    Can ‘permission in principle’ for new housing in England increase certainty, reduce ‘planning risk’, and accelerate housing supply?

    In this article we examine the probable impact of moving towards ‘up front’ planning permission for housing schemes in England, on development pace and future housing supply. That examination draws on interviews and focus groups with planning professionals, house builders, land promoters and others involved in land development. We begin by exploring the apparent effect of planning and ‘regulatory risk’ on development, before examining strategies, including upfront ‘permission in principle’ (PiP), that claim the potential to reduce that risk and deliver greater certainty for the development sector. The broader focus for this article is how those compliance-based strategies might operate in England’s otherwise discretionary planning system, in which the power to scrutinise and make decisions rests with local government and elected politicians, and what benefits they might bring

    Estudio del mojado y caracterización superficial por microscopía de barrido laser confocal de chapas de madera obtenidas por desenrollo

    El objetivo de este trabajo fue estudiar las propiedades superficiales de chapas obtenidas por desenrollo de cinco especies de maderas duras: okume, ayous, chopo, fromager y eucalipto, y comparar el comportamiento de ambas caras de las chapas. Se han determinado los ángulos de contacto de equilibrio y se ha determinado la energía libre superficial de la madera así como sus componentes polar y dispersa. Asimismo, mediante microscopía de barrido láser confocal se ha evaluado la rugosidad superficial. La energía libre superficial osciló entre 49-64 mJ/m2 y disminuyó en el orden okume > fromager > chopo > ayous ~ eucalipto, lo que indica un descenso de la capacidad de mojado en ese orden. El okume con el mayor valor de la energía libre superficial y los menores valores del ángulo de contacto de equilibrio se confirmó como la especie con las mejores propiedades de mojado, siendo además la que presentó los mayores valores de los parámetros de rugosidad superficial. Comparando las propiedades superficiales para las dos caras de las chapas se encontró que dependieron de la especie, pero en general, con la excepción de la rugosidad, las diferencias no fueron muy significativas siendo los mayores cambios para las chapas de okume que presentaron un mejor mojado en su cara interior. AbstractThe aim of this work was to study the surface properties of rotary-peeled veneers of five species of hardwoods: okume, ayous, poplar, fromager and eucalyptus and to compare the behavior of both sides of the veneers. The equilibrium contact angles were determined together with the wood surface free energy and its polar and disperse components. Additionally, surface roughness was evaluated using confocal laser scanning microscopy. Surface free energy ranged from 49-64 mJ/m2 and decreased in the order okume> fromager> poplar> eucalyptus ~ ayous, indicating a decrease in wettability in that order. Okume wood with the highest value of the surface free energy and the lowest values of the equilibrium contact angle was confirmed as the species with the best wetting properties, and also presented the highest values of surface roughness parameters. In general, there were not significant differences between the properties of both sides of the veneers, except for roughness. The largest differences were obtained for okume veneers whose interior side showed higher wettability

Moreover, cancer is a tissue growing in the context of a complex organism, thus it can be identified two main sources of variability responsible for the disease behavior: intrinsic and extrinsic factors which act, respectively, mainly inside the tumor cells and outside them at local or systemic levels. Our aim is to identify intrinsic factors to the tumor cells responsible for the different responses of breast cancer to chemotherapy with Doxorubicin and Docetaxel. For this purpose, we collected tumors developed in a cohort of genetically heterogeneous mice from a backcross between a resistant strain to breast cancer (C57BL/6) and a susceptible one (FVB) which overexpress the cNeu/ErbB2 protooncogene controlled by the MMTV promoter. The backcross mice were genotyped by SNP analysis. To identify tumor intrinsic factors controlling the response to chemotherapy, we transplanted 125 tumors collected from the backcross mice into singenic F1-C57/FVB mice to remove variability coming from the host compartments. Each tumor was transplanted into two F1 recipient mice; each one was treated with Doxorubicin or Docetaxel, and we studied tumor response to treatment. Linkage analysis permits us to identify QTL (Quantitative Trait Loci) controlling susceptibility to mammary cancer and evolution of the disease in the backcross population, and the specific intrinsic QTL associated with different chemotherapy responses in the F1 mice. Moreover, we are studying molecular and signalling pathways that control chemotherapy responses and the QTL associated with them. The identification of breast cancer susceptibility genes and their pathways associated with different response to chemotherapy will be important for the prediction of human breast cancer evolution during therapy, and to learn about the mechanisms involved in resistance to chemotherapy, thus it would help to develop new preventive and therapeutic strategies.Peer Reviewe

    Sex Differences in Hypothalamic Changes and the Metabolic Response of TgAPP Mice to a High Fat Diet

    The propensity to develop neurodegenerative diseases is influenced by diverse factors including genetic background, sex, lifestyle, including dietary habits and being overweight, and age. Indeed, with aging, there is an increased incidence of obesity and neurodegenerative processes, both of which are associated with inflammatory responses, in a sex-specific manner. High fat diet (HFD) commonly leads to obesity and markedly affects metabolism, both peripherally and centrally. Here we analyzed the metabolic and inflammatory responses of middle-aged (11–12 months old) transgenic amyloid precursor protein (TgAPP) mice of both sexes to HFD for 18 weeks (starting at 7–8 months of age). We found clear sex differences with females gaining significantly more weight and fat mass than males, with a larger increase in circulating leptin levels and expression of inflammatory markers in visceral adipose tissue. Glycemia and insulin levels increased in HFD fed mice of both sexes, with TgAPP mice being more affected than wild type (WT) mice. In the hypothalamus, murine amyloid β (Aβ) levels were increased by HFD intake exclusively in males, reaching statistical significance in TgAPP males. On a low fat diet (LFD), TgAPP males had significantly lower mRNA levels of the anorexigenic neuropeptide proopiomelanocortin (POMC) than WT males, with HFD intake decreasing the expression of the orexigenic neuropeptides Agouti-related peptide (AgRP) and neuropeptide Y (NPY), especially in TgAPP mice. In females, HFD increased POMC mRNA levels but had no effect on AgRP or NPY mRNA levels, and with no effect on genotype. There was no effect of diet or genotype on the hypothalamic inflammatory markers analyzed or the astrogliosis marker glial acidic protein (GFAP); however, levels of the microglial marker Iba-1 increased selectively in male TgAPP mice. In summary, the response to HFD intake was significantly affected by sex, with fewer effects due to genotype. Hypothalamic inflammatory cytokine expression and astrogliosis were little affected by HFD in middle-aged mice, although in TgAPP males, which showed increased Aβ, there was microglial activation. Thus, excess intake of diets high in fat should be avoided because of its possible detrimental consequences

    The Multiscenario Multienvironment BioSecure Multimodal Database (BMDB)

    A new multimodal biometric database designed and acquired within the framework of the European BioSecure Network of Excellence is presented. It is comprised of more than 600 individuals acquired simultaneously in three scenarios: 1) over the Internet, 2) in an office environment with desktop PC, and 3) in indoor/outdoor environments with mobile portable hardware. The three scenarios include a common part of audio/video data. Also, signature and fingerprint data have been acquired both with desktop PC and mobile portable hardware. Additionally, hand and iris data were acquired in the second scenario using desktop PC. Acquisition has been conducted by 11 European institutions. Additional features of the BioSecure Multimodal Database (BMDB) are: two acquisition sessions, several sensors in certain modalities, balanced gender and age distributions, multimodal realistic scenarios with simple and quick tasks per modality, cross-European diversity, availability of demographic data, and compatibility with other multimodal databases. The novel acquisition conditions of the BMDB allow us to perform new challenging research and evaluation of either monomodal or multimodal biometric systems, as in the recent BioSecure Multimodal Evaluation campaign. A description of this campaign including baseline results of individual modalities from the new database is also given.     SUMO regulates p21Cip1 intracellular distribution and with p21Cip1 facilitates multiprotein complex formation in the nucleolus upon DNA damage

    We previously showed that p21Cip1 transits through the nucleolus on its way from the nucleus to the cytoplasm and that DNA damage inhibits this transit and induces the formation of p21Cip1-containing intranucleolar bodies (INoBs). Here, we demonstrate that these INoBs also contain SUMO-1 and UBC9, the E2 SUMO-conjugating enzyme. Furthermore, whereas wild type SUMO-1 localized in INoBs, a SUMO-1 mutant, which is unable to conjugate with proteins, does not, suggesting the presence of SUMOylated proteins at INoBs. Moreover, depletion of the SUMO-conjugating enzyme UBC9 or the sumo hydrolase SENP2 changed p21Cip1 intracellular distribution. In addition to SUMO-1 and p21Cip1, cell cycle regulators and DNA damage checkpoint proteins, including Cdk2, Cyclin E, PCNA, p53 and Mdm2, and PML were also detected in INoBs. Importantly, depletion of UBC9 or p21Cip1 impacted INoB biogenesis and the nucleolar accumulation of the cell cycle regulators and DNA damage checkpoint proteins following DNA damage. The impact of p21Cip1 and SUMO-1 on the accumulation of proteins in INoBs extends also to CRM1, a nuclear exportin that is also important for protein translocation from the cytoplasm to the nucleolus. Thus, SUMO and p21Cip1 regulate the transit of proteins through the nucleolus, and that disruption of nucleolar export by DNA damage induces SUMO and p21Cip1 to act as hub proteins to form a multiprotein complex in the nucleolus
