    Use of water troughs by wild rabbits (Oryctolagus cuniculus) in a farmland area of north–west Spain

    Uso de bebederos por parte del conejo de monte (Oryctolagus cuniculus) en un paisaje agrícola en el noroeste de EspañaEn la península Ibérica, los bebederos son una herramienta de gestión de hábitat muy frecuente para incrementar las densidades de especies de caza menor, aunque el comportamiento de ingestión de agua de las especies “diana” no se ha estudiado en profundidad, como es el caso del conejo de monte (Oryctolagus cuniculus). Estudiamos el uso de bebederos por parte de conejos de monte durante 228 períodos de una semana en tres períodos consecutivos (junio–octubre) de 2008, 2009 y 2010 en un paisaje agrícola de noroeste de España, utilizando cámaras de fototrampeo. Los conejos utilizaron el 43% de los bebederos y se observó un número significativamente mayor de conejos bebiendo en bebederos rodeados por cobertura vegetal en comparación con bebederos situados en campos abiertos sin dicha cobertura vegetal. La mayoría de los conejos que bebieron fueron fotografiados de julio a septiembre (98%), si bien la utilización de bebederos no se relacionó claramente con la climatología. Los conejos bebieron principalmente durante la mañana (52% de los conejos) no tanto durante la tarde y noche, y raramente durante el mediodía. Los conejos se fotografiaron junto con perdices rojas (Alectoris rufa) en el 6% de las fotografías. Estos hallazgos sugieren que los bebederos son útiles para el conejo y otras especies con necesidades hídricas similares y que debieran ser colocados cerca de zonas con cobertura vegetal arbustiva.Installation of water troughs is a common approach to increase densities of small game species in the Iberian peninsula but little is known about the watering patterns of target species, such as the wild rabbit (Oryctolagus cuniculus). Using camera trapping, we monitored the use of water troughs by wild rabbits over 228 weeks in three consecutive periods, from June to October in 2008, 2009 and 2010, on farmland in north–west Spain. Wild rabbits used 43% of the water troughs. A significantly higher number of rabbits were observed drinking at troughs surrounded by shrub cover than at those in open fields. Most drinking events were recorded from July to September (98%), though the use of water troughs was not clearly related to weather. Wild rabbits drank mainly during the morning (52% of rabbits), less so in the evening and at night, and rarely in the afternoon. Wild rabbits were photographed together with red–legged partridges (Alectoris rufa) in 6% of photographs.     Cytosolic phosphorylated EGFR is predictive of recurrence in early stage penile cancer patients: A retropective study

    Background: Penile cancer (PC) is a rare tumor, and therapeutic options are limited for this disease, with an overall 5-year overall survival around 65-70%. Adjuvant therapy is not recommended for patients with N0-1 disease, despite up to 60% of these patients will die within 5 years from diagnosis. Methods: Medical records of all patients who underwent radical surgery at University Federico II of Naples and at National Tumor Institute "Pascale" of Naples for early squamous cell carcinoma of the penis from January, 2000 to December, 2011 were retrieved. Paraffin wax embedded tissue specimens were retrieved from the pathology archives of the participating Institutions for all patients. Expression of p-EGFR, EGFR and positivity to HPV were evaluated along with other histological variables of interest. Demographic data of eligible patients were retrieved along with clinical characteristics such as type of surgical operation, time of follow up, time of recurrence, overall survival. A multivariable model was constructed using a forward stepwise selection procedure. Results: Thirty eligible patients were identified. All patients were positive for EGFR by immunohistochemistry, while 13 and 16 were respectively positive for nuclear and cytosolic p-EGFR. No EGFR amplification was detected by FISH. Eight patients were positive for high-risk HPV by ISH. On univariable analysis, corpora cavernosa infiltration (OR 7.8; 95% CI = 0,8 to 75,6; P = 0,039) and positivity for cytosolic p-EGFR (OR 7.6; 95% CI = 1.49 to 50; P = 0.009) were predictive for recurrence, while only positivity for cytosolic p-EGFR (HR = 9.0; 95% CI 1.0-100; P = 0,0116) was prognostic for poor survival. Conclusion: It is of primary importance to identify patients with N0-1 disease who are at increased risk of recurrence, as they do not normally receive any adjuvant therapy. Expression of p-EGFR was found in this series to be strongly related to increase risk of recurrence and shorter overall survival. This finding is consistent with the role of p-EGFR in other solid malignancies. Integration of p-EGFR with classic prognostic factors and other histology markers should be pursued to establish optimal adjuvant therapy for N0-1 PC patients

    Human Mas-related G protein-coupled receptors-X1 induce chemokine receptor 2 expression in rat dorsal root ganglia neurons and release of chemokine ligand 2 from the human LAD-2 mast cell line

    Primate-specific Mas-related G protein-coupled receptors-X1 (MRGPR-X1) are highly enriched in dorsal root ganglia (DRG) neurons and induce acute pain. Herein, we analyzed effects of MRGPR-X1 on serum response factors (SRF) or nuclear factors of activated T cells (NFAT), which control expression of various markers of chronic pain. Using HEK293, DRG neuron-derived F11 cells and cultured rat DRG neurons recombinantly expressing human MRGPR-X1, we found activation of a SRF reporter gene construct and induction of the early growth response protein-1 via extracellular signal-regulated kinases-1/2 known to play a significant role in the development of inflammatory pain. Furthermore, we observed MRGPR-X1-induced up-regulation of the chemokine receptor 2 (CCR2) via NFAT, which is considered as a key event in the onset of neuropathic pain and, so far, has not yet been described for any endogenous neuropeptide. Up-regulation of CCR2 is often associated with increased release of its endogenous agonist chemokine ligand 2 (CCL2). We also found MRGPR-X1-promoted release of CCL2 in a human connective tissue mast cell line endogenously expressing MRGPR-X1. Thus, we provide first evidence to suggest that MRGPR-X1 induce expression of chronic pain markers in DRG neurons and propose a so far unidentified signaling circuit that enhances chemokine signaling by acting on two distinct yet functionally co-operating cell types. Given the important role of chemokine signaling in pain chronification, we propose that interruption of this signaling circuit might be a promising new strategy to alleviate chemokine-promoted pain

    How reliable is assessment of true vocal cord-arytenoid unit mobility in patients affected by laryngeal cancer? a multi-institutional study on 366 patients from the ARYFIX collaborative group

    Purpose: In clinical practice the assessment of the “vocal cord-arytenoid unit” (VCAU) mobility is crucial in the staging, prognosis, and choice of treatment of laryngeal squamous cell carcinoma (LSCC). The aim of the present study was to measure repeatability and reliability of clinical assessment of VCAU mobility and radiologic analysis of posterior laryngeal extension. Methods: In this multi-institutional retrospective study, patients with LSCC-induced impairment of VCAU mobility who received curative treatment were included; pre-treatment endoscopy and contrast-enhanced imaging were collected and evaluated by raters. According to their evaluations, concordance, number of assigned categories, and inter- and intra-rater agreement were calculated. Results: Twenty-two otorhinolaryngologists evaluated 366 videolaryngoscopies (total evaluations: 2170) and 6 radiologists evaluated 237 imaging studies (total evaluations: 477). The concordance of clinical rating was excellent in only 22.7% of cases. Overall, inter- and intra-rater agreement was weak. Supraglottic cancers and transoral endoscopy were associated with the lowest inter-observer reliability values. Radiologic inter-rater agreement was low and did not vary with imaging technique. Intra-rater reliability of radiologic evaluation was optimal. Conclusions: The current methods to assess VCAU mobility and posterior extension of LSCC are flawed by weak inter-observer agreement and reliability. Radiologic evaluation was characterized by very high intra-rater agreement, but weak inter-observer reliability. The relevance of VCAU mobility assessment in laryngeal oncology should be re-weighted. Patients affected by LSCC requiring imaging should be referred to dedicated radiologists with experience in head and neck oncology

    Synthesis and characterization of triangulene

    Triangulene, the smallest triplet-ground-state polybenzenoid (also known as Clar's hydrocarbon), has been an enigmatic molecule ever since its existence was first hypothesized1. Despite containing an even number of carbons (22, in six fused benzene rings), it is not possible to draw Kekulé-style resonant structures for the whole molecule: any attempt results in two unpaired valence electrons2. Synthesis and characterization of unsubstituted triangulene has not been achieved because of its extreme reactivity1, although the addition of substituents has allowed the stabilization and synthesis of the triangulene core3, 4 and verification of the triplet ground state via electron paramagnetic resonance measurements5. Here we show the on-surface generation of unsubstituted triangulene that consists of six fused benzene rings. The tip of a combined scanning tunnelling and atomic force microscope (STM/AFM) was used to dehydrogenate precursor molecules. STM measurements in combination with density functional theory (DFT) calculations confirmed that triangulene keeps its free-molecule properties on the surface, whereas AFM measurements resolved its planar, threefold symmetric molecular structure. The unique topology of such non-Kekulé hydrocarbons results in open-shell π-conjugated graphene fragments6 that give rise to high-spin ground states, potentially useful in organic spintronic devices7, 8. Our generation method renders manifold experiments possible to investigate triangulene and related open-shell fragments at the single-molecule level

    Comprehensive Pan-Genomic Characterization of Adrenocortical Carcinoma

    SummaryWe describe a comprehensive genomic characterization of adrenocortical carcinoma (ACC). Using this dataset, we expand the catalogue of known ACC driver genes to include PRKAR1A, RPL22, TERF2, CCNE1, and NF1. Genome wide DNA copy-number analysis revealed frequent occurrence of massive DNA loss followed by whole-genome doubling (WGD), which was associated with aggressive clinical course, suggesting WGD is a hallmark of disease progression. Corroborating this hypothesis were increased TERT expression, decreased telomere length, and activation of cell-cycle programs. Integrated subtype analysis identified three ACC subtypes with distinct clinical outcome and molecular alterations which could be captured by a 68-CpG probe DNA-methylation signature, proposing a strategy for clinical stratification of patients based on molecular markers
