Active Facilitation of Helper Dispersal by Parents and Siblings in the Cooperatively Breeding Acorn Woodpecker
Offspring that delay dispersal in cooperatively breeding species have been hypothesized to gain direct fitness benefits via parental facilitation—being passively tolerated on their natal territory by their parents—thereby enjoying enhanced survival and increased probability of acquiring a breeding position in the population. Here we describe active facilitation in the acorn woodpecker (Melanerpes formicivorus) by parents and siblings assisting the dispersal of helpers in their social group. Helpers in this species compete for reproductive opportunities in “power struggles” that take place when all breeders of one sex die or disappear, creating a reproductive vacancy. Individuals compete at power struggles in coalitions of relatives, in which larger coalitions are more likely to be victorious. Based on observations of banded birds, we found that an estimated 26% of individuals competing as part of a winning coalition at a power struggle returned to their home territory at its conclusion, suggesting that they were facilitating the dispersal of kin (generally parents or siblings) that stayed to become breeders on the new territory. In at least one group, sibling facilitation was reciprocated; that is, a bird that was helped at a power struggle by a sibling joined that same sibling as part of a coalition at a subsequent power struggle. Dispersal facilitation is a novel means by which parents can nepotistically enhance the direct fitness of offspring and siblings can enhance each other’s inclusive fitness in this highly social species
Wandering Woodpeckers: Foray Behavior in a Social Bird
In many cooperatively breeding taxa, nonbreeding subordinates, or helpers, use extra-territorial forays to discover dispersal opportunities. Such forays are considered energetically costly and foraying birds face aggression from conspecific members of the territories they visit. In contrast, breeders in cooperatively breeding taxa are expected to foray seldomly. We used novel tracking technologies to follow 62 acorn woodpeckers (Melanerpes formicivorus), a cooperatively breeding bird, to study extra-territorial foray behavior. Both helpers and breeders engaged in extra-territorial forays routinely and often several times per day. Helpers forayed earlier in the day and invested more time when foraying to high-quality territories. Unexpectedly, breeders forayed as often and as far as helpers. Breeders from high-quality territories forayed closer to their home territories than breeders from low-quality territories, reflecting a potential trade-off between foraying and territory defense. Such a routine pattern of extra-territorial forays in both helpers and breeders suggests that the motives behind forays differ by sex and social status and involve more than simply searching for dispersal opportunities
Inhibition of plasmin-mediated TAFI activation may affect development but not progression of abdominal aortic aneurysms
Objective: Thrombin-activatable fibrinolysis inhibitor (TAFI) reduces the breakdown of fibrin clots through its action as an indirect inhibitor of plasmin. Studies in TAFI-deficient mice have implicated a potential role for TAFI in Abdominal Aortic Aneurysm (AAA) disease. The role of TAFI inhibition on AAA formation in adult ApoE-/- mice is unknown. The aim of this paper was to investigate the effects of TAFI inhibition on AAA development and progression. Methods: Using the Angiotensin II model of AAA, male ApoE-/- mice were infused with Angiotensin II 750ng/kg/min with or without a monoclonal antibody inhibitor of plasmin-mediated activation of TAFI, MA-TCK26D6, or a competitive small molecule inhibitor of TAFI, UK-396082. Results: Inhibition of TAFI in the Angiotensin II model resulted in a decrease in the mortality associated with AAA rupture (from 40.0% to 16.6% with MA-TCK26D6 (log-rank Mantel Cox test p = 0.16), and 8.3% with UK-396082 (log-rank Mantel Cox test p = 0.05)). Inhibition of plasmin-mediated TAFI activation reduced the incidence of AAA from 52.4% to 30.0%. However, late treatment with MA-TCK26D6 once AAA were already established had no effect on the progression of AAA in this model. Conclusions: The formation of intra-mural thrombus is responsible for the dissection and early rupture in the angiotensin II model of AAA, and this process can be prevented through inhibition of TAFI. Late treatment with a TAFI inhibitor does not prevent AAA progression. These data may indicate a role for inhibition of plasmin-mediated TAFI activation in the early stages of AAA development, but not in its progression
Sulindac Sulfide Reverses Aberrant Self-Renewal of Progenitor Cells Induced by the AML-Associated Fusion Proteins PML/RARα and PLZF/RARα
Chromosomal translocations can lead to the formation of chimeric genes encoding fusion proteins such as PML/RARα, PLZF/RARα, and AML-1/ETO, which are able to induce and maintain acute myeloid leukemia (AML). One key mechanism in leukemogenesis is increased self renewal of leukemic stem cells via aberrant activation of the Wnt signaling pathway. Either X-RAR, PML/RARα and PLZF/RARα or AML-1/ETO activate Wnt signaling by upregulating γ-catenin and β-catenin. In a prospective study, a lower risk of leukemia was observed with aspirin use, which is consistent with numerous studies reporting an inverse association of aspirin with other cancers. Furthermore, a reduction in leukemia risk was associated with use of non-steroidal anti-inflammatory drug (NSAID), where the effects on AML risk was FAB subtype-specific. To better investigate whether NSAID treatment is effective, we used Sulindac Sulfide in X-RARα-positive progenitor cell models. Sulindac Sulfide (SSi) is a derivative of Sulindac, a NSAID known to inactivate Wnt signaling. We found that SSi downregulated both β-catenin and γ-catenin in X-RARα-expressing cells and reversed the leukemic phenotype by reducing stem cell capacity and increasing differentiation potential in X-RARα-positive HSCs. The data presented herein show that SSi inhibits the leukemic cell growth as well as hematopoietic progenitors cells (HPCs) expressing PML/RARα, and it indicates that Sulindac is a valid molecular therapeutic approach that should be further validated using in vivo leukemia models and in clinical settings
Toxin-Based Models to Investigate Demyelination and Remyelination.
Clinical myelin diseases, and our best experimental approximations, are complex entities in which demyelination and remyelination proceed unpredictably and concurrently. These features can make it difficult to identify mechanistic details. Toxin-based models offer lesions with predictable spatiotemporal patterns and relatively discrete phases of damage and repair: a simpler system to study the relevant biology and how this can be manipulated. Here, we discuss the most widely used toxin-based models, with a focus on lysolecithin, ethidium bromide, and cuprizone. This includes an overview of their respective mechanisms, strengths, and limitations and step-by-step protocols for their use
Meta Modeling for Business Process Improvement
Conducting business process improvement (BPI) initiatives is a topic of high priority for today’s companies. However, performing BPI projects has become challenging. This is due to rapidly changing customer requirements and an increase of inter-organizational business processes, which need to be considered from an end-to-end perspective. In addition, traditional BPI approaches are more and more perceived as overly complex and too resource-consuming in practice. Against this background, the paper proposes a BPI roadmap, which is an approach for systematically performing BPI projects and serves practitioners’ needs for manageable BPI methods. Based on this BPI roadmap, a domain-specific conceptual modeling method (DSMM) has been developed. The DSMM supports the efficient documentation and communication of the results that emerge during the application of the roadmap. Thus, conceptual modeling acts as a means for purposefully codifying the outcomes of a BPI project. Furthermore, a corresponding software prototype has been implemented using a meta modeling platform to assess the technical feasibility of the approach. Finally, the usability of the prototype has been empirically evaluated
Dilemas e desafios da função docente na sociedade atual: os sentidos da mudança
A abordagem propõe a constatação das exigências de mudança na atualidade, decorrentes do quadro complexo em que se dá a educação escolar hoje para pensar os rumos da atuação/formação do professor. Procurou-se analisar as questões presentes nas tensões vividas pelos docentes no processo pedagógico (curricular), analisando os reflexos do pensamento moderno e pós-moderno na trajetória docente. Em função dessa proposta, são apresentados resultados da investigação desenvolvida com alunos(as) da disciplina de Currículos e Programas do Curso de Pedagogia e posteriormente aprofundada em Projeto de Pesquisa (Prolicen-UFPR) no ano de 2001. Os pesquisadores investigaram professores de escolas públicas de Curitiba e região metropolitana. Buscou-se a leitura de significados, necessidades e opções presentes na condução e moldagem das práticas docentes na atualidade, considerando o conceito de habitus de BOURDIEU e as possibilidades de análise abertas por SACRISTÁN, GIROUX e GRAMSCI. Procurou-se desenredar a função docente da profusão de exigências que lhe são postas hoje, numa contribuição ao redirecionamento das propostas prescritivas de mudança e, sobretudo, de formação do professor
