    Using large volume samplers for the monitoring of particle bound micro pollutants in rivers

    The requirements of the WFD as well as substance emission modelling at the river basin scale require stable monitoring data for micro pollutants. The monitoring concepts applied by the local authorities as well as by many scientists use single sampling techniques. Samples from water bodies are usually taken in volumes of about one litre and depending on predetermined time steps or through discharge thresholds. For predominantly particle bound micro pollutants the small sample size of about one litre results in a very small amount of suspended particles. To measure micro pollutant concentrations in these samples is demanding and results in a high uncertainty of the measured concentrations, if the concentration is above the detection limit in the first place. In many monitoring programs most of the measured values were below the detection limit. This results in a high uncertainty if river loads were calculated from these data sets. The authors propose a different approach to gain stable concentration values for particle bound micro pollutants from river monitoring: A mixed sample of about 1000 L was pumped in a tank with a dirty-water pump. The sampling usually is done discharge dependant by using a gauge signal as input for the control unit. After the discharge event is over or the tank is fully filled, the suspended solids settle in the tank for 2 days. After this time a clear separation of water and solids can be shown. A sample (1 L) from the water phase and the total mass of the settled solids (about 10 L) are taken to the laboratory for analysis. While the micro pollutants can\u27t hardly be detected in the water phase, the signal from the sediment is high above the detection limit, thus certain and very stable. From the pollutant concentration in the solid phase and the total tank volume the initial pollutant concentration in the sample can be calculated. If the concentration in the water phase is detectable, it can be used to correct the total load. This relatively low cost approach (less costs for analysis because of small sample number) allows to quantify the pollutant load, to derive dissolved-solid partition coefficients and to quantify the pollutant load in different particle size classes

    Experimental analysis of suction on step-induced boundary-layer transition

    The influence of suction on step-induced boundary-layer transition has been experimentally investigated in the Cryogenic Ludwieg-Tube Goettingen at large chord Reynolds numbers (up to 16·106), Mach numbers from 0.35 to 0.77 and various streamwise pressure gradients by means of temperature-sensitive paint. Surface imperfections, implemented as combination of gap and forward-facing step, caused transition to occur at a location more upstream than in the case of a smooth surface (i.e. without gap and step). For this combination of imperfections, it was demonstrated for the first time in experiments that suction, achieved passively by exploiting the pressure difference between upper and lower side of the model, induced a movement of transition to a more downstream location than without suction, and in most cases even more downstream than on the smooth configuration at the same test conditions. Thus, the effect of suction was to even overcompensate the adverse effect of the combination of gap and forward-facing step on boundary-layer transition for the investigated test conditions

    System Simulation and Analysis of an LNG-Fueled SOFC System Using Additively Manufactured High Temperature Heat Exchangers

    A laboratory-scale solid oxide fuel cell (SOFC) system using liquefied natural gas (LNG) as a fuel is designed to be used as an energy converter on seagoing vessels (MultiSchIBZ project). The presented system design phase is supported by thermodynamic system simulation. As heat integration plays a crucial role with regard to fuel recirculation and endothermic pre-reforming, the heat exchanger and pre-reforming component models need to exhibit a high degree of accuracy throughout the entire operating range. Compact additively manufactured tube-bundle and plate-fin heat exchangers are designed to achieve high heat exchange efficiencies at low pressure losses. Their heat transfer correlations are derived from experimental component tests under operating conditions. A simulation study utilizing these heat exchanger characteristics is carried out for four configuration variants of pre-reforming and heat integration. Their system behaviour is analyzed with regard to the degree of pre-reforming and the outlet temperature of the fuel processing module. The combination of allothermal pre-reforming with additively manufactured plate-fin heat exchangers exhibits the best heat integration performance at nominal full load and yields a partial load capability to up to 60% electrical load at net electrical efficiencies of 58 to 60% (LHV). © 2022 by the authors.     Therapy of intracellular Staphylococcus aureus by tigecyclin

    BACKGROUND: In the fields of traumatology and orthopaedics staphylococci are the most frequently isolated pathogens. Staphylococcus aureus and Staphylococcus epidermidis are known to be the major causative agents of osteomyelitis. The increasing number of multiresistant Staphylococcus aureus and resistant coagulase-negative staphylococci as a trigger of complicated osteomyelitis and implant-associated infections is a major problem. Antibiotic therapy fails in 20% of cases. Therefore the development of novel antibiotics becomes necessary. METHODS: This study analyses tigecyclin, the first antibiotic of the glycylines, as a potential therapy for osteomyelitis caused by multiresistant Staphylococcus aureus. Therefore its intracellular activity and the potential use in polymethylmetacrylate-bone cement are examined. The intracellular activity of tigecyclin is determined by a human osteoblast infection model. The investigation of the biomechanical characteristics is conducted concerning the ISO 5833-guidelines. RESULTS: Tigecyclin shows in vitro an intracellular activity that ranges between the antimicrobial activity of gentamicin and rifampicin. A significant negative effect on the biomechanical characteristics with an impaired stability is detected after adding tigecyclin to polymethylmetacrylate-bone cement with a percentage of 1.225% per weight. CONCLUSIONS: This study shows that tigecyclin might be a potent alternative for the systemic therapy of osteomyelitis and implant-associated infections whereas the local application has to be reconsidered individually
