    Characteristics of Wetting-Induced Bacteriophage Blooms in Biological Soil Crust.

    Biological soil crusts (biocrusts) are photosynthetic "hot spots" in deserts and cover ∼12% of the Earth's terrestrial surface, and yet they face an uncertain future given expected shifts in rainfall events. Laboratory wetting of biocrust communities is known to cause a bloom of Firmicutes which rapidly become dominant community members within 2 days after emerging from a sporulated state. We hypothesized that their bacteriophages (phages) would respond to such a dramatic increase in their host's abundance. In our experiment, wetting caused Firmicutes to bloom and triggered a significant depletion of cyanobacterial diversity. We used genome-resolved metagenomics to link phage to their hosts and found that the bloom of the genus Bacillus correlated with a dramatic increase in the number of Caudovirales phages targeting these diverse spore-formers (r = 0.762). After 2 days, we observed dramatic reductions in the relative abundances of Bacillus, while the number of Bacillus phages continued to increase, suggestive of a predator-prey relationship. We found predicted auxiliary metabolic genes (AMGs) associated with sporulation in several Caudovirales genomes, suggesting that phages may influence and even benefit from sporulation dynamics in biocrusts. Prophage elements and CRISPR-Cas repeats in Firmicutes metagenome-assembled genomes (MAGs) provide evidence of recent infection events by phages, which were corroborated by mapping viral contigs to their host MAGs. Combined, these findings suggest that the blooming Firmicutes become primary targets for biocrust Caudovirales phages, consistent with the classical "kill-the-winner" hypothesis.IMPORTANCE This work forms part of an overarching research theme studying the effects of a changing climate on biological soil crust (biocrust) in the Southwestern United States. To our knowledge, this study was the first to characterize bacteriophages in biocrust and offers a view into the ecology of phages in response to a laboratory wetting experiment. The phages identified here represent lineages of Caudovirales, and we found that the dynamics of their interactions with their Firmicutes hosts explain the collapse of a bacterial bloom that was induced by wetting. Moreover, we show that phages carried host-altering metabolic genes and found evidence of proviral infection and CRISPR-Cas repeats within host genomes. Our results suggest that phages exert controls on population density by lysing dominant bacterial hosts and that they further impact biocrust by acquiring host genes for sporulation. Future research should explore how dominant these phages are in other biocrust communities and quantify how much the control and lysis of blooming populations contributes to nutrient cycling in biocrusts

    Archaean and Proterozoic diamond growth from contrasting styles of large-scale magmatism

    Precise dating of diamond growth is required to understand the interior workings of the early Earth and the deep carbon cycle. Here we report Sm-Nd isotope data from 26 individual garnet inclusions from 26 harzburgitic diamonds from Venetia, South Africa. Garnet inclusions and host diamonds comprise two compositional suites formed under markedly different conditions and define two isochrons, one Archaean (2.95 Ga) and one Proterozoic (1.15 Ga). The Archaean diamond suite formed from relatively cool fluid-dominated metasomatism during rifting of the southern shelf of the Zimbabwe Craton. The 1.8 billion years younger Proterozoic diamond suite formed by melt-dominated metasomatism related to the 1.1 Ga Umkondo Large Igneous Province. The results demonstrate that resolving the time of diamond growth events requires dating of individual inclusions, and that there was a major change in the magmatic processes responsible for harzburgitic diamond formation beneath Venetia from the Archaean to the Proterozoic

    Let n>2 and X be a Banach space of dimension strictly greater than n. We show there exists a directionally porous set P in X for which the set of C^1 surfaces of dimension n meeting P in positive measure is not meager. If X is separable this leads to a decomposition of X into a countable union of directionally porous sets and a set which is null on residually many C^1 surfaces of dimension n. This is of interest in the study of certain classes of null sets used to investigate differentiability of Lipschitz functions on Banach spaces

    The polymer, poly(diallyldimethylammonium chloride) (PDDA), is shown to undergo chemical change via a reduction mechanism during X-ray analysis. Examination of the N 1s spectrum of PDDA shows a time dependence on the degree of reduction, together with clear amplification of the extent of reduction through the charge compensation system and X-ray irradiation

    Formation of unusual yellow Orapa diamonds

    Twenty eclogitic diamonds from Orapa Mine (Botswana) with an unusual yellow colour are characterised for their growth structure, N systematics, and C isotope composition, and the major element composition of their silicate inclusions. The diamonds show complex luminescence with green, blue and non-luminescent zones and occasional sector zonation. All parts of the diamonds have low total N concentrations (<50 at.ppm, with one exception of <125 at.ppm) and a limited range in C isotope composition (−5.7 to −10.6‰). Fourier Transform Infrared spectra show bands at 1334, 1332, 1282, and 1240 cm−1 typical for Ib-IaA diamonds. Relict unaggregated N defects (Ns o and Ns +) are present and the preservation is likely caused by the low N concentrations and possible low mantle residence temperatures rather than young diamond formation (inclusion ages of 140, 1096, 1699 Ma; Timmerman et al. Earth Planet Sc Lett 463:178–188, 2017). Garnet and clinopyroxene inclusions extracted from 14 diamonds have an eclogitic composition with relatively low Ca contents and based on all characteristics, these diamonds form a distinct population from Orapa

    Detecting extreme mass ratio inspiral events in LISA data using the Hierarchical Algorithm for Clusters and Ridges (HACR)

    One of the most exciting prospects for the Laser Interferometer Space Antenna (LISA) is the detection of gravitational waves from the inspirals of stellar-mass compact objects into supermassive black holes. Detection of these sources is an extremely challenging computational problem due to the large parameter space and low amplitude of the signals. However, recent work has suggested that the nearest extreme mass ratio inspiral (EMRI) events will be sufficiently loud that they might be detected using computationally cheap, template-free techniques, such as a time-frequency analysis. In this paper, we examine a particular time-frequency algorithm, the Hierarchical Algorithm for Clusters and Ridges (HACR). This algorithm searches for clusters in a power map and uses the properties of those clusters to identify signals in the data. We find that HACR applied to the raw spectrogram performs poorly, but when the data is binned during the construction of the spectrogram, the algorithm can detect typical EMRI events at distances of up to 2.6\sim2.6Gpc. This is a little further than the simple Excess Power method that has been considered previously. We discuss the HACR algorithm, including tuning for single and multiple sources, and illustrate its performance for detection of typical EMRI events, and other likely LISA sources, such as white dwarf binaries and supermassive black hole mergers. We also discuss how HACR cluster properties could be used for parameter extraction.Comment: 21 pages, 11 figures, submitted to Class. Quantum Gravity.     Boronic acids for functionalisation of commercial multi-layer graphitic material as an alternative to diazonium salts

    A novel radical-based functionalisation strategy for the synthesis of functionalised commercially obtained plasma-synthesised multi-layer graphitic material (MLG) is presented herein. 4-(trifluoromethyl)phenyl boronic acid was utilised as a source of 4-(trifluoromethyl)phenyl radicals to covalently graft upon the graphitic surface of MLG. Such a methodology provides a convenient and safer route towards aryl radical generation, serving as a potential alternative to hazardous diazonium salt precusors. The structure and morphology of the functionalised MLG (Arf-MLG) has been characterised using XPS, Raman, TGA, XRD, SEM, TEM and BET techniques. The XPS quantitative data and Raman spectra provide evidence of successful covalent attachment of 4-(trifluoromethyl)phenyl groups to MLG
