR sequencer. In total, 382 mutations were found in 182.2Mb screened.
The average mutation density in the HorTILLUS, estimated as 1 mutation per 477 kb, is
among the highest mutation densities reported for barley. The majority of mutations were
G/C to A/T transitions, however about 8% transversions were also detected. Sixty-one
percent of mutations found in coding regions were missense, 37.5% silent and 1.1%
nonsense. In each gene, the missense mutations with a potential effect on protein
function were identified. The HorTILLUS platformis the largest of the TILLING populations
reported for barley and best characterized. The population proved to be a useful tool, both in functional genomic studies and in forward selection of barley mutants with
required phenotypic changes. We are constantly renewing the HorTILLUS population,
which makes it a permanent source of new mutations. We offer the usage of this valuable
resource to the interested barley researchers on cooperative basis

Comparative analysis of frameworks using TypeScript to build server applications
The subject of the research was a comparative analysis of programming frameworks that are intended for building applications. NestJS (version 8.1.1), FoalTS (version 2.5.0) and Ts.ED (version 6.69.1) were put together. An experiment was prepared based on scenarios that focused on the response time of applications to requests. Each of them had the same set of functionalities. NestJS turned out to be the most efficient of the compared skeletons. It achieved the best results. The worst results in each scenario were achieved by Ts.ED, especially with higher loads. The biggest differences in the comparison can be seen in studies conducted according to a scenario based on GET-type requests, and in particular with more objects in response
