    Peer Rejection and Friendships in Children with Attention-Deficit/Hyperactivity Disorder: Contributions to Long-Term Outcomes

    Even after evidence-based treatment, Attention- Deficit/Hyperactivity Disorder (ADHD) is associated with poor long-term outcomes. These outcomes may be partly explained by difficulties in peer functioning, which are common among children with ADHD and which do not respond optimally to standard ADHD treatments. We examined whether peer rejection and lack of dyadic friendships experienced by children with ADHD after treatment contribute to long-term emotional and behavioral problems and global impairment, and whether having a reciprocal friend buffers the negative effects of peer rejection. Children with Combined type ADHD (N0300) enrolled in the Multimodal Treatment Study of Children with ADHD (MTA) were followed for 8 years. Peer rejection and dyadic friendships were measured with sociometric assessments after the active treatment period (14 or 24 months after baseline; M ages 9.7 and 10.5 years, respectively). Outcomes included delinquency, depression, anxiety, substance use, and general impairment at 6 and 8 years after baseline (Mean ages 14.9 and 16.8 years, respectively). With inclusion of key covariates, including demographics, symptoms ofADHD, ODD, and CD, and level of the outcome variable at 24 months, peer rejection predicted cigarette smoking, delinquency, anxiety, and global impairment at 6 years and global impairment at 8 years after baseline. Having a reciprocal friend was not, however, uniquely predictive of any outcomes and did not reduce the negative effects of peer rejection. Evaluating and addressing peer rejection in treatment planning may be necessary to improve long-term outcomes in children with ADHD

    Competition between benthic cyanobacteria and diatoms as influenced by different grain sizes and temperatures

    An experimental laboratory set-up was used to study the influence of different grain size compositions and temperatures on the growth of benthic cyanobacteria and diatoms, and on the competition between these 2 groups. Monospecific cultures of 3 species of cyanobacteria (Merismopedia punctata, Microcoleus chthonoplastes, Oscillatoria limosa), and of 2 species of benthic diatoms (Phaeodactylum tricornutum and Nitzschia sp.) were used. The organisms were cultured in 100 ml flasks filled with medium and 3 different kinds of sediment: (1) Sand (fine sand, 63 to 200 µm), (2) Mud-I (mixed fine sand and mud <63 µm in the ratio 80:20 wt %), (3) Mud-II (mixed fine sand and mud in the ratio 50:50 wt %). Experimental temperatures were 10, 15 and 25°C. At 10°C and 15°C, both diatom species achieved the highest biomass on the sediments of the finest grain size (50 wt % < 63 µm) while cyanobacteria achieved low biomass levels. Coarsening of sediments at the same temperature levels revealed a gradually lower biomass of the diatoms. Particularly on sand, the diatoms never reached the same concentrations of chlorophyll a as on mud. The cyanobacteria, on the other hand, had the highest biomass on sand at 15°C. In the competition experiments the benthic diatom species Nitzschia sp. dominated all types of sediments at 10°C and 15°C. The experiments at 25°C were dominated by the filamentous cyanobacterium M. chthonoplastes. This indicates the importance of abiotic conditions for the distribution and abundance of benthic phototrophic micro-organisms

    Characteristics of drug-resistant tuberculosis in Abkhazia (Georgia), a high-prevalence area in Eastern Europe

    Although multidrug-resistant (MDR) tuberculosis (TB) is a major public health problem in Eastern Europe, the factors contributing to emergence, spread and containment of MDR-TB are not well defined. Here, we analysed the characteristics of drug-resistant TB in a cross-sectional study in Abkhazia (Georgia) between 2003 and 2005, where standard short-course chemotherapy is supplemented with individualized drug-resistance therapy. Drug susceptibility testing (DST) and molecular typing were carried out for Mycobacterium tuberculosis complex strains from consecutive smear-positive TB patients. Out of 366 patients, 60.4% were resistant to any first-line drugs and 21% had MDR-TB. Overall, 25% of all strains belong to the Beijing genotype, which was found to be strongly associated with the risk of MDR-TB (OR 25.9, 95% CI 10.2-66.0) and transmission (OR 2.8, 95% CI 1.6-5.0). One dominant MDR Beijing clone represents 23% of all MDR-TB cases. The level of MDR-TB did not decline during the study period, coinciding with increasing levels of MDR Beijing strains among previously treated cases. Standard chemotherapy plus individualized drug-resistance therapy, guided by conventional DST, might be not sufficient to control MDR-TB in Eastern Europe in light of the spread of "highly transmissible" MDR Beijing strains circulating in the community

    Dynamic changes in the higher-level chromatin organization of specific sequences revealed by in situ hybridization to nuclear halos

    A novel approach to study the higher level packaging of specific DNA sequences has been developed by coupling high-resolution fluorescence hybridization with biochemical fractionation to remove histones and distend DNA loops to form morphologically reproducible nuclear halos. Results demonstrate consistent differences in the organization of specific sequences, and further suggest a relationship to functional activity. Pulse-incorporated bromodeoxyuridine representing nascent replicating DNA localized with the base of the chromatin loops in discrete clustered patterns characteristic of intact cells, whereas at increasing chase times, the replicated DNA was consistently found further out on the extended region of the halo. Fluorescence hybridization to unique loci for four transcriptionally inactive sequences produced long strings of signal extending out onto the DNA halo or loop, whereas four transcriptionally active sequences remained tightly condensed as single spots within the residual nucleus. In contrast, in non-extracted cells, all sequences studied typically remained condensed as single spots of fluorescence signal. Interestingly, two transcriptionally active, tandemly repeated gene clusters exhibited strikingly different packaging by this assay. Analysis of specific genes in single cells during the cell cycle revealed changes in packaging between S-phase and non S-phase cells, and further suggested a dramatic difference in the structural associations in mitotic and interphase chromatin. These results are consistent with and suggestive of a loop domain organization of chromatin packaging involving both stable and transient structural associations, and provide precedent for an approach whereby different biochemical fractionation methods may be used to unravel various aspects of the complex higher-level organization of the genome

    Treatment of tuberculosis in a region with high drug resistance: Outcomes, drug resistance amplification and re-infection

    Introduction: Emerging antituberculosis drug resistance is a serious threat for tuberculosis (TB) control, especially in Eastern European countries. Methods: We combined drug susceptibility results and molecular strain typing data with treatment outcome reports to assess the influence of drug resistance on TB treatment outcomes in a prospective cohort of patients from Abkhazia (Georgia). Patients received individualized treatment regimens based on drug susceptibility testing (DST) results. Definitions for antituberculosis drug resistance and treatment outcomes were in line with current WHO recommendations. First and second line DST, and molecular typing were performed in a supranational laboratory for Mycobacterium tuberculosis (MTB) strains from consecutive sputum smear-positive TB patients at baseline and during treatment. Results: At baseline, MTB strains were fully drug-susceptible in 189/326 (58.0%) of patients. Resistance to at least H or R (PDR-TB) and multidrug-resistance (MDR-TB) were found in 69/326 (21.2%) and 68/326 (20.9%) of strains, respectively. Three MDR-TB strains were also extensively resistant (XDR-TB). During treatment, 3/189 (1.6%) fully susceptible patients at baseline were re-infected with a MDR-TB strain and 2/58 (3.4%) PDR-TB patients became MDR-TB due to resistance amplification. 5/ 47 (10.6%) MDR- patients became XDR-TB during treatment. Treatment success was observed in 161/189 (85.2%), 54/69 (78.3%) and 22/68 (32.3%) of patients with fully drug susceptible, PDR- and MDR-TB, respectively. Development of ofloxacin resistance was significantly associated with a negative treatment outcome. Conclusion: In Abkhazia, a region with high prevalence of drug resistant TB, the use of individualized MDR-TB treatment regimens resulted in poor treatment outcomes and XDR-TB amplification. Nosocomial transmission of MDR-TB emphasizes the importance of infection control in hospitals

    Paleotethys was a highly mobile oceanic realm pinching into the supercontinent Pangea between Gondwana and Eurasia in the late Paleozoic/early Mesozoic. Published Paleotethyan reconstructions reveal that the time of Paleotethys closure and the position of its suture are highly debated. We present new magmatic and detrital zircon ages, separated from pre-Alpine basement and Permian to Triassic cover rocks exposed in the External Hellenides of Crete. These age data reveal Variscan and Cimmerian docking of microplates along the southern margin of Laurasia and help to constrain the time of Paleotethys closure.46% of detrital zircons from quartzite in the Variscan basement, are Pennsylvanian with concordant ages at 321. ±. 2. Ma, 310. ±. 3. Ma, and 300. ±. 3. Ma. The basement is unconformably overlain by arc-related volcanics of the Tyros Unit, magmatic zircons of which yielded a concordant U-Pb zircon age at 285. ±. 2. Ma. Thus, the metasediments of the basement, interpreted as former trench sediments, were deposited, metamorphosed and exhumed in latest Carboniferous to early Permian times (302-283. Ma). Magmatic activity during this late Variscan phase is also indicated by igneous boulders within Olenekian (meta)conglomerates of the Tyros Unit, which yielded concordant U-Pb zircon ages at 291. ±. 2 and 310. ±. 2. Ma. The late Variscan orogenic phase is attributed to the collision of the Gondwana-derived southern Minoan terrane (SMT) with Laurasia subsequent to northward subduction of Paleotethys lithosphere and Viséan collision of the northern Minoan terrane (NMT).Magmatic activity ceased during the late Permian, but revived in the Lower Triassic as is indicated by felsic volcanics (249. ±. 2. Ma, concordant U-Pb zircon) and by detrital zircons (242. ±. 3, 240. ±. 5. Ma, 237. ±. 3. Ma concordant U-Pb zircon) of the Tyros Unit. At the same time the Variscan chain was exhumed and removed as is shown by the detritus in the Lower to Middle Triassic Tyros sediments, which includes high-grade metamorphic rocks and detrital zircons with U-Pb ages ranging from 280 to 335. Ma.A significant change in the detrital components occurred in the Ladinian when the Variscan basement with its Permo-Triassic cover was thrust on top of clastic sediments, today represented by the Phyllite-Quartzite Unit s.str. The Phyllite-Quartzite Unit s.str. shows Cadomian and older - but no Variscan - detritus because of its position along the northern margin of the Cimmerian ribbon continent. Thus, in the eastern Mediterranean, Paleotethys was closed during the Ladinian and the related suture in the External Hellenides is situated between the Variscan basement (active margin in the north) and the Phyllite-Quartzite Unit s.str (passive margin in the south). Carnian crustal extension led to subsidence of the Variscan/Cimmerian chain, most parts of which merged below sea level. This is the reason why 90% of the detritus of the Carnian Tyros Beds are not related to the Variscan, but to the Cadomian and Grenvillian basement of the E-Gondwana derived Cimmerian ribbon continent

    Cut-free Calculi and Relational Semantics for Temporal STIT Logics

    We present cut-free labelled sequent calculi for a central formalism in logics of agency: STIT logics with temporal operators. These include sequent systems for Ldm , Tstit and Xstit. All calculi presented possess essential structural properties such as contraction- and cut-admissibility. The labelled calculi G3Ldm and G3Tstit are shown sound and complete relative to irreflexive temporal frames. Additionally, we extend current results by showing that also Xstit can be characterized through relational frames, omitting the use of BT+AC frames

    Activation of myosin V–based motility and F-actin–dependent network formation of endoplasmic reticulum during mitosis

    It is widely believed that microtubule- and F-actin–based transport of cytoplasmic organelles and membrane fusion is down-regulated during mitosis. Here we show that during the transition of Xenopus egg extracts from interphase to metaphase myosin V–driven movement of small globular vesicles along F-actin is strongly inhibited. In contrast, the movement of ER and ER network formation on F-actin is up-regulated in metaphase extracts. Our data demonstrate that myosin V–driven motility of distinct organelles is differently controlled during the cell cycle and suggest an active role of F-actin in partitioning, positioning, and membrane fusion of the ER during cell division

    Origin and provenance of igneous clasts from late Palaeozoic conglomerate formations (Del Ratón and El Planchón) in the Andean Precordillera of San Juan, Argentina

    Late Palaeozoic conglomerate formations (Del Ratón and El Planchón) from the Andean Precordillera (Argentina) were studied to unravel their age, composition and provenance. The conglomerates from the Del Ratón Formation are formed by igneous clasts of acid, intermediate and basic compositions (volcanic and plutonic). Laser Ablation (ICP-MS) zircon U-Pb study has yielded an age of 348±2 Ma (late Tournaisian) for the crystallization of a granitic clast, interpreted as a maximum deposition age for the Del Ratón Formation. Geochemistry of these clasts (high LILE/HFSE and La/Yb ratios, negative Nb-Ta anomalies) suggests a calc-alkaline batholithic source, probably located along the Andean Frontal Cordillera currently to the west, where similar calc-alkaline igneous rocks have been described. The El Planchón Formation overlies the Del Ratón Formation and, in the studied conglomerates, there are only igneous clasts of mafic composition (volcanic/subvolcanic). These mafic clasts have a very similar petrography and geochemistry to the Late Ordovician mafic igneous rocks of the Western Precordillera (low LILE/HFSE and La/Yb ratios, no negative Nb-Ta anomalies). Therefore we suggest that the El Planchón conglomerate clasts were probably delivered mainly from northern sources within the Precordillera terrane. This change in clast provenance is tentatively related to a shift in mountain uplift from the Frontal Cordillera (in the west) to the Precordillera (in the east) after the early Visean.Las formaciones conglomeráticas del Paleozoico superior (Del Ratón y El Planchón) de la Precordillera Andina (Argentina) fueron es­tudiadas con el fin de determinar su edad, composición y procedencia. Los conglomerados de la Formación Del Ratón están constituidos por clastos de rocas ígneas (volcánicas y plutónicas) ácidas, intermedias y básicas. Un estudio en circones con espectrometría de masas con plasma acoplado por inducción con ablación láser (LA-ICP-MS) proporciona una edad de 348±2 Ma (Tournaisiense superior) para la cristalización de un clasto granítico, que interpretamos como la máxima edad del depósito de la Formación Del Ratón. La geoquímica de los clastos (altas relaciones LILE/HFSE y La/Yb, anomalías negativas de Nb-Ta) sugiere que probablemente derivan de batolitos calcoal­calinos descritos en algunos sectores de la Cordillera Frontal Andina localizada actualmente al oeste. La Formación El Planchón se super­pone a la Formación Del Ratón y los clastos estudiados en los conglomerados de esta formación son únicamente de rocas ígneas básicas (subvolcánicas/volcánicas). Estos clastos tienen una petrografía y geoquímica similar a las rocas ígneas máficas del Ordovícico Superior de la Precordillera Occidental (bajas relaciones LILE/HFSE y La/Yb, ausencia de anomalías negativas en Nb-Ta). Por ello nosotros suge­rimos que los clastos de los conglomerados de la Formación El Planchón proceden de materiales localizados al norte dentro de la propia Precordillera. Este cambio en la procedencia de los clastos es tentativamente relacionado con una transferencia de la deformación desde la Cordillera Frontal (al oeste) a la Precordillera (al este), provocando el levantamiento de la Precordillera a partir del Viseense inferior
