    Improving the Clinical Application of Natural Killer Cells by Modulating Signals Signal from Target Cells

    Relapsed acute myeloid leukemia (AML) is a significant post-transplant complication lacking standard treatment and associated with a poor prognosis. Cellular therapy, which is already widely used as a treatment for several hematological malignancies, could be a potential treatment alternative. Natural killer (NK) cells play an important role in relapse control but can be inhibited by the leukemia cells highly positive for HLA class I. In order to restore NK cell activity after their ex vivo activation, NK cells can be combined with conditioning target cells. In this study, we tested NK cell activity against KG1a (AML cell line) with and without two types of pretreatment-Ara-C treatment that induced NKG2D ligands (increased activating signal) and/or blocking of HLA-KIR (killer-immunoglobulin-like receptors) interaction (decreased inhibitory signal). Both treatments improved NK cell killing activity. Compared with target cell killing of NK cells alone (38%), co-culture with Ara-C treated KG1a target cells increased the killing to 80%. Anti-HLA blocking antibody treatment increased the proportion of dead KG1a cells to 53%. Interestingly, the use of the combination treatment improved the killing potential to led to the death of 85% of KG1a cells. The combination of Ara-C and ex vivo activation of NK cells has the potential to be a feasible approach to treat relapsed AML after hematopoietic stem cell transplantation

    Efecto de la densidad de siembra sobre el rendimiento de la okra (Abelmoschus esculentus (L.) Moench), en el valle de Lerma, Salta, Argentina

    La okra es una malvácea anual cultivada como hortaliza adaptada a climas tropicales y subtropicales. El manejo del cultivo es poco conocido en Salta y Argentina. El objetivo fue determinar la densidad de siembra más adecuada en el valle de Lerma evaluando su efecto sobre las componentes del rendimiento. Con un diseño de bloques completos al azar, con 12 réplicas, se compararon cuatro densidades de siembra de okra variedad Blue Sky que fueron 1, 2, 3 y 4 plantas por pozo, (T1, T2, T3 y T4), con filas a 70 cm y hoyos a 30 cm. Se sembró el 25/10/11 y la emergencia fue a los 21 días. Se cosecharon frutos inmaduros entre el 23/12/11 y el 28/03/12, con un tamaño entre 7-10 cm de largo. Se evaluó el nú- mero y peso total de frutos cosechados en todo el ciclo de producción y por fecha de cosecha. Para todas las variables analizadas, los menores valores promedio se observaron en T1, mientras que entre T2, T3 y T4, si bien se vio una tendencia en aumento, no existieron diferencias estadísticamente significativas. Por consiguiente y debido a la dificultad de conseguir la semilla en el país y por su alto costo, se recomienda sembrar dos semillas por pozo.Okra is an annual Malvaceae cultivated as a vegetable crop and adapted to tropical and subtropical climate. There is no much crop management information in Salta and Argentina. The goal of this research was to determine the best plant density in the Lerma valley, Salta, Argentina. Treatments were four okra plant populations, cultivar Blue Sky (1, 2, 3 and 4 plants per hill: T1, T2, T3 y T4) evaluating its effect on the yield components. Experimental design was a randomized complete block with 12 blocks. The experiment was sown on October 25, 2011 and seed emergency happen 21 days after sowing. Fruits were harvested at immature stage from 23/12/11 to 28/03/12, with a size length from 7 to 10 cm. The parameters measured were: number and total fruit weight harvested in the whole production cycle and for each harvest date. For all parameters analyzed, the lower average values were observed in T1, but there were not statiscally differences between T2, T3 and T4. Nevertheless, there was a growing trend, although not statistically significant.     Brd2/4 and Myc regulate alternative cell lineage programmes during early osteoclast differentiation in vitro

    Osteoclast (OC) development in response to nuclear factor kappa-Β ligand (RANKL) is critical for bone homeostasis in health and in disease. The early and direct chromatin regulatory changes imparted by the BET chromatin readers Brd2-4 and OC-affiliated transcription factors (TFs) during osteoclastogenesis are not known. Here, we demonstrate that in response to RANKL, early OC development entails regulation of two alternative cell fate transcriptional programmes, OC vs macrophage, with repression of the latter following activation of the former. Both programmes are regulated in a non-redundant manner by increased chromatin binding of Brd2 at promoters and of Brd4 at enhancers/super-enhancers. Myc, the top RANKL-induced TF, regulates OC development in co-operation with Brd2/4 and Max and by establishing negative and positive regulatory loops with other lineage-affiliated TFs. These insights into the transcriptional regulation of osteoclastogenesis suggest the clinical potential of selective targeting of Brd2/4 to abrogate pathological OC activation

    Chromatin-based, in cis and in trans regulatory rewiring underpins distinct oncogenic transcriptomes in multiple myeloma

    Multiple myeloma is a genetically heterogeneous cancer of the bone marrow plasma cells (PC). Distinct myeloma transcriptome profiles are primarily driven by myeloma initiating events (MIE) and converge into a mutually exclusive overexpression of the CCND1 and CCND2 oncogenes. Here, with reference to their normal counterparts, we find that myeloma PC enhanced chromatin accessibility combined with paired transcriptome profiling can classify MIE-defined genetic subgroups. Across and within different MM genetic subgroups, we ascribe regulation of genes and pathways critical for myeloma biology to unique or shared, developmentally activated or de novo formed candidate enhancers. Such enhancers co-opt recruitment of existing transcription factors, which although not transcriptionally deregulated per se, organise aberrant gene regulatory networks that help identify myeloma cell dependencies with prognostic impact. Finally, we identify and validate the critical super-enhancer that regulates ectopic expression of CCND2 in a subset of patients with MM and in chronic lymphocytic leukemia

    Reconstructing Haemodynamics Quantities of Interest from Doppler Ultrasound Imaging

    The present contribution deals with the estimation of haemodynamics Quantities of Interest by exploiting Ultrasound Doppler measurements. A fast method is proposed, based on the PBDW method. Several methodological contributions are described: a sub-manifold partitioning is introduced to improve the reduced-order approximation, two different ways to estimate the pressure drop are compared, and an error estimation is derived. A test-case on a realistic common carotid geometry is presented, showing that the proposed approach is promising in view of realistic applications.Comment: arXiv admin note: text overlap with arXiv:1904.1336

    Algorithm Engineering in Robust Optimization

    Robust optimization is a young and emerging field of research having received a considerable increase of interest over the last decade. In this paper, we argue that the the algorithm engineering methodology fits very well to the field of robust optimization and yields a rewarding new perspective on both the current state of research and open research directions. To this end we go through the algorithm engineering cycle of design and analysis of concepts, development and implementation of algorithms, and theoretical and experimental evaluation. We show that many ideas of algorithm engineering have already been applied in publications on robust optimization. Most work on robust optimization is devoted to analysis of the concepts and the development of algorithms, some papers deal with the evaluation of a particular concept in case studies, and work on comparison of concepts just starts. What is still a drawback in many papers on robustness is the missing link to include the results of the experiments again in the design

    Science Objectives for an X-Ray Microcalorimeter Observing the Sun

    We present the science case for a broadband X-ray imager with high-resolution spectroscopy, including simulations of X-ray spectral diagnostics of both active regions and solar flares. This is part of a trilogy of white papers discussing science, instrument (Bandler et al. 2010), and missions (Bookbinder et al. 2010) to exploit major advances recently made in transition-edge sensor (TES) detector technology that enable resolution better than 2 eV in an array that can handle high count rates. Combined with a modest X-ray mirror, this instrument would combine arcsecondscale imaging with high-resolution spectra over a field of view sufficiently large for the study of active regions and flares, enabling a wide range of studies such as the detection of microheating in active regions, ion-resolved velocity flows, and the presence of non-thermal electrons in hot plasmas. It would also enable more direct comparisons between solar and stellar soft X-ray spectra, a waveband in which (unusually) we currently have much better stellar data than we do of the Sun

    Dental Health and Mortality in People With End-Stage Kidney Disease Treated With Hemodialysis: A Multinational Cohort Study

    Background Dental disease is more extensive in adults with chronic kidney disease, but whether dental health and behaviors are associated with survival in the setting of hemodialysis is unknown. Study Design Prospective multinational cohort. Setting & Participants 4,205 adults treated with long-term hemodialysis, 2010 to 2012 (Oral Diseases in Hemodialysis [ORAL-D] Study). Predictors Dental health as assessed by a standardized dental examination using World Health Organization guidelines and personal oral care, including edentulousness; decayed, missing, and filled teeth index; teeth brushing and flossing; and dental health consultation. Outcomes All-cause and cardiovascular mortality at 12 months after dental assessment. Measurements Multivariable-adjusted Cox proportional hazards regression models fitted with shared frailty to account for clustering of mortality risk within countries. Results During a mean follow-up of 22.1 months, 942 deaths occurred, including 477 cardiovascular deaths. Edentulousness (adjusted HR, 1.29; 95% CI, 1.10-1.51) and decayed, missing, or filled teeth score ≥ 14 (adjusted HR, 1.70; 95% CI, 1.33-2.17) were associated with early all-cause mortality, while dental flossing, using mouthwash, brushing teeth daily, spending at least 2 minutes on oral hygiene daily, changing a toothbrush at least every 3 months, and visiting a dentist within the past 6 months (adjusted HRs of 0.52 [95% CI, 0.32-0.85], 0.79 [95% CI, 0.64-0.97], 0.76 [95% CI, 0.58-0.99], 0.84 [95% CI, 0.71-0.99], 0.79 [95% CI, 0.65-0.95], and 0.79 [95% CI, 0.65-0.96], respectively) were associated with better survival. Results for cardiovascular mortality were similar. Limitations Convenience sample of clinics. Conclusions In adults treated with hemodialysis, poorer dental health was associated with early death, whereas preventive dental health practices were associated with longer survival
