    Laboratory diagnosis and monitoring of desmopressin treatment of von Willebrand's disease by flow cytometry.

    Background and Objectives von Willebrand's disease (VWD) is a heterogeneous bleeding disorder caused by quantitative or qualitative defects in von Willebrand factor (VWF). The diagnosis of VWD requires several laboratory tests. The aim of our study was to validate a flow cytometric test for the diagnosis of VWD and for monitoring the effects of desmopressin therapy.Design and Methods Flow cytometric analysis of ristocetin-induced VWF binding to platelets was performed in platelet-rich plasma (PRP) samples from patients with VWD and from control subjects and in samples of formalin-fixed platelets in the presence of plasma from patients or controls. In 12 VWD patients the test was conducted before and 1 hour after desmopressin infusion. Results were compared with VWF:Ag, VWF:RCo, VWF:CB, RIPA, PFA-100® and the skin bleeding time.Results Ristocetin-induced VWF binding to platelets, evaluated by both flow cytometry-based assays, was significantly reduced in patients with type1, 2A and 2M VWD as compared with that in healthy subjects. Patients with type 2B VWD showed reduced binding of VWF to formalin-fixed platelets, but increased binding to autologous platelets in PRP, similar to RIPA. VWF binding to platelets assessed by both flow cytometric assays correlated significantly with VWF:Ag, VWF:RCo, VWF:CB, RIPA, PFA100® and bleeding time. VWF binding to platelets increased after desmopressin infusion.Interpretation and Conclusions The measurement of ristocetin-induced binding of VWF to platelets by flow cytometry is a sensitive, simple and rapid test for the diagnosis of VWD and for the monitoring of the effects of desmopressin therapy. The flow cytometric assay performed with autologous platelets is useful in the identification of type 2B VWD patients

    Analysis of changes in large-scale circulation patterns driving extreme precipitation events over the central-eastern China

    To an extent, large-scale circulation situations and moisture transport are responsible for extreme precipitation occurrence. The aim of our study is to investigate the possible modifications of circulation patterns (CPs) in driving extreme precipitation over the central-eastern China (CEC). The self-organizing map (SOM) and event synchronization methods are used to link the extreme precipitation events with CPs. Results show that 23% of rain gauges have a significant change point (at the 90% confidence level) in annual extreme precipitation from 1960 to 2015. Based on the identified change points, we classified the data into two periods, that is, 1960–1989 and 1990–2015. Overall, CPs characterized by obvious positive anomalies of 500 hPa geopotential height over the Eastern Eurasia continent and negative values over the surrounding oceans are highly synchronized with extreme precipitation events. During 1990–2015, the predominant CPs are more related to the extreme precipitation with enhanced event synchronization. We found that the CP changes produce an increase in extreme precipitation frequency from 1960–1989 to 1990–2015

    Impact Response of Nanofluid-Reinforced Antiballistic Kevlar Fabrics

    In this chapter the possibility to increase the resistance upon ballistic impact of typical Kevlar-based materials by means of silica nanofluids is investigated. Nanofluids are commonly known to have non-newtonian behavior, also indicated as shear thickening fluid (STF) property. STFs are very deformable materials in ordinary conditions (flowing like a liquid as long as no force is applied), but they turn into a very rigid solid-like material at high shear rates. The nanofluid optimization, mainly regarding its chemical and thermal stability, as well as the STF/fabric chemical coupling are crucial issues to tackle, in order to suitably exploit the STF property, i.e. to increase the fabric resistance upon impact. The present work summarizes some experimental results obtained on this subject. In particular, different kind of nanofluids realized by including several percentages of silica nanoparticles within polyethylene glycol are analyzed in terms of their morphological and rheological properties. Later on, the treatment with nanofluids of different typologies of Kevlar materials and the test panels manufacturing is described. Finally, the results of the ballistic characterization of the panels is presented and widely discussed. This latter is performed by means of an in-house built electromagnetic Coil Gun, able to explore a relatively large range of bullet velocity (up to 90m/s) with excellent precision and high degree of test reproducibility. The preliminary results obtained in such energy range show that STF impregnated fabrics have better penetration resistance compared to neat Kevlar, without affecting the fabric flexibility and, mainly, without overweighting the whole panel. That indicates that the addition of STFs to conventional ballistic fabrics enhance the material performances, thus suggesting further investigations in order to make such nanoreinforced materials effective for ballistic application

    Nuevos registros del raro Histiotus magellanicus (Chiroptera, Vespertilionidae) y otros murciélagos de Patagonia Central, Argentina

    Histiotus magellanicus (Philippi, 1866) is a poorly known Patagonian bat. Here we report on nine newly captured specimens and one unpublished locality from museum specimen that significantly improve our knowledge of this species' distribution, by incorporating two Argentine provinces and reducing the distributional gap between northern and southern historical records. The new records confirm H. magellanicus as a Subantarctic Forests endemic. In addition, we report new specimens and localities from four poorly known Patagonian bats for Chubut province: Histiotus macrotus, Myotis chiloensis, Lasiurus varius (Vespertilionidae) and Tadarida brasiliensis (Molossidae).Histiotus magellanicus (Philippi, 1866) es un murciélago patagónico muy pobremente conocido. Aquí reportamos nueve especímenes recientemente capturados y un espécimen de colección cuya localidad no había sido reportada hasta el momento. Las localidades aportadas mejoran considerablemente nuestro conocimiento de su distribución, incorporando a ella dos nuevas provincias argentinas y reduciendo considerablemente el vacío de datos entre los registros históricos del norte y sur de Patagonia. Los nuevos registros confirman a H. magellanicus como especie endémica de la ecorregión del Bosque Subantártico.
