    The Effects of Dasatinib in Experimental Acute Respiratory Distress Syndrome Depend on Dose and Etiology

    Background/Aims: Evidence suggests that tyrosine-kinase inhibitors may attenuate lung inflammation and fibrosis in experimental acute respiratory distress syndrome (ARDS). We hypothesized that dasatinib, a tyrosine-kinase inhibitor, might act differently depending on the ARDS etiology and the dose. Methods: C57/BL6 mice were divided to be pre-treated with dasatinib (1mg/kg or 10mg/kg) or vehicle (1% dimethyl-sulfoxide) by oral gavage. Thirty-minutes after pre-treatment, mice were subdivided into control (C) or ARDS groups. ARDS animals received Escherichia coli lipopolysaccharide intratracheally (ARDSp) or intraperitoneally (ARDSexp). A new dose of dasatinib or vehicle was administered at 6 and 24h. Results: Forty-eight hours after ARDS induction, dasatinib 1mg/kg yielded: improved lung morphofunction and reduced cells expressing toll-like receptor (TLR)-4 in lung, independent of ARDS etiology; reduced neutrophil and levels of interleukin (IL)-6, IL-10 and transforming growth factor (TGF)-β in ARDSp. The higher dose of dasatinib caused no changes in lung mechanics, diffuse alveolar damage, neutrophil, or cells expressing TLR4, but increased IL-6, vascular endothelial growth factor (VEGF), and cells expressing Fas receptor in lung in ARDSp. In ARDSexp, it improved lung morphofunction, increased VEGF, and reduced cells expressing TLR4. Conclusion: Dasatinib may have therapeutic potential in ARDS independent of etiology, but careful dose monitoring is required. © 2015 S. Karger AG, Basel

    Steps to build a DIY low-cost fixed-wing drone for biodiversity conservation

    Despite the proved usefulness of drones in biodiversity studies, acquisition costs and difficulties in operating, maintaining and repairing these systems constrain their integration in conservation projects, particularly for low-income countries. Here we present the steps necessary to build a low-cost fixed-wing drone for environmental applications in large areas, along with instructions to increase the reliability of the system and testing its performance. Inspired by DIY (Do It Yourself) and open source models, this work prioritizes simplicity and accounts for cost-benefit for the researcher. The DIY fixed-wing drone developed has electric propulsion, can perform pre-programmed flight, can carry up to 500 g payload capacity with 65 minutes flight duration and flies at a maximum distance of 20 km. It is equipped with a RGB (Red, Green and Blue) sensor capable of obtaining 2.8 cm per pixel Ground Sample Distance (GSD) resolution at a constant altitude of 100 m above ground level (AGL). The total cost was $995 which is substantially less than the average value of similar commercial drones used in biodiversity studies. We performed 12 flight tests in auto mode using the developed model in protected areas in Brazil, obtaining RGB images that allowed us to identify deforestation spots smaller than 5 m2 and medium-sized animals. Building DIY drones requires some technical knowledge and demands more time than buying a commercial ready-to-fly system, but as proved here, it can be less expensive, which is often crucial in conservation projects

    Asymptotically Extrinsic Tamed Submanifolds

    We study, from the extrinsic point of view, the structure at infinity of open submanifolds, ϕ : Mm → Mn(κ) isometrically immersed in the real space forms of constant sectional curvature κ ≤ 0.We shall use the decay of the second fundamental form of the so-called tamed immersions to obtain a description at infinity of the submanifold in the line of the structural results in Greene et al.     Repair of composite-to-masonry bond using flexible matrix

    The paper presents an experimental investigation on an innovative repair method, in which composite reinforcements, after debonding, are re-bonded to the substrate using a highly deformable polymer. In order to assess the effectiveness of this solution, shear bond tests were carried out on brick and masonry substrates within two Round Robin Test series organized within the RILEM TC 250-CSM: Composites for Sustainable strengthening of Masonry. Five laboratories from Italy, Poland and Portugal were involved. The shear bond performance of the reinforcement systems before and after repair were compared in terms of ultimate loads, load-displacement curves and strain distributions. The results showed that the proposed repair method may provide higher strength and ductility than stiff epoxy resins, making it an effective and cost efficient technique for several perspective structural applications

    Economic analysis of antenatal screening for human T-cell lymphotropic virus type 1 in Brazil: an open access cost-utility model

    BACKGROUND: Human T-cell lymphotropic virus type 1 (HTLV-1) is a retrovirus that causes severe diseases, such as aggressive cancer or progressive neurological disease. HTLV-1 affects mainly people in areas with low human development index and can be transmitted from mother to child, primarily through breastfeeding. Refraining from breastfeeding is an effective intervention to reduce the risk of infection in infants. However, HTLV-1 antenatal screening is not offered globally. According to WHO, the scarcity of cost-effectiveness studies is considered one of the major barriers to the implementation of policies to prevent HTLV-1 infection. Therefore, this study aimed to assess the cost-effectiveness of antenatal screening and postnatal interventions to prevent HTLV-1 mother-to-child transmission in Brazil and to develop an open-access, editable, mathematical model that can be used by other countries and regions to assess different scenarios. METHODS: In this cost-utility analysis, we constructed a decision tree and a Markov model to assess the cost-effectiveness of HTLV-1 antenatal screening and postnatal interventions (ie, avoidance of breastfeeding, by suppression of lactation with cabergoline, and provision of formula feed) to reduce transmission. For our model, we used data from Brazil and we took the perspective of the public health-care system to estimate costs. FINDINGS: The implementation of both screening and interventions would result in the prevention of 1039 infections in infants every year in Brazil with an incremental cost-effectiveness ratio (ICER) of US11 415perquality−adjustedlife−year(QALY).8811 415 per quality-adjusted life-year (QALY). 88% of all probabilistic sensitivity analysis simulations had ICER values lower than the Brazilian cost-effectiveness threshold (18 107·74 per QALY). HTLV-1 prevalence in pregnant women, the risk of HTLV-1 transmission when breastfeeding lasts for 6 months or more, and the cost of screening tests were the variables with the largest effect on ICER. INTERPRETATION: HTLV-1 antenatal screening is cost-effective in Brazil. An open-access model was developed, and this tool could be used to assess the cost-effectiveness of such policy globally, favouring the implementation of interventions to prevent HTLV-1 mother-to-child transmission worldwide.     CMB Telescopes and Optical Systems

    The cosmic microwave background radiation (CMB) is now firmly established as a fundamental and essential probe of the geometry, constituents, and birth of the Universe. The CMB is a potent observable because it can be measured with precision and accuracy. Just as importantly, theoretical models of the Universe can predict the characteristics of the CMB to high accuracy, and those predictions can be directly compared to observations. There are multiple aspects associated with making a precise measurement. In this review, we focus on optical components for the instrumentation used to measure the CMB polarization and temperature anisotropy. We begin with an overview of general considerations for CMB observations and discuss common concepts used in the community. We next consider a variety of alternatives available for a designer of a CMB telescope. Our discussion is guided by the ground and balloon-based instruments that have been implemented over the years. In the same vein, we compare the arc-minute resolution Atacama Cosmology Telescope (ACT) and the South Pole Telescope (SPT). CMB interferometers are presented briefly. We conclude with a comparison of the four CMB satellites, Relikt, COBE, WMAP, and Planck, to demonstrate a remarkable evolution in design, sensitivity, resolution, and complexity over the past thirty years.Comment: To appear in: Planets, Stars and Stellar Systems (PSSS), Volume 1: Telescopes and Instrumentatio

    The interaction between practice and performance pressure on the planning and control of fast target directed movement

    Pressure to perform often results in decrements to both outcome accuracy and the kinematics of motor skills. Furthermore, this pressure-performance relationship is moderated by the amount of accumulated practice or the experience of the performer. However, the interactive effects of performance pressure and practice on the underlying processes of motor skills are far from clear. Movement execution involves both an offline pre-planning process and an online control process. The present experiment aimed to investigate the interaction between pressure and practice on these two motor control processes. Two groups of participants (control and pressure; N = 12 and 12, respectively) practiced a video aiming amplitude task and were transferred to either a non-pressure (control group) or a pressure condition (pressure group) both early and late in practice. Results revealed similar accuracy and movement kinematics between the control and pressure groups at early transfer. However, at late transfer, the introduction of pressure was associated with increased performance compared to control conditions. Analysis of kinematic variability throughout the movement suggested that the performance increase was due to participants adopting strategies to improve movement planning in response to pressure reducing the effectiveness of the online control system
