When the East Goes to the Poll: Explaining the Regional Differences in AfD-Voting in the 2021 Federal Election
In the 2021 German federal election, the share of East Germans who voted for the Alternative for Germany (AfD), a radical right-wing party, was more than twice as high as the share of AfD voters in West Germany. While most scholars focus on the general determinants of AfD-voting, this paper sheds light on specific variables that could explain these east-west differences. To achieve this, I develop a two-sided argument which considers both the distribution of common AfD-voting determinants and the socialization experiences of East Germans. Multivariate regression with the 2021 GLES data reveals a remaining East bonus in AfD-voting, indicating that the distribution of voting determinants alone cannot explain the regional differences in AfD vote share. Nevertheless, this paper presents a novel structured approach to the issue.Bei der Bundestagswahl 2021 lag der Anteil der Alternative für Deutschland (AfD)-Wähler:innen in Ostdeutschland mehr als doppelt so hoch als der in Westdeutschland. Während Forschende sich häufig mit generellen Determinanten für eine AfD-Wahl beschäftigen, sucht diese Studie nach Variablen, die die Ost-West-Unterschiede erklären können. Hierfür wird ein Argument entwickelt, dass sowohl die Verteilung von allgemeinen Wahldeterminanten als auch Sozialisationseffekte von Ostdeutschen umfasst. Multivariate Regressionen mit den 2021 GLES Daten zeigen einen bestehenden Ostbonus für die AfD und verdeutlichen, dass die Verteilung von Wahldeterminanten allein die regionalen Unterschiede in den Wahlergebnissen nicht erklären kann. Use of subsidy resources for the implementation of the investment plan in the municipality
Bakalářská práce se zabývá využitím dotačních zdrojů v obci. Cílem práce je popsat investiční projekt a zhodnotit způsob jeho financování a vliv na rozvoj obce. V teoretické části bakalářské práce jsou popsány investice, možnosti financování veřejných investic a regionální politika Evropské unie a její dotační programy. Na závěr je popsán a zhodnocen veřejný projekt „Bezpečnost dopravy v obci Svésedlice“, který byl spolufinancován z dotací EU. Závěrem byla stanovena efektivnost tohoto projektu a bylo provedeno doporučení pro zlepšení efektivnosti.The present thesis deals with the use of subsidy sources in a municipality. Its objective is to describe the investment project and to evaluate the means of financing and influence on the development of the municipality. The theoretical part of the thesis delineates the investments, options to finance the public investments, and regional policy of the EU and its grant and subsidy schemes. The final part of the thesis outlines and evaluates the public project "Traffic Safety in the Municipality of Svésedlice“, which was co-financed from the EU subsidies. The efficiency of the project is assessed in the conclusion, along with recommendations to achieve improved efficiency.
Envelopa Office Center - selected parts of the construction technology plan
Diplomová práce se zaměřuje na vypracování vybraných částí stavebně technologického projektu pro realizaci projektu Envelopa Office Center Olomouc, které jsou vypracovány na základě projektové dokumentace použité se souhlasem projekční kanceláře. Hlavním stavebním objektem je polyfunkční budova s šesti nadzemními a jedním podzemním podlažím. Nosnou konstrukci tvoří železobetonový monolitický skelet založený na hlubinných pilotách. V diplomové práci je vypracována technická zpráva ke stavebně technologickému projektu, koordinační situace stavby, objektový finanční a časový plán. Pro realizaci hlavního stavebního objektu je zpracovaná studie realizace hlavních technologických etap, výkres zařízení staveniště s technickou zprávou, jsou navrženy a popsány hlavní stavební mechanismy a stroje a je zpracován časový plán výstavby hlavního stavebního objektu. Pro realizaci železobetonové monolitické konstrukce je navržen technologický předpis a kontrolní a zkušební plán.The diploma thesis focuses on the development of selected parts of the construction and technological project for the implementation of the Envelopa Office Center Olomouc project, which are developed on the basis of the project documentation used with the consent of the design office. The main construction object is a multifunctional building with six above-ground and one underground floor. The supporting structure consists of a reinforced concrete monolithic skeleton based on deep piles. The thesis includes a technical report for the construction and technological project, a coordination situation of the construction, the object financial and time plan. For the realization of the main construction object, a study of the realization of the main technological stages, a drawing of the site equipment with a technical report, the main construction mechanisms and machines are designed and described, and a time schedule for the construction of the main construction object is prepared. For the implementation of the reinforced concrete monolithic structure, a technological regulation and an inspection and testing plan are proposed.
Dickkopf-1 (DKK1) promotes tumor growth via Akt-phosphorylation and independently of Wnt-axis in Barrett’s associated esophageal adenocarcinoma
Esophageal adenocarcinoma (EAC) is still associated with poor prognosis, despite modern multi-modal therapies. New molecular markers, which control cell cycle and promote lymph node metastases or tumor growth, may introduce novel target therapies. Dickkopf-1 (DKK1) is a secreted glycoprotein that blocks the oncogenic Wnt/β-catenin signaling and its aberrant expression has been observed in many malignancies, including EAC. In this study, we investigated the biological role of DKK1 in EAC. Analysis of DKK1 and active β-catenin expression in human esophageal tissues confirmed a simultaneous DKK1-overexpression together with aberrant activation of β-catenin signaling in EAC in comparison with Barrett’s and healthy mucosa. To elucidate the molecular role of DKK1, the OE33 adenocarcinoma cells, which were found to overexpress DKK1, were subjected to functional and molecular assays following siRNA-mediated DKK1-knockdown. At the functional level, OE33 cell viability, proliferation, migration and invasion were significantly attenuated by the absence of DKK1. At the molecular level, neither DKK1-knockdown nor application of exogenous recombinant DKK1 were found to alter the baseline β-catenin signaling in OE33 cells. However, DKK1-knockdown significantly abrogated downstream Akt-phosphorylation. On the other hand, the Wnt-agonist, Wnt3a, restored the Akt-phorphorylation in the absence of DKK1, without, however, being able to further stimulate β-catenin transcription. These findings suggest that the β-catenin transcriptional activity in EAC is independent of Wnt3a/DKK1 site-of-action and define an oncogenic function for DKK1 in this type of malignancy via distinct activation of Akt-mediated intracellular pathways and independently of Wnt-axis inhibition. Taken together, DKK1 may present a novel therapeutic target in EAC
