    3-hydroxykynurenine suppresses CD4+ T-cell proliferation, induces T-regulatory-cell development, and prolongs corneal allograft survival

IDO (indoleamine 2,3-dioxygenase) modulates the immune response by depletion of the essential amino acid tryptophan, and IDO overexpression has been shown to prolong corneal allograft survival. This study was conducted to examine the effect of kynurenines, the products of tryptophan breakdown and known to act directly on T lymphocytes, on corneal graft survival. Methods. The effects of kynurenines on T-cell proliferation and death, T-regulatory-cell development, and dendritic cell function, phenotype, and viability were analyzed in vitro. The effect of topical and systemic administration of 3-hydroxykynurenine (3HK) on orthotopic murine corneal allograft survival was examined. Results. T-lymphocyte proliferation was inhibited by two of the four different kynurenines: 3HK and 3-hydroxyanthranilic acid (3HAA). This effect was accompanied by significant T-cell death. Neither 3HK nor 3HAA altered dendritic cell function, nor did they induce apoptosis or pathogenicity to corneal endothelial cells. Administration of systemic and topical 3HK to mice receiving a fully mismatched corneal graft resulted in significant prolongation of graft survival (median survival of control grafts, 12 days; of treated, 19 and 15 days, respectively; P < 0.0003). While systemic administration of 3HK was associated with a significant depletion of CD4+ T, CD8+ T, and B lymphocytes in peripheral blood, no depletion was found after topical administration. Conclusions. The production of kynurenines, in particular 3HK and 3HAA, may be one mechanism (in addition to tryptophan depletion) by which IDO prolongs graft survival.     Integrative taxonomy methods reveal high mealybug (Hemiptera: Pseudococcidae) diversity in southern Brazilian fruit crops.

    The Serra GaĂșcha region is the most important temperate fruit-producing area in southern Brazil. Despite mealybugs (Hemiptera: Pseudococcidae) infesting several host plants in the region, there is a lack of information about the composition of species damaging different crops. A survey of mealybug species associated with commercial fruit crops (apple, persimmon, strawberry and grapes) was performed in Serra GaĂșcha between 2013 and 2015, using both morphology and DNA analyses for species identification. The most abundant species were Pseudococcus viburni (Signoret), found on all four host plant species, and Dysmicoccus brevipes (Cockerell), infesting persimmon, vines and weeds. The highest diversity of mealybug species was found on persimmon trees, hosting 20 different taxa, of which Anisococcus granarae Pacheco da Silva & Kaydan, D. brevipes, Pseudococcus sociabilis Hambleton and Ps. viburni were the most abundant. A total of nine species were recorded in vineyards. Planococcus ficus (Signoret) and Pseudococcus longispinus (Targioni Tozzetti) were observed causing damage to grapes for the first time. A single species, Ps. viburni, was found associated with apples, while both Ps. viburni and Ferrisia meridionalis Williams were found on strawberry.     Novel yttrium and zirconium catalysts featuring reduced Ar-BIANH2 ligands for olefin hydroamination (Ar-BIANH2 = bis-arylaminoacenaphthylene)

    The novel class of bis-arylaminoacenaphthylenes (Ar-BIANH(2)) was employed for the preparation of zirconium and yttrium complexes to be used as catalysts for cyclohydroamination of a number of primary and secondary aminoalkenes. The complex [(2,6-iPr(2)C(6)H(3)-BIAN)Zr(NMe2)(2)(eta(1)-NHMe2)] was isolated and completely characterized, including X-ray diffraction analysis. Despite its easy and almost quantitative isolation, it showed only moderate catalytic performance in the intramolecular hydroamination, irrespective of the cyclization precursor used. On the other hand, in situ generated Y-III complexes obtained using the same class of ligands were found to be very active, leading to the hydroamination of substrates including those normally reluctant in undergoing cyclization such as those featuring an internal non-activated C=C double bond. Electron donating substituents and especially steric hindrance on the ligand improve the performance of the catalysts, allowing us to decrease the catalyst loading to 1 mol% in the latter case

    Using mixed analytical and numerical methods we investigate the development of singularities in the heat flow for corotational harmonic maps from the dd-dimensional sphere to itself for 3≀d≀63\leq d\leq 6. By gluing together shrinking and expanding asymptotically self-similar solutions we construct global weak solutions which are smooth everywhere except for a sequence of times T1<T2<...<Tk<∞T_1<T_2<...<T_k<\infty at which there occurs the type I blow-up at one of the poles of the sphere. We show that in the generic case the continuation beyond blow-up is unique, the topological degree of the map changes by one at each blow-up time TiT_i, and eventually the solution comes to rest at the zero energy constant map.Comment: 24 pages, 8 figures, minor corrections, matches published versio

    Knowledge-Attitudes-Practices About Malaria Among Communities in Southern Benin

    Malaria still remains the main public health problem in Benin. We explored the determinants that influenced malaria treatment as well as protective behaviors, to generate a framework of useful ideas as alternative strategies against malaria. A cross-sectional survey of the knowledge, attitudes and practices (KAP) was conducted at Hozin, Vakon and Agblangandan districts in southern region of Benin. Descriptive statistics were computed and mixed logistic regression helped evaluating the relationship between frequency of each category of severity of malaria and sex group, educational level, treatment, means of self-protection against mosquitoes and identification of the cause of malaria. A significant proportion 750 (81.3%) (p&lt;0.001) of participants stated that malaria was caused by mosquitoes. The respondents who mentioned sun as the cause of malaria, have trivialized more malaria in a proportion of about 59.30% (OR=2.67 [95% CI 1.61-4.44]) followed by those who have reported the cause of body weakness (43.68%) (OR=2.97 [95% CI 1.68-5.28]). Poor knowledge justifies the trivialization of the disease and poor management of malaria control means. National Malaria Control Programs should improve access to education, especially for women and could help improving prevention and control behaviours against malaria in communities
