An appropriate loading control for western blot analysis in animal models of myocardial ischemic infarction
An appropriate loading control is critical for Western blot analysis. Housekeeping proteins (HKPs), such as β-actin, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), and β-tubulin, are commonly used to normalize protein expression. But HKP expression can be impacted by certain experimental conditions, such as ischemic myocardial infarction. This study was undertaken to look for an appropriate loading control for western blot analysis of ischemic myocardium. Myocardial ischemic infarction was induced by left anterior descending coronary artery (LAD) ligation in Rhesus monkeys and C57BL/6 mice. The heart tissue samples from different areas and time points after surgery were subjected to western blot or gel staining. The level of β-actin, GAPDH, β-tubulin, and total protein were tested. The total protein level was consistent in all groups, whereas the protein level of β-tubulin and β-actin were different in all groups. However, the protein level of GAPDH was stable in the Rhesus monkey model. We concluded that total protein was the most appropriate internal control in different stages of myocardial ischemic disease of various animal models. GAPDH is a reliable internal control only for ischemic myocardium of Rhesus monkey
Noise Suppression in 94 GHz Radar-Detected Speech Based on Perceptual Wavelet Packet
A millimeter wave (MMW) radar sensor is employed in our laboratory to detect human speech because it provides a new non-contact speech acquisition method that is suitable for various applications. However, the speech detected by the radar sensor is often degraded by combined noise. This paper proposes a new perceptual wavelet packet method that is able to enhance the speech acquired using a 94 GHz MMW radar system by suppressing the noise. The process is as follows. First, the radar speech signal is decomposed using a perceptual wavelet packet. Then, an adaptive wavelet threshold and new modified thresholding function are employed to remove the noise from the detected speech. The results obtained from the speech spectrograms, listening tests and objective evaluation show that the new method significantly improves the performance of the detected speech
Rare Earth Element Yttrium Modified Mg-Al-Zn Alloy: Microstructure, Degradation Properties and Hardness
The overly-fast degradation rates of magnesium-based alloys in the biological environment have limited their applications as biodegradable bone implants. In this study, rare earth element yttrium (Y) was introduced into AZ61 magnesium alloy (Mg-6Al-1Zn wt %) to control the degradation rate by laser rapid melting. The results showed that the degradation rate of AZ61 magnesium alloy was slowed down by adding Y. This was attributed to the reduction of Mg17Al12 phase and the formation of Al2Y phase that has a more active potential, which decreased galvanic corrosion resulting from its coupling with the anodic matrix phase. Meanwhile, the hardness increased as Y contents increased due to the uniform distribution of the Al2Y and Mg17Al12 phases. However, as the Y contents increased further, the formation of excessive Al2Y phase resulted in the increasing of degradation rate and the decreasing of hardness due to its agglomeration
A Novel Dual-Fc Bispecific Antibody with Enhanced Fc Effector Function
Bispecific antibodies (BsAbs) are undergoing continued
development
for applications in oncology and autoimmune diseases. While increasing
activity by having more than one targeting arm, most BsAb engineering
employs single Fc engagement as monoclonal antibodies. Here, we designed
a novel immunoglobulin gamma-1 (IgG1)-derived dual-Fc BsAb containing
two Fc regions and two distinct asymmetric antigen binding arms comprising
a Fab arm and another VHH domain. In conjunction with the knob-into-hole
technology, dual-Fc BsAbs could be produced with a high yield and
good stability. We explore how Fc engineering effects on dual-Fc constructs
could boost the desired therapeutic efficacy. This new format enabled
simultaneous bispecific binding to corresponding antigens. Furthermore,
compared to the one-Fc control molecules, dual-Fc BsAbs were shown
to increase the avidity-based binding to FcγRs to result in
higher ADCC and ADCP activities by potent avidity via binding to two
antigens and Fc receptors. Overall, this novel BsAb format with enhanced
effector functionalities provides a new option for antibody-based
immunotherapy

Evidence of causal effect of major depression on alcohol dependence: findings from the psychiatric genomics consortium
BackgroundDespite established clinical associations among major depression (MD), alcohol dependence (AD), and alcohol consumption (AC), the nature of the causal relationship between them is not completely understood. We leveraged genome-wide data from the Psychiatric Genomics Consortium (PGC) and UK Biobank to test for the presence of shared genetic mechanisms and causal relationships among MD, AD, and AC.MethodsLinkage disequilibrium score regression and Mendelian randomization (MR) were performed using genome-wide data from the PGC (MD: 135 458 cases and 344 901 controls; AD: 10 206 cases and 28 480 controls) and UK Biobank (AC-frequency: 438 308 individuals; AC-quantity: 307 098 individuals).ResultsPositive genetic correlation was observed between MD and AD (rg = + 0.47, P = 6.6 × 10 ). AC-quantity showed positive genetic correlation with both AD (rg = + 0.75, P = 1.8 × 10 ) and MD (rg = + 0.14, P = 2.9 × 10 ), while there was negative correlation of AC-frequency with MD (rg = -0.17, P = 1.5 × 10 ) and a non-significant result with AD. MR analyses confirmed the presence of pleiotropy among these four traits. However, the MD-AD results reflect a mediated-pleiotropy mechanism (i.e. causal relationship) with an effect of MD on AD (beta = 0.28, P = 1.29 × 10 ). There was no evidence for reverse causation.ConclusionThis study supports a causal role for genetic liability of MD on AD based on genetic datasets including thousands of individuals. Understanding mechanisms underlying MD-AD comorbidity addresses important public health concerns and has the potential to facilitate prevention and intervention efforts
