    Predictability of Arctic sea ice drift in coupled climate models

    Abstract. Skillful sea ice drift forecasts are crucial for scientific mission planning and marine safety. Wind is the dominant driver of ice motion variability, but more slowly varying components of the climate system, in particular ice thickness and ocean currents, bear the potential to render ice drift more predictable than the wind. In this study, we provide the first assessment of Arctic sea ice drift predictability in four coupled general circulation models (GCMs), using a suite of “perfect-model” ensemble simulations. We find the position vector from Lagrangian trajectories of virtual buoys to remain predictable for at least a 90 (45) d lead time for initializations in January (July), reaching about 80 % of the position uncertainty of a climatological reference forecast. In contrast, the uncertainty in Eulerian drift vector predictions reaches the level of the climatological uncertainty within 4 weeks. Spatial patterns of uncertainty, varying with season and across models, develop in all investigated GCMs. For two models providing near-surface wind data (AWI-CM1 and HadGEM1.2), we find spatial patterns and large fractions of the variance to be explained by wind vector uncertainty. The latter implies that sea ice drift is only marginally more predictable than wind. Nevertheless, particularly one of the four models (GFDL-CM3) shows a significant correlation of up to −0.85 between initial ice thickness and target position uncertainty in large parts of the Arctic. Our results provide a first assessment of the inherent predictability of ice motion in coupled climate models; they can be used to put current real-world forecast skill into perspective and highlight the model diversity of sea ice drift predictability. </jats:p

    In silico characterization of the family of PARP-like poly(ADP-ribosyl)transferases (pARTs)

    BACKGROUND: ADP-ribosylation is an enzyme-catalyzed posttranslational protein modification in which mono(ADP-ribosyl)transferases (mARTs) and poly(ADP-ribosyl)transferases (pARTs) transfer the ADP-ribose moiety from NAD onto specific amino acid side chains and/or ADP-ribose units on target proteins. RESULTS: Using a combination of database search tools we identified the genes encoding recognizable pART domains in the public genome databases. In humans, the pART family encompasses 17 members. For 16 of these genes, an orthologue exists also in the mouse, rat, and pufferfish. Based on the degree of amino acid sequence similarity in the catalytic domain, conserved intron positions, and fused protein domains, pARTs can be divided into five major subgroups. All six members of groups 1 and 2 contain the H-Y-E trias of amino acid residues found also in the active sites of Diphtheria toxin and Pseudomonas exotoxin A, while the eleven members of groups 3 – 5 carry variations of this motif. The pART catalytic domain is found associated in Lego-like fashion with a variety of domains, including nucleic acid-binding, protein-protein interaction, and ubiquitylation domains. Some of these domain associations appear to be very ancient since they are observed also in insects, fungi, amoebae, and plants. The recently completed genome of the pufferfish T. nigroviridis contains recognizable orthologues for all pARTs except for pART7. The nearly completed albeit still fragmentary chicken genome contains recognizable orthologues for twelve pARTs. Simpler eucaryotes generally contain fewer pARTs: two in the fly D. melanogaster, three each in the mosquito A. gambiae, the nematode C. elegans, and the ascomycete microfungus G. zeae, six in the amoeba E. histolytica, nine in the slime mold D. discoideum, and ten in the cress plant A. thaliana. GenBank contains two pART homologues from the large double stranded DNA viruses Chilo iridescent virus and Bacteriophage Aeh1 and only a single entry (from V. cholerae) showing recognizable homology to the pART-like catalytic domains of Diphtheria toxin and Pseudomonas exotoxin A. CONCLUSION: The pART family, which encompasses 17 members in the human and 16 members in the mouse, can be divided into five subgroups on the basis of sequence similarity, phylogeny, conserved intron positions, and patterns of genetically fused protein domains

    Quality assessment of expert answers to lay questions about cystic fibrosis from various language zones in Europe: the ECORN-CF project

    Background: The European Centres of Reference Network for Cystic Fibrosis (ECORN-CF) established an Internet forum which provides the opportunity for CF patients and other interested people to ask experts questions about CF in their mother language. The objectives of this study were to: 1. develop a detailed quality assessment tool to analyze quality of expert answers, 2. evaluate the intra- and inter-rater agreement of this tool, and 3. explore changes in the quality of expert answers over the time frame of the project. Methods: The quality assessment tool was developed by an expert panel. Five experts within the ECORN-CF project used the quality assessment tool to analyze the quality of 108 expert answers published on ECORN-CF from six language zones. 25 expert answers were scored at two time points, one year apart. Quality of answers was also assessed at an early and later period of the project. Individual rater scores and group mean scores were analyzed for each expert answer. Results: A scoring system and training manual were developed analyzing two quality categories of answers: content and formal quality. For content quality, the grades based on group mean scores for all raters showed substantial agreement between two time points, however this was not the case for the grades based on individual rater scores. For formal quality the grades based on group mean scores showed only slight agreement between two time points and there was also poor agreement between time points for the individual grades. The inter-rater agreement for content quality was fair (mean kappa value 0.232+/-0.036, p<0.001) while only slight agreement was observed for the grades of the formal quality (mean kappa value 0.105+/-0.024, p<0.001). The quality of expert answers was rated high (four language zones) or satisfactory (two language zones) and did not change over time. Conclusions: The quality assessment tool described in this study was feasible and reliable when content quality was assessed by a group of raters. Within ECORN-CF, the tool will help ensure that CF patients all over Europe have equal possibility of access to high quality expert advice on their illness

    Influence of Slim Rod Material Properties to the Siemens Feed Rod and the Float Zone Process

    AbstractThe identification and understanding of material properties influencing the float zone process is important to crystallize high purity silicon for high efficiency solar cells. Also the knowledge of minimal requirements to crystallize monocrystalline silicon with the float zone process is of interest from an economic point of view. In the present study, feed rods for the float zone process composed of a central slim rod and the deposited silicon from the Siemens process are investigated. Previous studies have shown that the slim rod has a significant impact on the purity and suitability for further crystallization processes. In particular, contaminations like substitutional carbon and the presence of precipitates as well as the formation of oxide layers play an important role and are investigated in detail. For this purpose different slim rod materials were used in deposition and float zone crystallization experiments. Samples were prepared by cross sectioning and core drilling of Siemens rods, which were recrystallized with the float zone process. Recrystallized drilled cores are analyzed with FT-IR spectrometry concerning the carbon and oxygen content. To estimate the grain growth behavior on the slim rod surface in dependence of the used slim rod material, EBSD mappings inside a SEM are performed on squared and circular slim rods. TEM analysis was used to investigate the presence of an oxide layer at the interface between slim rod and deposited polycrystalline silicon. Additionally the influence of a nitrogen-containing gas atmosphere during the slim rod pulling is investigated by IR microscopy and ToF-SIMS regarding Si3N4 precipitation

    Interferometer readout-noise below the Standard Quantum Limit of a membrane

    Here we report on the realization of a Michelson-Sagnac interferometer whose purpose is the precise characterization of the motion of membranes showing significant light transmission. Our interferometer has a readout noise spectral density (imprecision) of 3E-16 m/sqrt(Hz) at frequencies around the fundamental resonance of a SiN_x membrane at about 100 kHz, without using optical cavities. The readout noise demonstrated is more than 16 dB below the peak value of the membrane's standard quantum limit (SQL). This reduction is significantly higher than those of previous works with nano-wires [Teufel et al., Nature Nano. 4, 820 (2009); Anetsberger et al., Nature Phys. 5, 909 (2009)]. We discuss the meaning of the SQL for force measurements and its relation to the readout performance and conclude that neither our nor previous experiments achieved a total noise spectral density as low as the SQL
