    Protective Yeasts Control V. anguillarum Pathogenicity and Modulate the Innate Immune Response of Challenged Zebrafish (Danio rerio) Larvae

    Indexación: Web of ScienceWe investigated mechanisms involved in the protection of zebrafish (Danio rerio) larvae by two probiotic candidate yeasts, Debaryornyces hansenii 97 (Dh97) and Yarrowia Iypolitica 242 (YI242), against a Vibrio anguillarum challenge. We determined the effect of different yeast concentrations (10(4)-10(7) CFU/mL) to: (i) protect larvae from the challenge, (ii) reduce the in vivo pathogen concentration and (iii) modulate the innate immune response of the host. To evaluate the role of zebrafish microbiota in protection, the experiments were performed in conventionally raised and germ free larvae. In vitro co-aggregation assays were performed to determine a direct yeast-pathogen interaction. Results showed that both yeasts significantly increased the survival rate of conventionally raised larvae challenged with V. anguillarum. The concentration of yeasts in larvae tended to increase with yeast inoculum, which was more pronounced for Dh97. Better protection was observed with Dh97 at a concentration of 106 CFU/mL compared to 104 CFU/mL. In germ-free conditions V anguillarum reached higher concentrations in larvae and provoked significantly more mortality than in conventional conditions, revealing the protective role of the host microbiota. Interestingly, yeasts were equally (Dh97) or more effective (YI242) in protecting germ-free than conventionally-raised larvae, showing that protection can be exerted only by yeasts and is not necessarily related to modulation of the host microbiota. Although none of the yeasts co aggregated with V anguillarum, they were able to reduce its proliferation in conventionally raised larvae, reduce initial pathogen concentration in germ-free larvae and prevent the upregulation of key components of the inflammatory/anti-inflammatory response (il1b, tnfa, c3, mpx, and il10, respectively). These results show that protection by yeasts of zebrafish larvae challenged with V anguillarum relates to an in vivo anti-pathogen effect, the modulation of the innate immune system, and suggests that yeasts avoid the host-pathogen interaction through mechanisms independent of co-aggregation. This study shows, for the first time, the protective role of zebrafish microbiota against V. anguillarum infection, and reveals mechanisms involved in protection by two non-Saccharomyces yeasts against this pathogen.http://journal.frontiersin.org/article/10.3389/fcimb.2016.00127/ful

    Metodologías de enseñanza-aprendizaje y su relación con la motivación e implicación del alumnado en las clases de Educación Física

    Teniendo en cuenta los bajos índices de motivación y participación del alumnado durante las clases de Educa-ción Física (EF), el objetivo del presente estudio es anali-zar la relación entre la percepción del alumnado sobre la metodología de enseñanza-aprendizaje empleada por los docentes (constructiva, productiva, reproductiva y de ac-ción libre) y la motivación e implicación mostrada durante las clases de EF. Los participantes fueron 465 estudiantes de sexo masculino (n = 223) y femenino (n = 242), con eda-des comprendidas entre los 10 y los 14 años (M = 11.36; DT = 0.91) pertenecientes a Educación Primaria (5º y 6º curso, n = 293) y Educación Secundaria (1º y 2º curso, n = 172). A través de un estudio con un diseño correlacional y de corte transversal, el alumnado rellenó cuestionarios que medían las metodologías de enseñanza, la motivación y la implicación en las clases. A través de un modelo de ecuaciones estructurales, los resultados obtenidos mos-traron que las metodologías constructiva y reproductiva se asociaron positivamente con la motivación autónoma. Además, las metodologías productiva y reproductiva se relacionaron con la motivación controlada, mientras que la metodología productiva se asoció con la desmotiva-ción. Por último, hay que señalar que únicamente la motivación autónoma se asoció con la implicación del alum-nado. Por tanto, el profesorado de EF debería apostar por metodologías más centradas en el alumnado en lugar de metodologías centradas en el docente, ya que son las que más se relacionan con una mayor calidad de la motivación e implicación en las clases de los estudiantesGiven the low motivation and engagements of some students during the Physical Education (PE) lessons, the main aim of this study is to analyze the relationship between the learning methods adopted by teachers (constructive, productive, reproductive, and laissez-faire) from the students’ perspective, and the motivational processes and the engagement of the students during the PE lessons. Participants were 465 male (n = 223) and female students (n = 242) aged between 10 and 14 years-old (M = 11.36; SD = 0.91) belonged to Elementary Education (fifth and sixth grade, n = 293) and Secondary Education (first and second grade, n = 172). Through a study with a correlational and cross-sectional design, students fulfillment self-reported questionnaires about teaching-learning methodologies, motivation and engagement. A structural equation modelling was used. The results showed that the constructive and reproductive methodologies were associated with the autonomous motivation. In addition, the productive and reproductive methodologies were related to the controlled motivation, whereas the productive methodology were associated with the amotivation. Finally, only the autonomous motivation was significant related to the engagement for PE lessons. Therefore, due to these obtained results, PE teachers should select more student-centered methodologies instead of teacher-centered methodologies, with the aim to get a higher quality student-motivation and engagement in the development of PE lessonsActividad Física y Deport

    Canagliflozin and renal outcomes in type 2 diabetes and nephropathy

    BACKGROUND Type 2 diabetes mellitus is the leading cause of kidney failure worldwide, but few effective long-term treatments are available. In cardiovascular trials of inhibitors of sodium–glucose cotransporter 2 (SGLT2), exploratory results have suggested that such drugs may improve renal outcomes in patients with type 2 diabetes. METHODS In this double-blind, randomized trial, we assigned patients with type 2 diabetes and albuminuric chronic kidney disease to receive canagliflozin, an oral SGLT2 inhibitor, at a dose of 100 mg daily or placebo. All the patients had an estimated glomerular filtration rate (GFR) of 30 to <90 ml per minute per 1.73 m2 of body-surface area and albuminuria (ratio of albumin [mg] to creatinine [g], >300 to 5000) and were treated with renin–angiotensin system blockade. The primary outcome was a composite of end-stage kidney disease (dialysis, transplantation, or a sustained estimated GFR of <15 ml per minute per 1.73 m2), a doubling of the serum creatinine level, or death from renal or cardiovascular causes. Prespecified secondary outcomes were tested hierarchically. RESULTS The trial was stopped early after a planned interim analysis on the recommendation of the data and safety monitoring committee. At that time, 4401 patients had undergone randomization, with a median follow-up of 2.62 years. The relative risk of the primary outcome was 30% lower in the canagliflozin group than in the placebo group, with event rates of 43.2 and 61.2 per 1000 patient-years, respectively (hazard ratio, 0.70; 95% confidence interval [CI], 0.59 to 0.82; P=0.00001). The relative risk of the renal-specific composite of end-stage kidney disease, a doubling of the creatinine level, or death from renal causes was lower by 34% (hazard ratio, 0.66; 95% CI, 0.53 to 0.81; P<0.001), and the relative risk of end-stage kidney disease was lower by 32% (hazard ratio, 0.68; 95% CI, 0.54 to 0.86; P=0.002). The canagliflozin group also had a lower risk of cardiovascular death, myocardial infarction, or stroke (hazard ratio, 0.80; 95% CI, 0.67 to 0.95; P=0.01) and hospitalization for heart failure (hazard ratio, 0.61; 95% CI, 0.47 to 0.80; P<0.001). There were no significant differences in rates of amputation or fracture. CONCLUSIONS In patients with type 2 diabetes and kidney disease, the risk of kidney failure and cardiovascular events was lower in the canagliflozin group than in the placebo group at a median follow-up of 2.62 years

    Omecamtiv mecarbil in chronic heart failure with reduced ejection fraction, GALACTIC‐HF: baseline characteristics and comparison with contemporary clinical trials

    Aims: The safety and efficacy of the novel selective cardiac myosin activator, omecamtiv mecarbil, in patients with heart failure with reduced ejection fraction (HFrEF) is tested in the Global Approach to Lowering Adverse Cardiac outcomes Through Improving Contractility in Heart Failure (GALACTIC‐HF) trial. Here we describe the baseline characteristics of participants in GALACTIC‐HF and how these compare with other contemporary trials. Methods and Results: Adults with established HFrEF, New York Heart Association functional class (NYHA) ≥ II, EF ≤35%, elevated natriuretic peptides and either current hospitalization for HF or history of hospitalization/ emergency department visit for HF within a year were randomized to either placebo or omecamtiv mecarbil (pharmacokinetic‐guided dosing: 25, 37.5 or 50 mg bid). 8256 patients [male (79%), non‐white (22%), mean age 65 years] were enrolled with a mean EF 27%, ischemic etiology in 54%, NYHA II 53% and III/IV 47%, and median NT‐proBNP 1971 pg/mL. HF therapies at baseline were among the most effectively employed in contemporary HF trials. GALACTIC‐HF randomized patients representative of recent HF registries and trials with substantial numbers of patients also having characteristics understudied in previous trials including more from North America (n = 1386), enrolled as inpatients (n = 2084), systolic blood pressure < 100 mmHg (n = 1127), estimated glomerular filtration rate < 30 mL/min/1.73 m2 (n = 528), and treated with sacubitril‐valsartan at baseline (n = 1594). Conclusions: GALACTIC‐HF enrolled a well‐treated, high‐risk population from both inpatient and outpatient settings, which will provide a definitive evaluation of the efficacy and safety of this novel therapy, as well as informing its potential future implementation

    The Effect of the Video Assistant Referee System Implementation on Match Physical Demands in the Spanish LaLiga

    The present study aimed to analyze the influence of the Video Assistant Referee (VAR) on match physical demands in the top Spanish professional football league. Match physical demand data from all the matches for two seasons (2017/2018 and 2018/2019) in the First Spanish Division (n = 1454) were recorded using an optical tracking system (ChyronHego®). Total distance, relative total distance covered per minute, distance covered between 14–21 km·h−1, distance covered between 21–24 km·h−1, and distance covered at more than 24 km·h−1 were analyzed; also, the number of sprints between 21–24 km·h−1 and more than 24 km·h−1 were taken into consideration. The times the VAR intervened in matches were also taken into account. Results showed that total distance and relative total distance significantly decreased in seasons with VAR compared to seasons without VAR. Finally, distance covered between 21–24 km·h−1, distance covered at more than 24 km·h−1, and the number of high-intensity efforts between 21–24 km·h−1 and more than 24 km·h−1 increased in seasons with VAR compared to seasons without VAR, but the differences were nonsignificant. Thus, these findings help practitioners to better understand the effects of the VAR system on professional football physical performance and to identify strategies to reproduce competition demands

    The Influence of Time Winning and Time Losing on Position-Specific Match Physical Demands in the Top One Spanish Soccer League

    The aim of the present study was to analyze the influence of time winning and time losing on position-specific match physical demands with and without ball possession in the top Spanish professional soccer league. All matches played in the First Spanish soccer league over four consecutive seasons (from 2015/16 to 2018/19) were recorded using an optical tracking system (i.e., ChyronHego), and the data were analyzed via Mediacoach®. Total distance (TD), and TD > 21 km·h−1 covered with and without ball possession were analyzed using a Linear Mixed Model, taking into account the contextual variables time winning and losing. Results showed that TD and TD > 21 km·h−1 covered by central midfielders (0.01 and 0.005 m/min, respectively), wide midfielders (0.02 and 0.01 m/min, respectively), and forwards (0.03 and 0.02 m/min, respectively) significantly increased while winning (p < 0.05). By contrast, TD and TD > 21 km·h−1 covered by central defenders (0.01 and 0.008 m/min, respectively) and wide defenders (0.06 and 0.008 m/min, respectively) significantly increased while losing (p < 0.05). In addition, for each minute that teams were winning, total distance with ball possession (TDWP) decreased, while, for each minute that teams were losing, TDWP increased. Instead, TDWP > 21 km·h−1 obtained opposite results. Total distance without ball possession increased when teams were winning, and decreased when teams were losing. Therefore, the evolution of scoreline significantly influences tactical–technical and physical demands on soccer matches
