Este capítulo ofrece una revisión de los factores que afectan a la regeneración natural de la vegetación, considerando la serie de etapas demográficas que determinan la dinámica demográfica de una pobla- ción. Son muy escasos los estudios disponibles que analizan toda la serie de eventos concatenados entre la producción de flores y frutos y el establecimiento exitoso de una planta adulta reproductiva. También son escasos los trabajos que permiten establecer los efectos aplazados que tienen las interacciones con animales a lo largo del ciclo de regeneración. La aproximación que proponemos cuantifica las pérdidas de propágulos en cada etapa demográfica e identifica “cuellos de botella” del reclutamiento que pue- den colapsar la regeneración natural de una especie. Revisamos una serie de casos de estudio que ilustran diversos procesos de limitación demográfica. El uso de técnicas explícitamente demográficas es fundamental para comprender la evolución de las especies forestales Mediterráneas y para diseñar actuaciones de preservación de sus poblaciones y de su extraordinaria diversidad.[EN]: We review the main factors influencing recruitment limitation in Mediterranean woody species by considering the sequential stages that determine the demographic cycle. Very few studies examine the whole set of demographic stages, from flower production to the successful establishment of adult reproductive plants, and their influence on recruitment. There are also few studies exploring the delayed effects of animal interactions throughout the regeneration cycle, but the information on stage-specific effects is more detailed. We propose an approach that quantifies the propagule losses at each sequential demographic stage and identifies demographic bottlenecks that might collapse population growth. We review a series of case studies illustrating different limitation processes.     Intrinsically disordered chromatin protein NUPR1 binds to the enzyme PADI4

    The nuclear protein 1 (NUPR1) is an intrinsically disordered protein involved in stress-mediated cellular conditions. Its paralogue nuclear protein 1-like (NUPR1L) is p53-regulated, and its expression down-regulates that of the NUPR1 gene. Peptidyl-arginine deiminase 4 (PADI4) is an isoform of a family of enzymes catalyzing arginine to citrulline conversion; it is also involved in stress-mediated cellular conditions. We characterized the interaction between NUPR1 and PADI4 in vitro, in silico, and in cellulo. The interaction of NUPR1 and PADI4 occurred with a dissociation constant of 18 ± 6 μM. The binding region of NUPR1, mapped by NMR, was a hydrophobic polypeptide patch surrounding the key residue Ala33, as pinpointed by: (i) computational results; and, (ii) site-directed mutagenesis of residues of NUPR1. The association between PADI4 and wild-type NUPR1 was also assessed in cellulo by using proximity ligation assays (PLAs) and immunofluorescence (IF), and it occurred mainly in the nucleus. Moreover, binding between NUPR1L and PADI4 also occurred in vitro with an affinity similar to that of NUPR1. Molecular modelling provided information on the binding hot spot for PADI4. This is an example of a disordered partner of PADI4, whereas its other known interacting proteins are well-folded. Altogether, our results suggest that the NUPR1/PADI4 complex could have crucial functions in modulating DNA-repair, favoring metastasis, or facilitating citrullination of other proteins

    The structure of 3,4-dihydropyrrolo[1,2-a]pyrazine and its N-protonated form is studied by ab initio calculations.     Association between exposure to air pollution and blood lipids in the general population of Spain.

    Background and Aims: We aimed to assess the associations of exposure to air pollutants and standard and advanced lipoprotein measures, in a nationwide sample representative of the adult population of Spain.Methods: We included 4647 adults (>18 years), participants in the national, cross- sectional, population- based [email protected] study, conducted in 2008– 2010. Standard lipid measurements were analysed on an Architect C8000 Analyzer (Abbott Laboratories SA). Lipoprotein analysis was made by an advanced 1H- NMR lipoprotein test (Liposcale®). Participants were assigned air pollution con-centrations for particulate matter <10 μm (PM10), <2.5 μm (PM2.5) and nitrogen dioxide (NO2), corresponding to the health examination year, obtained by mod-elling combined with measurements taken at air quality stations (CHIMERE chemistry- transport model).Results: In multivariate linear regression models, each IQR increase in PM10, PM2.5 and NO2 was associated with 3.3%, 3.3% and 3% lower levels of HDL- c and 1.3%, 1.4% and 1.1% lower HDL particle (HDL- p) concentrations (p< .001 for all associations). In multivariate logistic regression, there was a significant associa-tion between PM10, PM2.5 and NO2 concentrations and the odds of presenting low HDL- c (<40 mg/dL), low HDL- p (<p25) and higher LDL particle (LDL- p) concentrations (≥p75). In subgroup analyses there were stronger associations be-tween PM10 and NO2 and low HDL- p in men (p for interaction .008 and .034), and between NO2 and low HDL- p in individuals with obesity (p for interaction .015).Conclusions: Our study shows an association between the exposure to air pol-lutants and blood lipids in the general population of Spain, suggesting a link to atherosclerosisFunding for open access charge: Universidad de Málaga / CBU

    Asymptotics and zeros of Sobolev orthogonal polynomials on unbounded supports

    In this paper we present a survey about analytic properties of polynomials orthogonal with respect to a weighted Sobolev inner product such that the vector of measures has an unbounded support. In particular, we are focused in the study of the asymptotic behaviour of such polynomials as well as in the distribution of their zeros.     Fatty liver index as a predictor for type 2 diabetes in subjects with normoglycemia in a nationwide cohort study

    Our aim was to evaluate whether fatty liver index (FLI) is associated with the risk of type 2 diabetes (T2DM) development within the Spanish adult population and according to their prediabetes status; additionally, to examine its incremental predictive value regarding traditional risk factors. A total of 2260 subjects (Prediabetes: 641 subjects, normoglycemia: 1619 subjects) from the [email protected] cohort study were studied. Socio-demographic, anthropometric, clinical data and survey on habits were recorded. An oral glucose tolerance test was performed and fasting determinations of glucose, lipids and insulin were made. FLI was calculated and classified into three categories: Low ( 60). In total, 143 people developed diabetes at follow-up. The presence of a high FLI category was in all cases a significant independent risk factor for the development of diabetes. The inclusion of FLI categories in prediction models based on different conventional T2DM risk factors significantly increase the prediction power of the models when all the population was considered. According to our results, FLI might be considered an early indicator of T2DM development even under normoglycemic condition. The data also suggest that FLI could provide additional information for the prediction of T2DM in models based on conventional risk factors

    Adjuvant dabrafenib and trametinib for patients with resected BRAF-mutated melanoma: DESCRIBE-AD real-world retrospective observational study

    BRAF and MEK inhibitor, dabrafenib plus trametinib, adjuvant therapy is effective for high-risk resected melanoma patients with BRAF-V600 mutations. However, real-world evidence is limited. We aimed to determine the feasibility of this therapy in routine clinical practice. DESCRIBE-AD, a retrospective observational study, collected real-world data from 25 hospitals in Spain. Histologically confirmed and resected BRAF-mutated melanoma patients aged & GE;18 years who were previously treated with dabrafenib plus trametinib adjuvant therapy, were included. The primary objectives were treatment discontinuation rate and time to discontinuation. The secondary objectives included safety and efficacy. From October 2020 to March 2021, 65 patients were included. Dabrafenib and trametinib discontinuation rate due to treatment-related adverse events (TRAEs) of any grade was 9%. Other reasons for discontinuation included patients' decisions (6%), physician decisions (6%), unrelated adverse events (3%), disease progression (5%), and others (5%). The median time to treatment discontinuation was 9 months [95% confidence interval (CI), 5-11]. G3-4 TRAEs occurred in 21.5% of patients, the most common being pyrexia (3%), asthenia (3%), and diarrhoea (3%). Unscheduled hospitalisations and clinical tests occurred in 6 and 22% of patients, respectively. After 20-month median follow-up (95% CI, 18-22), 9% of patients had exitus due to disease progression, with a 12-month relapse-free survival and overall survival rates of 95.3% and 100%, respectively. Dabrafenib and trametinib adjuvant therapy proved effective for melanoma patients in a real-world setting, with a manageable toxicity profile.     Prognostic significance of FLT3-ITD length in AML patients treated with intensive regimens

    FLT3-ITD mutations are detected in approximately 25% of newly diagnosed adult acute myeloid leukemia (AML) patients and confer an adverse prognosis. The FLT3-ITD allelic ratio has clear prognostic value. Nevertheless, there are numerous manuscripts with contradictory results regarding the prognostic relevance of the length and insertion site (IS) of the FLT3-ITD fragment. We aimed to assess the prognostic impact of these variables on the complete remission (CR) rates, overall survival (OS) and relapse-free survival (RFS) of AML patients with FLT3-ITDmutations. We studied the FLT3-ITD length of 362 adult AML patients included in the PETHEMA AML registry. We tried to validate the thresholds of ITD length previously published (i.e., 39 bp and 70 bp) in intensively treated AML patients (n = 161). We also analyzed the mutational profile of 118 FLT3-ITD AML patients with an NGS panel of 39 genes and correlated mutational status with the length and IS of ITD. The AUC of the ROC curve of the ITD length for OS prediction was 0.504, and no differences were found when applying any of the thresholds for OS, RFS or CR rate. Only four out of 106 patients had ITD IS in the TKD1 domain. Our results, alongside previous publications, confirm that FLT3-ITD length lacks prognostic value and clinical applicability. © 2021, The Author(s)

    Antecedentes. Las lesiones por quemaduras se han convertidoen un problema de salud pública, especialmente enpaíses en desarrollo.Objetivo. Identificar la epidemiología del trauma por quemadurasen la población que consultó al servicio de urgenciasdel Hospital Infantil “Rafael Henao Toro” de la ciudadde Manizales en el período comprendido entre el año 2004y 2005.Material y métodos. Se realizó un estudio descriptivoretrospectivo con base en la revisión de 439 historias clínicas,evaluando las variables de edad, género, seguridad social,procedencia, área geográfica, causa, profundidad, gravedad,extensión en porcentaje, área corporal comprometida,estancia hospitalaria y compañía.Resultados. Se encontró que la mayoría de las quemadurasocurrieron en pacientes de un año de edad (21,6%),predominó el género masculino (59%). La mayoría notenía seguridad social (52,2%). El 44,9 por ciento de lospacientes residía en la ciudad de Manizales. La principaletiología fueron los alimentos en 194 pacientes (44,2%)predominando el grado I de quemadura en un 78,1 porciento, con gravedad moderada en 314 pacientes (71,5%).La media de estancia hospitalaria fue 11,99 días.Conclusiones. Se determinó una mayor frecuencia de quemadurasen niños, menores de cinco años, causadas poralimentos, en miembro superior y de gravedad moderada
