    Use of the FAO AquaCrop model in developing sowing guidelines for rainfed maize in Zimbabwe

    This paper presents a procedure in which the water-driven water productivity model AquaCrop was fine-tuned and validated for maize for the local conditions in Zimbabwe and then applied to develop sowing management options for decision support. Data from experiments of 2 seasons in Harare and from 5 other sites around Zimbabwe were used for the local calibration and validation of AquaCrop. Model parameters such as the reference harvest index (HIo); the canopy growth coefficient (CGC); early canopy decline and normalised biomass water productivity (WPb*) were adjusted during model calibration. Model performance was satisfactory after calibration with a Nash-Sutcliffe model efficiency parameter (EF = 0.81), RMSE = 15% and R2 = 0.86 upon validation. To develop sowing guidelines, historical climate series from 13 meteorological stations around Zimbabwe were used to simulate maize yield for 6 consecutive sowing dates determined according to criteria applicable in Zimbabwe. Three varieties and typical shallow and deep soil types were considered in the simulation scenarios. The simulated yield was analysed by an optimisation procedure to select the optimum sowing time that maximised long-term mean yield. Results showed that highest yields depended on the climate of the site (rainfall availability), variety (length of growing cycle) and soil depth (soil water storage capacity). The late variety gave higher mean yields for all sowing dates in the maize belt. Staggered sowing is recommended as a way of combating the effects of rainfall variability and as an answer to labour constraints.Keywords: biomass water productivity, AquaCrop, maize sowing dates, crop modellin

    Discovering Crystal Forms of the Novel Molecular Semiconductor OEG-BTBT

    This work is focused on a polymorphic and crystallographic study of a novel p-type organic semiconductor 2,7-bis(2-(2-methoxyethoxy)ethoxy)benzo[b]benzo[4,5]thieno[2,3-d]thiophene (OEG-BTBT). The well-known BTBT core is functionalized by eight-atom-long oligoethylene glycol side chains. Our results demonstrate the discovery of three crystal forms of the OEG-BTBT molecule, namely, Form I, Form II, and Form III, in different experimental conditions. Crystal structures of Form I and Form III are reported, while only unit cell indexing of Form II could be determined. Form I and Form II are enantiotropically related, and Form II is stable at temperatures higher than 127 °C. The kinetics of transformation to Form II was studied by the Avrami equation. Form III is a solvate crystal form which is rarely observed in the field of organic electronics, and upon release of dichloromethane, it converts to Form I. Furthermore, we studied the mechanical properties of the Form I crystals, which exhibit plastic bending upon applying mechanical stress in the [100] direction. This distinct mechanical behavior is rationalized by the slip layer topology, the intermolecular interactions energies from energy frameworks, and the Hirshfeld surface analysis

    Trypanosoma brucei gambiense-iELISA : a promising new test for the post-elimination monitoring of human African trypanosomiasis

    Background: The World Health Organization targeted Trypanosoma brucei gambiense human African trypanosomiasis (gHAT) for elimination as a public health problem and for elimination of transmission. To measure gHAT elimination success with prevalences close to zero, highly specific diagnostics are necessary. Such a test exists in the form of an antibody-mediated complement lysis test, the trypanolysis test, but biosafety issues and technological requirements prevent its large-scale use. We developed an inhibition ELISA with high specificity and sensitivity that is applicable in regional laboratories in gHAT endemic countries. Methods: The T. b. gambiense inhibition ELISA (g-iELISA) is based on the principle that binding of monoclonal antibodies to specific epitopes of T. b. gambiense surface glycoproteins can be inhibited by circulating antibodies of gHAT patients directed against the same epitopes. Using trypanolysis as reference test, the diagnostic accuracy of the g-iELISA was evaluated on plasma samples from 739 gHAT patients and 619 endemic controls and on dried blood spots prepared with plasma of 95 gHAT and 37 endemic controls. Results: Overall sensitivity and specificity on plasma were respectively 98.0% (95% CI 96.7 - 98.9) and 99.5% (95% CI 98.6-99.9). With dried blood spots, sensitivity was 92.6% (95% CI 85.4 - 97.0), and specificity was 100% (95% CI 90.5 - 100.0). The g-iELISA is stable for at least 8 months when stored at 2-8°C. Conclusion: The g-iELISA might largely replace trypanolysis for monitoring gHAT elimination and for post-elimination surveillance. The g-iELISA kit is available for evaluation in reference laboratories in endemic countries

    Relative transpiration as a decision tool in crop management: a case for rainfed maize in Zimbabwe

    Water stress has been considered to be the primary constraint to yield in water-limited arid and semi-arid environments. This paper describes the characterisation of the rainfall season using relative transpiration (Trel) of a maize crop at 13 climate stations in Zimbabwe. A soil water balance model was used to simulate relative crop transpiration for a maize crop over the duration of each rainfall season to assess its quality (severity of intraseasonal dry spells). The Trel and length of growing period (LGP) were subjected to frequency analyses and the results were interpolated (kriging) to form a GIS library of expected events in normal, wet and dry years. The normal LGP (50% PE) varied across the stations, with a range of 75 days, exposing opportunities for objective management of variety selection to match crop growth cycles to expected LGP. The time series of Trel showed the time variation of quality of the season with periods of high Trel identifying the high quality parts of the rainfall season suitable for crop production. Soil depth influenced quality of the season, with deeper soils improving quality.     Intravascular lymphoma: Clinical presentation, natural history, management and prognostic factors in a series of 38 cases, with special emphasis on the 'cutaneous variant'

    Despite its recognition as a distinct, extremely rare entity, no large studies of intravascular lymphoma (IVL) have been reported. The clinico-pathological characteristics of 38 human immunodeficiency virus-negative patients with IVL diagnosed in Western countries were reviewed to better delineate clinical presentation, clinical variants, natural history and optimal therapy. The IVL is an aggressive and usually disseminated disease (Ann Arbor stage IV in 68% of cases) that predominantly affects elderly patients (median age 70 years, range: 34-90; male:female ratio 0.9), resulting in poor Eastern Cooperative Oncology Group Performance Status (ECOG-PS >1 in 61%), B symptoms (55%), anaemia (63%) and high serum lactate dehydrogenase level (86%). The brain and skin are the most common sites of disease. In contrast to previous reports, hepatosplenic involvement (26%) and bone marrow infiltration (32%) were found to be common features in IVL, while nodal disease was confirmed as rare (11% of cases). Patients with disease limited to the skin ('cutaneous variant'; 26% of cases) were invariably females with a normal platelet count, and exhibited a significantly better outcome than the remaining patients, which deserves further investigation. Overall survival was usually poor; however, the early use of intensive therapies could improve outcome in young patients with unfavourable features. ECOG-PS >1, 'cutaneous variant', stage I and chemotherapy use were independently associated with improved survival

    Rivaroxaban versus enoxaparin for thromboprophylaxis after hip arthroplasty.

    This phase 3 trial compared the efficacy and safety of rivaroxaban, an oral direct inhibitor of factor Xa, with those of enoxaparin for extended thromboprophylaxis in patients undergoing total hip arthroplasty.In this randomized, double-blind study, we assigned 4541 patients to receive either 10 mg of oral rivaroxaban once daily, beginning after surgery, or 40 mg of enoxaparin subcutaneously once daily, beginning the evening before surgery, plus a placebo tablet or injection. The primary efficacy outcome was the composite of deep-vein thrombosis (either symptomatic or detected by bilateral venography if the patient was asymptomatic), nonfatal pulmonary embolism, or death from any cause at 36 days (range, 30 to 42). The main secondary efficacy outcome was major venous thromboembolism (proximal deep-vein thrombosis, nonfatal pulmonary embolism, or death from venous thromboembolism). The primary safety outcome was major bleeding.A total of 3153 patients were included in the superiority analysis (after 1388 exclusions), and 4433 were included in the safety analysis (after 108 exclusions). The primary efficacy outcome occurred in 18 of 1595 patients (1.1\%) in the rivaroxaban group and in 58 of 1558 patients (3.7\%) in the enoxaparin group (absolute risk reduction, 2.6\%; 95\% confidence interval [CI], 1.5 to 3.7; P<0.001). Major venous thromboembolism occurred in 4 of 1686 patients (0.2\%) in the rivaroxaban group and in 33 of 1678 patients (2.0\%) in the enoxaparin group (absolute risk reduction, 1.7\%; 95\% CI, 1.0 to 2.5; P<0.001). Major bleeding occurred in 6 of 2209 patients (0.3\%) in the rivaroxaban group and in 2 of 2224 patients (0.1\%) in the enoxaparin group (P=0.18).A once-daily, 10-mg oral dose of rivaroxaban was significantly more effective for extended thromboprophylaxis than a once-daily, 40-mg subcutaneous dose of enoxaparin in patients undergoing elective total hip arthroplasty.     Are hospitals delivering appropriate VTE prevention? The venous thromboembolism study to assess the rate of thromboprophylaxis (VTE start)

    The 7th conference of the American College of Chest Physicians (ACCP7) provides recommendations on the type, dose, and duration of thromboprophylaxis in hospitalized patients at risk of venous thromboembolism (VTE), but the extent to which hospitals follow these criteria has not been well studied. Discharge and billing records for patients admitted to any of 16 acute-care hospitals from January 2005 to December 2006 were obtained. Patients 18 years or older who had an inpatient stay ≥2 days and no apparent contraindications for thromboprophylaxis were grouped into the categories of critical care, surgery and medically ill before being assessed for additional VTE risk factors based on the diagnostic criteria outlined in ACCP7. For patients at risk, the recommended type (mechanical or pharmacologic), dose, and duration of thromboprophylaxis was identified based on the guidelines and compared to the regimen actually received, if any. Among the 258,556 hospitalized patients, 68,278 (26.4%) were determined to be at risk of VTE without apparent contraindications for thromboprophylaxis. The proportions of patients who received the appropriate type, dose, and duration of thromboprophylaxis were 10.5, 9.8, and 17.9% for critical care, medical, and surgical patients, respectively. Of those at risk, 36.8% received no thromboprophylaxis and an additional 50.2% received thromboprophylaxis deemed inappropriate for one or more reasons. The implementation of ACCP7 guidelines for type, dosage, and duration of thromboprophylaxis is low in patients at risk of VTE. There is a need for physicians and health systems to improve awareness and implementation of recommended thromboprophylaxis

    Sortase-mediated labelling of lipid nanodiscs for cellular tracing

    Lipid nanodiscs have broad applications in membrane protein assays, biotechnology and materials science. Chemical modification of the nanodiscs to expand their functional attributes is generally desirable for all of these uses. We present a method for site-selective labelling of the N-terminus of the nanodisc’s membrane scaffold protein (MSP) using the Sortase A protein. Labelling of the MSP was achieved when assembled within the lipid nanodisc architecture, demonstrating that this method can be used as a retrofit approach to modification of preformed nanodiscs before or during application. We label the MSP with a fluorescent fluorescein moiety and use them to image nanodisc uptake into HeLa cells. The Sortase A labelling method could be employed as a general approach to labelling nanodiscs with application-specific functionalities
