    Approximating Markov Chains for Bootstrapping and Simulation

    In this work we develop a bootstrap method based on the theory of Markov chains. The method moves from the two competing objectives that a researcher pursues when performing a bootstrap procedure: (i) to preserve the structural similarity -in statistical sense- between the original and the bootstrapped sample; (ii) to assure a diversification of the latter with respect to the former. The original sample is assumed to be driven by a Markov chain. The approach we follow is to implement an optimization problem to estimate the memory of a Markov chain (i.e. its order) and to identify its relevant states. The basic ingredients of the model are the transition probabilities, whose distance is measured through a suitably defined functional. We apply the method to the series of electricity prices in Spain. A comparison with the Variable Length Markov Chain bootstrap, which is a well established bootstrap method, shows the superiority of our proposal in reproducing the dependence among data

    A Tabu Search Heuristic Procedure in Markov Chain Bootstrapping

    Markov chain theory is proving to be a powerful approach to bootstrap nite states processes, especially where time dependence is non linear. In this work we extend such approach to bootstrap discrete time continuous-valued processes. To this purpose we solve a minimization problem to partition the state space of a continuous-valued process into a nite number of intervals or unions of intervals (i.e. its states) and identify the time lags which provide \memory" to the process. A distance is used as objective function to stimulate the clustering of the states having similar transition probabilities. The problem of the exploding number of alternative partitions in the solution space (which grows with the number of states and the order of the Markov chain) is addressed through a Tabu Search algorithm. The method is applied to bootstrap the series of the German and Spanish electricity prices. The analysis of the results conrms the good consistency properties of the method we propose

    Polyimide hollow fiber membranes for CO2 separation from wet gas mixtures

    MatrimidÂź5218 hollow fiber membranes were prepared using the dry-wet spinning process. The transport properties were measured with pure gases (H2, CO2, N2, CH4 and O2) and with a mixture (30% CO2 and 70% N2) in dry and wet conditions at 25 ÂșC, 50 ÂșC, 60 ÂșC and 75 ÂșC and up to 600 kPa. Interesting values of single gas selectivity up to 60 ÂșC (between 31 and 28 for CO2/N2 and between 33 and 30 for CO2/CH4) in dry condition were obtained. The separation factor measured for the mixture was 20% lower compared to the single gas selectivity, in the whole temperature range analyzed. In saturation conditions the data showed that water influences the performance of the membranes, inducing a reduction of the permeance of all gases. Moreover, the presence of water caused a decrease of single gas selectivity and separation factor, although not so significant, highlighting the very high water resistance of hollow fiber membrane modules

    PoN-S : a systematic approach for applying the Physics of Notation (PoN)

    Visual Modeling Languages (VMLs) are important instruments of communication between modelers and stakeholders. Thus, it is important to provide guidelines for designing VMLs. The most widespread approach for analyzing and designing concrete syntaxes for VMLs is the so-called Physics of Notation (PoN). PoN has been successfully applied in the analysis of several VMLs. However, despite its popularity, the application of PoN principles for designing VMLs has been limited. This paper presents a systematic approach for applying PoN in the design of the concrete syntax of VMLs. We propose here a design process establishing activities to be performed, their connection to PoN principles, as well as criteria for grouping PoN principles that guide this process.     Sphingosine-1 phosphate induces cAMP/PKA-independent phosphorylation of the cAMP response element-binding protein (CREB) in granulosa cells

    Background and aims: Sphingosine-1 phosphate (S1P) is a lysosphingolipid present in the ovarian follicular fluid. The role of the lysosphingolipid in gonads of the female is widely unclear. At nanomolar concentrations, S1P binds and activates five specific G protein-coupled receptors (GPCRs), known as S1P1-5, modulating different signaling pathways. S1P1 and S1P3 are highly expressed in human primary granulosa lutein cells (hGLC), as well as in the immortalized human primary granulosa cell line hGL5. In this study, we evaluated the signaling cascade activated by S1P and its synthetic analogues in hGLC and hGL5 cells, exploring the biological relevance of S1PR-stimulation in this context. METHODS AND RESULTS. hGLC and hGL5 cells were treated with a fixed dose (0.1 \u3bcM) of S1P, or by S1P1- and S1P3-specific agonists SEW2871 and CYM5541. In granulosa cells, S1P and, at a lesser extent, SEW2871 and CYM5541, potently induced CREB phosphorylation. No cAMP production was detected and pCREB activation occurred even in the presence of the PKA inhibitor H-89. Moreover, S1P-dependent CREB phosphorylation was dampened by the mitogen-activate protein kinase (MEK) inhibitor U0126 and by the L-type Ca2+ channel blocker verapamil. The complete inhibition of CREB phosphorylation occurred by blocking either S1P2 or S1P3 with the specific receptor antagonists JTE-013 and TY52156, or under PLC/PI3K depletion. S1P-dependent CREB phosphorylation induced FOXO1 and the EGF-like epiregulin-encoding gene (EREG), confirming the exclusive role of gonadotropins and interleukins in this process, but did not affect steroidogenesis. However, S1P or agonists did not modulate granulosa cell viability and proliferation in our conditions. Conclusions: This study demonstrates for the first time that S1P may induce a cAMP-independent activation of pCREB in granulosa cells, although this is not sufficient to induce intracellular steroidogenic signals and progesterone synthesis. S1P-induced FOXO1 and EREG gene expression suggests that the activation of S1P\u2013S1PR axis may cooperate with gonadotropins in modulating follicle development
