    Selective aqueous acetylation controls the photoanomerization of α-cytidine-5â€Č-phosphate

    Nucleic acids are central to information transfer and replication in living systems, providing the molecular foundations of Darwinian evolution. Here we report that prebiotic acetylation of the non-natural, but prebiotically plausible, ribonucleotide α-cytidine-5â€Č-phosphate, selectively protects the vicinal diol moiety. Vicinal diol acetylation blocks oxazolidinone formation and prevents C2â€Č-epimerization upon irradiation with UV-light. Consequently, acetylation enhances (4-fold) the photoanomerization of α-cytidine-5â€Č-phosphate to produce the natural ÎČ-pyrimidine ribonucleotide-5â€Č-phosphates required for RNA synthesis

    A New Ant Species of the Genus Tetramorium Mayr, 1855 (Hymenoptera: Formicidae) from Saudi Arabia, with a Revised Key to the Arabian Species

    Tetramorium amalae sp. n. is described and illustrated from Saudi Arabia based on two worker caste specimens collected in Al Bahah region. The new species belongs to the T. shilohense group and appears to be closely related to T. dysderke Bolton from Nigeria. T. amalae is distinguished by having well-developed frontal carinae, smaller eyes, greater head length and width, greater pronotal width, and the petiole node is longer than broad. Tetramorium latinode Collingwood & Agosti is recorded for the first time from Saudi Arabia and for only the second time since the original description. The worker caste of T. latinode is redescribed and illustrated using scanning electron micrographs to facilitate recognition and the gyne is described for the first time with observations given on species relationships, biology and habitat. A revised key to the nineteen Tetramorium species recorded from Arabian Peninsula based on worker castes is provided. Tetramorium bicarinatum (Nylander) is recorded for the first time from Saudi Arabia. It is suggested that T. amalae and T. latinode are endemic to the Arabian Peninsula

    Monitoring trends in HIV prevalence among young people, aged 15 to 24 years, in Manicaland, Zimbabwe

    BACKGROUND: In June 2001, the United Nations General Assembly Special Session (UNGASS) set a target of reducing HIV prevalence among young women and men, aged 15 to 24 years, by 25% in the worst-affected countries by 2005, and by 25% globally by 2010. We assessed progress toward this target in Manicaland, Zimbabwe, using repeated household-based population serosurvey data. We also validated the representativeness of surveillance data from young pregnant women, aged 15 to 24 years, attending antenatal care (ANC) clinics, which UNAIDS recommends for monitoring population HIV prevalence trends in this age group. Changes in socio-demographic characteristics and reported sexual behaviour are investigated. METHODS: Progress towards the UNGASS target was measured by calculating the proportional change in HIV prevalence among youth and young ANC attendees over three survey periods (round 1: 1998-2000; round 2: 2001-2003; and round 3: 2003-2005). The Z-score test was used to compare differences in trends between the two data sources. Characteristics of participants and trends in sexual risk behaviour were analyzed using Student's and two-tailed Z-score tests. RESULTS: HIV prevalence among youth in the general population declined by 50.7% (from 12.2% to 6.0%) from round 1 to 3. Intermediary trends showed a large decline from round 1 to 2 of 60.9% (from 12.2% to 4.8%), offset by an increase from round 2 to 3 of 26.0% (from 4.8% to 6.0%). Among young ANC attendees, the proportional decline in prevalence of 43.5% (from 17.9% to 10.1%) was similar to that in the population (test for differences in trend: p value = 0.488) although ANC data significantly underestimated the population prevalence decline from round 1 to 2 (test for difference in trend: p value = 0.003) and underestimated the increase from round 2 to 3 (test for difference in trend: p value = 0.012). Reductions in risk behaviour between rounds 1 and 2 may have been responsible for general population prevalence declines. CONCLUSIONS: In Manicaland, Zimbabwe, the 2005 UNGASS target to reduce HIV prevalence by 25% was achieved. However, most prevention gains occurred before 2003. ANC surveillance trends overall were an adequate indicator of trends in the population, although lags were observed. Behaviour data and socio-demographic characteristics of participants are needed to interpret ANC trends

    Nonlinear digital signal processing in mental health: characterization of major depression using instantaneous entropy measures of heartbeat dynamics

    Nonlinear digital signal processing methods that address system complexity have provided useful computational tools for helping in the diagnosis and treatment of a wide range of pathologies. More specifically, nonlinear measures have been successful in characterizing patients with mental disorders such as Major Depression (MD). In this study, we propose the use of instantaneous measures of entropy, namely the inhomogeneous point-process approximate entropy (ipApEn) and the inhomogeneous point-process sample entropy (ipSampEn), to describe a novel characterization of MD patients undergoing affective elicitation. Because these measures are built within a nonlinear point-process model, they allow for the assessment of complexity in cardiovascular dynamics at each moment in time. Heartbeat dynamics were characterized from 48 healthy controls and 48 patients with MD while emotionally elicited through either neutral or arousing audiovisual stimuli. Experimental results coming from the arousing tasks show that ipApEn measures are able to instantaneously track heartbeat complexity as well as discern between healthy subjects and MD patients. Conversely, standard heart rate variability (HRV) analysis performed in both time and frequency domains did not show any statistical significance. We conclude that measures of entropy based on nonlinear point-process models might contribute to devising useful computational tools for care in mental health

    Statistical Signatures of Photon Localization

    The realization that electron localization in disordered systems (Anderson localization) is ultimately a wave phenomenon has led to the suggestion that photons could be similarly localized by disorder. This conjecture attracted wide interest because the differences between photons and electrons - in their interactions, spin statistics, and methods of injection and detection - may open a new realm of optical and microwave phenomena, and allow a detailed study of the Anderson localization transition undisturbed by the Coulomb interaction. To date, claims of three-dimensional photon localization have been based on observations of the exponential decay of the electromagnetic wave as it propagates through the disordered medium. But these reports have come under close scrutiny because of the possibility that the decay observed may be due to residual absorption, and because absorption itself may suppress localization. Here we show that the extent of photon localization can be determined by a different approach - measurement of the relative size of fluctuations of certain transmission quantities. The variance of relative fluctuations accurately reflects the extent of localization, even in the presence of absorption. Using this approach, we demonstrate photon localization in both weakly and strongly scattering quasi-one-dimensional dielectric samples and in periodic metallic wire meshes containing metallic scatterers, while ruling it out in three-dimensional mixtures of aluminum spheres.Comment: 5 pages, including 4 figure

    Prebiotic synthesis of cysteine peptides that catalyze peptide ligation in neutral water

    Peptide biosynthesis is performed by ribosomes and several other classes of enzymes, but a simple chemical synthesis may have created the first peptides at the origins of life. a-Aminonitriles—prebiotic a–amino acid precursors—are generally produced by Strecker reactions. However, cysteine’s aminothiol is incompatible with nitriles. Consequently, cysteine nitrile is not stable, and cysteine has been proposed to be a product of evolution, not prebiotic chemistry. We now report a high-yielding, prebiotic synthesis of cysteine peptides. Our biomimetic pathway converts serine to cysteine by nitrile-activated dehydroalanine synthesis. We also demonstrate that N-acylcysteines catalyze peptide ligation, directly coupling kinetically stable—but energy-rich—a-amidonitriles to proteinogenic amines. This rare example of selective and efficient organocatalysis in water implicates cysteine as both catalyst and precursor in prebiotic peptide synthesis

    International audienceAbstractBackground Metafounders are pseudo-individuals that encapsulate genetic heterozygosity and relationships within and across base pedigree populations, i.e. ancestral populations. This work addresses the estimation and usefulness of metafounder relationships in single-step genomic best linear unbiased prediction (ssGBLUP).ResultsWe show that ancestral relationship parameters are proportional to standardized covariances of base allelic frequencies across populations, such as Fst\documentclass[12pt]{minimal}\usepackage{amsmath}\usepackage{wasysym} \usepackage{amsfonts} \usepackage{amssymb} \usepackage{amsbsy}\usepackage{mathrsfs}\usepackage{upgreek}\setlength{\oddsidemargin}{-69pt}\begin{document}FstF_{\text{st}}\end{document} fixation indexes. These covariances of base allelic frequencies can be estimated from marker genotypes of related recent individuals and pedigree. Simple methods for their estimation include naïve computation of allele frequencies from marker genotypes or a method of moments that equates average pedigree-based and marker-based relationships. Complex methods include generalized least squares (best linear unbiased estimator (BLUE)) or maximum likelihood based on pedigree relationships. We derived a genomic relationship matrix, compatible with pedigree relationships, that is constructed as a cross-product of {−1,0,1} codes and that is equivalent (apart from scale factors) to an identity-by-state relationship matrix at genome-wide markers. Using a simulation with a single population under selection in which only males and youngest animals are genotyped, we observed that generalized least squares or maximum likelihood gave accurate and unbiased estimates of the ancestral relationship parameter (true value: 0.40) whereas the naïve method and the method of moments were biased (average estimates of 0.43 and 0.35). We also observed that genomic evaluation by ssGBLUP using metafounders was less biased in terms of estimates of genetic trend (bias of 0.01 instead of 0.12), resulted in less overdispersed (0.94 instead of 0.99) and as accurate (0.74) estimates of breeding values than ssGBLUP without metafounders and provided consistent estimates of heritability.ConclusionsEstimation of metafounder relationships can be achieved using BLUP-like methods with pedigree and markers. Inclusion of metafounder relationships reduces bias of genomic predictions with no loss in accuracy

    Solidification thermal parameters affecting the columnar-to-equiaxed transition

    Experiments were conducted to analyze the columnar-to-equiaxed transition (CET) during the upward unsteady-state directional solidification of Al-Cu and Sn-Pb alloys, under different conditions of superheat and heat-transfer efficiencies at the metal/mold interface. A combined theoretical and experimental approach is developed to quantitatively determine the solidification thermal parameters: transient heat-transfer coefficients, tip growth rates, thermal gradients, and cooling rates. The observed results do not give support to CET criteria based individually either on tip growth rate or temperature gradients ahead of the liquidus isotherm. Rather, the analysis has indicated that a more convenient criterion should encompass both thermal parameters through the tip cooling rate. The columnar growth is expected to prevail throughout the casting for a tip cooling rate higher than a critical value, which depends only on the alloy system and was observed to be about 0.2 K/s for Al-Cu alloys and 0.01 K/s for Sn-Pb alloys in the present investigation.3372107211

    Correlation between unsteady-state solidification conditions, dendrite spacings, and mechanical properties of Al-Cu alloys

    The wide range of operational conditions existing in foundry and casting processes generates as a direct consequence a diversity of solidification microstructures. Structural parameters such as grain size and interdendritic spacings are strongly influenced by the thermal behavior of the metal/mold system during solidification, imposing, as a consequence, a close correlation between this system and the resulting microstructure. Mechanical properties depend on the microstructural arrangement defined during solidification. Expressions correlating the mechanical behavior with microstructure parameters should be useful for future planning of solidification conditions in terms of a determined level of mechanical strength, which is intended to be attained. In the present work, analytical expressions have been developed describing thermal gradients and tip growth rate during one-dimensional unsteady-state solidification of alloys. Experimental results concerning the solidification of Al-4.5 wt pet Cu and Al-15 wt pet Cu alloys and dendritic growth models have permitted the establishment of general expressions correlating microstructure dendrite spacings with solidification processing variables. The correlation of these expressions with experimental equations relating mechanical properties and dendrite spacings provides an insight into the preprogramming of solidification in terms of casting mechanical properties.31123167317
