    Incubation Behaviour of Common Eiders Somateria Mollissima in the Central Baltic: Nest Attendance and Loss in Body Mass

    Here we present the recording of body mass change and weight loss during incubation in a Common Eider Somateria mollissima colony at Christiansø in the Central Baltic (55°19'N 15°11'E). The study was conducted during April and May 2015 and a total number of four birds were followed (two were lost due to predation and three due to power outages). Body mass and nesting behaviour was recorded electronically over a period of 26-27 days using automatic poultry scales and a surveillance video camera. During incubation, the eiders underwent a 28-37% loss in body mass and left the nest on average 13 times (range: 7-17 times) for a period of 7-70 min. In general, birds with high initial body mass left their nest for a shorter total time than birds with lower initial body mass. The recorded daily changes in body mass indicate that the eiders foraged during the incubation period, not just leaving the nest for rehydration or in response to disturbance, which improve our current understanding of eider incubation behaviour. Such information is important to fully understanding of eider breeding biology in order to better conserve and manage the species during its breeding seasons where individual birds undergo extreme stress that may affect reproductive outcome and adult survival

    New insights into the wheat chromosome 4D structure and virtual gene order, revealed by survey pyrosequencing

    AbstractSurvey sequencing of the bread wheat (Triticum aestivum L.) genome (AABBDD) has been approached through different strategies delivering important information. However, the current wheat sequence knowledge is not complete. The aim of our study is to provide different and complementary set of data for chromosome 4D. A survey sequence was obtained by pyrosequencing of flow-sorted 4DS (7.2×) and 4DL (4.1×) arms. Single ends (SE) and long mate pairs (LMP) reads were assembled into contigs (223Mb) and scaffolds (65Mb) that were aligned to Aegilops tauschii draft genome (DD), anchoring 34Mb to chromosome 4. Scaffolds annotation rendered 822 gene models. A virtual gene order comprising 1973 wheat orthologous gene loci and 381 wheat gene models was built. This order was largely consistent with the scaffold order determined based on a published high density map from the Ae. tauschii chromosome 4, using bin-mapped 4D ESTs as a common reference. The virtual order showed a higher collinearity with homeologous 4B compared to 4A. Additionally, a virtual map was constructed and ∼5700 genes (∼2200 on 4DS and ∼3500 on 4DL) predicted.     Phase 3 Trial of 177Lu-Dotatate for Midgut Neuroendocrine Tumors

    Background Patients with advanced midgut neuroendocrine tumors who have had disease progression during first-line somatostatin analogue therapy have limited therapeutic options. This randomized, controlled trial evaluated the efficacy and safety of lutetium-177 (177Lu)-Dotatate in patients with advanced, progressive, somatostatin-receptor-positive midgut neuroendocrine tumors. Methods We randomly assigned 229 patients who had well-differentiated, metastatic midgut neuroendocrine tumors to receive either 177Lu-Dotatate (116 patients) at a dose of 7.4 GBq every 8 weeks (four intravenous infusions, plus best supportive care including octreotide long-acting repeatable [LAR] administered intramuscularly at a dose of 30 mg) (177Lu-Dotatate group) or octreotide LAR alone (113 patients) administered intramuscularly at a dose of 60 mg every 4 weeks (control group). The primary end point was progression-free survival. Secondary end points included the objective response rate, overall survival, safety, and the side-effect profile. The final analysis of overall survival will be conducted in the future as specified in the protocol; a prespecified interim analysis of overall survival was conducted and is reported here. Results At the data-cutoff date for the primary analysis, the estimated rate of progression-free survival at month 20 was 65.2% (95% confidence interval [CI], 50.0 to 76.8) in the 177Lu-Dotatate group and 10.8% (95% CI, 3.5 to 23.0) in the control group. The response rate was 18% in the 177Lu-Dotatate group versus 3% in the control group (P<0.001). In the planned interim analysis of overall survival, 14 deaths occurred in the 177Lu-Dotatate group and 26 in the control group (P=0.004). Grade 3 or 4 neutropenia, thrombocytopenia, and lymphopenia occurred in 1%, 2%, and 9%, respectively, of patients in the 177Lu-Dotatate group as compared with no patients in the control group, with no evidence of renal toxic effects during the observed time frame. Conclusions Treatment with 177Lu-Dotatate resulted in markedly longer progression-free survival and a significantly higher response rate than high-dose octreotide LAR among patients with advanced midgut neuroendocrine tumors. Preliminary evidence of an overall survival benefit was seen in an interim analysis; confirmation will be required in the planned final analysis. Clinically significant myelosuppression occurred in less than 10% of patients in the 177Lu-Dotatate group.     Lead concentrations in blood from incubating common eiders (Somateria mollissima) in the Baltic Sea

    Here we investigate if lead may be a contributing factor to the observed population decline in a Baltic colony of incubating eiders (Somateria mollissima). Body mass and blood samples were obtained from 50 incubating female eiders at the Baltic breeding colony on Christianso during spring 2017 (n = 27) and 2018 (n = 23). All the females were sampled twice during early (day 4) and late (day 24) incubation. The full blood was analysed for lead to investigate if the concentrations exceeded toxic thresholds or changed over the incubation period due to remobilisation from bones and liver tissue. Body mass, hatch date and number of chicks were also analysed with respect to lead concentrations. The body mass (mean +/- SD g) increased significantly in the order: day 24 in 2018 (1561 +/- 154 g) < day 24 in 2017 (1618 +/- 156 g) < day 4 in 2018 (2183 +/- 140 g) < day 4 in 2017 (2359 +/- 167 g) (all p < 0.001). The lead concentrations increased significantly in the opposite order i.e. day 4 in 2017 (41.7 +/- 67.1 mu g/L) < day 24 in 2017 (55.4 +/- 66.8 mu g/L) < day 4 in 2018 (177 +/- 196 mu g/L) < day 24 in 2018 (258 +/- 243) (all p < 0.001). From day 4 to 24, the eider females had a 1.33-fold increase in blood lead concentrations in 2017 and a 1.46-fold increase in 2018. Three of the birds (13%) sampled in 2018 had lead concentrations that exceeded concentrations of clinical poisoning (500 mu g/L) and eleven (48%) had concentrations that exceeded the threshold for subclinical poisoning (200 mu g/L). In 2017, none of the birds exceeded the high toxic threshold of clinical poisoning while only one (4%) exceeded the lower threshold for subclinical poisoning. Three of the birds (6%) sampled in 2018 had lead concentrations that exceeded those of clinical poisoning while 12 birds (24%) resampled in both years exceeded the threshold for subclinical poisoning. In addition, lead concentrations and body mass on day 4 affected hatch date positively in 2018 (both p < 0.03) but not in 2017. These results show that bioavailable lead in bone and liver tissue pose a threat to the health of about 25% of the incubating eiders sampled. This is particularly critical because eiders are largely capital breeding which means that incubating eiders are in an energetically stressed state. The origin of lead in incubating eiders in the Christianso colony is unknown and it remains an urgent priority to establish the source, prevalence and mechanism for uptake. The increase in lead from day 4 to day 24 is due to bone and liver remobilization; however, the additional lead source(s) on the breeding grounds needs to be identified. Continued investigations should determine the origin, uptake mechanisms and degree of exposure to lead for individual birds. Such research should include necropsies, x-ray, lead isotope and stable C and N isotope analyses to find the lead sources(s) in the course of the annual cycle and how it may affect the population dynamics of the Christianso colony which reflects the ecology of the Baltic eiders being suitable for biomonitoring the overall flyway

    The effect of collagen organization on tensile strength loss in anterior cruciate ligament grafts post-reconstruction surgery

    Gemstone Team LEGSGrafts used for anterior cruciate ligament (ACL) reconstructions fall short of restoring native mechanics. This study investigated a morphological cause for tension loss by comparing native ACL and two common grafts, bone-patellar tendonbone (BPTB) and semitendinosus/gracilis hamstring tendon (ST/G), in a cadaveric system. Tension loss during continuous passive motion was quantified via force transducer. Microstructural changes were assessed by measuring collagen crimp angles. No significant differences were found for rates of percent tension loss relative to total tension loss among grafts. However, all groups displayed exponential decay, implying rapid tension loss. The crimp angles for the unstressed grafts were significantly different from each other, suggesting innate differences. The percent change experienced by stressed grafts, normalized to their unstressed baselines, showed that ST’s crimp behavior was significantly different from that of ACL and BPTB, implying the BPTB graft is superior for ACL reconstruction because it better mimics the ACL’s morphological behavior

    Barriers and incentives to orphan care in a time of AIDS and economic crisis: a cross-sectional survey of caregivers in rural Zimbabwe

    BACKGROUND: Africa is in an orphan-care crisis. In Zimbabwe, where one-fourth of adults are HIV-positive and one-fifth of children are orphans, AIDS and economic decline are straining society's ability to care for orphans within their extended families. Lack of stable care is putting thousands of children at heightened risk of malnourishment, emotional underdevelopment, illiteracy, poverty, sexual exploitation, and HIV infection, endangering the future health of the society they are expected to sustain. METHODS: To explore barriers and possible incentives to orphan care, a quantitative cross-sectional survey in rural eastern Zimbabwe asked 371 adults caring for children, including 212 caring for double orphans, about their well-being, needs, resources, and perceptions and experiences of orphan care. RESULTS: Survey responses indicate that: 1) foster caregivers are disproportionately female, older, poor, and without a spouse; 2) 98% of non-foster caregivers are willing to foster orphans, many from outside their kinship network; 3) poverty is the primary barrier to fostering; 4) financial, physical, and emotional stress levels are high among current and potential fosterers; 5) financial need may be greatest in single-orphan AIDS-impoverished households; and 6) struggling families lack external support. CONCLUSION: Incentives for sustainable orphan care should focus on financial assistance, starting with free schooling, and development of community mechanisms to identify and support children in need, to evaluate and strengthen families' capacity to provide orphan care, and to initiate and support placement outside the family when necessary

    Lipid Classes and Fatty Acid Patterns are Altered in the Brain of γ-Synuclein Null Mutant Mice

    The well-documented link between α-synuclein and the pathology of common human neurodegenerative diseases has increased attention to the synuclein protein family. The involvement of α-synuclein in lipid metabolism in both normal and diseased nervous system has been shown by many research groups. However, the possible involvement of γ-synuclein, a closely-related member of the synuclein family, in these processes has hardly been addressed. In this study, the effect of γ-synuclein deficiency on the lipid composition and fatty acid patterns of individual lipids from two brain regions has been studied using a mouse model. The level of phosphatidylserine (PtdSer) was increased in the midbrain whereas no changes in the relative proportions of membrane polar lipids were observed in the cortex of γ-synuclein-deficient compared to wild-type (WT) mice. In addition, higher levels of docosahexaenoic acid were found in PtdSer and phosphatidylethanolamine (PtdEtn) from the cerebral cortex of γ-synuclein null mutant mice. These findings show that γ-synuclein deficiency leads to alterations in the lipid profile in brain tissues and suggest that this protein, like α-synuclein, might affect neuronal function via modulation of lipid metabolism
