    Sensing Technologies in the Evaluation of the Mechanical Properties of the Plantar Flexors by Using the Free Vibration Technique

    The use of free vibration techniques to evaluate the responsiveness of a one-degree-of-freedom dynamic system linked to musculo-articular stiffness (MAS) of the plantar flexors has become an accepted procedure in scientific literature. However, the applicability and widespread use of this parameter have been limited thus far for different issues. On the one hand, the measurement of this parameter requires a noninvasive, nonstress laboratory test, which implies the use of appropriate in-lab infrastructure, ad hoc mechanical devices, and specific signal sensing units. On the other hand, it requires an efficient treatment and processing of the signal data obtained from laboratory tests through different signal filtering techniques and mathematical transformations. In the present manuscript, the fundamentals of the free vibration technique, the measurement device, the adjustment procedure, and finally the software based on java platform are presented in order to show the applicability of the MAS as parameter to evaluate the functional response of one subject based on his mechanical response of the lower part of the body once the whole procedure is addressed and controlled to ensure the reproducibility and the reliability of the test

    Room Temperature In-plane <100> Magnetic Easy Axis for Fe3O4/SrTiO3(001):Nb Grown by Infrared PLD

    We examine the magnetic easy-axis directions of stoichiometric magnetite films grown on SrTiO3:Nb by infrared pulsed-laser deposition. Spin-polarized low-energy electron microscopy reveals that the individual magnetic domains are magnetized along the in-plane film directions. Magneto-optical Kerr effect measurements show that the maxima of the remanence and coercivity are also along in-plane film directions. This easy-axis orientation differs from bulk magnetite and films prepared by other techniques, establishing that the magnetic anisotropy can be tuned by film growth.Comment: 3 pages, 3 figure

    Ganadería y ambiente: una experiencia de trabajo con escuelas rurales de Tandil

    Con el objetivo de dar a conocer los impactos ambientales de la ganadería y en particular de su emisión de metano, un gas con efecto invernadero, se trabajó durante tres años con alumnos de cinco escuelas primarias del medio rural en Tandil. Se hizo un trabajo preparativo con las maestras, y luego las presentaciones de los temas, una visita al campus universitario, y por último un juego de mesa para discutir y afianzar los contenidos. Se logró un cambio en la percepción de los chicos sobre la posibilidad de que los bovinos afecten el ambiente, sobre las posibles causas del cambio climático, y sobre la necesidad de pensar en estrategias para la adaptación y mitigación en los sistemas ganaderos.In order to make the environmental impact of livestock known and, particularly, methane emission as a greenhouse gas, we worked along three years with students from five elementary schools in rural areas of Tandil. The process consisted in preparatory works with teachers as a start point, then the topic presentation to the students, a visit to the University Campus and, finally, a grupal board game to discuss and consolidate contents. A change was achieved in children´s perception about the possibility that cattle affects the environment, on possible causes of climate change and in the need to think strategies for adaptation and mitigation in livestock systems.Relato de experiencias del desarrollo de proyectos de extensió

    A chemogenomic screening identifies CK2 as a target for pro-senescence therapy in PTEN-deficient tumours

    Enhancement of cellular senescence in tumours triggers a stable cell growth arrest and activation of an antitumour immune response that can be exploited for cancer therapy. Currently, there are only a limited number of targeted therapies that act by increasing senescence in cancers, but the majority of them are not selective and also target healthy cells. Here we developed a chemogenomic screening to identify compounds that enhance senescence in PTEN-deficient cells without affecting normal cells. By using this approach, we identified casein kinase 2 (CK2) as a pro-senescent target. Mechanistically, we show that Pten loss increases CK2 levels by activating STAT3. CK2 upregulation in Pten null tumours affects the stability of Pml, an essential regulator of senescence. However, CK2 inhibition stabilizes Pml levels enhancing senescence in Pten null tumours. Taken together, our screening strategy has identified a novel STAT3-CK2-PML network that can be targeted for pro-senescence therapy for cancer

    Global DNA Methylation levels analysis in a serie of Hematological, Breast and Colorectal cancer samples from Argentina

    Unlike their normal counterparts, tumor cells exhibited highly variable CpG methylation levels in a large proportion of the genome, which can lead to malignant cell transformation through multiple pathways. This prompted us to assess the extent of LINE1 methylation, a surrogate marker of global DNA methylation, of samples derived from controls and cancer patients from Argentina. Preliminary DNA methylation results from selected samples were replicated in a large serie of 146 controls (blood) and various cancer types: 112 oncohematological cancer (HemCa), 70 colorectal cancer (CRC) and 68 breast cancer (BrCa) samples. Further, we evaluated correlation with biological, clinical and demographic features. Blood samples were available in all cases, and for solid tumors paired tumoral/non-tumoral adjacent tissues (T/N) were available too. LINE1 methylation level was analyzed by MS-MLPA method. HemCa cases showed statistically significant higher LINE1 methylation level (p>0.001) compared to controls (mean 0.93 and 0.84, respectively). This variation could be a consequence of chemotherapy. Methylation status in blood (0.86) and N tissue (0.87) from BrCa cases did not differ from controls, while levels in T tissue (0.88) were significantly higher than controls (p<0.05). No differences between N and T tissues were found. CRC cases showed hypomethylation for LINE1 when comparing T (0.81) to blood (0.87) or N tissues (0.88), reaching statistical significance of p<0.05 and p<0.001, respectively. This is in line with reported results. We found a negative correlation between individual age and methylation level in controls (-0.17, p=0.04), and BrCa T tissue (-0.33, p=0.03). Finally, no relevant associations between global methylation and mitochondrial genome variation (copy number and ancestry) were found for controls and HemCa sample sets.     Efectos de cadmio en compuestos fenolicos totales y flavonoides de Euglena gracilis

    In the present study the production of phenolic acids and flavonoid compounds by E. gracilis exposed to two cadmium concentrations (0.02 and 0.14 mM) was evaluated using high-performance liquid chromatography (HPLC). The results showed that E. gracilis exposed to 0.02 mM Cd+2 increased significantly the total content of phenolic compounds (798.46 ± 12.61 ?g GA/g) and total flavonoids (241.34 ± 47.63 ?g QE/g) with respect to the control (137.34 ± 19.80 ?g QE/g DW and 549.00 ± 8.57 ?g GA/g DW, respectively). However, no significant increase in the total content of phenolic compounds (568.54 ± 17.42 ?g GA/g DW) and total flavonoids (141.11 ± 9.36 ?g QE/g DW) were observed in E. gracilis exposed to 0.14 mM Cd+2. Further research is necessary to determine the specific role of flavonoids in E. gracilis exposed to high concentrations of Cd+2.En el presente estudio la producción de compuestos fenolicos y flavonoides producidos por E. gracilis expuesto a dos concentraciones de cadmio fue evaluado usando cromatografía liquida de alta precisión (HPLC). Los resultados mostraron que 0.02 mM de Cd+2 incrementaba significativamente el contenido total de compuestos fenolicos (798.46 ± 12.61 ?g GA/g) y flavonoides (241.34 ± 47.63 ?g QE/g) con respecto al control (137.34 ± 19.80 ?g QE/g DW y 549.00 ± 8.57 ?g GA/g DW) respectivamente.     Breakthrough COVID-19 in vaccinated patients with hematologic malignancies: results from EPICOVIDEHA survey

    Limited data have been published on the epidemiology and outcomes of breakthrough COVID-19 in patients with hematological malignancy (HM) after anti-SARS-CoV-2 vaccination. Adult HM who received at least one dose of anti-SARS-CoV-2 vaccine and diagnosed with breakthrough COVID-19 between January 2021 and March 2022 and registered in EPICOVIDEHA were included in this analysis. A total of 1548 cases were included, mainly with lymphoid malignancies (1181 cases, 76%). After viral genome sequencing in 753 cases (49%), Omicron variant was prevalent (517, 68.7%). Most of the patients received at least two vaccine doses before COVID-19 (1419, 91%), mostly mRNA-based (1377, 89%). Overall, 906 patients (59%) received specific treatment for COVID-19. After 30-days follow-up from COVID-19 diagnosis, 143 patients (9%) died. The mortality rate in patients with Omicron variant was of 7.9%, comparable to that reported for the other variants. The 30-day mortality rate was significantly lower than in the pre-vaccine era (31%). In the univariable analysis, older age (p<0.001), active HM (p<0.001), severe and critical COVID-19 (p=0.007 and p<0.001, respectively) were associated with mortality. Conversely, patients receiving monoclonal antibodies, even for severe or critical COVID-19, had a lower mortality rate (p<0.001). In the multivariable model, older age, active disease, critical COVID-19 and at least 2-3 comorbidities were correlated with a higher mortality, whereas the administration of monoclonal antibodies, alone (p<0.001) or combined with antivirals (p=0.009), was observed protective. While mortality is significantly lower than in the pre-vaccination era, breakthrough COVID-19 in HM is still associated with considerable mortality. Death rate was lower in patients who received monoclonal antibodies, alone or in combination with antivirals.     Plasma and stool metabolomic biomarkers of non-alcoholic fatty liver disease in Argentina

    Background: Non-invasive biomarkers are urgently needed to identify patients with non-alcoholic fatty liver disease (NAFLD) at risk of disease progression, particularly in high prevalence areas such as Latin America. In this regard, targeted metabolomics is a powerful technology for discovering new gut microbiome-derived metabolites. Thus, we aimed to identify potential metabolomic biomarkers related to NAFLD stage in Argentina, and to assess their relationship with clinical and host genetic factors. Methods: Adult healthy volunteers (HV) and biopsy-proven simple steatosis (SS) or non-alcoholic steatohepatitis (NASH) patients were recruited. Demographic, clinical and food frequency consumption data, as well as plasma and stool samples were collected. SNP rs738409 (PNPLA3 gene) was determined in all volunteers. HPLC and flow injection analysis with MS/MS in tandem was applied for metabolomic studies using the MxP Quant 500 Kit (Biocrates Life Sciences AG, Austria). Significantly different metabolites among groups were identified with MetaboAnalyst v4.0. Bivariate and multivariate analyses were used to identify variables that were independently related to NAFLD stage. Forward stepwise logistic regression models were constructed to design the best feature combination that could distinguish between study groups. Receiver Operating Characteristic (ROC) curves were used to evaluate models? accuracy.Results: 19 HV, 12 SS and 22 NASH patients were recruited. Diet was similar between groups. The concentration of 33 out of 424 detected metabolites (25 in plasma and 8 in stool) was significantly different among study groups. Levels of triglycerides (TG) were higher among NAFLD patients, whereas levels of phosphatidylcholines (PC) and lysoPC were higher among HV. The PNPLA3 risk genotype for NAFLD and NASH (GG) was related to higher plasma levels of eicosenoic acid FA(20:1) (p<0.001). Plasma metabolites showed a higher accuracy for diagnosis of NAFLD and NASH when compared to stool metabolites (Table 1). Body mass index (BMI) and plasma levels of PC aa C24:0, FA(20:1) and TG(16:1_34:1) showed high accuracy for diagnosis of NAFLD; whereas the best AUROC for discriminating NASH from SS was that of plasma levels of PC aa C24:0 and PC ae C40:1 (Table 1).Conclusions: Gut microbiome-derived metabolomic biomarkers were identified in plasma and stool, but plasma metabolites were better diagnostic biomarkers of NAFLD and NASH in Argentina.     Aplikasi Herbisida 2,4-d Dan Penoxsulam Pada Pertumbuhan Dan Hasil Tanaman Padi Sawah (Oryza Sativa L.)

    Salah satu teknik budidaya untuk meningkatkan produksi tanaman padi sawah yaitu dengan mengurangi persaingan antara tanaman dengan gulma. Pengendalian dengan kimiawi merupakan salah satu cara mengurangi pertumbuhan gulma di pertanaman padi. Cara kimiawi merupakan cara yang praktis, efektif dan efisien untuk mengendalikan gulma. Penelitian ini bertujuan untuk mempelajari pengaruh dari aplikasi herbisida 2,4-D dan penoxsulam dalam meningkatkan pertumbuhan dan hasil padi sawah serta menentukan dosis aplikasi herbisida 2,4-D dan penoxsulam baik secara tunggal maupun campuran dalam mengendalikan gulma pada tanaman padi sawah. Penelitian telah dilaksanakan pada bulan Maret-Juli 2014 di Desa Campurasri, Ngawi. Penelitian menggunakan Rancangan Acak Kelompok sederhana, dengan menempatkan 11 perlakuan yaitu H1 : kontrol herbisida 2,4-D; H2 : 2,4-D 11,25 kg ha-1; H3 : 2,4-D 22,5 kg ha-1; H4 : 2,4-D 33,75 kg ha-1; H5 : kontrol herbisida penoxsulam; H6 : penoxsulam 200 ml ha-1; H7 : penoxsulam 400 ml ha-1; H8 : penoxsulam 600 ml ha-1; H9 : 2,4-D 11,25 kg ha-1 dan penoxsulam 200 ml ha-1; H10 : 2,4-D 22,5 kg ha-1 dan penoxsulam 400 ml ha-1; H11 : 2,4-D 33,75 kg ha-1 dan penoxsulam 600 ml ha-1. Hasil penelitian menunjukkan bahwa perlakuan herbisida 2,4-D 11,25 kg ha-1 dan penoxsulam 200 ml menghasilkan bobot kering total tanaman dengan peningkatan sebesar 34,62 % dibandingkan dengan kontrol. Pada produksi tanaman padi peningkatan terjadi sebesar 29,77 % pada perlakuan herbisida 2,4-D 33,75 kg ha-1 dan penoxsulam 600 ml dibandingkan dengan kontrol
