    Stretch Intensity vs. Inflammation: A Dose-dependent Association?

    The intensity of stretching is rarely reported in scientific literature. In this study, we examined the effects of stretching intensities at 30%, 60%, and 90% of maximum range of movement (mROM) on the inflammatory response of the right hamstring muscle. Methods: A randomised within-subject trial was conducted with 11 healthy recreationally active males over a three week period. Participants were strapped into an isokinetic dynamometer in the supine position, with the right knee fastened in a knee immobilizer. After randomising the ROM percentages, the hamstring muscle was moved to one of the three chosen ROM percentages for that week and held there for 5 x 60 seconds followed by a 10 second rest between repetitions. A 5ml blood sample was collected pre-, immediately post, and at 24 hours post intervention for high sensitivity C-reactive protein (hsCRP) assessments. Results: Significant increases in hsCRP levels were observed between 30% mROM and 90% mROM (p=0.004) and 60% mROM and 90% mROM (p=0.034), but not between 30% and 60% (p>0.05). Conclusions: Muscle stretching at submaximal levels does not elicit a significant systemic inflammatory responses

    A programmable, multi-format photonic transceiver platform enabling flexible optical networks

    Development of programmable photonic devices for future flexible optical networks is ongoing. To this end, an innovative, multi-format QAM transmitter design is presented. It comprises a segmented-electrode InP IQ-MZM to be fabricated in InP, which can be directly driven by low-power CMOS logic. Arbitrary optical QAM format generation is made possible using only binary electrical signals, without the need for high-performance DACs and high-swing linear drivers. The concept enables a host of Tx-side DSP functionality, including the spectral shaping needed for Nyquist-WDM system concepts. In addition, we report on the development of an optical channel MUX/DEMUX, based on arrays of microresonator filters with reconfigurable bandwidths and center wavelengths. The device is intended for operation with multi-format flexible transceivers, enabling Dense (D)WDM superchannel aggregation and arbitrary spectral slicing in the context of a flexible grid environment

    Closely coupled metallodielectric electromagnetic band-gap structures formed by double-layer dipole and tripole arrays

    The concept of closely coupled metallodielectric electromagnetic band-gap (CCMEBG) structures is introduced and investigated using two-dimensional (2-D) double-layer dipole and tripole arrays. An efficient numerical method based on a set of coupled integral equations is used to simulate the double-layer array response. The arrays are placed in close proximity to each other and shifted appropriately in order to produce maximum element coupling. Measurements are presented for oblique plane wave and surface wave incidences. A substantial decrease of the stopband center frequency is observed with the CCMEBG design for both element geometries. Furthermore, wider bandwidth and improved angular stability as compared to single-layer MEBG is obtained. The tripole arrays arranged on a hexagonal lattice exhibit common stopband for any polarization of the incident field due to the symmetry of the element in conjunction with the lattice. The lowering of the resonance for up to 4 to 1 in simulation results emerges as the layers are separated by less than λ/1200 (0.1 mm at 2.5 GHz)

    The Effects of Different Passive Static Stretching Intensities on Recovery from Unaccustomed Eccentric Exercise - A Randomized Controlled Trial

    Effects of passive static stretching intensity on recovery from unaccustomed eccentric exercise of right knee extensors was investigated in 30 recreationally active males randomly allocated into three groups: high-intensity (70-80% maximum perceived stretch), low-intensity (30-40% maximum perceived stretch), and control. Both stretching groups performed 3 sets of passive static stretching exercises of 60s each for hamstrings, hip flexors, and quadriceps, over 3 consecutive days, post-unaccustomed eccentric exercise. Muscle function (eccentric and isometric peak torque) and blood biomarkers (CK and CRP) were measured before (baseline) and after (24, 48, and 72h) unaccustomed eccentric exercise. Perceived muscle soreness scores were collected immediately (time 0), and after 24, 48, and 72h post-exercise. Statistical time x condition interactions observed only for eccentric peak torque (p=.008). Magnitude-based inference analyses revealed low-intensity stretching had most likely, very likely, or likely beneficial effects on perceived muscle soreness (48-72h and 0-72h) and eccentric peak torque (baseline-24h and baseline-72h), compared with high-intensity stretching. Compared with control, low-intensity stretching had very likely or likely beneficial effects on perceived muscle soreness (0-24h and 0-72h), eccentric peak torque (baseline-48h and baseline-72h), and isometric peak torque (baseline-72h). High-intensity stretching had likely beneficial effects on eccentric peak torque (baseline-48h), but likely harmful effects eccentric peak torque (baseline-24h) and CK (baseline-48h and baseline-72h), compared with control. Therefore, low-intensity stretching is likely to result in small-to-moderate beneficial effects on perceived muscle soreness and recovery of muscle function post-unaccustomed eccentric exercise, but not markers of muscle damage and inflammation, compared with high-intensity or no stretching.The accepted manuscript in pdf format is listed with the files at the bottom of this page.     Generalized Holographic Cosmology

    We consider general black hole solutions in five-dimensional spacetime in the presence of a negative cosmological constant. We obtain a cosmological evolution via the gravity/gauge theory duality (holography) by defining appropriate boundary conditions on a four-dimensional boundary hypersurface. The standard counterterms are shown to renormalize the bare parameters of the system (the four-dimensional Newton's constant and cosmological constant). We discuss the thermodynamics of cosmological evolution and present various examples. The standard brane-world scenarios are shown to be special cases of our holographic construction.Comment: 15 pages, 5 figure

    Development of methamphetamine conjugated vaccine through hapten design: in vitro and in vivo characterization

    Background: Methamphetamine (METH) substance-use disorder is an ever-growing global health issue with no effective treatment. Anti-METH vaccines are under investigation as an alternative to existing psychological interventions. This platform has made significant progress over past decades mainly in preclinical stages, and efforts to develop an anti-METH vaccine with a high antibody response are of utmost importance. Methodology: A novel conjugated anti-METH vaccine was developed using METH HCl as the starting material for the design of hapten, a peptide linker consisting of five lysines and five glycines, and finally immunogenic carrier mannan, which is novel to this platform. All the chemical reaction steps were confirmed by several analytical techniques, and the immunogenicity of the developed vaccine was investigated in a mouse model. Results: Thin-layer chromatography and gas chromatography confirmed the reaction between METH and peptide linker. UV, NMR and color tests were used to confirm the presence of the aldehyde groups in oxidized mannan (OM). The final conjugated vaccine was confirmed by UV and LC-MS. The stability of mannan, the METH hapten, and the final vaccine was evaluated by UV and LC-MS and demonstrated satisfactory stability over 3 months in various storage conditions. Animal studies supported the immunogenicity of the novel vaccine. Conclusions: We successfully developed and characterized a novel METH vaccine in vitro and in vivo. The present study findings are encouraging and will form the basis of further exploration to assess its effectiveness to prevent METH addiction in preclinical models

    Design of Linear and Cyclic Mutant Analogues of Dirucotide Peptide (MBP82–98) against Multiple Sclerosis: Conformational and Binding Studies to MHC Class II

    Background: Multiple sclerosis (MS) is an autoimmune disorder of the central nervous system. MS is a T cell-mediated disease characterized by the proliferation, infiltration, and attack of the myelin sheath by immune cells. Previous studies have shown that cyclization provides molecules with strict conformation that could modulate the immune system. Methods: In this study, we synthesized peptide analogues derived from the myelin basic protein (MBP)82⁻98 encephalitogenic sequence (dirucotide), the linear altered peptide ligand MBP82⁻98 (Ala91), and their cyclic counterparts. Results: The synthesized peptides were evaluated for their binding to human leukocyte antigen (HLA)-DR2 and HLA-DR4 alleles, with cyclic MBP82⁻98 being a strong binder with the HLA-DR2 allele and having lower affinity binding to the HLA-DR4 allele. In a further step, conformational analyses were performed using NMR spectroscopy in solution to describe the conformational space occupied by the functional amino acids of both linear and cyclic peptide analogues. This structural data, in combination with crystallographic data, were used to study the molecular basis of their interaction with HLA-DR2 and HLA-DR4 alleles. Conclusion: The cyclic and APL analogues of dirucotide are promising leads that should be further evaluated for their ability to alter T cell responses for therapeutic benefit against MS
