    MALDI-MS argininyl bufadienolide esters fingerprint from parotoid gland secretions of Rhinella arenarum : age, gender and seasonal variation

    In many amphibians, the granular glands can be grouped in special regions forming macroglands. This is the case of toads, characterized by the presence of a pair of parotoid macroglands, strategically located to give protection by poison release in case of attacks. The product secreted consists of a wide variety of chemical compounds including proteins, peptides, biogenic amines, toxic steroidal bufadienolides, and various alkaloids, depending on the species. In this work, using Rhinella arenarum, we have performed, for the first time, the matrix assisted-ultraviolet laser desorption/ionization mass spectrometry and tandem mass spectrometry characterization of the components of the secretion used as crude material, just suspended in MeOH (or MeCN). The crude sample as a whole (whole suspension) was spotted on the matrix assisted-ultraviolet laser desorption plate for analysis. Electrospray ionization-Orbitrap was used for cross-checking experiments. The pattern of signals obtained at m/z ranges 600 to 800 and 1200 to 1600 could be assigned as the argininyl bufadienolide esters fingerprint characteristic of female and male.     Supporting the search for the CEP location with nonlocal PNJL models constrained by Lattice QCD

    We investigate the possible location of the critical endpoint in the QCD phase diagram based on nonlocal covariant PNJL models including a vector interaction channel. The form factors of the covariant interaction are constrained by lattice QCD data for the quark propagator. The comparison of our results for the pressure including the pion contribution and the scaled pressure shift ΔP/T4\Delta P / T^4 vs T/TcT/T_c with lattice QCD results shows a better agreement when Lorentzian formfactors for the nonlocal interactions and the wave function renormalization are considered. The strength of the vector coupling is used as a free parameter which influences results at finite baryochemical potential. It is used to adjust the slope of the pseudocritical temperature of the chiral phase transition at low baryochemical potential and the scaled pressure shift accessible in lattice QCD simulations. Our study, albeit presently performed at the meanfield level, supports the very existence of a critical point and favors its location within a region that is accessible in experiments at the NICA accelerator complex.Comment: 7 pages, 7 Figures.     Coping with verbal and social bullying in middle school

    Becoming a victim of verbal and social bullying in middle school can lead to illness, psychological stress, and maladjustment. The coping strategies that students utilize when they are bullied may influence the likelihood and severity of these negative effects. In this study, we examined the predictions made by students in two middle schools about the ways that they would cope with becoming a victim of verbal and social bullying. We also analyzed influences for coping strategies and student willingness to seek help with bullying at school. The results show that middle school students generally expect that they will utilize adaptive approach strategies in trying to solve the problem or obtain support from others, but those who had been victimized in the last month were more likely than those not involved in bullying, to predict that they would engage in maladaptive avoidance coping strategies if victimized in the future. Willingness to seek help was found to be enhanced by approach coping strategies, less aggressive attitudes, and lower perceptions of school bullying. Policy implications for efforts to encourage approach coping strategies in middle school students through educational interventions and school counseling are discussed.peer-reviewe

    Functionalisation of Ti6Al4V and hydroxyapatite surfaces with combined peptides based on KKLPDA and EEEEEEEE peptides

    Surface modifications are usually performed on titanium alloys to improve osteo-integration and surface bioactivity. Modifications such as alkaline and acid etching, or coating with bioactive materials such as hydroxyapatite, have previously been demonstrated. The aim of this work is to develop a peptide with combined titanium oxide and hydroxyapatite binders in order to achieve a biomimetic hydroxyapatite coating on titanium surfaces. The technology would also be applicable for the functionalisation of titanium and hydroxyapatite surfaces for selective protein adsorption, conjugation of antimicrobial peptides, and adsorption of specialised drugs for drug delivery. In this work, functionalisation of Ti6Al4V and hydroxyapatite surfaces was achieved using combined titanium-hydroxyapatite (Ti-Hap) peptides based on titanium binder (RKLPDA) and hydroxyapatite binder (EEEEEEEE) peptides. Homogeneous peptide coatings on Ti6Al4V surfaces were obtained after surface chemical treatments with a 30 wt % aqueous solution of H2O2 for 24 and 48 hours. The treated titanium surfaces presented an average roughness of Sa=197 nm (24 h) and Sa=128 nm (48 h); an untreated mirror polished sample exhibited an Sa of 13 nm. The advancing water contact angle of the titanium oxide layer after 1 hour of exposure to 30 wt % aqueous solution of H2O2 was around 65°, decreasing gradually with time until it reached 35° after a 48 hour exposure, suggesting that the surface hydrophilicity increased over etching time. The presence of a lysine (L) amino acid in the sequence of the titanium binder resulted in fluorescence intensity roughly 16 % higher compared with the arginine (R) amino acid analogue and therefore the lysine containing titanium binder was used in this work. The Ti-Hap peptide KKLPDAEEEEEEEE (Ti-Hap1) was not adsorbed by the treated Ti6Al4V surfaces and therefore was modified. The modifications involved the inclusion of a glycine spacer between the binding terminals (Ti-Hap2) and the addition of a second titanium binder (KKLPDA) (Ti-Hap3 and Ti-Hap4). The Ti-Hap peptide aptamer which exhibited the strongest intensity after the titanium dip coating was KKLPDAKKLPDAEEEEEEEE (Ti-Hap4). On the other hand, hydroxyapatite surfaces, exhibiting an average roughness of Sa=1.42 µm, showed a higher fluorescence for all peptides compared with titanium surfaces

    A study of the effects of exercise on the urinary metabolome using normalisation to individual metabolic output

    Aerobic exercise, in spite of its multi-organ benefit and potent effect on the metabolome, has yet to be investigated comprehensively via an untargeted metabolomics technology. We conducted an exploratory untargeted liquid chromatography mass spectrometry study to investigate the effects of a one-h aerobic exercise session in the urine of three physically active males. Individual urine samples were collected over a 37-h protocol (two pre-exercise and eight post-exercise). Raw data were subjected to a variety of normalization techniques, with the most effective measure dividing each metabolite by the sum response of that metabolite for each individual across the 37-h protocol expressed as a percentage. This allowed the metabolite responses to be plotted on a normalised scale. Our results highlight significant metabolites located in the following systems: purine pathway, tryptophan metabolism, carnitine metabolism, cortisol metabolism, androgen metabolism, amino acid oxidation, as well as metabolites from the gastrointestinal microbiome. Many of the significant changes observed in our pilot investigation mirror previous research studies, of various methodological designs, published within the last 15 years, although they have never been reported at the same time in a single study

    Small and medium-sized enterprises play a major role in national economy. However, their lifetime is limited by the micro-economic climate. This paper discusses whether credit exposure has a major impact on the rate of bankruptcy and this hypothesis is verified by means of statistical methods, based on official statistical data and selected published studies and researches. The hypothesis was not fully confirmed in our investigation. The output of the paper was an evaluation of the dependency of bankruptcy of SMEs on their credit exposure and the presentation of other factors that might have an impact of SMEs, making them either successful or going to bankruptcy.peer-reviewe

    Estructura, dinámica y vigencia de los convenios colectivos de trabajo sectoriales del ámbito privado (1975-2014)

    El artículo propone debatir con aquellos argumentos que consideran que, a pesar del intenso desarrollo de la negociación colectiva sucedida en Argentina entre 2003-2014, no tuvo lugar una significativa renegociación de los Convenios Colectivos de Trabajo (CCT) sectoriales acordados entre 1991-2002, etapa caracterizada por la flexibilización laboral. Metodológicamente, se analizaron los CCT sectoriales firmados entre 1975 y 2014. Esta muestra cubre el 95% de los asalariados formales del sector privado no agrícola. Los resultados de la investigación indican que actualmente el 64% de los CCT sectoriales negociados entre 1991 y 2002 se encuentran inactivos. Así, se demuestra que gran parte de los CCT sectoriales establecidos en la década de los '90 no muestran continuidad en su aplicación post 2003, evidencia que pone en cuestión otros estudios sobre la temática.The article discuss with those arguments that consider that, despite the intense development of collective bargaining occurred in Argentina between 2003-2014, it did not take place a significant renegotiation of sectoral Collective Bargaining Agreements (CBA) established between 1991-2002, a period characterized by labor flexibility. Methodologically, we analyzed the sectoral CBA signed between 1975 and 2014. This sample covers the 95% of formal workers in the non-agricultural private sector. The results arising from research indicate that currently the 64% of the sectoral CBA negotiated between 1991 and 2002 are inactive. Therefore, it is shown that much of the sectoral CBA established in the early 90's did not have continuity in their application, proving questionable the results of other studies on the same subject.Fil: Trajtemberg, David.     Effects of photoperiod extension on clock gene and neuropeptide RNA expression in the SCN of the Soay sheep

    In mammals, changing daylength (photoperiod) is the main synchronizer of seasonal functions. The photoperiodic information is transmitted through the retino-hypothalamic tract to the suprachiasmatic nuclei (SCN), site of the master circadian clock. To investigate effects of day length change on the sheep SCN, we used in-situ hybridization to assess the daily temporal organization of expression of circadian clock genes (Per1, Per2, Bmal1 and Fbxl21) and neuropeptides (Vip, Grp and Avp) in animals acclimated to a short photoperiod (SP; 8h of light) and at 3 or 15 days following transfer to a long photoperiod (LP3, LP15, respectively; 16h of light), achieved by an acute 8-h delay of lights off. We found that waveforms of SCN gene expression conformed to those previously seen in LP acclimated animals within 3 days of transfer to LP. Mean levels of expression for Per1-2 and Fbxl21 were nearly 2-fold higher in the LP15 than in the SP group. The expression of Vip was arrhythmic and unaffected by photoperiod, while, in contrast to rodents, Grp expression was not detectable within the sheep SCN. Expression of the circadian output gene Avp cycled robustly in all photoperiod groups with no detectable change in phasing. Overall these data suggest that synchronizing effects of light on SCN circadian organisation proceed similarly in ungulates and in rodents, despite differences in neuropeptide gene expression
